  • 우수한 자생식물자원인 산국의 수량과 유효성분을 높이기 위해 식물생장조절제 ABA와 SA를 처리한 결과를 요약하면 다음과 같다. 1. 꽃의 건물수량은 ABA, SA과 ABA+SA 처리구에서 무처리구보다 증가되었으며, ABA 처리구보다는 SA 처리구가 증수되었다. 2. 무기성분의 함량변화는 SA 처리구가 다른 처리구들에 비하여 질소, 인산, 마그네슘의 함량이 증가하였으나 칼륨의 흡수는 오히려 감소하였다. 3. 아미노산함량은 SA 처리구와 ABA+SA 처리구는 ABA 처리구보다 높았다. 4. 생장조절제처리는 정유함량을 증가시켰으며, 그 반응은 monoterpenoid계 화합물보다는 sesquiterpennoid계 화합물에서 더 민감하였다. 5. Cumambrin A의 함량은 ABA 처리구에서 증가하였고, SA 처리구에서는 차이가 없었다.

열처리에 따른 씀바귀 침출차의 품질특성 (Quality Characteristics of Tea Thermally Processed from Dried Ixeris dentata Root)

  • 씀바귀뿌리를 이용하여 침출차를 제조하고자 건조온도 및 처리조건을 달리하여 유리당, 유기산, cynaroside 및 총폴리페놀성물질 함량, 유리아미노산, 색도 및 관능적 특성을 검토하였다. 총유리당, 유기산, 유리아미노산 및 cynaroside 의 함량은 천일건조한 것이 가장 높았으며 총폴리페놀성 함량은 열풍건조 후 볶음 처리한 것이 가장 높았다. 건조한 씀바귀뿌리의 유리당 중 sucrose, glucose, fructose 및 galactose는 열풍건조에 의하여 감소하였고 천일건조 시 검출되지 않았던 maltose가 열풍건조에 정량되었으며 증숙 후 열풍건조 시 1.67%의 높은 함량을 보여주었다. 씀바귀뿌리 내에 함유되어있는 유리아미노산함량 중 arginine이 669.2 mg%으로 가장 높았고 뒤이어 serine이 379.9 mg%으로 높은 함량을 보여주었다. 씀바귀뿌리 건조분말의 색도는 열풍건조 후 볶음처리를 한 것이 밝기, 적색도 및 황색도 모두 가장 높았으나 열수침출 후의 색도는 적색도와 황색도는 타처리구에 비하여 가장 높았지만 밝기는 가장 낮게 나타났다. 씀바귀뿌리를 이용한 침출차의 관능검사결과 열풍건조 후 볶음처리한 처리구가 가장 높은 점수를 보였다.

The Effect of Extract from Sea Buckthorn on DNCB-induced Atopic Dermatitis in NC/Nga Mice

  • Sea Buckthorn (Hippophae rhamnoides L.) has been used in traditional medicine for the treatment of cough, indigestion, circulatory problems and pain. The associated anti-inflammatory effect of this agent is achieved via the inhibition of Nf-${\kappa}B$ signaling, a property that has been demonstrated to effectively control the symptoms of various skin disorders, including atopic dermatitis. Accordingly, the purpose of this study was to assess the efficacy of Sea Buckthorn in reducing the production of lipopolysaccharide (LPS) activated nitric oxide (NO) by inhibiting the Nf-${\kappa}B$ pathway, as measured by the symptoms of atopic dermatitis (AD) occurring secondarily to inflammation and immune dysregulation. Our data demonstrate that Sea Buckthorn significantly decreased the LPS-induced production of NO (p<0.001). Atopic dermatitis was induced by repeated application of 2,4-dinitrochlorobenzene to the dorsal skin of mice. Topical application of 5% Sea Buckthorn extract improved the symptoms of AD, specifically reducing disease severity scores, scratching behaviors and epidermal thickness. When compared to the control group, animals treated with Sea Buckthorn exhibited increased serum IL-12 levels and decreased serum TNF-${\alpha}$, IL-4 and IL-5 levels. Such a modulation of biphasic T-helper (Th)1/Th2 cytokines may result in a reduction in serum IgE levels. Our findings suggest that mechanism of action of Sea Buckthorn in the treatment of AD is associated with a marked anti-inflammatory effect as well as an inhibition of Th2-mediated IgE overproduction via the modulation of biphasic Th1/Th2 cytokines. Such results suggest that topical Sea Buckthorn extract may prove to be a novel therapy for AD symptoms with few side effects.

주요종자전염병 억제를 위한 항균성 약용식물탐색 (Screening of Medicinal Plants with Antifungal Activity on Major Seedborne Disease)

  • 50종의 약용식물 추출액을 공시하여 생체외(in vitro) 및 생체내 (in vivo)에서 몇 가지 주요 종자전염성병 억제를 위한 항균활성식물을 선발하고 선발된 식물에 대한 항균활성을 검정한 결과는 다음과 같다. 가) 공시된 50종의 약용식물추출액을 공시병원균(벼도열병균, 참깨검은무늬병균, 고추탄저병균, 무·배추검은무늬병균)을 접종한 종자에 대한 항균활성을 습지법과 물한천법으로 정정한 결과 마늘, 주목, 대황, 무슬, 감초, 달맞이꽃 등에서 항균활성이 우수한 것으로 나타났다. 나) 1차 선발된 식물추출액을 종자에 희석 처리하여 감자한천법으로 검정한 결과, 마늘 추출액이 10배에서도 항균활성을 보였고 종자침적 시간에 따른 효과를 보면 긴 시간 침적이 가장 높은 항균활성을 나타냈으며 종자침적온도에 따른 항균효과를 보면 $25^{\circ}C$ 처리가 대체적으로 우수했다. 다) 식물 추출액의 종자발아및 초기생육에 미치는 영향은 마늘 추출액의 경우 무처리에 비하여 종자발아에 차이가 없었고 초기생육은 오히려 양호하였다. 주목의 추출액은 무와 배추에서 약간의 영향이 있었고 우슬, 감초, 달맞이꽃 등에서는 종자 발아 및 초기 생육을 심하게 저해했다.

짝자래나무[Rhamnus yoshinoi] 가지 추출물에 의한 전립선암세포의 Wnt/β-catenin 분해 유도 활성 및 GC/MS 분석 (Extract from the branches of Rhamnus yoshinoi exerts anti-cancer effects on human prostate cancer cells through Wnt/β-catenin proteasomal degradation and identification of compounds by GC/MS)

  • 본 연구에서는 갈매나무과에 속하는 Rhamnus yoshinoi (RY, 짝자래나무) 가지 부위의 70% 에탄올 추출물을 이용하여 전립선암세포의 항암 활성을 규명하고자 하였다. 짝자래 나무 가지 추출물을 전립선암 세포 PC-3에 처리하여 β-catenin과 TCF4의 단백질 수준 감소를 확인하였다. 이후, β-catenin과 TCF4의 mRNA 발현을 조사한 결과 β-catenin은 감소하지 않았고, TCF4는 감소하였다. 이를 통해, β-catenin mRNA의 발현에는 영향이 없지만, TCF4 mRNA 발현은 억제하는 것으로 나타났다. 단백질 분해효소억제제인 MG132를 처리한 전립선암 세포 PC-3에서 단백질 수준 확인을 통해 β-catenin의 단백질 분해를 유도할 수 있음을 확인하였다. 또한 전립선암 세포 PC-3에서 짝자래나무 가지 추출물의 GSK-3β 유도 β-catenin 단백질 분해 Kinase 구명과 β-catenin 인산화에 영향을 미치는 것을 확인하였다. 이상의 연구 결과로 짝자래나무 가지 추출물은 GSK-3β 의존성 Wnt/β-catenin 단백질의 분해를 통해 전립선암의 생육 억제와 관련이 있는 것으로 확인된다. 또한 짝자래나무 가지 추출물에서 항암활성과 관련된 활성물질이 있는 것으로 확인되었다. 본 결과는 전립선암의 항암제 개발을 위한 소재로 짝자래나무 가지 추출물의 활용이 가능할 것으로 판단된다.

약사법규 중의 기존한약서 활용에 대한 고찰 (Study on the Application of Established Korean Medical Classics in the Korean Pharmaceutical Affairs Act)

  • Purpose: There is an issue in applying various principles introduced in established Korean medical classics to "Pharmaceuticals Approval, Notification and Review" of "herbal medicinal preparations" and "new drugs from natural products" that are used for western forms of medical treatment. Thus, an analysis of the origin, purpose, and application of established Korean medical classics in the Korean Pharmaceutical Affairs Act is essential. Methods: We collected data regarding the origin, purpose, and application of established Korean medical classics in the Korean Pharmaceutical Affairs Act, and classified them by periodical change and subjects. Results: Established Korean medical classics are applied as follows: 1) as criteria for Korean medicine distributors' sales of mixed herbal drugs (Pharmaceutical Affairs Act; since 1953), 2) as official compendiums for pharmacists' preparation of Korean medicine (Ministry of Health and Welfare's authoritative interpretation; from early 1970s to 1993), 3) as standards for oriental pharmacists' quality measurement of preparations (notification of the Ministry of Health and Welfare; since 1995), 4) as criteria for "Pharmaceuticals Approval, Notification and Review" of herbal medicinal preparations and crude drug preparations (notifications regarding drug approval process by the Ministry of Health and Welfare and the Ministry of Food and Drug Safety; since 1978), and 5) as standards for the quality of materials of health functional food (from 2004 to 2011). Conclusion: The application of Korean medical classics has been closely related with the change of the laws, regulations, and systems that are relevant to Korean medicine, and it seems to be more favorable for pharmacists than oriental pharmacists. Meanwhile, regulations that apply prescriptions that are recorded in Korean medical classics - dosage, indications, and preparation methods - as criteria for the approval of crude drug preparations for western medical treatment should be abolished.

  • Cinnamyl alcohol dehydrogenase (CAD, EC 1.1.1.95), catalyzes the reduction of hydroxycinnamaldehydes to give hydroxycinnamyl alcohols, or "monolignols," the monomeric precursors of lignin. Lignins are important components of cell walls and lignified secondary cell walls play crucial roles in long distance transport of water and nutrients during plant growth and development and in plant defense against biotic and abiotic stresses. Here a cDNA clone containing a CAD gene, named as PgCAD, was isolated from a commercial medicinal plant Panax ginseng. PgCAD is predicted to encode a precursor protein of 177 amino acid residues, and its sequence shares high homology with a number of other plant CADS. The expression of PgCAD in adventitious roots and hairy roots of P. ginseng was analyzed using reverse transcriptase (RT)-PCR under various abiotic stresses such as salt, salicylic acid, wounding and chilling treatment that triggered a significant induction of PgCAD at different time points within 2-48 h post-treatment. This study revealed that PgCAD may help the plants to survive against various abiotic stresses.

Effect of Nelumbo nucifera on Proliferation, Migration and Expression of MMP-2 and MMP-9 of rSMC, A431 and MDA-MB-231

  • Nelumbo nucifera Gaertn.(family Nymphaeaceae) has been used for summer heat syndrome as home remedy in Japan, China and Korea. Although whole plant parts are edible, root is commonly consumed. It has been reported that rhizome extract showed anti-diabetic and anti-inflammatory effects. However, in spite of usefulness for treatment of various diseases, the effect of Nelumbo nucifera rhizome(NNR) on proliferation, migration and matrix degrading enzymes-matrix metalloproteinsases(MMPs), the expression of which degrades extracellular matrix(ECM) leading to metastasis, has not been fully elucidated. We examined the effect of hot water extract of NNR on the proliferation, migration and secretion of MMP-2 and MMP-9 in rat smooth muscle cells(rSMC), epidermoid cancer cells(A431) and breast cancer cells(MDA-MB-231). The proliferation assay was carried out using MTT assay, the principle of which depends upon the conversion of MTT by mitochondrial dehydrogensases of viable cells to formazan crystals. The effect of NNR on migration of cells was examined using wound healing assay. Our results showed that there was inhibition in the proliferation, migration and expression of MMP-2 and MMP-9 in dose dependent fashion in all the cells used. Thus, we concluded that NNR could be used as traditional medicine in the treatment of various diseases where proliferation, migration and MMPs' expression plays a pathological role like in restenosis and metastasis.
