Melanin-dispersing Effect of Vasopressin on Frog Skin Melanocyte (Vasopressin의 멜라닌 확산작용에 대하여)

  • It has been reported that vasopressin disperse the melanophore granule of frog skin. The author used hypophysectomized and adrenergic receptor blockaded animals in order to define the mechanism of vasopressin on the melanopore pigment of frog skin. The Rana niglomaculata which could be found in the Seoul area were used on this experiment. The amount of the following drugs were injected into the lymphatic sac of the frog; vaospressin $(0.05\;{\mu}g/g\;B.W.)$, dibenzylin $(0.05\;{\mu}g/g\;B.W.)$, and propranolol $(0.01\;{\mu}g/g\;B.W.)$. The following results were observed; 1. Vasopressin dispersed the melanin granules of melanocyte of frog skin. 2. The melanin granule dispersion activity of vasopressin was observed on the hypophysectomized frog. 3. The melanin granule dispersion was observed on the adrenergic receptor blockaded frog with dibenzylin or propranolol respectively, especially the later one was found to be more obvious. 4. The melanin granule dispersion was observed on the frog which was injected with vasopressin after alpha-receptor or beta-receptor blockade and the later one was found to be more obvious. 5. The melanin granule dispersion was more effective with the blockade of beta-receptor after the treatment with vasopressin on hypophysectomized frog.

Observation of the Structure of Melanin Granule in Mummy's Hair Shaft Using High-Voltage Electron Microscopy (초고압전자현미경을 이용한 미라 모발 멜라닌과립의 구조 관찰)

  • Three-dimensional melanin granules of mummy's hair analyzed using high voltage electron microscopy at 1250kV. The melanin granules in the cortex of mummy's hair located in close proximity to the outer of cortex in the adjacent place of the cuticles. A lot of melanin granule performed in this area. It is a distinguished difference from modern human. While it rotated up to 60 degree counter-clockwise and taken a photo per one degree using high voltage electron microscopy. The melanin granule displayed with long elliptical and variable at the size. The size of granule is measured from $0.3{\sim}0.6{\mu}m$ in minimum diameter and $0.5{\sim}1{\mu}m$ in maximum diameter. Conclusionally, high voltage electron microscope has higher resolution and penetration power than conventional transmission electron microscope that could be load biological thick specimen.

  • The physicochemical properties of damaged hair by irradiation of ultraviolet-B (UV-B) have been investigated by using transmission electron microscope and scanning electron microscope. The range of irradiation of hair irradiated for expectative 6 hours, 12 hours, 24 hours and 48 hours with stimulated ultraviolet ray. The treated hairs showed characteristic morphological damage pattern in the cornified cell of matrix and the cuticle following time past. The various sized vacuoles in the endocuticle of the cuticular cells was formed. The statistically significant differences in diameter of cuticular cell were observed in terms of tranverse swelling by formation of vacuoles. The hair cortex and matrix undergo long term exposure to UV-B radiation. The macrofibrils of cortex appeared to be affected most by UV-B, although the morphology and volume of melanin granule was not changed. The physicochemical destruction of hair matrix and cuticular cells is largely accelerated by long term irradiation of UV-B.

Morphological Change of Men's Hair Shaft by Weathering (풍화작용에 의한 정상모발의 형태학적 변화)

  • The morphological changes in normal and weathering hair shafts of the human scalp were investigated by using the transmission and scanning electron microscopes. The hair shaft composed of cuticular layer, cortex and medula. The surface of normal hairs are smooth and covered by imbricated cuticular scales. The cuticular layer consists of five to seven cuticle cells. These cells, which are flat and thin, measuring about $100{\mu}m$ long and $0.4{\mu}m$ thick, appears intercellular membrane complex in diameter 25 nm. The cortex composed of melanin granules and cornified cells, which multicomponent concentric microfibrils in diameter about 8 nm give rise to macrofibrils in diameter $0.5{\mu}m$ to $0.8{\mu}m$ encased in limiting membrane. The melanin granules are spherical shaped about $0.5{\mu}m$ in size and scattered between macrofibrils. The medulla in the normal hairs are $16{\mu}m$ in diameter centrally region of cortex. Normal hair shafts undergo progressive degenerative changes due to a variety of environmental insults. In the initial weathering process of hair, the cuticular scales became irregularly raised and broken, and then cuticle cells formed cytoplasmic vacuolation, following dissociated intercellular membrane complex, ultimately entirely lost and nuded cortex. Occasionally, transverse fissures were seen at hair shafts indicating that the hairs were deteriorated. Complete removal of the cuticular layer in the heavily damaged cortex portions appeared splitting of the cortical cell into its macrofibrils and scattering of melanin granules.

  • The ultrastructure of the cutaneous chromatophores and the origin of pigment granules in the goldfish, Carassius auratus L., are studied with electron microscope. Leucophores contain amorphous pigment granules, leucosomes and melanosomes contain oval shaped electron dense melanin pigment granules. These pigment granules are synthesized from the Golgi complexes of each chromatophores and released into the cytoplasm. Among the dermal chromatophores, another kind of dermal cells, named rER rich cells, are also observed. And pinocytotic vesicles are being formed only the inner surface of the chromatophores adhering to the rER rich cells. From these observations, such hypothesis are being possible that pigment materials are synthesized and transfered from the rER rich cells to dermal chromatophores by the pinocytosis. And next remodelizing mechanisms are carried out by the Golgi complexes of each chromatophores.

Melanotrichia in Poodles : Clinical and Histopathological Findings (푸들에서 발생한 melanotrichia: 임상적 그리고 조직병리학적 특징)

  • A 5-year-old, female Poodle and her daughter, 2-year-old, were referred to Konkuk University Veterinary Teaching Hospital (KVTH) due to discoloration of hair on dorsum, and generalized alopecia following clipping. Some papules and pustules are found on patch area. Complete blood counts and serum chemistry profiles were not remarkable. Trichogram revealed that melanin-granule was pigmented on hair bulb and shaft in affected area and no melanin pigmentation was noted in non-patch area. And skin biopsy showed mild epidermal hyperplasia and mild inflammation in dermis of the patch area. This case report indicates that melanotrichia can be occurred by postclipping inflammation. However, the mechanism of melanocyte proliferation and rates of melanogenesis in response to postclipping remains to be unclear in these cases.

  • Most of the electromagnetic agents used in physical therapy rely for their effects on tissue heating and photoreaction of the body. Infrared and untraviolet light on the other hand, owns its place in medicine because it produces direct photochemial reaction when it interacts with the body. This study was carried out to investigate and review for photoreaction of the akin. The results were an follows. 1. The effects of the infrared are heat production, increasing metabolism, increasing circulation, vasodilatation and pigmentation. 2. Directed photoreactions are divided into acute reaction and chronic reaction, and the acute reaction makes pigmentation from $290\~320nm$ of ultraviolet ray. 3. Ultraviolet ray formated pigmentations, which are melanoblasts excited from ultraviolet ray and received chemical stimulation, that make melanin granule. 4. If exposured with long duration, at ultraviolet ray, it makes skin thickening and epithelioma. 5. Indirected photoreaction is made by existenced hypersensitivity of photoreaction or lack of photodefence structure. 6. The phototoxic reactions are synthesized by chemical reaction of excitement from ultraviolet ray also this time analysis, synthesis and polymerization from energy of a circumferenced substance. 7. Sunscreen substances are P-amino benzoic acid and oxidate titan.

The Fine Structure of Human Fetal Nail Matrix (한국인 태아 조기질의 미세구조에 관한 연구)

  • The differentiation of nail matrix and fine structure of matrix cells were studied with light and electron microscope using specimens from nails of thumb finger in Korean fetuses 14 to 24 weeks old. Fetal nail matrix consisted of two horizontal layers, thicker ventral and thinner dorsal matrices, originating from invagination of epidermis in proximal nail field. Matrix being generally thicker in its distal region than the apex became gradually thickened with increase of the fetal age. Each matrix consisted of single layer of basal cells and multiple layers of squamous cells which are arranged close to and parallel to the central axis of the nail mairix. The process of keratinization of fetal nail matrix was noted to be occured concurrently in the ventral and dorsal matrices along the central axis of matrix toward distal and dorsal direction. Squamous cells became matured with accumulation of tonofilaments, increase of keratohyalin granules, discharge of membrane coating granules, and narrowing of intercellular spaces, thickening of plasma membrane and finally being transformed into horny cells of nail plate. Horny cells of nail plate filled with fibrous elements in the electron dense amorphous substance. These findings of keratinization process of fetal nail matrix appeared to be similar to those of keratinization in epidermis and inner root sheath of the hair. In the nail matrix, however, corresponding region to the keratogenous zone of growing hair follicle was not observed. Vacuolated squamous cells of nail matrix seen on light microscopy was considered to be artefactual product, but squamous cells with condensed small nuclei rarely found adjacent nail plate was considered to be one of the squamous cells with unknown function. Proximal end of nail plate was observed on dorsal surface of nail field distal to the proximal nail fold at 14 and 16 weeks old human embryos. Proximal prolongation of the proximal end of nail plate was occured with advancing fetal age and afterward 21 weeks nail plate invaded into nail matrix. Melanin granule containing cells and Merkel cells were present only on the basal layer of dorsal nail matirx.
