Relationships in Cd, Hg, Pb and Zn Concentrations Between Hair and Internal Organs of Korean (한국인 모발과 내부장기조직간 Cd, Hg, Pb 및 Zn함량의 상호관련성)

  • In order to study the relationships between trace element concentrations of hair and internal organs, autopsy samples of hair, liver, kidney cortex, kidney medulla, heart, lung, spleen, cerebrum, testis and bone from 162 Korean subjects were analyzed for Cd, Hg, Pb and Zn by inductively coupled plasma atomic emission spectrometry. High significant positive correlations were found between the Cd concentration in hair and bone and Pb in hair and lung. Hair Hg levels were highly significantly correlated with organ Hg levels in the liver, kidney cortex, kidney medulla, heart, lung, spleen, cerebrum, testis and bone. And hair Zn levels were highly significantly correlated with organ Zn levels in the kidney cortex, kidney medulla, spleen, testis and bone.

  • To determine the optimal transplanting time of wanggol in southern areas of Korea, wanggol c.v. Gangsan the highest yielding variety was grown under three different tansplanting times. Yield components such as stem length, number of tillers, number of heading stem and stems diameters were the highest at the transplanting time of May 20. Plants sown of Mar 30 showed the highest dry cortex and medulla yield. Judging from the results reported above, at optimum transplanting time combined with sowing time toy yield of wanggol (cyperus iwassaki Makino) seemed to be the time of May 20 and Mar. 30.

  • It has been demonstrated the presence of leptin receptors (Ob-Ra) on epinephrine-secreting chromaffin cells in rat adrenal medulla, suggesting that leptin may directly affect the adrenal medulla (Cao et al., 1997). Leptin is found to stimulate catecholamine (CA) synthesis in cultured bovine adrenal medullary cells (Utsumomiya et al., 2001; Shibuya et al., 2002)and cultured porcine adrenal medullary cells (Takekoshi et al., 2001). Thus, the present study was designed to examine the effect of leptin on CA release from the isolated perfused rat adrenal gland, and to establish its mechanism of action. (omitted)

Gintonin facilitates catecholamine secretion from the perfused adrenal medulla

  • The present study was designed to investigate the characteristics of gintonin, one of components isolated from Korean Ginseng on secretion of catecholamines (CA) from the isolated perfused model of rat adrenal gland and to clarify its mechanism of action. Gintonin (1 to $30{\mu}g/ml$), perfused into an adrenal vein, markedly increased the CA secretion from the perfused rat adrenal medulla in a dose-dependent fashion. The gintonin-evoked CA secretion was greatly inhibited in the presence of chlorisondamine ($1{\mu}M$, an autonomic ganglionic bloker), pirenzepine ($2{\mu}M$, a muscarinic $M_1$ receptor antagonist), Ki14625 ($10{\mu}M$, an $LPA_{1/3}$ receptor antagonist), amiloride (1 mM, an inhibitor of $Na^+/Ca^{2+}$ exchanger), a nicardipine ($1{\mu}M$, a voltage-dependent $Ca^{2+}$ channel blocker), TMB-8 ($1{\mu}M$, an intracellular $Ca^{2+}$ antagonist), and perfusion of $Ca^{2+}$-free Krebs solution with 5mM EGTA (a $Ca^{2+}$chelater), while was not affected by sodium nitroprusside ($100{\mu}M$, a nitrosovasodialtor). Interestingly, LPA ($0.3{\sim}3{\mu}M$, an LPA receptor agonist) also dose-dependently enhanced the CA secretion from the adrenal medulla, but this facilitatory effect of LPA was greatly inhibited in the presence of Ki 14625 ($10{\mu}M$). Moreover, acetylcholine (AC)-evoked CA secretion was greatly potentiated during the perfusion of gintonin ($3{\mu}g/ml$). Taken together, these results demonstrate the first evidence that gintonin increases the CA secretion from the perfused rat adrenal medulla in a dose-dependent fashion. This facilitatory effect of gintonin seems to be associated with activation of LPA- and cholinergic-receptors, which are relevant to the cytoplasmic $Ca^{2+}$ increase by stimulation of the $Ca^{2+}$ influx as well as by the inhibition of $Ca^{2+}$ uptake into the cytoplasmic $Ca^{2+}$ stores, without the increased nitric oxide (NO). Based on these results, it is thought that gintonin, one of ginseng components, can elevate the CA secretion from adrenal medulla by regulating the $Ca^{2+}$ mobilization for exocytosis, suggesting facilitation of cardiovascular system. Also, these findings show that gintonin might be at least one of ginseng-induced hypertensive components.

Effects of Losartan on Catecholamine Release in the Isolated Rat Adrenal Gland

  • The aim of this study was to determine whether losartan, an angiotensin II (Ang II) type 1 ($AT_1$) receptor could influence the CA release from the isolated perfused model of the rat adrenal medulla. Losartan (5${\sim}$50 ${\mu}$M) perfused into an adrenal vein for 90 min produced dose- and time-dependent inhibition of the CA secretory responses evoked by ACh (5.32 mM), high $K^+$ (56 mM, a direct membrane depolarizer), DMPP (100 ${\mu}$M) and McN-A-343 (100 ${\mu}$M). Losartan failed to affect basal CA output. Furthermore, in adrenal glands loaded with losartan (15 ${\mu}$M) for 90 min, the CA secretory responses evoked by Bay-K-8644 (10 ${\mu}$M, an activator of L-type $Ca^{2+}$ channels), cyclopiazonic acid (10 ${\mu}$M, an inhibitor of cytoplasmic $Ca^{2+}$ -ATPase), veratridine (100 ${\mu}$M, an activator of $Na^+$ channels), and Ang II (100 nM) were markedly inhibited. However, at high concentrations (150${\sim}$300 ${\mu}$M), losartan rather enhanced the CA secretion evoked by ACh. Collectively, these experimental results suggest that losartan at low concentrations inhibits the CA secretion evoked by cholinergic stimulation (both nicotininc and muscarinic receptors) as well as by membrane depolarization from the rat adrenal medulla, but at high concentration it rather inhibits ACh-evoked CA secretion. It seems that losartan has a dual action, acting as both agonist and antagonist to nicotinic receptors of the rat adrenal medulla, which might be dependent on the concentration. It is also thought that this inhibitory effect of losartan may be mediated by blocking the influx of both $Na^+$ and $Ca^{2+}$ into the rat adrenomedullary chromaffin cells as well as by inhibiting the $Ca^{2+}$ release from the cytoplasmic calcium store, which is thought to be relevant to the $AT_1$ receptor blockade, in addition to its enhancement of the CA release.

Immunohistochemical study on endocrine cells of the thymus of duck(Anas platyrhynchos platyrhyncos, Linne) (청둥오리의 가슴샘 내분비세포에 대한 면역조직화학적 연구)

  • Endocrine cells in the thymus of duck(Anas platyrhynchos platyrhyncos, Linne) were studied immunohistochemically from 23 days of incubation to 32 weeks of age. Somatostatin-, growth hormone(GH)-, gastrin/cholecystokinin(Gas/CCK)-, polypeptide YY(PYY)-, S-100 protein(S-100 P)-, dopamine-, serotonin(5-HT)-, and bovine chromogranin (BCG)-immunoreactive cells were detected in the duck thymus by the PAP techniques. These immunoreactive cells were observed in the medulla and in the juxtacortical medulla. No immunoreactivity of calcitonin and bovine pancreatic polypeptide(BPP) antiserum were observed. The argyrophil cells by Grimelius techniques were observed from 23 days of incubation to 32 weeks of age and peaked in 5 weeks of age. In the early develpmental stage, the argyrophil cells were distributed only in the medulla, while these cells were distributed mainly in the medulla and a few cells were distributed in the corticomedullary junction from 3 weeks of age. These immunoreactive cells were generally round, oval and elliptical and occasionally spindle, polygonal and polymorphous with the long cytopslasmic processes in shape. The present study suggests that the intrathymic endocrine cells may associate with the functional maturation of T-lymphocytes on the establishment of immunity. The further study will be needed to elucidate the function of these thymic endocrine cells.

Surgical Outcome of Adult Idiopathic Chiari Malformation Type 1

  • Objective : The pathophysiology of idiopathic Chiari malformation (CM) type 1 is disturbance of free cerebrospinal fluid (CSF) flow and restoration of normal CSF flow is the mainstay of treatment. Additional migration of the medulla oblongata in pediatric patients is referred to as CM type 1.5, but its significance in adult patients is unknown. This study is to compare surgical outcomes of adult idiopathic CM type 1.5 with that of type 1. Methods : Thirty-eight consecutive adult patients (M : F=11 : 27; median, 33.5; range, 18-63) with syringomyelia due to idiopathic CM type 1 were reviewed. Migration of the medulla oblongata was noted in 13 patients. The modified McCormick scale (MMS) was used to evaluate functional status before and one year after surgery. All patients underwent foramen magnum decompression and duroplasty. Factors related to radiological success (${\geq}50%$ decrease in the diameter of the syrinx) were investigated. The follow-up period was $72.7{\pm}55.6$ months. Results : Preoperative functional status were MMS I in 11 patients and MMS II in 14 of CM type 1 and MMS I in 8 and II in 5 of CM type 1.5. Of patients with MMS II, 5/14 patients in group A and 3/5 patients in group B showed improvement and there was no case of deterioration. Radiological success was achieved in 32 (84%) patients and restoration of the cisterna magna (p=0.01; OR, 46.5) was the only significant factor. Conclusion : Migration of the medulla oblongata did not make a difference in the surgical outcome when the cisterna magna was restored.

Ultrasonographic Evaluation of Renal Cortex and Outer Medulla Thickness in Dogs with Chronic Kidney Disease

  • This study was performed to retrospectively pursue any correlation between renal cortex thickness (RCT), outer medulla thickness (OMT) on ultrasonography (US) and chronic kidney disease (CKD) with International Renal Interest Society (IRIS) stage. Medical records and US findings of the dogs diagnosed CKD were reviewed for comparing to those of the clinically healthy dogs from March. 2015 through June. 2016. To evaluate the correlation about normal and CKD patients, RCT and the OMT were measured on US images. RCT and OMT were adjusted by dividing body surface area for standardization (RCTS, OMTS). Also the ratio of RCT/OMT were calculated and these indices were evaluated to investigate any tendency between 5 groups (normal and 4 IRIS stages) using Jonckheere-Terpstra trend test (J-T test). The RCTS showed a declining tendency (p < 0.02) and the OMTS showed an inclining tendency from normal through each IRIS stage (p < 0.01). The RCT/OMT showed also declining tendency (p < 0.01). Although the gold standard for renal function including GFR is lack, it is thought that differentiation between normal and CKD patients could be possible on US measurement of renal cortex and outer medulla thickness, which could be an alternative index for kidney function in diagnostic imaging.

Influence of SKF81297 on Catecholamine Release from the Perfused Rat Adrenal Medulla

  • The aim of the present study was to investigate the effects of 6-chloro-7,8-dihydroxy-1-phenyl-2,3,4,5-tetrahydro-1H-3-benzazepine(SKF81297), a selective agonist of dopaminergic $D_1$ receptor, on the secretion of catecholamines(CA) evoked by cholinergic stimulation and membrane-depolarization in the isolated perfused rat adrenal gland, and also to elucidate the mechanism involved. SKF81297($10{\sim}100{\mu}M$) perfused into an adrenal vein for 60 min produced dose- and time-dependent inhibition of CA secretory responses evoked by ACh(5.32 mM), high $K^+$(56 mM), DMPP($100{\mu}M$) and McN-A-343($100{\mu}M$). Also, in adrenal glands loaded with SKF81297($30{\mu}M$), the CA secretory responses evoked by Bay-K-8644($10{\mu}M$), an activator of L-type $Ca^{2+}$ channels and cyclopiazonic acid($10{\mu}M$), an inhibitor of cytoplasmic $Ca^{2+}$-ATPase were also inhibited. However, in the presence of the dopamine $D_1$ receptor antagonist, (R)-(+)-8-chloro-2,3,4,5-tetrahydro-3-methyl-5-phenyl-1H-benzazepine-7-ol(SCH23390, $3{\mu}M$), which is a selective antagonist of dopaminergic $D_1$ receptor, the inhibitory responses of SKF81297($30{\mu}M$) on the CA secretion evoked by ACh, high $K^+$, DMPP, McN-A-343, Bay-K-8644, and cyclopiazonic acid were significantly reduced. Collectively, these experimental results suggest that SKF81297 inhibits the CA secretion from the rat adrenal medulla evoked by cholinergic stimulation(both nicotininc and muscarinic receptors) and membrane depolarization. This inhibitory of SKF81297 seems to be mediated by stimulation of dopaminergic $D_1$ receptors located on the rat adrenomedullary chromaffin cells, which are relevant to extra- and intracellular calcium mobilization. Therefore, it is thought that the presence of the dopaminergic $D_1$ receptors may be involved in regulation of CA release in the rat adrenal medulla.

Characteristics of Central Noradrenergic Nervous System Response in SHR to Stimulation of Central ${\alpha}_2-Adrenoceptor$ (중추 ${\alpha}_2$-수용체 자극에 대한 선천성 고혈압쥐의 중추 노르아드레날린성 신경계 반응의 특성)

  • It has been postulated that abnormal characteristics of central noradrenergic nervous system has been implicated in the development and maintenance of hypertension in several modes of experimental hypertension including spontaneously hypertensive rats (SHR). In the present study, we attempt to determine if abnormal characteristics of central noradrenergic nervous system in SHR is caused by genetic factors or hypertensive phenomena by evaluating the changes of central adrenoceptors after long-term treatment of clonidine. Animals were divided into three groups; (1) 14 week-old SHR; (2) age-matched normotensive Wistar rats (NW); (3) DOCA-Salt induced hypertensive rats (DS). Clonidine (100 ug/kg) or vehicle was injected intraperitonealy twice a day for 15 days. Changes of ${\alpha}_1-$ and ${\alpha}_2-receptor$ desities following clonidine treatment were determiend in frontal corte, medulla oblongata and hypothalamus using 3H-WB4101 and 3H-clonidine, respectively. Densities of ${\alpha}_1$ and ${\alpha}_2-receptors$ following clonidine treatment were not changed in frontal cortex and medulla oblongate of SHR as well as DS, but increased in frontal cortex of NW and decreased in medulla oblongata of NW. On the other hand, densities of ${\alpha}_1-receptors$ were increased and densities of ${\alpha}_2-receptors$ were not changed in hypothalamus of SHR but densities of ${\alpha}_1-$ and ${\alpha}_2-receptors$ were decreased in hypothalamus of DS as well as NW. These results suggest that such differences in frontal cortex and medulla oblongata of SHR may be results of hypertensive phenomena whereas those in hypothalamus may be relevant to genetic factors of SHR.
