신생 흰쥐와 성숙 흰쥐 각막의 미세구조 변화에 관한 연구 (An Ultrastructral changes of the new-born rats and adult rat's cornea)

  • 본 연구는 발생과정에 따른 흰쥐 각막의 변화를 주사 및 투과형 전자현미경으로 관찰하였다. 생후 1일된 안검이 개검되지 않은 흰쥐의 각막 60안, 생후 10주된 성체 흰쥐의 각막 40안을 적출하여 각막의 변화를 비교 연구 하였다. 연령증가에 따라 각막의 미세구조의 변화가 나타났으며 각막의 상피는 오각형의 모양이며, 세포간 경계는 분명하였으나 성장할수록 탈락세포는 짙은 전자밀도를 나타냈으며, 새로 생성된 세포는 밝은 전자밀도를 나타내어 명암의 차이가 뚜렷하였다. 보우만층은 연령의 증가에 따라 콜라겐섬유가 불균질한 구조에서 균질한 구조로 변화하였다. 실질층의 각막세포는 세포 소기관의 밀도가 성체로 갈수록 감소하였고, 콜라겐층은 연령증가에 따라 종횡으로 평행배열을 나타내었다. 데스멧층은 성체로 갈수록 "banded layer"에서 "non-banded layer"의 형태로 변화하였으며 층의 두께가 현저히 두꺼워진것이 관찰되었다. 내피층은 단층으로 구성되어 있으며, 성체로 갈수록 내피 세포수는 감소하는 반면 감소된 세포수의 자리는 세포 크기가 확대됨으로써 보상되었다.

cis-Dichlorodiammineplatinum (II) 이 생쥐 폐포간중격의 미세구조에 미치는 영향 (Effects of the cis-Dichlorodiammineplatinum on the Fine Structures of the Interalveolar Septum in the Mouse)

  • cis-Dichlorodiammineplatinum (II) (cis-Platin), a metallic compound, has widely been used as an effective anticancer chemotherapeutic agent. The precise mechanism of action of this agent is still unknown, but it is postulated that cis-Platin may act on the cancer cell like bifunctional alkylating agents. Although this agent is very beneficial to the patients with cervical cancer, germinoma of testis, neuroblastoma and others, it may also damage to the normal cell so that many side effects; severe hemorrhagic enterocolitis, bone marrow depression, renal damage and liver damage will develope. This experiment has been undertaken to pursue the cytotoxic effects of the cis-Platin on the ultrastructures of the interalveolar septum in the mouse lung. A total of 55 healthy male mice of ICR strain were used as experimental animals and divided into 5 mice of normal control group and 50 mice of cis-Platin treated group. The mice of cis-Platin treated group were sacrificed by carotid exsanguination at 6, 12, 24 hours, 3 days and 7 days after intraperitoneal injection of 6.0 mg of cis-Platin ($Abiplatin^R$ Abic Co. Ltd.) per kg of mouse body weight. The specimen obtained from the lower lobe of left lung were sliced into $1mm^3$ and prefixed with 2% glutaraldehyde -2.5% paraformaldehyde solution prepared with Millonig's phosphatae buffer solution (pH 7.4) at $4^{\circ}C$ for 3-4 hours. After postfixation with 1% osmium tetroxide solution all specimens were embedded in Epon 812. Ultrathin sections about $600-800{\AA}$ in thickness were stained with uranyl acetate and lead citrate and observed with Hitachi-600 electron microscope. The results obtained were as follows: 1. Local swellings with increase of electron density and number of pinocytic vesicles in the cytoplasms of the type I pneumocyte and endothelial cell of the blood air barrier in interalveolar septum of cis-platin treated mice were observed. 2. Cisternae of rough endoplasmic reticulum were dilated and sacculated in association with detachment of membrane bound ribosomes of the type II pneumocyte in interalveolar septum of cis-Platin treated mice. 3. Swollon mitochondria with uneven electron density of their matrix were observed in the type II pneumocyte of interalveolar septum in the cis-Platin treated mice. 4. The lamellae of lammelar bodies in type II pneumocyte of interalveolar septum in cis-Platin treated mice were devoided or transformed into homogeneous electron dense material. It is consequently suggested that cis-Platin would induce the cellular edema of type I pneumocyte and endothelial cell, and degenerative changes of cytoplasmic organelles of the type II pneumocyte in the interalveolar septum of the mouse lung.

오플록사신 및 리도카인 함유 수분 감응성 구강점막 패취제의 제조 및 방출 특성 (Preparation and Release Properties of Oromucosal Moisture-activated Patches Containing Lidocaine or Ofloxacin)

  • This study was aimed to design and formulate the moisture-activated patches containing ofloxacin and lidocaine for antibacterial and local anesthetic action. The solubility of lidocaine at $32^{\circ}C$ in various vehicles decreased in the rank order of PG $759.5{\pm}44.5\;mg/mL$ > PGL > IPM > PEG 300 > PEG 400 > Ethanol > PGMC > DGME > PGML > OA > $Captex^{\circledR}\;300$ > $Captex^{\circledR}\;200$ > water $(4.0{\pm}0.1\;mg/mL)$. Ofloxacin revealed very low solubility, which the highest solubility was obtained from PEG 400 $(18.7{\pm}6.3\;mg/mL)$ among the vehicles used. The addition of lactic acid increased the solubility of ofloxacin dramatically; the solubility at 5% lactic acid was $133.7{\pm}9.7\;mg/mL$. As $2-hydroxypropyl-{\beta}-cyclodextrin$ was added at the concentrations of 40, 80, 120, 160 and 200 mM, the solubilities of lidocaine and ofloxacin were enhanced up to three and two times, respectively, with concentration-dependent pattern. Gel intermediates for filmtype patches were prepared with mucoadhesive polymer, viscosity builders, lidocaine or ofloxacin at pH values from 5 to 7. Gels were cast onto a release liner and dried at room temperature. Dried patch was attached onto an adhesive backing layer, thus forming a patch system. Patches containing a single drug component were characterized by in vitro measurement of drug release rates through a cellulose barrier membrane. The release study was carried out at $37^{\circ}C$ using a Franz-type cell. Receptor solutions were isotonic phosphate buffers (pH 7.4). Samples $(100\;{\mu}L)$ were taken over 24 hours and quantitated by a verified HPLC method. The releases from all tested were proportional to the square root of time. The release rates were 0.9, 157.3 and $281.7\;{\mu}g/cm^{2}/min^{1/2}$ for the lidocaine patches and 19.8,37.2 and $50.7\;{\mu}g/cm^{2}/min^{1/2}$ for the ofloxacin patches at the concentrations of 0.3, 0.5 and 1 %, respectively. The release rates were dose dependent in both drug patches $(R^{2}\;=\;0.9077\;for\;lidocaine;\;R^{2}\;=\;0.9949\;for\;ofloxacin)$ and those were also thickness-dependent $(R^{2}\;=\;0.9246\;for\;lidocaine;\;R^{2}\;=\;0.9512\;for\;ofloxacin)$.

A New Strategy to Fabricate a Colloidal Array Templated $TiO_2$ Photoelectrode for Dye-sensitized Solar Cells

  • Nanocrystalline titanium dioxide ($TiO_2$) materials have been widely used as an electron collector in DSSC. This is required to have an extremely high porosity and surface area such that the dye can be sufficiently adsorbed and be electronically interconnected, resulting in the generation of a high photocurrent within cells. In particular, their geometrical structures and crystalline phase have been extensively investigated as important issues in improving its photovoltaic efficiency. In this study, we present a new strategy to fabricate a photoelectrode having a periodic structured $TiO_2$ film templated from 1D or 3D polystyrene (PS) microspheres array. Monodisperse PS spheres of various radiuses were used for colloidal array on FTO glasses and two types of photoelectrode structures with different $TiO_2$ materials were investigated respectively. One is the igloo-shaped electrode prepared by $TiO_2$ deposition by RF-sputtering onto 2D microsphere-templated substrates. At the interface between the film and substrate, there are voids formed by the decomposition of PS microspheres during the calcination step. These holes might be expected to play the predominant roles as scattering spherical voids to promote a light harvesting effect, a spacious structure for electrolytes with higher viscosity and effective paths for electron transfer. Additionally the nanocrystalline $TiO_2$ phase prepared by the RF-sputtering method was previously reported to improve the electron drift mobility within $TiO_2$ electrodes. This yields solar cells with a cell efficiency of 2.45% or more at AM 1.5 illumination, which is a very remarkable result, considering its $TiO_2$ electrode thickness (<2 ${\mu}m$). This study can be expanded to obtain higher cell efficiency by higher dye loading through the increase of surface area or multi-layered stacking. The other is the inverse opal photonic crystal electrode prepared by titania particles infusion within 3D colloidal arrays. To obtain the enlargement of ordered area and high quality of crystallinity, the synthesis of titania particles coated with a organic thin layer were applied instead of sol-gel process using the $TiO_2$ precursors. They were dispersed so well in most solvents without aggregates and infused successfully within colloidal array structures. This ordered mesoporous structure provides the large surface area leading to the enough adsorption of dye molecules and have an light harvesting effect due to the photonic band gap properties (back-and-forth reflection effects within structures). A major advantage of this colloidal array template method is that the pore size and its distribution within $TiO_2$ photoelectrodes are determined by those of latex beads, which can be controlled easily. These materials may have promising potentials for future applications of membrane, sensor and so on as well as solar cells.

치은부에 이식한 이중인공진피의 조직학적 치유 (Histological healing after grafting of bilayer artificial dermis in the oral environment)

  • The autogenous free gingival graft is the most predictable procedure currently used to increase the width of the attached gingiva in periodontics. But the major disadvantage of the procedure is to create the multiple surgical wounds at both a donor site and a recipient site. The other problem is the limited amount of available graft material in oral cavity. Therefore, recent researches have been focused to develop the biomaterial to substitute the autogenous gingival tissue. The purpose of this study was to evaluate the histologic healing after grafting of bilayer artificial dermis, compared to the free gingival graft. Four non-smoking subjects (mean age, 32.5 years) in systemically healthy state and good oral hygiene were selected according to their particular needs for correction of mucogingival problems as suggested by Nabers(1966). The recipient sites were prepared through the procedure for the free gingival graft and were grouped according to the graft materials: Experimental group(n=5) - bilayer artifcia1 dermis ($Terudermis^{(R)}$; Terumo Co. Japan) and Control group(n=6) - free gingival graft with autogenous palatal mucosa. Biopsies were harvested at 1,2,3 and 6 weeks postsurgery to evaluate histologically. At the third week in the experimental group and at the second week of in the control group, the grafts has been clinically stabilized on the recipient bed and the graft border has been blended into the surrounding tissue. In the experimental group after 1 week of grafting, the epithelial migration from the adjacent tissue to graft material was seen and after 3 weeks of grafting, the : nflmmation decreased, collagen layer of the artificial dermis was lost and the basement membrane of epithelium was formed. After 6 weeks of grafting, both groups demonstrated orthokeratinized epithelium and increased thickness of epithelial tissue and the rete peg formation, similar to the adjacent tissue, Histologic evaluation revealed a biologic acceptance and incorporation of the collagen layers of the graft tissue to the host tissue, without foreign body reaction. In conclusion, a bilayer artificial dermis is essentially similar to autogeneous free gingival graft in the correction of mucogingival problems, and has the advantages of decreased patient morbidity (no donor site) and availability of abundant amounts of graft material when needed.

Evaluation of tissue ingrowth and reaction of a porous polyethylene block as an onlay bone graft in rabbit posterior mandible

  • Purpose: A new form of porous polyethylene, characterized by higher porosity and pore interconnectivity, was developed for use as a tissue-integrated implant. This study evaluated the effectiveness of porous polyethylene blocks used as an onlay bone graft in rabbit mandible in terms of tissue reaction, bone ingrowth, fibrovascularization, and graft-bone interfacial integrity. Methods: Twelve New Zealand white rabbits were randomized into 3 treatment groups according to the study period (4, 12, or 24 weeks). Cylindrical specimens measuring 5 mm in diameter and 4.5 mm in thickness were placed directly on the body of the mandible without bone bed decortication, fixed in place with a titanium screw, and covered with a collagen membrane. Histologic and histomorphometric analyses were done using hematoxylin and eosin-stained bone slices. Interfacial shear strength was tested to quantify graft-bone interfacial integrity. Results: The porous polyethylene graft was observed to integrate with the mandibular bone and exhibited tissue-bridge connections. At all postoperative time points, it was noted that the host tissues had grown deep into the pores of the porous polyethylene in the direction from the interface to the center of the graft. Both fibrovascular tissue and bone were found within the pores, but most bone ingrowth was observed at the graft-mandibular bone interface. Bone ingrowth depth and interfacial shear strength were in the range of 2.76-3.89 mm and 1.11-1.43 MPa, respectively. No significant differences among post-implantation time points were found for tissue ingrowth percentage and interfacial shear strength (P>0.05). Conclusions: Within the limits of the study, the present study revealed that the new porous polyethylene did not provoke any adverse systemic reactions. The material promoted fibrovascularization and displayed osteoconductive and osteogenic properties within and outside the contact interface. Stable interfacial integration between the graft and bone also took place.

Y-doped BaZrO3을 이용한 저온형 박막 연료전지 연구 (Study on Low-Temperature Solid Oxide Fuel Cells Using Y-Doped BaZrO3)

  • 본 연구에서는 저온형 연료전지와 고온형 연료전지의 작동 및 구성 요소 측면 단점들을 보완하기 위해 중온 영역에서 작동하는 박막 연료전지를 제작하였다. 박막 연료전지는 이트륨이 도핑된 바륨 지르코네이트(BYZ) 전해질과 백금 수소극/공기극으로 이루어져 있으며, 성능은 $350^{\circ}C$에서 측정하였다. 350nm의 두께를 가지는 백금 수소극은 다공성 기판 위에 스퍼터링 기법을 이용하여 증착하였다. BYZ전해질은 펄스레이저 기법을 이용하여 $1{\mu}m$ 증착하였고, 상부에 스퍼터링 기법을 이용하여 200nm의 두께를 가지는 백금 공기극을 증착하였다. 개회로 전압은 약 0.81V이었고, 최대 출력 성능은 11.9mW/$cm^2$이었다.

동절기 피해의 이해와 겨울철 골프장 관리: 리뷰 (Winterkill and Strategy of Golf Course Management: A Review)

  • 동절기 피해는 겨울철에 나타나는 피해의 형태로 유형에 따라 저온피해, 건조피해, 관부수화, 그리고 동절기 병 발생등이 있다. 저온피해는 대기온도와 토양온도에 의해 그 피해가 발생하는데 토양온도가 관부의 생육에 더 큰 영향을 미치기 때문에 그 피해의 정도가 더 심하게 발생한다. 관부수화는 세포내결빙에 의해 발생하는 피해로 세포막을 파괴하며 결과적으로 세포내 조직을 탈수시키게 된다. 저온건조는 동절기 기간동안 식물체의 잎 혹은 식물전체에 건조로 인한 피해가 나타나 고사에 이르게 되는 피해를 말한다. 동절기 피해를 예방하기 위해 가을철 인산과 칼륨의 시비가 동절기 피해에 대한 내성을 증진시키는 역할을 하게 된다. 또한 피복재의 사용이 동절기 피해를 최소화 하는 기능을 하는데 그 종류는 여러가지가 있으며 겨울철 잔디위에 쌓인 눈도 대기온도로부터 토양온도를 유지하는 피복재 역할을 하게 된다. 동절기 피해를 최소화 하기 위한 방법으로 통기와 배토의 방법이 있다. 통기와 배토는 모두 주로 대취층의 두께를 줄이기 위한 방법으로 사용이 되고 또한 겨울철 배토를 위해 사용되는 검은색 모래는 토양에 잔존해 있는 얼음을 녹이고 토양온도를 유지해주는 역할을 하게 되므로 피복재와 같이 동절기 피해를 최소화 하는 역할을 하게 된다.

p-수렴 완전층별모델에 의한 일면패치로 보강된 원공 적층판의 휨효과 (Bending Effect of Laminated Plates with a Circular Hole Repaired by Single-Sided Patch Based on p-Convergent Full Layerwise Model)

  • 기존에 설치되어 있는 구조물의 양면대칭 패치보강은 항상 면내거동만을 유발하나 시공상 어려움이 있다. 반면에 일면 패치보강의 경우 인장력의 증가에 따라 중립축의 위치가 대칭이 아니므로 휨에 대한 강성도가 증가하게 되며, 결과적으로 적층판의 휨을 심화시키게 된다. 이 연구에서는 일면 패치보강된 적층판의 두께방향은 물론이고 원공주위의 응력집중계수를 산정하기 위해 p-수렴 완전층별모델을 제안하였다. 가정된 변위장의 정의를 위해, 임의의 층에서 변위-변형률 관계와 3차원 구성방정식은 2차원 및 3차원 계층적 형상함수의 조합이 사용된다. 원형경계의 기하형상을 나타내기 위해 초유한사상기법이 사용되며, 다른 외삽법을 사용하지 않고 각 층마다 절점에서의 응력값을 직접적으로 얻기위해 가우스-로바토 수치 적분이 수행되었다. 제안된 모델의 정확도와 단순성은 기존의 3차원 유한요소해석과 실험에 의해 구해진 결과들과의 비교를 통해 검증되었다. 또한 정사각형, 원형, 고리형 형상의 다양한 패치보강에 따른 휨효과를 조사하였다.

볼락, Sebastes inermis 산출 후 초기 성장 및 눈의 조직학적 발달 특성 (Early Growth and Characteristic of Histological Eye Development in Post Parturition Dark banded Rockfish, Sebastes inermis)

  • 어류의 시각 구조와 시각 기능 연구에 있어서 행동적 요인이나 환경적 요인의 중요성은 시각생태학에서 매우 중요하게 다루어지고 있으며, 볼락, Sebastes inermis에 관하여는 아직까지 초기 눈의 성장 및 발달에 관한 발생생물학적 연구가 이루어진 바가 없음을 고려하여 이에 대한 연구를 수행하였다. 볼락의 전장, 두장, 두고, 눈의 직경과 수정체 직경은 산출 직후부터 산출 후 60일까지의 단계에서 양성의 상관관계(positive allometry)를 보여주었다. 전장과 관련된 두장과 두고에서의 증가와, 두고와 관련된 눈의 직경과 수정체 직경의 성장은 거의 동형(isometric)성장을 나타내었다. 눈은 산출단계에 완전히 형성되었다. 산출 단계의 눈은 시신경 섬유층, 신경절 세포층, 내망상층, 내과립층, 외망상층, 외과립층, 외경계막, 간체, 추체층 및 상피층을 가지며, 산출 후 60일의 망막 두께는 산출 직후의 망막 두께보다 더 크다. 본 실험기간 동안, 망막의 간체와 추체층의 비율 및 외과립층과 시신경의 비율은 상당히 증가한 반면, 망막의 외망상층과 내과립층, 신경절 세포층의 비율은 현저하게 감소하였다(P<0.05). 차후, 본 연구에 사용된 볼락 망막에 대한 빛 민감성과 공간 해상력에 관한 부수 연구가 필요하리라 사료된다.
