Preparation and Hypoglycemic Effect of Reconstituted Grain Added with Selected Medicinal Herb Extract (한방 생약재 추출물을 첨가한 재성형 당뇨쌀의 제조 및 혈당강하 효과)

  • Hot-water extract from selected medicinal herbs of diabetes prescription was mixed with brown rice and pearled barley flours and extruded to prepare a reconstituted grain for diabetes millitus patients. Even though the cooked reconstituted grain containing medicinal herb extract was somewhat different from cooked ordinary rice in textural properties measured by texture analyzer, it was estimated to be organoleptically acceptable in sensory parameters. The reconstituted grain added with medicinal herb extract reduced the rate of dialysis of glucose evaluated by in vitro dialysis experiment. The effects of reconstituted grain diet on blood glucose levels in diabetes patients were studied during 3-week period. Compared to normal diet, the reconstituted grain diet for 2 weeks significantly decreased the fasting and 2-hr postprandial blood glucose levels in diabetes patients by 14% and 10%, respectively. These results suggested that the reconstituted grain could be used as an effective therapeutic diet for the control of diabetes mellitus.

Effect of Medicinal Plant Extract Incorporated Carrageenan Based Films on Shelf-Life of Chicken Breast Meat

  • This study was performed to examine the possibility of water extracts for several medicinal plants, such as Amomum tsao-ko, Alpinia oxyphylla, and Citrus unshiu, as an active packaging ingredient for prevention of lipid oxidation. Chicken breast meats were packed with medicinal plant extracts incorporated carrageenan based films and their physico-chemical and microbial properties during storage at $5^{\circ}C$ were investigated. In chicken meat samples packed with A. tsao-ko (TF) or A. oxyphylla (OF) extract incorporated carrageenan based films, pH value, thiobarbituric acid reactive substances (TBARS), and the population of total microbes were significantly lower than those of the negative control (film of no extract was incorporated, CF) after 5 d of storage (p<0.05). Especially, TBARS value of TF ($0.12{\pm}0.01$ mg malonaldehyde/kg meat) was significantly lower than chicken meat samples packed with positive control (ascorbic acid incorporated film, AF, $0.16{\pm}0.01$ mg malonaldehyde/kg meat) at 3 d of storage, and it means TF has enough antioxidative activity to prevent the lipid oxidation of chicken meat. However, there was no consistent effect on VBN values of chicken meats packed with medicinal plant extracts incorporated films during storage. Based on the obtained results, it is considered that A. tsao-ko extract has potential for being used as a natural antioxidant ingredient in active packaging areas.

Enhancing effect of Panax ginseng on Zip4-mediated zinc influx into the cytosol

  • Background: Zinc homeostasis is essential for human health and is regulated by several zinc transporters including ZIP and ZnT. ZIP4 is expressed in the small intestine and is important for zinc absorption from the diet. We investigated in the present study the effects of Panax ginseng (P. ginseng) extract on modulating Zip4 expression and cellular zinc levels in mouse Hepa cells. Methods: Hepa cells were transfected with a luciferase reporter plasmid that contains metal-responsive elements, incubated with P. ginseng extract, and luciferase activity was measured. Using 65ZnCl2, zinc uptake in P. ginseng-treated cells was measured. The expression of Zip4 mRNA and protein in Hepa cells was also investigated. Finally, using a luciferase reporter assay system, the effects of several ginsenosides were monitored. Results: The luciferase activity in cells incubated with P. ginseng extract was significantly higher than that of control cells cultured in normal medium. Hepa cells treated with P. ginseng extract exhibited higher zinc uptake. P. ginseng extract induced Zip4 mRNA expression, which resulted in an enhancement of Zip4 protein expression. Furthermore, some ginsenosides, such as ginsenoside Rc and Re, enhanced luciferase activity driven by intracellular zinc levels. Conclusion: P. ginseng extract induced Zip4 expression at the mRNA and protein level and resulted in higher zinc uptake in Hepa cells. Some ginsenosides facilitated zinc influx. On the basis of these results, we suggest a novel effect of P. ginseng on Zip4-mediated zinc influx, which may provide a new strategy for preventing zinc deficiency.

Development of Drink from Composition with Medicinal Plants and Evaluation of its Physiological Function in Aorta Relaxation (한약재를 이용한 음료의 개발 및 혈관개선에 미치는 기능성 평가)

  • The purpose of this study was to investigate the possibility of medicinal plants application as an edible functional food resources. We carried out to develop a traditional functional beverage by using hot-water extraction of 4 medicinal plants(Polygonatun sibiricum, Ophiopogonis radix, Lycii fructus, Schizandriae fructus) and we examined the effects of drink on physiological function in aorta relaxation. Thus, the effect of developed beverage on phenylephrine induced contraction of isolated rat thoracic aorta. Contractile force was measured with force displacement transducer under 1.5g loading tension. Brix, pH and titratable acidity of developed drink were 9.5%, 3.3 and 0.22%. The approximate nutritional composition of beverage was carbohydrate, 5.98%, crude protein, 0.70%, crude fat, 0.20% and crude ash, 0.20%. Developed beverage contained K(4.00 ㎎%), Na(3.68 ㎎%), Ca(2.54 ㎎%), Mg(1.60 ㎎%) and Fe(0.29㎎%). The contraction forces by injection of phenylephrine in isolated thoracic aorta were significantly low in each experimental groups compared with control groups, These results that developed drink with medicinal plants can be used as a functional material to decrease aorta contraction.

  • This study analyzed surface-chemical transitions in manufacturing process of traditional acupuncture as proposed method by Dongeuibogam. The manufacturing process of traditional acupuncture from a used iron for a long time was divided by primary medicinal herbs treatment, secondary medicinal herbs treatment and tertiary treatment using by dog meat. The traditional acupuncture research process was measured according to the characteristics and changes of the specimens at each processing step of the manufacturing process. The following devices were used to Surface analysis. Scanning electron microscopy (SEM), Energy Dispersive Spectrometer (EDS), X-ray diffraction analyzer (XRD). As a result, medicinal herbs removed impurities on the surface and raised the antibacterial effect in the manufacturing process of traditional acupuncture. Furthermore, the ingredients of medicinal herbs were coated on the surface of the iron. Dog meat influenced to prevent surface corrosion, reduce friction when the acupuncture was inserted. Although the process empirically obtained, a glimpse of the wisdom of our ancestors was revealed. These ancestral wisdom can be expected to apply today, when used in manufacturing process of a modern stainless steel acupuncture to compensate for the defective part.

Antibacterial Activity of Some Medicinal Plants against Propionibacterium acnes (여드름 원인균 Propionibacterium acnes에 대한 생약 추출물의 항균활성 측정)

  • Acne, one of the most common skin disease, is a chronic inflammatory disease and is caused by uncertain and multiple factors. Propionibacterium acnes belongs to the human cutaneous normal flora and is a major etiologic agent of acne vulgaris. In this study, we evaluated the antibacterial effect of 19 medicinal plants. Antibacterial activity of extracts prepared from the 19 medicinal plants was investigated against bacteria related to acnes, Propionibacterium acnes. Among them, some medicinal plants inhibited the growth of the P. acnes. Minimum inhibitory concentration (MIC) of Polygonum aviculare (Herb) was 0.2 mg/ml against P. acnes. MIC of Dianthus chinensis (Aerial Part), Forsythia viridissima (Fruit), Lygodium japonicum (Spore) and Sophora flavescens (Root) were 0.4 mg/ml. Based on these results, Polygonum aviculare (Herb), Dianthus chinensis (Aerial Part), Forsythia viridissima (Fruit), Lygodium japonicum (Spore) and Sophora flavescens (Root) may be considered as a candidate for a good medicine for acne.

Antibacterial Activity of Caryophylli Flos on the Growth of Dental Caries Bacteria, Streptococcus mutans KDJ-50 (구강균 Streptococcus mutans KDJ-50에 대한 정향의 향균효과)

  • This study was conducted to find antibacterial agent against growth of dental caries bacteria. St. mutans KDJ-50 from various medicinal plants of which safety was indetified. Medicinal plants used in this study was dried and grinded after purchased at Daegu yakryung sijang and Kyungsan jungang sijang. Medicinal plants extracted with 80% ethanol at boiling point for 3 hrs was used as antibacterial agent after freeze dried. This study was conducted to find antibacterial agent against growth of dental caries bacteria. St. mutans KDJ-50 from 32 medicinal plants of which safety was indetified. The result of using paper disc method. Caryophylli Flos. Coptidis Rhizoma. Phellodendri Cortex. Schizamdrae Fructs. Myristicae Semen, Crataegi Fructus and Acori Graminei Rhizoma was selected as antibacterial agent. The result of viable cell counting method. the antibacterial activity of Caryophylli Flos was highest among tested 7 medicinal plants followed by Phellodendri Cortex and Coptidis Rhizoma. The extinction effect of the St. mutans by Caryophylli Flos was shown with the addition of 0.5 (w/v) in the medium. The high antibacterial activity was acquired at high extraction temperature and long extraction temperature. The antibacterial activity of Caryophylli Flos was not effected by the concentration of ethanol.

The effect of medicinal herb administration to arthritis induced rabbit arthritistreatment (생약복합제제가 토끼의 퇴행성관절염 개선에 미치는 영향)

  • The purpose of this study was is to investigate the arthritis treatment and prevention of the effects the herbral administration to experimental animals induces arthritis 4 weeks for medicinal herbs (Scutellaria baicalensis Georgi, Acanthopanacis cortex, Achyranthis Radix). To investigate the prevention of histopathological examination and molecular biological examinations for arthritis improvement in vivo New Zealand, white rabbits were divided into a normal (Normal, n = 7), or a collagen-induced arthritis (CIA, n = 7), or a collagenase injection and medicinal herbs (SAA, n = 7). It was confirmed that induced arthritis was treated with Gross examination, and the measurement of average arthritis index (MAI) and improved arthritis by medicinal herbs. TNF-${\alpha}$ and IL-$1{\beta}$ were significantly increased in the CIA and SAA groups, compared to the normal group (p < 0.05). MMP-1 and TIMP-2 were significantly increased in the CIA, compared to the other groups (p < 0.05). MMP-1 and TIMP-2 were significantly decreased in the SAA group, compared to the CIA group (p < 0.05). As a result, Herbal administration used in this study might be able to help in the treatment of arthritis induced by inflammatory and immunosuppressive effects, that which can be expected.

Immune-Enhancing Effect and Anti-Obesity Activit of Kadsura japonica Fruits

  • Under the COVID-19 pandemic, interest in immune enhancement and anti-obesity is increasing. Thus, in this study, we investigated whether Kadsura japonica fruits (KJF) exhibits immunostimulatory activity and anti-obesity activity. KJF increased the production of immunostimulatory factors and phagocytosis in RAW264.7 cells. Inhibition of TLR2 and TLR4 blocked KJF-mediated production of immunostimulatory factors in RAW264.7 cells. In addition, the inhibition of MAPK and PI3K/AKT signaling pathway reduced KJF-mediated production of immunostimulatory factors, and the activation of MAPK and PI3K/AKT signaling pathway by KJF suppressed the inhibition of TLR2/4. KJF attenuated the lipid accumulation and the protein expression such as CEBPα, PPARγ, perilipin-1, adiponectin, and FABP4 related to the lipid accumulation in 3T3-L1 cells. In addition, KJF inhibited excessive proliferation of 3T3-L1 cells and protein expressions such as β-catenin and cyclin D1 related to cell growth. These findings indicate that KJF may have immunostimulatory activity and anti-obesity activity.

Mechanization for Labor-Saving in Seeding and Harvesting of Bupleurum falcatum L. (시호(柴胡) 파종(播種) 및 수확(收穫)의 성력기계화(省力機械化))

  • This experiment was conducted to know the labor saving effect and reducing production cost by agricultural mechanization in the cultivation of Bupleurum falcatum. Labor reducing effects of the drilling seeder by hand and the machine attached to two wheel tiller were 97%, but emergency rate was highest in the former. Dry root yield per plant was increased by low amount of seed sowing but that yield per unit area was increased at much seeding amount in the seeder attached to the tiller. The drilling seeder by hand was showed highest standing ratio of seedling and produced yield to 84.1kg of root yield per 10a. Labor saving effect was the best at the multipurposes mechanized harvester and labor saving and famer's income ratio were increased to 69% and 50% respectively. Labor time and cost were reduced to 74% and 69% respectively by mechanization of sowing and harvest cultivation practice on Bupleurum falcatum.
