Effects of Medicinal Plant Extracts on Antioxidant System in Ethanol-intoxicated Rats

  • Four medicinal plants selected from preliminary screening study were evaluated in the aspects of their antioxidant activities in alcohol-intoxicated rats. Rats fed 1% ${\alpha}-tocopherol-supplemented$ diet as positive control and ones done ${\alpha}-tocopherol-deficient$ diet as negative control were compared with ones done the plant extract-supplemented diet (n=8). After the administration of the experimental diets for 4 weeks, typical increments in activities of manganese-superoxide dismutase (Mn-SOD) and glutathione peroxidase (GSH-px) indicated in alcohol-intoxicated rats, were not observed in ones fed Lagerstroemia and Ulmus extract-supplemented diet. The content of thiobarbituric acid reactive substance (TBARS), the product of lipid peroxidation, did not increased in rats fed plant extracts-supplemented diet except for Terminalia. From the results, it is concluded that Lagerstroemia and Ulmus have physiologically efficient antioxidant activities.

Anti-allergic Activity of Oriental Medicinal Herbs

  • The effects of extracts from various Oriental medicinal herbs on mast-cell-mediated allergic reactions were investigated in this study. Out of these extracts, the medicinal herb Atractylodis rhizoma alba exhibited the most potent activity in the cells, with $IC_50$ values of $20.5\;{\mu}g/ml$, by DNP-BSA and $33.04\;{\mu}g/ml$, by compound 48/80. Similar to the in vitro activity, Atractylodis rhizoma alba inhibited compound-48/80-induced systemic anaphylaxis by 31.3% at 300 mg/kg in mice. It also suppressed the secretion of $TNF-{\alpha}$, a major pro-inflammatory cytokine in the cells. These results may suggest that Atractylodis rhizoma alba shows anti-allergic activity in compound 48/80-induced anaphylactic mice through the inhibition of mast cells.

소청룡탕 한약 농축분말을 첨가한 초콜릿의 제조 및 산화방지 특성 (Antioxidant Characteristics and Preparation of Chocolate Added with Sochungryong-Tang (Oriental Medicinal Plants Extract))

  • The purpose of this study was to investigate the effects of oriental medicinal plant extracts added to chocolates on the moisture content, color, radical scavenging activity and sensory characteristics. The oriental medicinal plant extracts were added at a weight percentage of 0, 4 and 8%. As the ratio of the extracts in the chocolates increased, the moisture content, color, and radical scavenging activity increased. In sensory evaluation, significant differences (p<0.05) were shown in taste, bitterness, and overall acceptability depending on the addition of oriental medicinal plant extracts but there were no significant differences in chocolate aroma and texture.

Inhibitory Effects of Methanol Extracts from Korean Medicinal Plants against HIV-1 Protease Activity

  • Korean medicinal plants were screened for their inhibitory activity against HIV-1 protease. The inhibitory activity of protease was determined by incubating the extracts in reaction mixtures containing protease and substrate $His-Lys-Ala-Arg-Val-Leu-(p-NO_{2}-Phe)-Glu-Ala-Nle-Ser-NH_{2}$ to perform proteolytic cleavage reactions. In this study the twenty six extracts from medicinal plants were investigated. Of the extracts tested, the extracts from the stem of Morus alba. exhibited the strongest activity with inhibition of 81% at a concentration of $100{\mu}g/ml$. The extracts of the flower of Saxjfraga stolonifera, and stems of Euonymus japonica and Castanea crenata showed appreciable inhibitory activity (>50%) against HIV-1 protease at same concentration.

피부각질형성세포에서 동의보감 충부약재 14 종이 염증성 사이토카인 생성에 미치는 영향 (Effects of 14 Chung-bu Medicinal Materials Described in the Dongui Bogam on Inflammatory Cytokines Production in HaCaT Keratinocytes)

  • 본 연구에서는 피부각질형성세포에서 동의보감에 기록된 14 종의 충부약재에 대한 염증성 사이토카인 생성에 미치는 영향을 조사하였다. 이를 확인하기 위하여 멀티플렉스 사이토카인 분석으로 17 종에 대한 사이토카인을 스크리닝 하였다. 충부약재 14 종 중, 제조 및 전갈 추출물은 IL-5 생성을 억제하였고, 제조, 상표초 및 수질 추출물은 IL-6 생성을, 제조, 상표초, 지룡, 수질 및 문합 추출물은 IL-8 생성에 억제 효과를 보였다. 합개, 잠사, 선퇴 및 문합 추출물은 IL-13 생성을 억제하였으며, 귀판, 잠사 및 지룡 추출물은 MIP-1β 생성에 유의적인 효과를 확인하였다. 이러한 충부약재는 피부염증 억제를 위한 항염증제 활용 가능성에 의미를 둘 수 있지만, 그 가치를 증명하기 위해서는 피부개선을 위한 기전연구를 포함하여 인체피부모사판 모델에서 추가 실험을 통한 검증이 선행 되어야 한다.

한약재 첨가사료 급여가 비육돈의 생산형질과 도체품질에 미치는 영향 (Effects of feeding medicinal herbs on growth performance and carcass quality in finishing pigs)

  • This study was performed to evaluate the effects of medicinal herbs on characteristics and qualities of pork for 6 months using 102 post-weaning piglets, To improve the bod gain and feed efficiency rate, post-weaning piglets were fed with medicinal herbs from 4 to 10 weeks after birth, while grower pigs for 60 days before slaughter. Daily body gain of piglets of experimental group was higher(493.1g) than control(447.8g), with feed demand index 1.49 versus 1.6 times in control, indicating. The more pigs were fed with herbs, the higher body gain was observed. Among carcass characteristics, dressing percentage and backfat thickness were higher in experimental group(68.26% and 14.52mm) than control(67.39% and 14.36mm). Experimental group(0.5% medicinal herb-fed) produced more A class carcass. In fatty acid analysis, meats of experimental group contained less saturated(more unsaturated) fatty acids than control. Especially, palmitic acid, a precursor of cholesterol, was less and oleic acid was more contained than control. In amino acid analysis, essential amino acids(methionine and isoleucine) were increased in meats of experimental group. In concolusion, feeding medicinal herbs has effects such as increased body gain, improved feed demand, and carcass class on piglets and improved meat quality on growers.

Ginsenoside Rk1 ameliorates paracetamol-induced hepatotoxicity in mice through inhibition of inflammation, oxidative stress, nitrative stress and apoptosis

  • Background: Frequent overdose of paracetamol (APAP) has become the major cause of acute liver injury. The present study was designed to evaluate the potential protective effects of ginsenoside Rk1 on APAP-induced hepatotoxicity and investigate the underlying mechanisms for the first time. Methods: Mice were treated with Rk1 (10 mg/kg or 20 mg/kg) by oral gavage once per d for 7 d. On the 7th d, allmice treated with 250mg/kg APAP exhibited severeliverinjury after 24 h, and hepatotoxicitywas assessed. Results: Our results showed that pretreatment with Rk1 significantly decreased the levels of serum alanine aminotransferase, aspartate aminotransferase, tumor necrosis factor, and interleukin-$1{\beta}$ compared with the APAP group. Meanwhile, hepatic antioxidants, including superoxide dismutase and glutathione, were elevated compared with the APAP group. In contrast, a significant decrease in levels of the lipid peroxidation product malondialdehyde was observed in the ginsenoside Rk1-treated group compared with the APAP group. These effects were associated with a significant increase of cytochrome P450 E1 and 4-hydroxynonenal levels in liver tissues. Moreover, ginsenoside Rk1 supplementation suppressed activation of apoptotic pathways by increasing Bcl-2 and decreasing Bax protein expression levels, which was shown using western blotting analysis. Histopathological observation also revealed that ginsenoside Rk1 pretreatment significantly reversed APAP-induced necrosis and inflammatory infiltration in liver tissues. Biological indicators of nitrative stress, such as 3-nitrotyrosine, were also inhibited after pretreatment with Rk1 compared with the APAP group. Conclusion: The results clearly suggest that the underlying molecular mechanisms in the hepatoprotection of ginsenoside Rk1 in APAP-induced hepatotoxicity may be due to its antioxidation, antiapoptosis, anti-inflammation, and antinitrative effects.

Cytotoxic effects of ethanol extracts of Saussurea lappa mediated by mitochondrial apoptotic pathway

  • Saussurea lappa and Taraxacum mongolicum have been used for herbal medicinal treatments against cancers in East Asia. We performed this study to understand the molecular basis underlying the anti-tumor effects of two herbs and analyzed the effects of these medicinal herbs on proliferation and on expression of cell growth/apoptosis related molecules by using an AGS gastric cancer cell line. The treatments of Saussurea lappa dramatically reduced cell viabilities in a dose and time-dependent manner, but Taraxacum mongolicum did not. FACS analysis and Annexin V staining assay also showed that Saussurea lappa induces apoptotic cell death of AGS. Expression analyses via RT-PCR and Western blots revealed that Saussurea lappa increased expression of the p53 and its downstream effector p21/sup Waf1/, and that the both increased expression of apoptosis related Bax and cleavage of active caspase-3 protein. We also confirmed the translocation of Bax to mitochondria Collectively, our data demonstrate that Saussurea lappa induces growth inhibition and apoptosis of human gastric cancer cells, and these effects are correlated with down- and up-regulation of growth-regulating apoptotic and tumor suppressor genes, respectively.

  • In recent years, several therapeutic drugs have been rationally designed and synthesized based on the novel knowledge gained from investigating the actions of biologically active chemicals derived from foods, plants, and medicinal herbs. One of the major advantages of these naturalistic chemicals is their ability to interact with multiple targets in the body resulting in a combined beneficial effect. Ginseng is a perennial herb (Araliaceae family), a species within the genus Panax, and a highly valued and popular medicinal plant. Evidence for the medicinal and health benefits of Panax ginseng and its components in preventing neurodegeneration has increased significantly in the past decade. The beneficial effects of P. ginseng on neurodegenerative diseases have been attributed primarily to the antioxidative and immunomodulatory activities of its ginsenoside components. Mechanistic studies on the neuroprotective effects of ginsenosides revealed that they act not only as antioxidants but also as modulators of intracellular neuronal signaling and metabolism, cell survival/death genes, and mitochondrial function. The goal of the present paper is to provide a brief review of recent knowledge and developments concerning the beneficial effects as well as the mechanism of action of P. ginseng and its components in the treatment and prevention of neurodegenerative diseases.
