Alteration of Neural Activity and Effect of Yanggyuksanhwa-tang(Lianggesanhuo-tang) on Cerebral Ischemia of Aged BCAO Rats; [$^{14}C$]2-Deoxyglucose Autoradiography Study (노령 흰쥐의 뇌허혈 손상시 뇌대사활성의 변화 및 량격산화탕의 영향에 대한 [$^{14}C$2-Deoxyglucose Autoradiography 연구)

  • Objective : This study investigated the alteration of neural activity and effect of Yanggyuksanhwa-tang (Lianggesanhuo-tang) on cerebral ischemia of rats. Methods : Considering age-related impact on cerebral ischemia, aged rats (18 months old) were used for this study. Ischemic damage was induced by the transient occlusion of bilateral common carotid arteries (BCAO) with hypotension. Yanggyuksanhwa-tang (Lianggesanhuo-tang) was administered twice a day orally. Then alterations of neural activities in the brain of aged BCAO rats were measured by the [$^{14}C$]2-deoxyglucose autoradiography method. Results : The BCAO in aged rats led to significant decrease of neural activity in the whole brain. Treatment with Yanggyuksanhwa-tang (Lianggesanhuo-tang) significantly attenuated the decrease of neural activity in the whole brain following BCAO ischemia. Treatment significantly attenuated the decrease of neural activity in the CA1, CA2, CA3, dentate gyrus of the hippocampus, activated barrel, barrel cortex, somatosensory cortex, cingulate cortex, caudate putamen, and medial septal nucleus following BCAO in aged rats. Treatment with Yanggyuksanhwa-tang (Lianggesanhuo-tang) also significantly attenuated the decrease of neural activity in the anteroventral thalamic nucleus, ventral anterior thalamic nucleus, arcuate nucleus, posterior hypothalamic area, medial mammillary nucleus, lateral periaqueductal gray, dorsal raphe nucleus, interpeduncular nucleus, median raphe nucleus, and medial pontine nucleus. Conclusion : It can be suggested that Yanggyuksanhwa-tang (Lianggesanhuo-tang) has a neuroprotecuve effect on cerebral ischemia through the control of glucose metabolic rate and cerebral blood flow.

Effect of pear extracts containing herbal medicine(Lycii Fructus, Coicis Semen, Alimatis Rhizoma and Astragali Radix) on body weight, lipid metabolism and immune responses in rats fed high fat diets (II) (배추출물과 구기자, 의이인, 택사, 황기 추출물 배합이 고지방식이로 유발된 비만 백서의 체중, 지질대사, 염증 및 면역기능에 미치는 영향 (II))

  • Objectives : This study was conducted to 9% compared with the previous experimental obesity-induced metabolic function in rats that it was a mixture of pear extract and herbal drugs(Lycii Fructus, Coicis Semen, Alimatis Rhizoma and Astragali Radix) on high fat diet-induce obesity rats. Methods : The animals were used in male rats. Control group fed high-fat diet only. Experimental groups were divided four, ExpI group was fed high-fat diet with a mixture pear extract and Lycii Fructus 9%, Exp II group was fed high-fat diet with a mixture pear extract and Coicis Semen 9%, Exp III group was fed high-fat diet with a mixture pear extract and Alimatis Rhizoma 9%, and Exp IV group was fed high-fat diet with a mixture pear extract and Astragali Radix 9% for 4 weeks. Results : The body weight gain increased in all groups, but attenuated gradually in the experimental groups compared to the control group. The food intakes were significantly lower in all Exp groups than the control group. The concentrations of serum total cholesterol (TC), LDL-cholesterol, and free fatty acid were significantly lower in Exp II than in the control group. Also the concentration of serum free fatty acid was significantly lower in the Exp IV group than in the control group. In inflammatory activities, the Exp II, IV group was significantly lower than the control group. Conclusion : The results indicated that Exp I group (administered a mixture of pear extract and Lycii Fructus) reduced fat accumulation, body weight and the highest elevated lipid metabolism, while the Exp II group (administered the mixture of pear extract and Coicis Semen) and Exp IV group (administered the mixture of pear extract and Astragali Radix) had the highest elevated immune activity.

Microbial Diversity in Korean Traditional Fermenting Starter, Nuruk, Collected in 2013 and 2014

  • A total of sixty-six samples of Nuruk, a fermention starter used to make the Korean traditional rice wine, Makgeolli, were collected from central and southern regions of Korea in 2013 and 2014. We classified two groups of the Nuruk samples, "commercial" and "home-made", according to the manufacturing procedure and purpose of use. Commercial Nuruks were made in a controlled environment where the temperature and humidity are fixed and the final product is supplied to Makgeolli manufacturers. Home-made Nuruks were made under uncontrolled conditions in the naturally opened environment and were intended for use in the production of small amounts of home-brewed Makgeolli. We obtained more than five hundred isolates including filamentous fungi and yeasts from the Nuruk samples followed by identification of fungal species. Also we stored glycerol stocks of each single isolate at $-70^{\circ}C$. We identified the species of each isolate based on the sequences of ITS regions amplified with two different universal primer pairs. We also performed morphological characterization of the filamentous fungi and yeast species through observations under the microscope. We investigated the major fungal species of commercial and home-made Nuruks by counting the colony forming units (CFU) and analyzing the occurrence tendency of fungal species. While commercial Nuruks contained mostly high CFU of yeasts, home-made Nuruks showed relatively high occurrence of filamentous fungi. One of the representative Nuruk manufacturers used both domestic wheat bran and imported ones, mainly from US, as raw material. Depending on the source of ingredient, the fungal diversity was somewhat different. Another commercial Nuruk sample was collected twice, once in 2013 and again in 2014, and showed different diversity of fungal species in each year. Nuruks obtained from the southern regions of Korea and Jeju island showed high frequency of yeast such as Saccharomycopsis fibuligera and Pichia species as well as unique filamentous fungus, Monascus species. S. fibuligera was easily found in many Nuruk samples with high CFU. The major filamentous fungi were Aspergillus, Lichtheimia, Mucor and Penicillium species. In order to further our understanding of the isolates and their potential industrial applications, we assayed three enzymes, alpha amylase, glucoamylase and acid protease from 140 isolates out of about five hundred isolates and selected about 10 excellent strains with high enzyme activities. With these fungal isolates, we will perform omics analyses including genomics, transcriptomics, metabolic pathway analyses, and metabolomics followed by whole genome sequencing of unique isolates associated with the basic research of Nuruk and that also has applications in the Makgeolli making process.

Enzymatic Characterization of Bacillus cereus Lactate Dehydrogenase Isozymes Expressed in Escherichia coli (Bacillus cereus에서 유래한 Lactate Dehydrogenase 동질효소 유전자의 대장균 내 발현 및 효소특성 규명)

  • Lactate dehydrogenases (LDHs) have been highly focused for long time, due to their important roles in biochemical and metabolic pathways of cells. On the basis of genome-wide searching results, three putative LDH genes from Bacillus cereus ATCC 14579 genome have been PCR-amplified, cloned, and well-expressed in E. coli. All three BcLDH isozymes are supposed to share highly conserved catalytic amino acid residues in common $NAD^+$-dependent LDHs. Meanwhile, BcLDH1 consisting of 314 amino acids shares 86 and 49% of identities with BcLDH2 and 3, respectively. Interestingly, only BcLDH1 showed the converting activities between L-lactate and pyruvate in the presence of $NAD^+$ coenzyme, while the other isozymes are likely to have almost no activity. As a result, it was revealed that BcLDH1 can be a typical $NAD^+$-dependent L-lactate-specific dehydrogenase.

The Experimental Study of the Effects of Sungyangikgibuja'ang and Kwangyebujalijungt' and of Soŭm-In on Yang-Insufficient Syndrome (소음인(少陰人) 승양익기부자탕(升陽益氣附子湯)과 관계부자리중탕(官桂附子理中湯)이 양허증(陽虛證)에 미치는 영향(影響)에 관(關)한 실험적(實驗的) 연구(硏究))

  • In order to investigate experimentally the effects of Sung-yangikgibujat'ang (SIT) and Kwangyebujalijungt'ang (KBT) on Yang-insufficient syndrome (陽虛證) induced by Hydrocortisone acetate (H.A.) in experimental animals (Mice and Rat), the author experimented various activities. Delayed type hypersensitivity (DTH), Rosette forming cells (RFC), hemagglutinin (HA) titers, Hemolysin (HL) titers, Body weight, Whole blood viscosity, Plasma Viscosity, Hematocrit, RBC, Albumin, Total protein, Triglyceride, cholesterol and Glucose were measured. The results summerized as follows; 1. In DTH and RFC all the experimental groups were increased significantly in comparison to the H.A.-treated group. 2. In HA titers SIT_treated group were increased significantly and KBT-treated group showed increasing tendancy, but showed no significance. 3. In HL titers all the experimental groups showed increasing tendancy, but showed no significance. 4. In body weight all the experimental groups showed increasing tendancy, but showed no significance. 5. In whole blood viscosity all the experimental groups were decreased significantly in comparison to the H.A.-treated group. 5. In whole blood viscosity all the experimental groups were decreased significantly in comparison to the H.A.-treated group. 6. In plasma viscosity KBT-treated group were decreased significantly and SIT-treated group showed decreasing tendancy, but showed no significance. 7. In Hematocrit SIT-treated group were decreased significantly and KBT-treated group showed decreasing tendancy, but showed no significance. 8. In RBC, albumin and cholesterol all the experimental groups were decreased significantly in comparison to the H.A.-treated group. 9. In total protein and triglyceride KBT-treated group were decreased significantly and SIT-treated group showed decreasing tendancy, but showed no significance. 10. In Glucose SIT-treated group were decreased significantly and KBT-treated group showed decreasing tendancy, but showed no significance. From above findings, it has been demonstrated that Sungyangikgibujat'ang and Kwangyebujalijungt'ang seem to produce the effectiveness on the recovery from depression of the cell-mediated immune response, blood circulation and energy metabolic rate, induced by Hydrocortisone acetate, and in the humoral immune response Sungyangikgibujat'ang have the effectiveness on the recovery, and in cellular component of blood Sungyangikgibujat'ang was more effective than Kwangyebujalijungt' ang, and in plasma of blood Kwangyebujalijungt'ang was more effective than Sungyangikgibujat'ang. Therefore it is suggested that Sungyangikgibujat'ang and Kwangyebujalijungt'ang have the effectiveness on the recovery from Yang-insufficient syndrome more or less.

  • Although microorganisms are, in fact, the most diverse and abundant type of organism on Earth, the ecological functions of microbial populations remains poorly understood. A variety of bacteria including marine Vibrios encounter numerous ecological challenges, such as UV light, predation, competition, and seasonal variations in seawater including pH, salinity, nutrient levels, temperature and so forth. In order to survive and proliferate under variable conditions, they have to develop elaborate means of communication to meet the challenges to which they are exposed. In bacteria, a range of biological functions have recently been found to be regulated by a population density-dependent cell-cell signaling mechanism known as quorum-sensing (QS). In other words, bacterial cells sense population density by monitoring the presence of self-produced extracellular autoinducers (AI). N-acylhomoserine lactone (AHL)-dependent quorum-sensing was first discovered in two luminescent marine bacteria, Vibrio fischeri and Vibrio harveyi. The LuxI/R system of V. fischeriis the paradigm of Gram-negative quorum-sensing systems. At high population density, the accumulated signalstrigger the expression of target genes and thereby initiate a new set of biological activities. Several QS systems have been identified so far. Among them, an AHL-dependent QS system has been found to control biofilm formation in several bacterial species, including Pseudomonas aeruginosa, Aeromonas hydrophila, Burkholderia cepacia, and Serratia liquefaciens. Bacterial biofilm is a structured community of bacterial cells enclosed in a self-produced polymeric matrix that adheres to an inert or living surface. Extracellular signal molecules have been implicated in biofilm formation. Agrobacterium tumefaciens strain NT1(traR, tra::lacZ749) and Chromobacterium violaceum strain CV026 are used as biosensors to detect AHL signals. Quorum sensing in lactic acid bacteria involves peptides that are directly sensed by membrane-located histidine kinases, after which the signal is transmitted to an intracellular regulator. In the nisin autoregulation process in Lactococcus lactis, the NisK protein acts as the sensor for nisin, and NisR protein as the response regulator activatingthe transcription of target genes. For control over growth and survival in bacterial communities, various strategies need to be developed by which receptors of the signal molecules are interfered with or the synthesis and release of the molecules is controlled. However, much is still unknown about the metabolic processes involved in such signal transduction and whether or not various foods and food ingredients may affect communication between spoilage or pathogenic bacteria. In five to ten years, we will be able to discover new signal molecules, some of which may have applications in food preservation to inhibit the growth of pathogens on foods.

Effects of Silt and Clay on the Rates of Respiration, Filtration and Nitrogen Excretion in Shellfish, Mactra veneriformis (동죽의 호흡률, 여수율 및 질소배설에 미치는 부니의 영향)

  • The effects of silt and clay, water temperature and salinity on the metabolic activities: rates of respiration, filtration and nitrogen excretion in the clam, Mactra veneriformis, were investigated in ranges of $0{\sim}250ppm,\;15{\sim}25^{\circ}C\;and\;10{\sim}30\%0$ respectively. The weight specific respiration rate decreased with shell length and significantly declined at lower temperatures and lower salinity levels. Average respiration rates, filtration rates and ammonia excretion rates of the clam groups declined as water temperature was lowered. The respiration rate(R) and the filtration rate(F) decreased as the concentration of silt and clay(C) increased, the relationships between the two parameters were expressed as a regression line, R=a+b LogC and LogF=a+b LogC, respectively. On the rearing experiment treated with 50ppm of silt and clay, the relationship between the accumulative mortality(M) and the elapsed day(T) was $M=0.0186T^{2.7965}$, and the median lethal time was calculated at 16.8 days. From the rearing experiment, both rate of respiration and ammonia nitrogen excretion of experimental groups and control groups decreased with elapsed days, the rates of experimental groups, especially declined more compared with those of the control groups.

Anti-skin Aging Potential of Alcoholic Extract of Phragmites communis Rhizome

  • Chronological aging and photoaging affect appearance, causing wrinkles, pigmentation, texture changes, and loss of elasticity in the skin. Phragmites communis is a tall perennial herb used for its high nutritional value and for medicinal purposes, such as relief from fever and vomiting and facilitation of diuresis. In this study, we investigated the effects of ethanol extract of P. communis rhizome (PCE) on skin aging. The total flavonoid and total phenolic content in PCE were 2.92 ± 0.007 ㎍ of quercetin equivalents (QE) and 231.8 ± 0.001 ㎍ of gallic acid equivalents (GAE) per 100 mg of dried extract (n = 3). The half-maximal inhibitory concentration (IC50) values of PCE for 2,2-diphenyl-1-picrylhydrazyl radical (DPPH) and hydrogen peroxide scavenging activities were 0.96 and 0.97 mg/mL, respectively. PCE showed inhibitory effects on tyrosinase when L-tyrosine (IC50 = 1.25 mg/mL) and L-3,4-dihydroxyphenylalanine (IC50 = 0.92 mg/mL) were used as substrates. PCE treatment up to 200 ㎍/mL for 24 h did not cause any significant cytotoxicity in B16F10 melanocytes, human dermal fibroblasts (HDFs), and HaCaT keratinocytes. In B16F10 melanocytes, PCE (25 and 50 ㎍ /mL) inhibited melanin production and cellular tyrosinase activity after challenge with α-melanocyte-stimulating hormone (α-MSH; p < 0.05). In HDFs, PCE suppressed the mRNA expression of matrix metalloproteinase-1 (MMP-1) and reduced the activity of elastase (p < 0.05). In addition, ultraviolet B (UVB)-mediated downregulation of hyaluronic acid synthase-2 gene expression in HaCaT keratinocytes was also effectively suppressed by PCE treatment. Overall, our results showed that PCE has potential anti-skin aging activity associated with the suppression of hyperpigmentation, wrinkle formation, and reduction in dryness. PCE is a promising candidate for the development of an anti-skin aging cosmetic ingredient.

Short Heterodimer Partner as a Regulator in OxLDL-induced Signaling Pathway

  • Oxidized low-density lipoprotein (oxLDL) has been shown to modulate transactivations by the peroxisome proliferator activated receptor (PPAR)$\gamma$ and nuclear factor-kappa B (NF$\kappa$B). In this study, the oxLDL signaling pathways involved with the NF$\kappa$B transactivation were investigated by utilizing a reporter construct driven by three upstream NF$\kappa$B binding sites, and various pharmacological inhibitors. OxLDL and its constituent lysophophatidylcholine (lysoPC) induced a rapid and transient increase of intracellular calcium and stimulated the NF-KB transactivation in resting RAW264.7 macrophage cells in an oxidation-dependent manner. The NF$\kappa$B activation by oxLDL or lysoPC was inhibited by protein kinase C inhibitors or an intracellular calcium chelator. Tyrosine kinase or PI3 kinase inhibitors did not block the NF$\kappa$B transactivation. Furthermore, the oxLDL-induced NF$\kappa$B activity was abolished by the PPAR$\gamma$ ligands. When the endocytosis of oxLDL was blocked by cytochalasin B, the NF$\kappa$B transactivation by oxLDL was synergistically increased, while PPAR transactivation was blocked. These results suggest that oxLDL activates NF-$\kappa$B in resting macrophages via protein kinase C- and/or calcium-dependent pathways, which does not involve the endocytic processing of oxLDL. The endocytosis-dependent PPAR$\gamma$ activation by oxLDL may function as an inactivation route of the oxLDL induced NF$\kappa$B signal. Short heterodimer partner (SHP), specifically expressed in liver and a limited number of other tissues, is an unusual orphan nuclear receptor that lacks the conventional DNA-binding domain. In this work, we found that SHP expression is abundant in murine macrophage cell line RAW 264.7 but suppressed by oxLDL and its constituent I3-HODE, a ligand for peroxisome proliferator-activated receptor y. Furthermore, SHP acted as a transcription coactivator of nuclear factor-$\kappa$B (NF$\kappa$B) and was essential for the previously described NF$\kappa$B transactivation by lysoPC, one of the oxLDL constituents. Accordingly, NF$\kappa$B, transcriptionally active in the beginning, became progressively inert in oxLDL-treated RAW 264.7 cells, as oxLDL decreased the SHP expression. Thus, SHP appears to be an important modulatory component to regulate the transcriptional activities of NF$\kappa$B in oxLDL-treated, resting macrophage cells.

The Effects of Static Magnetic Field and Pulsed Electromagnetic Field on Alkaline Phosphatase and DNA synthetic Activity of ME3T3-E1 Cells (정자기장과 맥동전자기장이 MC3T3-E1 세포의 ALP 및 DNA 활성도에 미치는 영향)

  • The purpose of this study was to evaluate the effects of magnetic field on cellular activity of MC3T3-E1 cells. The cellular activity was monitored by alkaline phosphatase and DNA synthetic activity in control, static magnetic field and pulsed electromagnetic field groups. A static magnetic field was applied to the cell by placing one, two, three, foue, and five samarium-cobalt magnets above and below each cell plate for 24hours per day. A pulsed electromagnetic field with a frequency of 100 herz was applied for 10 hours per day. After 10 days of magnetic field exposure, there were increase of alkaline phosphatase activity in static magnetic field groups consisted of one, two and three magnetic groups. Alkaline phosphatase activities were not significantly increased in four and five magnetic groups. Application of pulsed electromagnetic field did not result in significant increase in alkaline phosphatase activity compared to control. DNA synthetic activity in both static and pulsed electromagnetic field group were not significantly different from that in control group. The result of this study suggest that magnetic field could have effect on the metabolism of bone cells related to the cellular metabolic process.
