융복합 시대의 12시간 공복저항운동이 대사효율성 및 피로물질에 미치는 영향 (Effect of 12 Hours Fasting Resistance Exercise on Metabolic Efficiency and Fatigue in Convergence Ages)

  • 본 연구는 8주간 12시간의 공복을 유지한 후 저항운동을 통해 대사효율성과 피로물질에 미치는 영향을 살펴보고자 하였다. 이를 위해 50대 중년여성을 대상으로 사전 동질성 검정을 통해 운동군(EX)과 대조군(CON) 두 집단으로 구분 선정하였다. 운동군(EX)은 8주간 주 4회 12시간 공복 유지 후 60분간의 저항운동을 실시하였으며, 대조군(CON)은 운동군(EX)의 비교군으로 일반적인 생활을 할 수 있도록 하였다. 운동군(EX)에 적용된 저항운동은 상체와 하체를 중심으로 하는 프리웨이트와 고정식 기구를 사용하여 대근육 중심의 운동프로그램을 적용하였으며, 운동강도는 1RM을 기준으로 4주까지는 70%, 5-8주는 80%로 설정하여 처치프로그램을 시행하였다. 자료 분석은 8주간의 공복저항운동 전과 후의 안정 시에 혈액채취 결과를 바탕으로 기술통계와 두 집단의 비교를 위한 변량분석을 실시하여 검증하였다. 이와 같은 절차를 통해 다음과 같은 결론을 얻었다. 첫째, 12시간 공복저항운동은 대사효율성 변인인 혈압(SBP, DBP), 지방, 공복혈당, 공복인슐린에서 처치프로그램에 대한 의미 있는 상호작용효과를 나타냈다(p<.01). 둘째, 12시간 공복저항운동은 피로물질 변인인 젖산, 요산에서 처치프로그램에 대한 의미 있는 상호작용효과를 나타냈다(p<.01). 본 연구의 결과를 바탕으로 8주간의 12시간 공복저항운동은 대사효율성 및 피로물질 개선에 긍정적 영향을 미치는 것으로 사료된다.

저항전분이 흰쥐의 에너지 영양소 이용효율에 미치는 영향 (Effect of Resistant Starch on Availability of Energy Nutrients in Rats)

  • The purpose of this study was to compare the nutritional effects of resistant starch, cellulose and pectin on growth rate, metabolic energy and apparent digestibility in rats. Forty eight male Sprague-Dawley rats were randomly assigned to six dietary treatments, Rats were fed, ad libitum one of six experimental diets: 5% of resistant starch (RS), 5% of cellulose (CE), 5% of pection(PT), 2.5% resistant starch and 2.5% cellulose(RS+CE), 2.5% resistant starch and 2.5% pectin(RS+PT), and 2.5% cellulose and 2.5% pectin (CE+PT) during 4 weeks. The results obtained were as follows: body weight gain were higher in groups RS and CE, compared with that in the group PT, where other groups such as RS+CE, RS+PT and CE+PT showed no significant differences to the RS group. Food intake and food efficiency into showed the same patterns. Metabolic energy were similar in the RS and CE, and was the lowest the metabolic energy in the group CE+PT was significantly low. Apparent digestibility of protein was significantly higher in the group RS, compared with the groups CE and PT. Apparent digestibility of mixed dietary fiber groups such as the RS+CE, RS+PT and CE+PT group were also higher compared with the group PT. Apparent digestibility of lipid were significantly higher in the groups RS, CE and PT. Apparent digestibility of lipid in the mixed pectin groups such as the RS+PT and CE+PT growth rate, metabolic energy and apparent digestibility of lipid in the mixed pectin groups such as the RS+PT and CE+PT recorded the lowest, while that of the group RS+CE was significantly high. From above results, following was concluded: The growth rate, metabolic energy and apparent digestibility of protein and lipid of group RS were significantly higher in comparision with the group PT, while availability of energy nutrients of group CE was similar to that of the group RS. Also resistant starch fed with pectin(RS+PT) seemed to have increased the growth rate and the apparent digestibility of protein compared with those of the group PT fed with pectin(PT) alone, however RS+PT scemed to have the apparent digestibility of lipid.

Enteric methane emissions, energy partitioning, and energetic efficiency of zebu beef cattle fed total mixed ration silage

  • Objective: The main objective of this study was to evaluate the effect of different feeding levels of a total mixed ration silage-based diet on feed intake, total tract digestion, enteric methane emissions, and energy partitioning in two beef cattle genotypes. Methods: Six mature bulls (three Thai natives, and three Thai natives - Charolais crossbreeds) were assigned in a replicated $3{\times}3$ Latin square design, with cattle breed genotype in separate squares, three periods of 21 days, and three energy feeding above maintenance levels (1.1, 1.5, and 2.0 MEm, where MEm is metabolizable energy requirement for maintenance). Bulls were placed in a metabolic cage equipped with a ventilated head box respiration system to evaluate digestibility, record respiration gases, and determine energy balance. Results: Increasing the feeding level had no significant effect on digestibility but drastically reduced the enteric methane emission rate (p<0.05). Increasing the feeding level also significantly increased the energy retention and utilization efficiency (p<0.01). The Thai native cattle had greater enteric methane emission rate, digestibility, and energy utilization efficiency than the Charolais crossbred cattle (p<0.05). The daily metabolizable energy requirement for maintenance in Thai native cattle ($388kJ/kg\;BW^{0.75}$, where $BW^{0.75}$ is metabolic body weight) was 15% less than that in Charolais crossbred cattle ($444kJ/kg\;BW^{0.75}$). Conclusion: Our results suggested that the greater feeding level in zebu beef cattle fed above maintenance levels resulted in improved energy retention and utilization efficiency because of the reduction in enteric methane energy loss. The results also indicated higher efficiency of metabolisable energy utilization for growth and a lower energy requirement for maintenance in Bos indicus than in Bos taurus.

Fabrication of a Partial Genome Microarray of the Methylotrophic Yeast Hansenula polymorpha: Optimization and Evaluation of Transcript Profiling

  • The methylotrophic yeast Hansenula polymorpha has been extensively studied as a model organism for methanol metabolism and peroxisome biogenesis. Recently, this yeast has also attracted attention as a promising host organism for recombinant protein production. Here, we describe the fabrication and evaluation of a DNA chip spotted with 382 open reading frames (ORFs) of H. polymorpha. Each ORF was PCR-amplified using gene-specific primer sets, of which the forward primers had 5'-aminolink. The PCR products were printed in duplicate onto the aldehyde-coated slide glasses to link only the coding strands to the surface of the slide via covalent coupling between amine and aldehyde groups. With the partial genome DNA chip, we compared efficiency of direct and indirect cDNA target labeling methods, and found that the indirect method, using fluorescent-labeled dendrimers, generated a higher hybridization signal-to-noise ratio than the direct method, using cDNA targets labeled by incorporation of fluorescence-labeled nucIeotides during reverse transcription. In addition, to assess the quality of this DNA chip, we analyzed the expression profiles of H. polymorpha cells grown on different carbon sources, such as glucose and methanol, and also those of cells treated with the superoxide­generating drug, menadione. The profiles obtained showed a high-level induction of a set of ORFs involved in methanol metabolism and oxidative stress response in the presence of methanol and menadione, respectively. The results demonstrate the sensitivity and reliability of our arrays to analyze global gene expression changes of H. polymorpha under defined environmental conditions.

Secretory Expression, Functional Characterization, and Molecular Genetic Analysis of Novel Halo-Solvent-Tolerant Protease from Bacillus gibsonii

  • A novel protease gene from Bacillus gibsonii, aprBG, was cloned, expressed in B. subtilis, and characterized. High-level expression of aprBG was achieved in the recombinant strain when a junction was present between the promoter and the target gene. The purified recombinant enzyme exhibited similar N-terminal sequences and catalytic properties to the native enzyme, including high affinity and hydrolytic efficiency toward various substrates and a superior performance when exposed to various metal ions, surfactants, oxidants, and commercial detergents. AprBG was remarkably stable in 50% organic solvents and retained 100% activity and stability in 0-4 M NaCl, which is better than the characteristics of previously reported proteases. AprBG was most closely related to the high-alkaline proteases of the subtilisin family with a 57-68% identity. The secretion and maturation mechanism of AprBG was dependent on the enzyme activity, as analyzed by site-directed mutagenesis. Thus, when taken together, the results revealed that the halo-solvent-tolerant protease AprBG displays significant activity and stability under various extreme conditions, indicating its potential for use in many biotechnology applications.

A CRISPR/Cas9 Cleavage System for Capturing Fungal Secondary Metabolite Gene Clusters

  • More and more available fungal genome sequence data reveal a large amount of secondary metabolite (SM) biosynthetic 'dark matter' to be discovered. Heterogeneous expression is one of the most effective approaches to exploit these novel natural products, but it is limited by having to clone entire biosynthetic gene clusters (BGCs) without errors. So far, few effective technologies have been developed to manipulate the specific large DNA fragments in filamentous fungi. Here, we developed a fungal BGC-capturing system based on CRISPR/Cas9 cleavage in vitro. In our system, Cas9 protein was purified and CRISPR guide sequences in combination with in vivo yeast assembly were rationally designed. Using targeted cleavages of plasmid DNAs with linear (8.5 kb) or circular (8.5 kb and 28 kb) states, we were able to cleave the plasmids precisely, demonstrating the high efficiency of this system. Furthermore, we successfully captured the entire Nrc gene cluster from the genomic DNA of Neosartorya fischeri. Our results provide an easy and efficient approach to manipulate fungal genomic DNA based on the in vitro application of Cas9 endonuclease. Our methodology will lay a foundation for capturing entire groups of BGCs in filamentous fungi and accelerate fungal SMs mining.

A Novel System with EMG-controlled FES Enhanced Gait Function and Energy Expenditure for Older Adults

  • Objective: This study was conducted to analyze the effect of wearable Electromyography-controlled functional electrical stimulation (EMG-controlled FES) System on Gait Function and cardiopulmonary metabolic efficiency during walking in older adults. Design: Cross-section study Methods: Total 22 older adult participants suitable to selection criteria of this study participated in this study. The EMG-controlled FES System, which functions as a wearable physical activity assist FES system was used. All participations performed randomly assigned two conditions (Non-FES assist [NFA], FES assist [FA]) of walking. In all conditions, spatio-temporal parameters and kinematics and kinetics parameters during walking was collected via 3D motion capture system and 6 minutes walking test (6MWT) and metabolic cost during walking and stairs climbing was collected via a portable metabolic device (COSMED K5, COSMED Srl, Roma, Italy). Results: In Spatio-temporal parameters aspects, The EMG-controlled FES system significantly improved gait functions measurements of older adults with sarcopenia at walking in comparison to the NFA condition (P<0.05). Hip, knee and ankle joint range of motion increased at walking in FA condition compared to the NFA condition (P<0.05). In the FA condition, moment and ground reaction force was changed like normal gait during walking of older adults in comparison to the NFA condition (P<0.05). The EMG-controlled FES system significantly reduced net cardiopulmonary metabolic energy cost, net energy expenditure measurement at stairs climbing (P<0.05). Conclusions: This study demonstrated that EMG-controlled FES is a potentially useful gait-assist system for improving gait function by making joint range of motion and moment properly.

Metabolic Flux Analysis of a Poly-${\beta}$-hydroxybutyrate Producing Cyanobacterium, Synechococcus sp. MA19, Grown under Photoautotrophic Conditions

  • To understand the utilization property of light energy, Synechococcus sp. MA19, a poly-${\beta}$-hydroxybutyrate (PHB) producer, was cultivated at the different incident light intensities of 15.3, 50.0 and 78.2 W/$m^2$ using media with and without phosphate. From the results of metabolic flux analysis, it was found that the cell yield based on ATP synthesis was estimated as $3.5{\times}10^{-3}$ kg-biomass/mol-ATP in these cultures. Under the examined conditions, there were no significant differences in the efficiency of light energy conversion to chemical energies estimated as ATP synthesis and reducing potential (NADH + NADPH) formation whether the PHB synthesis took place or not. The energy converted from light to ATP was kept relatively high around the energy absorbed by the cells of $2.5-3.0{\times}10^{6} J\;h^{-1}\;kg^{-1}$, whereas the energy of reducing potential was hardly changed in the examined range of the energy absorbed by the cells.

Aged garlic extract enhances exercise-mediated improvement of metabolic parameters in high fat diet-induced obese rats

  • Aged garlic extract (AGE) is known to have a protective effect against immune system, endothelial function, oxidative stress and inflammation. We examined the effects of exercise with and without aged garlic extract administration on body weight, lipid profiles, inflammatory cytokines, and oxidative stress marker in high-fat diet (HFD)-induced obese rats. Forty-five Sprague-Dawley rats were fed either a HFD (HFD, n = 40) or a normal diet (ND, n = 5) for 6 weeks and thereafter randomized into ND (n = 5), HFD (n = 10), HFD with AGE (n = 10), HFD with Exercise (n = 10), or HFD with Exercise+AGE (n = 10) for 4 weeks. AGE groups were administered at a dose of 2.86 g/kg body weight, orally. Exercise consisted of running 15-60 min 5 days/week with gradually increasing intensity. AGE (P<0.01), Exercise, and Exercise+AGE (P<0.001) attenuated body weight gain and food efficiency ratio compared to HFD. Visceral fat and liver weight gain were attenuated (P<0.05) with all three interventions with a greater effect on visceral fat in the Exercise+AGE than AGE (P<0.001). In reducing visceral fat (P<0.001), epididymal fat (P<0.01) and liver weight (P<0.001), Exercise+AGE was effective, but exercise showed a stronger suppressive effect than AGE. Exercise+AGE showed further additive effects on reducing visceral fat and liver weight (P<0.001). AGE significantly attenuated the increase in total cholesterol and low-density lipoprotein-cholesterol compared with HFD (P<0.05). Exercise+AGE attenuated the increase in triglycerides compared with HFD (P<0.05). Exercise group significantly decrease in C-reactive protein (P<0.001). These results suggest that AGE supplementation and exercise alone have anti-obesity, cholesterol lowering, and anti-inflammatory effects, but the combined intervention is more effective in reducing weight gain and triglycerides levels than either intervention alone.

Current Status and Applications of Adaptive Laboratory Evolution in Industrial Microorganisms

  • Adaptive laboratory evolution (ALE) is an evolutionary engineering approach in artificial conditions that improves organisms through the imitation of natural evolution. Due to the development of multi-level omics technologies in recent decades, ALE can be performed for various purposes at the laboratory level. This review delineates the basics of the experimental design of ALE based on several ALE studies of industrial microbial strains and updates current strategies combined with progressed metabolic engineering, in silico modeling and automation to maximize the evolution efficiency. Moreover, the review sheds light on the applicability of ALE as a strain development approach that complies with non-recombinant preferences in various food industries. Overall, recent progress in the utilization of ALE for strain development leading to successful industrialization is discussed.
