The Effect of Lysine to Protein Ratio on Growth Performance and Efficiency of Nitrogen Utilization in Pigs

  • One feeding trial and two metabolic trials were conducted to investigate the effects of lysine to protein ratio in practical swine diets on growth performance and efficiency of nitrogen retention and utilization in different growing phases. In Trial one (the feeding trial), 90 mixed sex pigs weighing $9.1{\pm}1.4kg$ (Duroc ${\times}$ Landrance ${\times}$ Beijing Black) were used to study the effects of concentrations of 5.2, 5.3, 5.8, 6.4 and 7.2 g lysine/100 g CP in diets containing 1.2% lysine on growth performance and serum urea nitrogen. The results showed that feed conversion efficiency and economic efficiency were best for pigs fed the diet containing the lysine concentration of 5.8 g /100 g crude protein. Serum urea nitrogen concentration decreased linearly (p=0.0009) and serum free lysine content increased linearly (p=0.0017) as the lysine to protein ratio in diets increased from 5.2 to 7.2 g/100 g. In Trials two and three (the metabolic trials), five growing barrows (Duroc ${\times}$ Landrance ${\times}$ Beijing black), with initial body weights of approximately $26{\pm}2.4kg$ and $56.3{\pm}3.5kg$, respectively, were allotted to five dietary treatments according to a $5{\times}5$ Latin square design. Trial two contained 5.2, 5.7, 6.1, 6.7 and 6.8 g lysine/100 g CP treatments. Trial three contained 4.6, 5.0, 5.6, 6.1 and 6.6 g lysine/100 g CP treatments. The results showed that nitrogen retention in growing pigs decreased linearly (p=0.0011 in Trial two; p=0.0099 in Trial three) as the lysine to protein ratio in diets increased. The ratio of lysine to protein in diets resulting in maximum nitrogen retention was 5.2 g/100 g and 5.0 g/100 g in Trial two and Trial three, respectively. In Trial two, apparent biological value and gross nitrogen efficiency increased linearly (p=0.0135 and p=0.0192, respectively) as the lysine to protein ratio increased from 5.2 to 6.8 g lysine/100 g CP. In summary, we concluded that the optimal Lysine to Protein Ratios for 8-20 kg and 20-80 kg pigs were 5.8 g/100 g and 5.0 to 5.2 g/100 g, respectively.

Influence of Sodium Nitrate (NaNO$_3$) of Different Feeds on Growth and Bioenergetics of Bivoltine NB$_4$D$_2$ Race of the Silkworm, Bombyx mori L.

  • Dietary supplementation of sodium nitrate with different concentrations 50, 100, 200, 500, 700 and 1000 $\mu\textrm{g}$/ml of single, two, three and four feeds to fifth instar larvae of biovoltine NB$_4$D$_2$ race of the silkworm, B. mori resulted in significant increase in the food conversion, conversion rate and conversion efficiency $K_1$ and $K_2$. However, there were significant decrease in the food assimilation, assimilation rate and assimilation efficiency in the sodium nitrate treated groups as compared with that of the corresponding parameters of the carrier control. This indicates that the administration of sodium nitrate may stimulate metabolic activities, thereby influencing conversion of food into body weight in the bivoltine silkworm, B. mori.

칡 열수추출물이 흰쥐의 알콜올 대사효소계에 미치는 영향 (Effects of Pueraria thunbergiana Bentham Water Extracts on Hepatic Alcohol Metabolic Enzyme System In Rats)

  • 갈화와 갈근이 알코올성 간손상 흰쥐의 알코올 대사효소계에 미치는 영항을 구명하기 위하여 알코올을 투여한 흰쥐에게 갈화와 갈근 열수추출물을 2수준으로 나누어 5주간 급여한 후 그 효과를 관찰하였다. 체중증가량, 식이섭취량과 식이효율은 알코올 투여시 감소하였으며 갈화와 갈근추출물 급여로 회복되는 경향을 보였다. 장기 무게에 미치는 칡추출물의 영향은 알코올 투여로 증가된 간, 뇌와 심장 무게를 감소시켰는데 갈화가 갈근에 비하여 뇌와 심장 무게의 감소효과가 큰 것으로 나타났다. 알코올 대사효소 활성 변화에서 알코올 투여시 유의적으로 증가된 ADH와 MEOS 활성은 칡추출물 급여시 감소된 반면 AlDH 활성은 대조군에서 유의적으로 감소되었는데 칡추출물 급여시 그 활성이 증가되었다. MAO 활성과 과산화수소 함량도 알코올 투여시 증가되었으며 갈화와 갈근 열수추출물 급여시 유의적으로 감소되었다. 이상의 결과에서 갈화와 갈근 열수추출물 급여는 알코올 중독된 흰쥐의 알코올 대사계 효소 활성 변화에 영향을 미쳐 알코올성 간손상 완화에 효과적일 것으로 사료된다.

식이중 단백질과 지방수준이 흰쥐의 성장, 질소와 에너지 이용 및 체조성에 미치는 영향 (Effect of Dietary Protein and Fat Levels on the Utilization of Nitrogen and Energy and Body Composition in Rats)

  • In order to investigate the effect of dietary protein and fat levels on the growth and the utilization of nitrogen and energy and body composition in rats, Sprague\ulcornerDawley 48 male rats of 8 weeks old weighing approxijIlately 215-220g were subjected to feeding trials for 8 weeks and then subsequently to metabolic trials for 2 weeks. Four dietary protein levels (4, 8, 16, 32%) and each protein level contained two fat levels(3.9, 11.7%=1O,3J% of 3600kcal ME/kg) by addition of an appropriate amount of carbo\ulcornerhydrate and the following results were obtained. The body weight gain and food efficiency ratio of the rats to which a diet of 16% protein and 3.9% fat was fed were significantly higher than in either case of 8% pro\ulcornertein diet or of 32% protein diet. The digestibility of protein in the experimental diets was 73.3 -93.4%. The digesti\ulcornerbility of energy ( energy absorption) in the experimental diets was 83.2 -91.5%. The utilization of protein and the metabolic energy efficiency in the experimental diets was highest at the diet of 8% protein and 3.9% fat. The analysis of the body composition after feeding trials for 8 weeks has shown that the content of body water and protein were not affected by protein and fat levels in diet. The content of body fat in the rats to which 3.9% fat diet was fed was high\ulcornerer than that in those to which 11.7% fat diet was fed. From the above experimental results it may be suggested that the best formula of diet for the 8 weeks old rats may be composed of the 8% protein and 3.9% fat.

The Effect of Spent Medium Recycle on Cell Proliferation, Metabolism and Baculovirus Production by the Lepidopteran Se301 Cell Line Infected at Very Low MOI

  • The aim of this paper was to study the effect of spent medium recycle on Spodoptera exigua Se301 cell line proliferation, metabolism, and baculovirus production when grown in batch suspension cultures in Ex-Cell 420 serum-free medium. The results showed that the recycle of 20% of spent medium from a culture in mid-exponential growth phase improved growth relative to a control culture grown in fresh medium. Although both glucose and glutamine were still present at the end of the growth phase, glutamate was always completely exhausted. The pattern of the specific glucose and lactate consumption and production rates, as well as the specific glutamine and glutamate consumption rates, suggests a metabolic shift at spent medium recycle values of over 60%, with a decrease in the efficiency of glucose utilization and an increase in glutamate consumption to fuel energy metabolism. Baculovirus infection provoked a change in the metabolic pattern of Se301 cells, although a beneficial effect of spent medium recycle was also observed. Both growth rate and maximum viable cell density decreased relative to uninfected cultures. The efficiency of glucose utilization was dramatically reduced in those cultures containing the lowest percentages of spent medium, whereas glutamine and glutamate consumption was modulated, thereby suggesting that infected cells were devoted to virus replication, retaining their ability to incorporate the nutrients required to support viral replication. Recycle of 20% of spent medium increased baculovirus production by around 90%, thus showing the link between cell growth and baculovirus production.

Metabolic engineering of Lilium ${\times}$ formolongi using multiple genes of the carotenoid biosynthesis pathway

  • Lilium ${\times}$ formolongi was genetically engineered by Agrobacterium-mediated transformation with the plasmid pCrtZW-N8idi-crtEBIY, which contains seven enzyme genes under the regulation of the CaMV 35S promoter. In the transformants, ketocarotenoids were detected in both calli and leaves, which showed a strong orange color. In transgenic calli, the total amount of carotenoids [133.3 ${\mu}g/g$ fresh weight (FW)] was 26.1-fold higher than in wild-type calli. The chlorophyll content and photosynthetic efficiency in transgenic orange plantlets were significantly lowered; however, after several months of subculture, they had turned into plantlets with green leaves that showed significant increases in chlorophyll and photosynthetic efficiency. The total carotenoid contents in leaves of transgenic orange and green plantlets were quantified at 102.9 and 135.2 ${\mu}g/g$ FW, respectively, corresponding to 5.6- and 7.4-fold increases over the levels in the wild-type. Ketocarotenoids such as echinenone, canthaxanthin, 3'-hydroxyechinenone, 3-hydroxyechinenone, and astaxanthin were detected in both transgenic calli and orange leaves. A significant change in the type and composition of ketocarotenoids was observed during the transition from orange transgenic plantlets to green plantlets. Although 3'-hydroxyechinenone, 3-hydroxyechinenone, astaxanthin, and adonirubin were absent, and echinenone and canthaxanthin were present at lower levels, interestingly, the upregulation of carotenoid biosynthesis led to an increase in the total carotenoid concentration (+31.4%) in leaves of the transgenic green plantlets.

  • 본 연구에서는 심혈관계 질환 합병증을 동반하지 않은 국내 본태성 고혈압 환자의 안정시 심박수에 따른 대사증후군 세부요인 변화에 대한 12개월 추적 조사 결과를 확인해 보고자 한다. 2013년부터 국내 25개 대학병원에서 진행되어 온 대규모 전향적 관찰 연구인 KorHR(Korean Hypertension Registry) 자료를 이용하였고 총 951명의 데이터를 R 통계 프로그램을 이용하여 분석하였다. 본 연구결과 초진과 재진 고혈압 환자의 안정시 심박수 그룹에 따라 대사증후군 세부요인의 변화 및 대사증후군 세부요인 증가 위험이 다른 것으로 나타났다. 따라서 초진과 재진 고혈압 환자에 따른 맞춤형 안정시 심박수 조절을 위한 지침 마련과 안정시 심박수 조절을 위한 다양한 중재방안 마련이 필요하다고 생각된다. 이를 통해 대표적인 만성질환인 고혈압에 따른 심혈관계 질환 발생을 예방하여 환자 및 가족의 삶의 질을 향상시킬 뿐만 아니라 보건 의료 분야에서의 효율성을 증대시키는데 기여하고자 한다.

지방조직과 면역체계의 상호작용 및 관련 염증물질에 관한 고찰 (The Interaction of Adipose Tissue with Immune System and Related Inflammatory Molecules)

  • Background: Adipose tissues were initially introduced as energy storages, but recently they have become famous as an endocrine organ which produces and secretes various kinds of molecules to make physiologic and metabolic changes in human body. It has been studied that these molecules are secreted in abundance as the adipose tissue becomes bigger along with obesity. Furthermore, it has been found that they are mediating systemic inflammation and generation of metabolic diseases such as type 2 diabetes and atherosclerosis. On the basis of these, we studied previous papers which have been researched about the interaction between preadipocytes and macrophages, adipose tissues and lymph nodes, and adipose tissue secreting molecules. Results: Firstly, preadipocytes and macrophages are expressing similar transcriptomes and proteins, and preadipocytes can be converted to mature macrophages which have phagocytic activity. Moreover, the monocytes, which initially located in the bone marrow, are filtrated to the adipose tissue by monocyte chemotatic protein-1 and are matured to macrophages by colony stimulating factor-1. Secondly, adipose tissues and their associated lymph nodes are interacting each other in terms of energy efficiency. Lymph nodes promote lipolysis in adipose tissues, and polyunsaturated fatty acids in adipocytes become energy sources for dendritic cells. Lastly, adipose tissues produce and secrete proinflammatory molecules such as leptin, adiponectin, TNF-${\alpha}$, IL-6, and acute phase proteins, which induce the inflammation and potentially generate metabolic diseases. Conclusion: According to these, we can link adipose tissues to inflammation, but we need to affirm the actual levels and roles of adipose tissue-derived proinflammatory molecules in human body.

마늘이 고지방 식이를 섭취한 UCP2 형질전환 마우스의 대사성 조직에서 UCP2 전사 조절에 미치는 영향 (Effects of Garlic on Uncoupling Protein 2 (UCP2) Transcriptional Regulation in Metabolic Tissues of UCP2 Transgenic Mice Fed on a High-Fat Diet)

  • This study was performed to investigate the effects of garlic on uncoupling protein 2 (UCP2) transcriptional regulation of UCP2-luciferase transgenic mice fed on a high fat diet to induce obesity. To examine the transcriptional regulation of UCP2, we generated transgenic mice with a UCP2 promoter (-1,830/+30 bp) containing luciferase as a reporter gene. UCP2-luciferase transgenic mice were fed a 45% high-fat diet for 8 weeks to induce obesity. Subsequently, mice were maintained on either a high-fat control diet (TG-CON), or high-fat diets supplemented with 2% (TG-GL2) or 5% (TG-GL5) garlic for a further 8 weeks. Dietary garlic reduced body weight and energy efficiency ratio in the TG-GL5 group, compared to the TG-CON group. Furthermore, garlic supplementation significantly decreased white adipose tissue fat mass and plasma levels of triglycerides, total cholesterol, and leptin in the TG-GL2 and TG-GL5 groups, compared to the TG-CON group. Specifically, UCP2 promoter activity in metabolic tissues such as liver, white adipose tissue, brown adipose tissue, and skeletal muscle was increased by garlic supplementation. These results suggest that dietary garlic was partially associated with an increase of UCP2 transcriptional activity in metabolic tissues for decreasing obesity.

Cow Residual Feed Intake(RFI) monitoring and metabolic abnormality prediction system using wearable device for Milk cow and Beef

  • 본 논문에서는 소의 사료 취식량(Feed Intake), 반추(Rumination), 발정기(In Heat) 모니터링 기술을 이용하여, RFI(Residual Feed Intake) 모니터링 및 신진대상 이상을 예측하는 웨어러블 디바이스 및 PC용 웹과 스마트폰 어플리케이션을 이용한 모니터링 시스템을 설계하고 구현하였다. 본 시스템의 개발로 농장주는 경제적 효율성의 증가가 기대된다. 사료 섭취량을 분석하면, 소의 체중에 근거한 추천 사료량과 소가 섭취하는 사료량과의 차이를 확인할 수 있으며, Metabolic disorder(신진대사 이상)에 대한 조기 발견이 가능할 것으로 예상된다. 본 논문의 결과물을 사용하는 농장주는 가장 효율적인 성과를 나타내는 소를 구별할 수 있으며, 소의 표피(목)에 부착하는 웨어러블 장치로부터 입력되는 6축 모션 센서 신호와 웨어러블 장치에 부착된 마이크를 통해 입력되는 소의 목넘김 소리를 통해서 소의 반추와 사료섭취량을 측정할 수 있다. 향후에는 심박, 호흡 등의 추가적인 생체신호를 측정할 수 있도록 개선 작업을 진행할 예정이다.
