The incividual and combined effects of the chloroacetanilide herbicide pretilachlor and of the safener fenclorim on the growth and selected physiological processes of rice (Oryza sativa L., var 'Lemont')were evaluated under greenhouse and laboratory conditions. Fenclorim applied at rates ranging from 50 to 300 g a.i./ha antagonized the injurious effects caused by 150 to 900 g a.i./ha of pretilachlor on 15-day old wet-sown rice grown under greenhouse conditions. When used rates of 150 g/ha or higher, fenclorim reversed completely the effects of all doses of pretilachlor on rice. When the two compounds were given simultaneously, fenclorim enhanced the uptake of $^{14}C$pretilachlor into rice leaf mesophyll protoplasts measured for 1 hr, indicating that competition for uptake at the protoplast level is not involved in the protective action of this safener. The safener-induced stimulation of pretilachlor uptake was particularly evident when fenclorim was used at concentrations of 10, 20 and $40{\mu}M$. Following 4 hr of incubation, individual treatments with pretilachlor inhibited the in vitro incorporation of radiolabeled precursors into proteins, DNA, and lipids of rice leaf protoplasts only when used at the high concentration of $100{\mu}M$M. Individual treatments with high concentrations (10 or $100{\mu}M$) of the safener fenclorim inhibited the incorporation of radiolabeled precursors into proteins and lipids of rice protoplasts, but had no DNA synthesis. The combined effects of pretilachlor and fenclorim on the incorporation of radiolabeled precursors into these macromolecules of isolated rice mesophyll protoplasts appeared to be additive or slightly synergistic rather than antagonistic. Fenclorim at $1{\mu}M$ antagonized the effects of pretilachlor on total lipids of rice leaf protoplasts. In addition, individual and combined treat-menu with pretilachlor and fenclorim influenced the incoroporation of$^{14}C$acetate into polar lipids, triglycerides and steryl esters of rice leaf protoplas causing a redistribution of carbon in these lipid fractions. However, these effects were not large enough to explain the herbicidal activity of pretilachlor or to account for the protective action of the safener fenclorim. Overall, the uesults of the present study idnicate that the safener fenclorim does not seem to protect rice against pretilachlor injury by antagonizing its effects on protein, DNA, or lipid syntheses.

This study was conducted to find out the influence of elevated atmospheric $CO_2$ concentrations and air temperature on photosynthesis and fruit quality of 'Fuji'/M.9 apple trees and to investigate these to the effects of climate change during the last four years (2009-2012). The treatments employed were: 'Ambient' (ambient temperature + ambient $CO_2$ concentration); 'High $CO_2$' (ambient temperature + elevated $CO_2$ concentration); 'High Temp'. (elevated temperature + ambient $CO_2$ concentration); and 'High $CO_2$ + High Temp'. (elevated temperature + elevated $CO_2$ concentration). The elevated temperature plots were maintained at $4^{\circ}C$ higher than ambient air temperature, while the elevated $CO_2$ plots were maintained at 700 ${\mu}mol{\cdot}mol^{-1}$. Annual treatment period was applied from end of April to beginning of November for four years. Results showed that elevated $CO_2$ decreased stomatal conductance and leaf SPAD value, but increased photosynthetic rate, intercellular $CO_2$ concentration (Ci), and starch content of mesophyll tissue. In the vegetative growth, elevated temperature increased total number of shoot and total shoot growth per tree, but elevated $CO_2$ decreased average shoot length. In the fruit quality, elevated $CO_2$ increased soluble solid content, fruit red color, and ethylene production. In conclusion, elevated $CO_2$ increased photosynthetic rate of apples during the early growth, but effect of increased photosynthetic rate due to elevated $CO_2$ was decreased during latter growth stage. Elevated temperature, on the other hand, tended to decrease photosynthetic rate of apples during the early growth, but that tended to increase during latter growth stage. Both elevated $CO_2$ and temperature tended to decrease the degree of decreased photosynthetic rate due to each factor.
Photoinhibition studies were conducted with ginseng (Panax ginseng) grown under shade and tobacco (N. tobaccum cv. Bulgaria) grown under full sunlight. The plants were exposed to light intensity of 580, 1280, 1770, 2580 ${\mu}$E/㎡/sec in normal air for 3.6 9 hours. Light saturation of ginseng was observed at 550 ${\mu}$E/㎡/sec and that of tobacco was at 1600 ${\mu}$E/㎡/sec. Tobacco exposed to high light intensity and long duration of light irradiation didn't show entire reduction in photosynthetic capacity. But in ginseng, 20.3% of photosynthesis was reduced in light intensity of 2,580 ${\mu}$E/㎡/sec during 9 hours. Light response in photosynthesis differed considerably between tobacco and ginseng, and ginseng exposed to high light intensity showed remarkable reduction in photosynthesis. The extent of photoinhibition of ginseng was dependent on the length of exposure to the high light intensity. Stomatal resistance of ginseng seemed not to be changed by photoinhibition but mesophyll resistance was increased.
Photosynthetic inhibition to temperature were conducted with ginseng(4 year old) and tobacco(var. Bulgaria). The plants were kept under various temperature conditions from 1$0^{\circ}C$ to 4$0^{\circ}C$ and 440$\mu$E/$m^2$/sec for 3 and 6hrs, and net $CO_2$ uptake were measured after 2hrs at $25^{\circ}C$. Photosynthetic optimal leaf temperature of ginseng was 21$^{\circ}C$ and tobacco was $25^{\circ}C$. Stomatal resistance and mesophyll resistance increased at high temperature. Especially, stomatal resistance seemed to have a significant role in determining the temperature responses of photosynthesis. In tobacco photosynthetic capacity was not changed by temperature treatment for 3hrs. However, 6hrs exposure reduced 8% of net photosynthesis at 4$0^{\circ}C$ and 12% at 1$0^{\circ}C$. Ginseng plants exposed for 6hrs at 4$0^{\circ}C$ lost photosynthetic capacity by 83%. Temperature responses of ginseng were very sensitive at above-optimum temperature resulting greater thermal inhibition other than photoinhibition.

The origins and the functions of the multi vesicular bodies and the various structures of the membranes related to the cytolysomes were studied in the mycelium cells of Rhizopus nigricans, Aspergillus niger and A. ochraceus, in the hymenium and basidium cells of Agricus bisporus sand Rhizopogon rubesecens, in the cells of assimilation tissue of Marchtantia polymorpha and Pogonalum inflexum and in the mesophyll cells of Pteridium aqiulinum, Pinus densiflora, Ginkgo biloba and Panax ginseng fixed with glutaraldehyde-paraformaldehyde-$ OsO_4$. In Rhizopus nigricans, Aspergillus niger, A. ochraceus, Agricus bisporus sand Rhizopogon rubescens, the concentric multilamellar, multivesicular, myelin-vesicle-tubular and concentric parallel-lamellar complexes were originated from the plasmalemma, while in Marehantia polymorpha, Pogonatum inflexum, Pteridium aquilinum, Pinus densiflora, Ginkgo biloba and Panax ginseng, they were originated from plasmalemma and the cytoplasm. The structures originated from the plasmalemma may be grouped into multi vesicular body and myelin-like structure, both forming the secondary vacuoles or protruding into the central vacuoles and finally degrading, In some cases, endoplasmic reticulum within the cytoplasm encloses some part of the cytoplasm to form a circle where the membranous lamellae increase in number, while the enclosed cytoplasm decrease to be eventually replaced by the multilamellar structure which is released into the vacuoles and subsquently degraded. The structures originated from the cytoplasm are believed to be the cytosegresomes or cytolysomes closely related to the differentiation of the vacuoles. The possible fate of these structures are also discussed.
Patterns of tannin accumulation in leaves of C-4 Euphorbia maculata have been examined using electron microscopy. Tannins, which are secondary metabolite phenolic compounds, were found to be deposited conspicuously in vacuoles of certain tissues regardless of their stage in development. However, patterns of deposit accumulation were distinguishable by their cell type during leaf differentiation. The deposits appeared most concentrated in the concentric bundle sheath cells enclosing veins, while little or no density was detected mostly in the mesophyll cells close to the epidermis. An ultrastructural study revealed that the deposits were restricted to the vacuoles at an early stage of leaf development; during which the vacuoles were almost completely filled with the tanniferous substances. The deposits themselves took different forms ranging from granules to huge globules while expanding leaf blade. As the leaf matured, the deposits accumulated either centripetally adjacent to the inner tangential tonoplast or by penetration into the cytoplasm amongst various cellular organelles, resulting in an extremely dense cytoplasm. Electron micrographs frequently showed the delineation of each organelle by the presence of dense deposits within the cytoplasm. Some large depository vacuoles filled with tannins had a corrugated appearance on the sectioned surface. The pattern and potential role of the deposits have been discussed.
Major contributions has been made in cellular ultrastructure studies with the use of high voltage electron microscopy (HVEM) and tomography. Applications of HVEM, accompanied by appropriate image processing, have provided great improvements in the analysis of three-dimensional cellular structures. In the present study, structural patterns of the crystalline bodies that are distinguished in mesophyll plastids of CAM-performing Sedum rotundifolium L., have been investigated using HVEM and tomography. Tilting, and diffraction pattern analysis were performed during the investigation. The titlting was performed at ${\pm}60^{\circ}\;with\;2^{\circ}$ increments while examining serial sections ranging from 0.125 to $1{\mu}m$ in thickness. The young plastids exhibited crystalline inclusion bodies that revealed a peculiar structural pattern. They were irregular in shape and also variable in size. Their structural attributes affected the plastid morphology. The body consisted of a large number of tubular elements, often reaching up to several thousand in number. The tubular elements typically aggregated to form a fluster The elements demonstrated either a parallel or lattice arrangement depending on the sectioning angle. The distance between the elements was approximately 20nm as demonstrated by the diffraction analysis. HVEM examination of the serial sections revealed an occasional fusion or branching of elements within the inclusion bodies. Finally, a three-dimensional reconstruction of the plastid crystalline bodies has been attempted using two different image processing methods.
The shoots of balloon flower (Platycodon grandiflorum A. DC.) in vitro germinated from seeds were cultured on MS basal medium containing 0.1 mg/L NAA under the various sucrose concentrations and with/without membrane filter (MF) on the lid of vessel. The growth responses were checked to obtain healthy plantlets. The $CO_2$ and $C_2$H$_4$ concentration in vessel without MF were higher than those with MF. The $CO_2$ concentration without MF was increased as days in culture went by whereas the $C_2$H$_4$ concentration was decreased. The plant growth with MF and high sucrose concentration was good. Fresh and dry weight of plantlets cultured in sucrose 4.5% with MF were higher than those in no sucrose without MF. Also the content of chlorophyll of plantlets cultured with MF was high and the content of sugar was shown a similar results and a remarkable difference between MF treatments, especially. Stomata cultured with MF was closer than that without MF and mesophyll of leaf were more developed with MF or in high sucrose concentration. When the plantlets were transplanted in the pot at 25$\pm$2$^{\circ}C$, 75% relative humidity and low PPFD (photosynthetic photon flux density), the percentage of survival after 13 days without MF was 0% but it was 100% with MF regardless of sucrose concentrations.

Histopathological changes in the witches'-broom mycoplasma (MLO) infected jujube trees after trunk injection of oxytetracycline HCl (OTC) solution were investigated by using light microscope, fluorescence microscope and electron microscope. In contrast to the healthy leaves, adaxial and abaxial bundle sheath extensions in the major veins of diseased leaves were highly developed, but mesophyll cells were very small, loose and necrosed. In the diseased leaves, excessive phloem was formed, xylem differentiation was supressed and polyphenols were accumulated. Jujube trees treated with 500ml of 2000ppm solution of OTC showed complete remission of witches'-broom symptoms above the injection site, but cells in the sheath extention were hypertrophied and sieve tube necrosis occurred in the secondary phloem of major veins. MLO-specific fluorescence was not observed in the samples of diseased jujube trees treated with OTC, while those from non-treated infectedstress distinctly showed a moderate amount of MLO-specific fluorescence. In the observation under the electron microscope, sieve element of diseased tissues and its associated companion cells were found to have collapsed. Parenchymas cells contained many large starch filled plastids and most samples of diseased and OTC-treated tissues showed an abnormal accumulation of starch compared to the healthy controls. Mycoplasma-like organisms were observed only within mature sieve elements in diseased tissues, but never in OTC-treated tissues.
The effects of selenium (Se) and germanium (Ge) fertilization on the growth and quality of Chinese cabbages cultivated in spring and autumn and peppers cultivated in spring were investigated. $Se\;(Na_2SeO_4)\;and\;Ge\;(GeO_2)$ were supplied 5, 10, or 20 times in an aqueous solution of 0, 2, 4, or $8mg{\cdot}L^{-1}$ during the cultivation of Chinese cabbages and peppers. The fresh weight of Chinese cabbages increased by Ge fertilization with high concentration. But it was not affected by Se fertilization. The content of vitamin C increased by 10 times application with $4mg{\cdot}L^{-1}$ of Se or Ge. The concentration of Se in Chinese cabbage increased according to increasing concentration of Se fertilization. Se concentration was higher in the outer leaves than in the inner leaves. Se concentration in the mesophyll was higher than that in the midrib. Ge fertilization increased the uptake and concentration of Ge in autumn-cultivated Chinese cabbages. Se and Ge fertilization did not affect the fresh weight of peppers. The content of vitamin C in pepper increased by 20 times application of $2mg{\cdot}L^{-1}$ of Se. Vitamin C content in red peppers was twice as much as in green peppers.
