Metabolomics Approach for Classification of Medicinal Plants

  • Selection of specific medicinal sources as well as bioactive compounds is important for the preparation of medicine and related products with good quality. It is necessary to pay close attention for choosing correct medicinal sources, particularly in case of medicinal plants, because of their diversity, which can affect the quality and efficacy of medicine. Discrimination of plants based on morphological or genetic characteristics has been used as a conventional classification method of pharmaceutical sources so far; however, more need demands more general methods for accurate quality assessment of medicinal plants. In this study, ultra performance liquid chromatography/quadrupole time-of-flight mass spectrometry (UPLC/Q-TOF MS) technique applied to this metabolic profiling is a powerful tool due to its higher sensitivity, resolution, and speed compared to conventional HPLC technique. The metabolite profiling of several medicinal plants including Panax ginseng was carried out using UPLC/Q-TOF MS and total metabolites were then subsequently applied to various statistical tools to compare the patterns. The developed metabolomics tool with UPLC/Q-TOF MS successfully identified and classified the samples tested according to their origins.

Genetic diversity of the threatened Saussurea dorogostaiskii (Asteraceae) in the Khuvsgul region of Mongolia

  • Saussurea dorogostaiskii Palib. (Asteraceae) is a critically endangered medicinal plant in Mongolia and Russia. We studied the genetic variation of S. dorogostaiskii from three mountains of northern Mongolia. The genetic profile was assessed in 70 individuals from eight populations using five inter-simple sequence repeat markers, producing 53 loci with 96.4% polymorphism across all bands. Shannon's index (I) and Nei's gene diversity (H) value at the species level of S. dorogostaiskii are 0.25 and 0.17, respectively. An AMOVA showed high genetic variation among the populations (22% of populations and 32% of mountains), consistent with the high genetic differentiation (GST = 0.49) and low gene flow (Nm = 0.51) in S. dorogostaiskii populations. Eight populations were clustered into two groups, corresponding to their geographic locations. The low within-population genetic diversity and high genetic differentiation among S. dorogostaiskii populations factor into their endangered designation. This genetic analysis reveals that all populations are equally threatened, and community-based conservation is appropriate for these species.

Medicinal plant diversity in the southern and eastern Gobi Desert region, Mongolia

  • Background: The southern and eastern parts of the Gobi Desert area are a unique dry ecosystem with a diverse regional desert, semi-desert, and mountain dry steppe flora. This area habitat is located at the overlap of different floristic regions; on its northeast side, Central Asian desert flora is dominating, and on the eastern side, East Asian flora is observed. The comprehensive survey was carried out to find the floral diversity of the medicinal plants on the region. Methods: All recorded species in this study were based on the collected voucher specimens between June and August in the year 2017. Results: We recorded 23 families, 57 genera, and 78 species of vascular plants. The families Asteraceae (15 species), Fabaceae (10 species), and Amaranthaceae (10 species) were represented most in the study area, while Caragana (5 species), Salsola (4 species), and Arnebia (3 species) were the most common genera found. Conclusion: Conservation status for remarkable species was also reviewed based on the literature. Around the study area, 24 species as "sub-endemic," 10 species as "very rare," 4 species as "rare," 1 species as "alien," 13 species as "relict," 10 species as "Red Book," 2 species as "endangered (EN)," 3 species as "vulnerable (VU)," 3 species as "near threatened (NT)," and 2 species as "least concern (LC)" plants are growing.

태안반도 꽃지 약용해조의 계절적 군집구조 및 수직분포 (Seasonal Community Structure and Vertical Distribution of Medicinal Seaweeds at Kkotji in Taean Peninsula, Korea)

  • Marine algal flora and community structure of medicinal seaweeds were examined at Kkotji of Taean Peninsula, Korea from May 2005 to January 2006. Seventy-nine seaweeds including 42 medicinal algae and one marine plant were identified. Sargassum thunbergii was the representative alga occurred at all seasons and shore levels. The dominant medicinal seaweeds were perennial S. thunbergii, Neorhodomela aculeata, and Corallina pilulifera, and ephemeral Monostroma grevillei, Porphyra yezoensis, and Ulva pertusa. Their vertical distribution were N. aculeata – P. yezoensis, M. grevillei, and U. pertusa – C. pilulifera from high to low intertidal zone. The average biomass of medicinal seaweeds varied from 34.17 g m–2 in spring to 56.41 g m–2 in summer. At Kkotji shore, the opportunistic species (Enteromorpha, Ulva, and Cladophora) and turf-forming algae (Caulacanthus okamurae and Gelidium divaricatum) were easily observed. Such fast growing ESG II (ecological state group) was 87.50% and slow growing perennial algae, ESG I was only 12.15%. Also, diversity index (H’) and dominance index (DI) indicate that the seaweed community of Kkotji is unstable. Therefore, Kkotji rocky shore should be more protected from human activities such as turbulence and eutrophication in order to maintain species diversity and abundance of medicinal seaweeds.

Analysis of common and characteristic actions of Panax ginseng and Panax notoginseng in wound healing based on network pharmacology and meta-analysis

  • In recent years, an increasing number of reports have explored the wound healing mechanism of these two traditional Chinese herbal medicines- Panax ginseng and Panax notoginseng, but there is no systematic research on the related core functions and different mechanisms in the treatment of wound healing up to now. Based on network pharmacology and meta-analysis, the present work aimed to comprehensively review the commonality and diversity of P. ginseng and P. notoginseng in wound healing. In this study, a wound healing-related "ingredients-targets" network of two herbs was constructed. Thereafter, meta-analysis of the multiple target lists by Metascape showed that these two medicines significantly regulated blood vessel development, responses to cytokines and growth factors and oxygen levels, cell death, cell proliferation and differentiation, and cell adhesion. To better understand the discrepancy between these two herbs, it was found that common signaling pathways including Rap1, PI3K/AKT, MAPK, HIF-1 and Focal adhesion regulated the functions listed above. In parallel, the different pathways including renin-angiotensin system, RNA transport and circadian rhythm, autophagy, and the different metabolic pathways may also explained the discrepancies in the regulation of the above-mentioned functions, consistent with the Traditional Chinese Medicine theory about the effects of P. ginseng and P. notoginseng.

차세대염기서열분석법을 이용한 잔대의 SSR 마커 개발 (Development of Simple Sequence Repeat Markers from Adenophora triphylla var. japonica (Regel) H. Hara using Next Generation Sequencing)

  • Background: Adenophora triphylla var. japonica (Regel) H. Hara shows vegetative growth with radical leaves during the first year and shows reproductive growth with cauline leaves and bolting during the second year. In addition, the shape of the plant varies within the same species. For this reason, there are limitations to classifying the species by visual examination. However, there is not sufficient genetic information or molecular tools to analyze the genetic diversity of the plant. Methods and Results: Approximately 34.59 Gbp of raw data containing 342,487,502 reads was obtained from next generation sequencing (NGS) and these reads were assembled into 357,211 scaffolds. A total of 84,106 simple sequence repeat (SSR) regions were identified and 14,133 primer sets were designed. From the designed primer sets, 95 were randomly selected and were applied to the genomic DNA which was extracted from five plants and pooled. Thirty-nine primer sets showing more than two bands were finally selected as SSR markers, and were used for the genetic relationship analysis. Conclusions: The 39 novel SSR markers developed in this study could be used for the genetic diversity analysis, variety identification, new variety development and molecular breeding of A. triphylla.

Genetic Diversity of Wild and Cultivated Populations of American Ginseng (Panax Quinquefolium) from Eastern North America Analyzed by RAPD Markers

  • The objective of this study was to assess genetic diversity among 6 different wild ginseng populations from New York, Kentucky, North Carolina, Pennsylvania, Tennessee and Virginia, and to compare these wild populations to one cultivated population. RAPD markers were used to estimate the genetic difference among samples from the 7 populations. The 64 random primers were screened, and 15 primers were selected which exhibited the 124 highly reproducible polymorphic markers. The ratio of discordant bands to total bands scored was used to estimate the genetic distance within and among populations. Multidimensional scaling (MDS) of the relation matrix showed distinctive separation between wild and cultivated populations. The MDS result was confirmed using pooled chi-square tests for fragment homogeneity. This study suggests that RAPD markers can be used as population-specific markers for American ginseng.

구지뽕 나무의 엽록체 TrnL-F 영역 염기서열 분석을 통한 특이적 SNP 분자마커의 확인 (Identification of specific SNP molecular marker from Cudrania tricuspidata using DNA sequences of chloroplast TrnL-F region)

  • 구지뽕 나무(Cudrania tricuspidata Bureau)는 일반적으로 이용되는 대표적인 다년생의 약용식물이다. 최근 국제적 추세에 따라 자국의 유전자원의 발굴, 보존 등이 강화됨에 따라 인접국가와 국내 자생 꾸지뽕 계통을 판별 할 수 있는 기준설정에 관한 연구의 필요성이 대두되고 있지만, 분자생물학적 판별 기술의 개발은 아직 미흡한 실정이다. 본 연구에서는 국내 토종과 중국 계통의 구지뽕 나무의 기원을 판별하기 위해 엽록체에 존재하는 trnL-trnF 유전자단편에서 SNP를 이용한 판별 프라이머를 확보하였으며 이를 보완하여 보다 신속하게 판별하기 위하여 ARMS-PCR기술을 이용한 판별마커와 그 조건을 확립하였다. 그러므로, 본 연구에서 개발된 SNP 마커는 다양한 지역에서 서식하는 구지뽕 계통들의 신속한 확인을 위해 매우 유용하게 이용될 것으로 생각된다.
