액아배양에 의한 희귀 수종 미선나무의 기내번식 (Micropropagation of a Rare Species, Abeliophyllum distichum Nakai. via axillary bud culture)

  • 미선나무의 기내번식을 위하여 MS배지에 세 가지 싸이토키닌 (zeatin, kinetin 및 BA)을 세 가지 농도로 처리하여 액아 배양을 실시하였다. 줄기유도는 BA가 가장 효과적이었고, 생장은 zeatin이 좋았다. Kinetin은 2.0 및 5.0 mg/L 처리에서 줄기발생 효과가 있었으나 그 효과는 zeatin, BA에 비해 저조했다. 500 lux정도의 약광 하에서 배양은 싸이토키닌 처리에 관계없이 줄기의 생장을 촉진하였다. 발근은 IBA가 첨가된 1/2 GD배지에서 주효했다. 발근된 개체는 인공배양토에서 100% 활착되었고, 정상 생장이 가능했다. 이상의 결과에서 미선나무의 기내 번식이 가능함을 보여 주었다.

Research on the relationship between the thermal characteristics and the type of land cover in Beijing urban area by ASTER data

  • The study utilizes remote sensing as the main monitoring means. With different spatial high-resolution, multichannel ASTER remote sensing image as the main information in Beijing city zone; with regional border and statistical data as auxiliary factor a study between the thermal space distribution character and the underground medium is analyzed based on the GIS logical algorithm and synthetic analysis technology. Results show thermal forming mechanism and the rule of distribution is mainly related to the underground medium and the change of the city distribution. Different underground medium has different degree and intensity influence on the thermal space distribution. Furthermore, urban greenbelt and water areas can reduce the thermal effect and large-scale greenbelt creates green island effect. In addition, Road net, residential area, population density, heat resources and so on have some positive effect on the thermal distribution, which increase the local temperature and intensity on the other hand. It is important to study the thermal distribution and its related factors, which contributes to the plan, construction and development of the city.

Effects of astaxanthin supplementation in fertilization medium and/or culture medium on the fertilization and development of mouse oocytes

  • Objective: We investigated the effect of supplementing fertilization medium and/or culture medium with astaxanthin (AST) on the two phases of in vitro fertilization: gamete fertilization and embryo development. Methods: Mouse cumulus-oocyte complexes were divided into four groups with 5 µM AST added to the fertilization medium (group 3, n=300), culture medium (group 2, n=300), or both media (group 4, n=290). No AST was added to the control group (group 1, n=300). Results: The fertilization rate was significantly higher (p<0.001) in the groups using AST supplemented fertilization medium (group 3, 79.0%; group 4, 81.4%) than those without AST (group 1, 56.3%; group 2, 52.3%). The blastocyst rate calculated from the two-cell stage was significantly lower (p<0.001) in the groups using AST-supplemented embryo culture medium (group 2, 58.0%; group 4, 62.3%) than in those without AST (group 1, 82.8%; group 3, 79.8%). The blastocyst rate calculated from the number of inseminated oocytes was highest in group 3 (189/300, 63.0%) and lowest in group 2 (91/300, 30.3%) with statistical significance compared to other groups (p<0.001). There were significantly higher numbers of cells in the inner cell mass and trophectoderm, as well as significantly higher total blastocyst cell counts, in group 3 than in the control group. Conclusion: An increased blastocyst formation rate and high-quality blastocysts were found only in the fertilization medium that had been supplemented with AST. In contrast, AST supplementation of the embryo culture medium was found to impair embryo development.

Effect of supplement of SCM in culture medium for in vitro development of bovine in vitro fertilized oocytes

  • Background: The successful production of superior or transgenic offspring from in vitro produced embryos in cattle relies heavily on the quality of blastocyst stage embryos. In order to enhance the developmental competency of these embryos, a novel culture method was devised. Methods: This study utilized stem cell culture medium (SCM) from hESCs as a supplement within the culture medium for bovine in vitro produced embryos. To gauge the efficacy of this approach, in vitro fertilized embryos were subjected to culture in CR1aa medium enriched with one of three supplements: 0.3% BSA, 10% FBS, or 10% SCM. Results: The blastocyst development and hatching rates of one-cell zygotes cultured in CR1aa medium supplemented with SCM (23.9% and 10.2%) surpassed those cultured in CR1aa medium supplemented with BSA (9.3% and 0.0%) or FBS (3.1% and 0.0%) (p < 0.05). Furthermore, post-zygotic gene activation, cleaved embryos cultured in CR1aa medium supplemented with SCM (57.8% and 34.5%) exhibited notably higher rates (p < 0.05) compared to those cultured with BSA (12.9% and 0.0%) or FBS (45.7% and 22.5%) supplementation. Furthermore, the microinjection of SCM into the cytoplasm or pronucleus of fertilized zygotes resulted in elevated blastocyst development and hatching rates, particularly when the microinjected embryos were subsequently cultured in CR1aa medium supplemented with SCM from the 8-cell embryo stage onwards (p < 0.05), in contrast to those cultured with FBS supplementation. Conclusions: In conclusion, this study conclusively demonstrated that the incorporation of SCM into the culture medium significantly enhances the developmental progress of preimplantation embryos.

Vitrification법에 의한 돼지 난자의 동결-응해 후 생존능력에 있어서 동해보호제와 Superoxide Dismutase의 영향 (Effect of Superoxide Dismutase and Cryoprotectants on Viability of Frozen-thawed Porcine Oocytes by Vitrification Method)

  • 본 연구에서는 vitrification방법을 사용하여 돼지 미수정란의 동결-융해시 난자생존능력에 대한특정 동해방지제 사용과 superoxide dismutase(SOD) 첨가의 영향을 검토하고자 수행하였다. 그 결과 미성숙 난자를 ethylene glycol과 DMSO 노출 후 성숙율(M-I에서 19.9%)이 glycerol과 DMSO 노출 후 성숙율(M-I에서 6.5%)보다 더 높았으며 ethylene glycol에 노출 후에는 M-I기로 성숙발달한 난자는 관찰되지 않았다. 한편. 미성숙 난자의 동결-융해 후 성숙배양시 SOD l unit/$m\ell$의 첨가시 (3.0%) 무첨가시(3.0%)와 성숙율이 같았으나 M-I에서 M-II 단계까지의 성숙율은 l unit/$m\ell$ 첨가 (65.0%)가 무첨가(54.0%)보다 높은 경향을 보였다. 또한, 성숙난자의 동결-융해 후 수정 시 정자 침입율은 lunit/$m\ell$ SOD 첨가(6.0%)의 경우 무첨가시 (3.7%)보다 더 높게 나타났다. 본 연구의 결과로부터 vitrifiration 방법을 이용 한 돼지 미수정란의 동결- 융해시 ethylenc glycol과 DMSO를 동해방지제로 사용하는 것이 효과적이며, 지질산화를 방지하기 위해 배양액에 l unit/$m\ell$의 SOD를 첨가하는 것이 동결-융해된 미수정란의 생존능력을 향상시키는 것으로 생각된다.

  • 본 연구는 중소기업의 기술 사업화 역량에 영향을 미치는 요인들에 대한 연구의 흐름을 다양화하기 위해 중소벤처 기업의 제조 역량과 시험 및 검사 역량이 외부 정보 네트워크의 다양성과 기술 사업화 역량 사이의 관계에 미치는 매개 효과를 실증 분석하였다. IBM SPSS 23을 사용하여 한국의 683개 중소벤처기업에 대해 실시한 최소자승 회귀분석, Sobel 검정과 Baron 및 Kenny 검정 결과를 통해 본 연구는 중소기업의 제조 역량과 시험 및 검사 역량의 역할에 대한 후속 연구에 유용한 세 가지 실증 분석 결과를 제공한다. 첫째, 중소벤처기업의 제조 역량은 중소벤처기업의 외부 정보 네트워크의 다양성이 기술 사업화 역량에 미치는 양(+)의 효과를 부분 매개하는 것으로 분석되었다. 둘째, 중소벤처기업의 시험 및 검사 역량은 이러한 양(+)의 효과를 부분 매개하는 것으로 밝혀졌다. 셋째, 중소기업의 제조 역량과 시험 및 검사 역량은 이러한 양(+)의 효과를 공동으로 부분 매개하는 것으로 분석되었다.

공배양의 작용기전에 관한 연구 (Mechanism for the Action of Co-culture)

  • Objective: A number of studies to improve in vitro culture conditions have been tried over past ten years by using co-culture system with helper somatic cells. However, the mechanism of coculture is poorly understood. This study was designed to understand the mechanism for the mode of actual action of co-culture using co-culture system of ICR strain's 1-cell embryos with human oviduct epithelial cells by examining the effect of conditioned medium and contactless coculture using a cell culture insert on the embryo development and by measuring the level of superoxide anion from conditioned medium after co-culture. Methods: ICR strain's zygote embryos were cultured in medium alone (control), coculture, conditioned medium, or contactless coculture system for 6 days. Conditioned media (CM) were prepared as following 5 groups. All CM were collected after culturing oviduct cells for 2 days. CM-1 was stored at $-20^{\circ}C$ until use, and CM-2 was prepared just before use as a culture medium. CM-3 was cocultured with embryos and retrieved just before use. CM-4 and CM-5 were derives from the microfilteration of CM-2 and CM-3, respectively, using Microcon-10 (10 kDa molecular weight cut-off). The percentage of the embryos developed to hatched blastocyst stage and the level of superoxide anion in supernatant from medium alone culture (control), coculture, and contactless coculture were measured. Results: The rates of embryo development to the hatched blastocyst stage were significantly higher in coculture (43%) than in control (0%) (p<0.05). The CM-1 group had no embryo development since 2-cell embryonic stage, whereas the CM-2, CM-3, CM-4 and CM-5 groups had the improved development to 4 or 8 cell embryo stage, but the similar rate of development to hatched blastocyst compared to control. The effect of coculture on embryo development was disappeared in the contactless coculture group. The level of superoxide anion was significantly reduced in coculture group compared to control. Conclusion: It is concluded that the present coculture system overcomes the 2-cell block in vitro and improves the embryo development. This beneficial effect may be due to the direct cell-cell contact between embryo and helper cells or the removal of deleterious components from medium rather than the embryotrophic factors.

배지 성분이 Streptomyces toxytricini에서의 lipstatin 발효에 미치는 영향 (Effect of Medium Components on the Lipstatin Production by Streptomyces toxytricini)

  • Streptomyces toxytricini로부터 lipstatin생산을 최적화하기 위해, 배지 성분이 lipstatin생산에 미치는 영향을 조사하였다. 이를 위해 tryptic soy broth (TSB) 배지를 기본 배지로 하여 $28^{\circ}C$, 200 rpm 으로 3일간 배양하여 총 배양액(seed culture) 을 만들고, 여기에 다양한 탄소원, 질소원, 지질 및 지방산등을 함유한 TSB 배지에 2% 접종한 후 60시간 동안 주 발효를 실시한 후, 배지 중의 lipstatin양을 측정하였다. 탄소원 중에서 glucose와 glycerol을 첨가한 배지에서 균체가 가장 잘 성장하였지만, lipstatin생산에는 lactose나 sucrose가 가장 우수한 것으로 나타났다. 한편, 질소원으로는 yeast extract가 균체 성장에 가장 좋았지만, 1.7% casitone과 0.3% soytone으로 구성된 TSB 배지에서 lipstatin생산량이 가장 높게 나타났다. 또한 lipstatin생산을 증가시키기 위해 triolein을 발효 배지에 첨가한 결과, 균체 성장은 증가하였지만, lipstatin의 생산은 현저히 감소하는 경향을 보였다. 한편, lipstatin의 생합성 원료로 추정되는 지방산들을 발효 배지에 0.5% 첨가하여 발효를 실시한 결과, 불포화 지방산인 linoleic acid나 oleic acid를 첨가한 경우 S. toxytricini의 성장이 억제되었으나, 포화 지방산인 stearic acid를 첨가한 경우에는 균체성장 뿐만 아니라 lipstatin 생산량도 증가하였다.

It is actively researching into a small and medium enterprise as the mean to create new employment, new industry, as means to from integration of a all and medium enterprise,as source of high competitive power. The status quo of rapid changing into informationization have been realized at considerable level in Korea. Information society is defined as information technology is main key to determine individuals competitiveness, which can solve effectively the side effect result from industrialization. It cleary imply that information technology is the most promising and important industry in 21st century. Therefore, We should seek to foster independent a small and medium enterprise and develop them corresponding to new concept of a small and medium enterprise in 21st century. The main frame of policy should be new economic system, which can contribute establishment of a small and medium enterprise, management innovation. It also attribute a small and medium enterprise to reveal their creative. New economic paradigm in 21st will be expanded with organization, market, technology. So far, a small and medium enterprise has been acknowledge as economic weaker and the one should be protected. However, In 21st century a small and medium enterprise will be considered as active majority or a source of creative. Development of technology to produce a small quantity with variety product and acceleration of knowledge and informationization will result in comparative merits of a small and medium enterprise. Hereby, The role and relative importance of a small and medium enterprise in our economic will getting be larger and It will be developed as the main force to activate the economic.
