In the present study, the effect of cysteine and NT or bisphenol A(BP) on in vitro aturation(IVM) of porcine oocytes were examined. COCs was cultured in NCSU-23 medium supplement with 10% FCS which had previously been covered with mineral oil and equilibrated in a humidified atmosphere of 5% $CO_2$ and 95% air at $38^{\circ}C$. The IVM rate of oocytes cultured for 48 hrs in NCSU-23 medium supplement with 0.5~10.0 mM cysteine were $34.0{\pm}3.2%$, $36.0{\pm}3.5%$, $48.0{\pm}3.8%$, $22.0{\pm}3.2%$, respectively. The IVM rate of oocytes cultured in NCSU-23 medium supplement with 0.5~5.0mM NT for 48 hrs were $24.0{\pm}4.2%$, $18.0{\pm}4.9%$, $8.0{\pm}2.2%$, respectively. NT affects oocyte in vitro maturation rate in a dose-dependent. This result were significantly lower than the control group. The IVM rate of oocytes cultured for 48 hrs in NCSU-23 medium supplement with 1.0 mM NT+5.0 mM cysteine($38.0{\pm}4.3%$) were significantly higher than that of NT treatment. The IVM rate of oocytes cultured in NCSU-23 medium supplement with 0.05~5.0 mM BP for 48 hrs were $20.0{\pm}4.7%$, $10.0{\pm}5.3%$, $6.0{\pm}3.2%$, respectively. The IVM rate of oocytes cultured in NCSU-23 medium supplement with BP was significantly lower cultured non supplement of BP ($44.0{\pm}3.5%$). BP affects porcine oocyte maturation rate in a dose-dependent manner. The IVM rate of oocytes cultured for 48 hrs in NCSU-23 medium supplement with 1.0 mM BP+5.0 mM cycteine ($32.0{\pm}3.2%$) were increased than that of BP treatment.
This study derived measures to reduce exposure doses by identifying factors which affect the external radiation dose rate of patients treated with radiopharmaceuticals for PET-CT tests. The external radiation dose rates were measured on three parts of head, thorax and abdomen at a distance of 50cm from the surface of 60 PET-CT patients. It showed there are changes in factors affecting the external radiation dose rate over time after the administration of F-18 FDG. The external radiation dose rate was lower in the patients with more water intake than those with less water intake before the injection of radiopharmaceuticals at all three points: right after the injection of radiopharmaceuticals (average 4.17 mins), after the pre-PEET-CT urination step (average 77.47 mins), and right after the PET-CT test (average 114.15 mins). The study also found there is a need to increase the amount of water intake before the injection of radiopharmaceuticals in order to maintain a low external radiation dose rate in patients. This strategy is only possible under the assumption that the quality of the video has not changed after conducting this study on the relations between the image and quality. This study also found a need to use radiopharmaceuticals with the minimum amount needed for each patient because F-FDG doses affects the external radiation dose rate at the point right after the injection of radiopharmaceuticals. Urination frequency was the most significant factor to affect the external radiation dose rates at the point right after the PET-CT test and the point after the pre-PET-CT urination step. There is a need to realize the strategy to increase the urination frequency of patients to maintain the external radiation dose rate low (average 77.47 mins) before and after the injection of radiopharmaceuticals. In addition, at this point, there is a need to take advantage of personal strategies because the external radiation dose rate is lower if the fasting time is shorter, the contrast medium is used, and the amount of water intake is increased after the administration of radiopharmaceuticals. Finally this study found the need to be able to generalize these findings through an in-depth research on the factors affecting the external radiation dose rate, which includes radiopharmaceutical dose, urination frequency, the amount of water intake, fasting time and the use of contrast medium.
Objectives of this study were to investigate influencing factors of detention time, particle size distribution, and filter medium characteristics for waterworks sludge dewatering. The stepped pressure filtration was carried out with lab scale apparatus and the filter press pilot test for dewatering was conducted at the water treatment plant. Effects of filter medium and polymer dose were examined through observing water content and dewatering velocity and cyclic dewatering rate with filter press pilot test. Relationships among detention time, particle size distribution and filtration resistance were analyzed. Prolongation of sludge detention time was found to cause blinding phenomenon in cake and filter medium and to decrease dewatering process efficiency. The average specific resistance increased according to detention time. In pilot test of dewatering for thickened sludge with Nylon Multi-NY840D and Nylon Mono-100% filter media, dewatering velocities were 0.92 and $0.93kg\;DS/m^2{\cdot}hr$ according to 0.1% polymer dose of dried solids weight base. And cyclic dewatering rates were 2.45 and $2.50kg\;DS/m^2{\cdot}cycle$ cycle for the Nylon Multi-NY840D and Nylon Mono-100% media. Dewatering velocity of polymer dosed sludge was observed to be higher than that of non-polymer sludge.
Late effects of chronic exposure to gamma radiation are potential hazards to worker in radiation facilities as well as to the general public. Recently, chronic gamma radiation exposure effects have become a serious concern. Using a total of 60 mice, we studied the biological effects of medium-dose chronic exposure to gamma radiation. Sixty female 6-week-old specific pathogen free Balb/c mice were randomly divided into six groups (five groups irradiated and one non-irradiated control group). Irradiation was carried out for 7 days using gamma rays at dose rates of 119.65, 238.10, 357.14, 476.19 and $595.24mGy\;h^{-1}$ with total doses 20, 40, 60, 80 and 100 Gy. After irradiation, we determined survival rate of gamma radiation exposed mice during 1 week and 476.19 and $595.24mGy\;h^{-1}$ exposed group mice showed less 10% of survival rate. Otherwise, 119.65, 238.10 and $357.14mGy\;h^{-1}$ exposed group mice were survived each 100%, 80% and 70%. Half of survived mice after 1 week are immediately sacrifice and counted body and spleen weights. Compared with control non-irradiated group, total body weights and spleen weights isolated from 119.65, 238.10 and 357.14 irradiated group mice showed significant decreased. However, no significant alteration was observed between 119.65, 238.10 and $357.14mGy\;h^{-1}$ irradiated group. Overall, our results show for the first time that medium-dose chronic gamma radiation has the potential to stimulation of biological effects.
코발트-60 근접조사선원을 대체할 Ir-l92 선원주변의 2차원적 선량분포를 얻기 위하여, 조사선량률과, 조직감쇠계수를 구하였다. 조직감쇠계수는 선원에서 20 cm 지점까지 실험식을 구하였다. ?보기 방사능은 조사선량상수를 사용하여 결정하였으며, 2.5mm 직경에 두께 2.5 mm 의 선원은 조직선량을 정하기 위해 선원을 4401 개로 분할하여 선원 자체의 흡수효과와 ?슐벽의 차폐 효과와 조직감쇠계수를 적용하였다. 조직감쇠계수는 4차식을 사용하여 1% 오차범위내에서 실험값을 얻을 수 있었으며, Meisberger 상수는 선원에서 많이 떨어질수록 오차가 커서, 10 cm 위치서 7%, 20 cm 에서 33%의 오차를 발견할 수 있었으나, 겉보기방사능과 선원모양 및 크기가 달라 다른 결과를 가져올 수 있다고 본다. 발표된 Ir-192 선원의 에너지스펙트럼을 이용한 선량률상수는 절삭에너지 10 keV인 경우 4.69 R$cm^2$/mCi-hr을 얻었으며 Air Kerma는 0.973 을 구하였다. 이 실험에서 고안 선원의 분할선원에 의한 선원자체흡수와 ?슐벽에 의한 감쇠는 실선원의 54.6%가 겉보기방사능으로 나타남을 알 수 있었으며, 선량계획에 이용하기 위해 단위 ?보기 방사능에 대한 2차원적 선량표를 준비함으로써 기하학적선량과 선량 비등방성을 쉽게 이용할 수 있도록 하였다.
This study was performed to evaluate the subacute toxicity of CJ-50002 (Vibrio Vaccine) in SPF Spraqur-Dawley (SD) rats. Vibrio vaccine was administered orally at a dose level of high (167mg/kg/day), medium (16.7mg/kg/day), and low (16.7mg/kg/day) once a day and repeated fro 4 weeks. Ten males and female rats were assigned to each group. After 4 week administration, no significant dose-dependent changes in body weight, water and food consumption rate or organ weight were noted dependent changes in body weight, water and food consumption rate or organ weight were noted among 4 groups. Urinanalysis, hematology, and serum chemistry, also fail to detect any dose-related change among 4 groups tested. During necropsy and histopathological examination, no specific toxicity related to treated material was found. The result of this study demonstrated that vibrio vaccine when administered orally for 4 weeks at a high dose of 167mg/kg/day, no dose-related toxicity was found in treated make and female rats.
The effects of water extracts and powder of Korean ginseng on the division of Saccharomyces cerevisiae were studied. 1. The addition of several doses of water extracts and powder of ginseng to the yeast medium of Moyer and Coghill showed various promoted division of Saccharomyces. 2. The optimal dose of ginseng on tile division of Saccharomyces (0.08% dry ginseng medium solution per $10\;cells/mm^3$) could be recognized. 3. On the culture for 24 hours at $18^{\circ}C$, the cell number of control group was $13.25{\times}10^3\;cells/mm^3$ and that of the optimal dose group of water extracts of ginseng was $23.20{\times}10^3\;cells/mm^3$. On the culture, for 24 hours at $25^{\circ}C$, the cell number of control group was $16.85{\times}10^3\;cells/mm^3$ and that of the optimal dose group was $30.20{\times}10^3\;cells/mm^3$. The increasing rate of cell divison by the ginseng was about twice than that of control group. The optimal dose treatment of ginseng at $18^{\circ}C$ was more effective than control group at $25^{\circ}C$. 4. On the culture for 24 hours at $18^{\circ}C$, the increasing rate of water extracts of ginseng was 75.1%, and the rate of ginseng powder was 7.6%. On the culture for 24 hours at $25^{\circ}C$, the rate of water extracts of ginseng was 79.8%, and the rate of ginseng powder was 57.2%. Therefore water extracts of ginseng was more effective than ginseng powder of same dry weight, and the promoted effect of ginseng powder at $25^{\circ}C$ was more effective than at $18^{\circ}C$.
Using conidia of Neurospora, changes in respiratory activities and the sensitivity to the ultraviolet light of the cells at different growing stages were measured by manometric methods, and the correlation between them was observed. Efficiency in the utilization of various carbon sources, such as, glucose, sucrose, maltose, starch and sodium acetate, in growth and exogenous respiration of N. crassa was also determined. Growth rate of N. crassa was decreased considerably in the medium containing sodium acetate than in the glucose medium and was almost zero in the lactose medium, whereas the utilization of sucrose, maltose and starch was ve교 high, as that of glucose. Respiratory activities of the cells veried considerably depending upon their different growing stages. Actively growing hyphae exhibited the greatest activity in exogenous glucose respiration, followed by germinating and activated conidia in decreasing order. There was no proportional relationship between the dose of ultraviolet light irradiated and its effect on the respiratory activity of the cells, though the more the dose of ultraviolet light, the more the injury. The sensitivity of the cells to ultraviolet light varied with the different respiratory activities of the cells linked to the developmental stages. In general, the more actively growing cells having high respiratory activities exhibited the more serious injury.
The effect of loading dose, plasticiser and PVA molecular weight on naproxen release from hydrophilic polymer matrix was examined. Hydrophilic polymer matrix was prepared with PVA and PVP by adding glycerine as plasticiser. The release of naproxen from polymer matrix was determined in phosphate buffer medium. The release rate of naproxen from the polymer matrix increased as drug loading dose and plasticiser percentage increased. Raproxen released from the polymer matrix showed the time square root kinetics. Without changing the release-pattern, the release rate of naproxen could not be changed by varying molecular weight of PVA. Linearly released time range increased as drug loading dose increased, whereas decreased as plasticiser percentage increased up to 30%.
The purpose of this study was performed to investigate the mineralization and differentiation of osteobalsts for bone regeneration in vitro and the effect of rate of the composition in periodontal cells on mineralization. For this study, healthy gingival tissues were surgically obtained from the patients during 1st premolar extraction for the purposes of orthodontic treament. Gingival tissue was washed several time with Phosphate buffered saline contained high concentration of antibiotics and antifungal agent, and cultured in Dulbecco's Modified Eagle's Medium(DMEM, Gibco, U.S.A.). Every cell were cultured in state at $37^{\circ}C$, 100% of humidity, 5% of $CO_2$ incubator. Bone marrow stromal cells were isolated from 5-clay-old rat femur with using medium irrigation mathod by syringe. Cell suspension medium were centrifuged at 1500 rpm for 5 min and then cultured in the petri dish. Two kinds of cell were freezed and stocked in the liquid nitrogen tank until experiment. Cell were incubated into the 24 multi-well plate with $5{\times}10^4$cell/well of medium at $37^{\circ}C$, 100% of humidity 5% $CO_2$ incubator for 24 hours. After discarded of the supernatent of medium, O.5ml of medium were reapplied and incubated. And counted the number of cell using the hemocytometer and inverted light microscope. We have measured the number of mineralized nodule with using Alizarin red S. staining in microscope. Furthermore every cell were observed the morphological change between every rate of co-culture of the two kinds of cell. The results were as follows; The rate of proliferation of co-culture cell revealed high rate tendency compared the bone marrow stromal cell only and low growth rate to compared with gingival fibroblast only. The tendency of formation of the mineralized nodule were observed dose-depend pattern of bone marrow stromal cell. It is concluded that the gingival fibroblast may inhibit the formation of mineralized nodule in the culture of the bone marrow stromal cell.
