Study of the Microbial and Chemical Properties of Goat Milk Kefir Produced by Inoculation with Taiwanese Kefir Grains

  • One of the prerequisites for the successful implementation of industrial-scale goat kefir production is to understand the effects of different kefir grains and culture conditions on the microbial and chemical properties of the goat kefir. Thus, the objectives of the present study were to evaluate the characteristics of kefir grains in Taiwan on the microbial and chemical properties of goat milk kefir, as well as to understand the influence of culture conditions on production of medium chain-length triglycerides (MCT). Kefir grains were collected from households in northern Taiwan. Heat-treated goat milk was inoculated with 3-5% (V/W) kefir grains incubated at 15, 17.5, 20 or 22.5$^{\circ}C$ for 20 h, and the microflora count, ethanol content, and caproic (C6), caprylic (C8), and capric acid (C10) levels measured at 4 h intervals. Our results indicate that incubation with kefir grains results in 10$^6$-10$^7$ CFU/ml microflora count and 1.18 g/L of ethanol content at 20 h of fermentation. Incubation with 5% kefir grain at 20-22.5$^{\circ}C$ produces the highest MCT levels.

The Influence of Lipids on Exocrine Pancreatic Secretions in Pigs - Review -

  • The characteristics of the exocrine pancreatic secretions in pigs and its hormonal regulation as influenced by dietary lipids are reviewed. There is clear evidence that the secretion of lipolytic enzymes is positively correlated with the amount of fat consumed by the pig. For example, there was an increase in the specific lipase activity by 83% after the dietary fat content was increased from 5% to 25%. Moreover, it was shown that also the quality of fat has an influence on exocrine pancreatic secretions. Peroxidized canola oil stimulated total lipase secretion much more than non-peroxidized oil. The influence of fatty acid composition on exocrine pancreatic secretions is discussed equivocally. Some authors showed that saturated fats stimulated the exocrine pancreatic secretions more than unsaturated. Others showed that the chain length of fatty acids had a strong influence on pancreatic secretions as well. Due to the different surgical methods used for sampling of pancreatic juice and wide variety of fats and oils used in these studies, direct comparisons between studies are extremely difficult to make. Plasma levels of hormones such as cholecystokinin (CCK), neurotensin (NT) and peptide YY (PYY) are influenced by the nutrient composition of the diet. With increasing amounts of fat present in the small intestine, the release of these hormones was stimulated. There is evidence that CCK release is dependent on the chain length of the fatty acids. Medium chain triglycerides stimulated the CCK release more than long chain triglycerides. Neurotensin was released more by unsaturated than by saturated fatty acids; similar results were observed for the PYY release. However, results are contradictory and further investigations are warranted that focus on the underlying mechanisms involved in the regulatory response of the exocrine pancreas to lipids of different origin.

Staphylococcus haemolyticus Lipase; High-Level Expression in Escherichia coli and Activation of Nonionic Detergent

  • A high level of Staphylococcus haemolyticus L62 lipase was expressed in an Escherichia coli transformant. The expressed lipase activity in the cell-free extract was 70,800 U/l, which corresponded to 30% of the total cellular protein. Pre-mixing of the l62 lipase with some nonionic detergents enhanced its hydrolytic activity towards olive oil: Tween detergents activated the L62 lipase by 3 fold. Gel filtration chromatography of the Tween-80-L62 lipase mixture demonstrated a polymerized complex (∼180 kDa) formed exclusively between Tween-80 and the L62 lipase. The lipase enzyme in the complex showed a higher specific activity towards most triacylglycerols than the intact L62 lipase. The activity enhancement towards each substrate was quite different depending on the acyl chain length; the activity towards tributyrin, trilinolein, and trilinolenin was much more enhanced than the towards the medium and the long-chain saturated triglycerides.
