Triterpenoid Saponin Contents of the Leaf, Stem and Root of Codonopsis lanceolata (더덕 잎, 줄기, 뿌리 부위의 Triterpenoid 사포닌 함량)

  • Codonopsis lanceolata (Campanulaceae) has been used in traditional medicines, as its roots contain several kinds of 3,28-bidesmosidic triterpenoid saponin with high medicinal values. In this study, we induced hairy root-derived transgenic plants of C. lanceolata and analyzed triterpenoid saponins from the leaf, stem and root. Transgenic plants were regenerated from the hairy roots via somatic embryogenesis. The saponins are lancemaside A, B and E, foetidissimoside A, and aster saponin Hb. Transgenic plants contained richer triterpenoids saponin than wild-type plants. Major saponin lancemaside A was the most abundant saponin in the stem from transgenic-plant, $4.76mg{\cdot}1^{-1}dry$ stem. These results suggest that transgenic plants of C. lanceolata could be used as medicinal materials for the production of triterpene saponins.

  • 잇꽃(Carthamus tinctorius L.)은 국화과에 속하는 두해살이 초본 식물로 한자명은 홍화(紅花), 영명으로는 Safflower를 사용하고 있다. 잇꽃의 원산지는 이집트, 아프카니스탄 등의 서남아시아가 원산지로 알려져 있으며, 우리나라의 잇꽃 재배면적은 2010년 39 ha 수준이 재배되다가 2016년에는 67 ha로 재배면적이 증가한 것으로 보고되고 있다. 우리나라에서 잇꽃 이용은 잇꽃 종실을 위주로 이용되고 있으나 일본의 경우 잇꽃의 경관적 가치를 활용하여 산형현(山形縣)에서는 홍화축제가 매년 개최되고 있으며, 미국 등에서는 잇꽃의 저온압착유가 기능성 건강식품류로 판매되고 있다. 본 연구는 일조시수가 짧은 표고 500 m의 준고랭지에서 잇꽃의 경관적 가치 활용도를 평가하기 위해 차광조건에 따른 개화특성 및 종실 수량성을 평가하기 위해 수행되었다. 비가림 하우스에서 5월 9일 파종후 생육 54일차인 7월 5일 개화기에 지상부 발육량을 조사한 결과 차괄비율이 강해질수록 엽장폭비가 감소하여 세장형에서 장타원형으로 성장함을 알수 있었고, 분지수는 30% 차광조건에서 증가하는 경향을 보였다. 같은 시기에 조사한 엽면적에서는 30% 차광에서 증가하다가 차광비율이 높아질수록 감소하였으며, 식물체당 총 착화수는 30% 차광조건에서 20.3화/주로 가장 많았고 2차 화경의 착화수도 많은 경향을 보였다. 착생된 꽃의 개화단계를 3등급으로 구분하여 비율을 조사한 결과 차광 조건에서는 개화 2, 3단계의 비율이 무차광에 비해 낮아 개화가 지연됨을 알수 있었고, 건조한 관상화의 색도에서는 차광비율이 높아질수록 b 값이 증가하여 황색값이 높아지는 것을 알수 있었다. 파종 후 생육 94일 차인 8월 13일 종실을 수확하여 식물체당 100립중을 조사한 결과 무차광 4.9 g에 비해 차광조건에서는 감소하는 경향을 보였고, 종실수량성에서도 무차광에서 201.1 kg/10 a로 가장 높아 차광조건에서 잇꽃 재배는 개화지연에 의한 등숙률의 감소로 종실수량성이 감소하는 것으로 판단되었다.

  • 고사리(Pteridium aquilinum Kuhn)는 고사리 속(Pteridium spp.)에 속하는 양치식물의 총칭으로서 우리나라에는 22과 70속 272종이 분포되어 있는 것으로 보고되었다. 고사리의 어린 순에는 가식부 100g 당 칼슘 15.0mg, 칼륨 185.0mg 등이 함유되어 있으며 골다공증, 심혈관 질환 등에 효과가 있고 식이섬유로 인해 변비 예방에도 유용한 것으로 알려져 있다. 우리나라의 고사리 재배면적은 2019년 기준 5,349ha 수준을 보이고 있으며 전체 산채류 재배면적에서 26%의 비중을 차지하고 있으나 재배면적의 증가에 따라 고사리 재배의 부가가치를 높일수 있는 방안이 요구되고 있는 실정으로 그 중 고사리 뿌리에서 추출한 전분은 중국 및 일본 등지에서 면류나 제과용으로 이용되고 있어 고사리 재배의 부가가치를 향상시킬 수 있는 방안으로 기대되고 있다. 본 연구에서는 고사리 뿌리의 채취시기에 따른 뿌리 전분의 생산성과 이화학적 특성을 조사하기 위해 2020년 3월 26일부터 10월 28일 까지 5회에 걸쳐 뿌리를 수확하여 세척한 후 고무망치로 파쇄하고 수침 방법을 이용해 추출한 전분의 추출수율과 이화학적 특성을 조사하였다. 채취시기별 고사리 뿌리의 생산성은 7월 27일 수확에서 3,783.3kg/10a로 유의하게 높은 수량을 보였다. 파쇄한 고사리 뿌리를 24시간 sodium metabisulfite 0.4% V/W 용액에 수침하여 전분을 추출한 후 24시간 간격으로 3회에 걸쳐 진탕, 침전과 세척의 과정을 거친 후 조사한 전분 추출 수율은 3월 26일 수확한 뿌리에서 7.6%로 가장 높았고 7월 27일과 9월 24일 채취한 경우에서는 전분이 검출되지 않았으며 10월 28일에는 4.9%의 유의하게 낮았다. 추출한 전분의 색도분석에서 L값은 채취시기가 늦어질수록 높아지는 경향을 보였고 a, b값은 낮아지는 경향을 보여 전분의 색도가 향상되는 경향을 보였으며, 신속점도측정기로 조사한 전분의 호화특성에서는 채취시기가 늦어질수록 전분의 최고점도는 높아졌으나 최저점도 및 최종점도는 낮아지는 경향을 나타냈다. 전분의 무기물 함량은 3월 26일과 5월 25일 채취하는 경우 Fe > Ca > Mg > K > P의 순으로 함량이 높았으나 10월 28일 채취한 경우에서는 Fe와 Ca이 감소되고 K 함량은 증가하여 Ca > Fe > Mg > K > P의 순으로 무기물 함량이 높게 나타났다.

Vitamin U in Medicinal Food Plants

  • Vitamin U levels in 26 kinds of food plants are well known to healthy vegetables in Asian or Western countries were determined. Spinach showed the highest level of 452.04 mg/kg and in order Pack-choi (343.18 mg/kg) > Kale (234.18 mg/kg) > Somssukbujaengi (197.66 mg/kg) > leaf mustard (196.21 mg/kg) > aralia bud (192.50 mg/kg)> broccoli (189.03 mg/kg) >Asparagus (187.35 mg/kg). Among Korean wild medicinal plants, Sumssukbujaengi showed the highest value followed by Sanmanul (a kind of wild garlic) level of 143.46 mg/kg. For Chinese cabbages and cabbages, vitamin U showed different levels according to the parts of plant such as core, middle, outward leaves. In both samples, middle parts of leaves including midribs contained the highest level of vitamin U. The level of vitamin U was dependent on the part of the plant sample and cultivars. Leaf parts of turnip and white radish showed higher value of 84.82 mg/kg and 124.62 mg/kg than those of roots which were in order of middle (112.39 mg/kg), top (84.84 mg/kg) and bottom (84.61 mg/kg) portions in the white radish. In the analysis of amino acids, we didn't find either distinctive relationship between methionine and vitamin U synthesis or significant connection various free amino acids and vitamin U level in food plants.

In orther to analyze the vegetation of Wol-chulmountain area, medical wild plants structure and distribution. Medical wildplants of Wol-chul mourltain consisted of 11'0 familis, 338 species in all. Effects of Interactions Among Age, Cultivation Method (Location) and Population on Ginsenoside Content of Wild Panax Quinquefolium L. One Year after Transplanting from Wild

  • To evaluate the effects of cultivar, environment, age and cultivation times on ginsenoside content among 8 wild populations of American ginseng (Panax quinquefolium), the concentrations of 6 ginsenosides in root were determined at the time of collection (T0) of plants from the wild and 1 year after (T1) transplanting the roots to each of two different forest garden locations. Both location and population had significant effects on root and shoot growth. Overall, ginsenoside Rb1 was most abundant. The second most abundant ginsenoside were Re and Rg1, however the contents of them were not significantly different from each other. Concentrations of Rg1 and Re were inversely related. Ginsenoside Re was influenced by population and location. Ginsenoside Rg1, Rb1, Rc, Rb2 and Rd were influenced by population, location and age. Ginsenoside levels were consistently lower but growth was consistently higher at the more intensively managed garden location.

  • There shoul be high possibility of preventive and immune eficacy aqainst cancer when the anti-cancer plants are properly utilized in human dietary life as it is evident that considerable part of wild plantd or both medicinal use and ood are growing naturally in the mountainous area in korea, some of which have ben proved to be anti-cancer plants. this study, at this initial stage, has been done to acquire the fundamentals of 235 kinds of ante-cancer plants growing naturally in korea. from the results o thisresearch, the anti-cancer plants have been sorted out by amily and its number. and also the status of regional distribution of those plants and the kinds of anti-cancer plants used for both medicine and food were studied and experimented to see the medical efficacy against anti-tumor. this experiments were conduced in accodance with the total packed cell volume method and cyto toxicity method.

Isolation and Tyrosinase Inhibitory Activity of Wild Yeasts Obtained from Soil in the Fields of Medicinal Plants, Ginseng and Korean Angelica (인삼과 당귀 재배 토양으로부터 야생효모들의 분리 및 미백성 Tyrosinase 저해활성)

  • The goal of this study was to isolate wild yeasts from the fields of medicinal plants and investigate its tyrosinase inhibitory activities. Wild yeasts isolated from soil in the ginseng and Korean angelica fields of Geumsan, Chungcheongnam-do, Korea were identified by comparing the nucleotide sequences of the D1/D2 domain of 26S rDNA. In total, 43 yeast strains belonging to 21 species were isolated from 50 soil samples obtained from two medicinal plant fields. From the ginseng field, six strains of Rhodotorula glutinis and four strains of Sampaiozyma ingeniosa were isolated, out of which Rhodotorula glutinis strains were dominant. In the Korean angelica field, six strains of Cyberlindnera saturnus, three strains of Piskurozyma taiwanensis, and three strains of Saitozyma podzolica were isolated, out of which Cyberlindnera saturnus strains were dominant. We prepared cell-free extracts of the isolated wild yeasts and their tyrosinase inhibitory activities were investigated. Among 43 yeast strains, cell-free extracts of Naganishia globosa G1-7 showed the highest tyrosinase inhibitory activity (28.0%).

Effects of processing method on the pharmacokinetics and tissue distribution of orally administered ginseng

  • Background: The use of different methods for the processing of ginseng can result in alterations in its medicinal properties and efficacy. White ginseng (WG), frozen ginseng (FG), and red ginseng (RG) are produced using different methods. WG, FG, and RG possess different pharmacological properties. Methods: WG, FG, and RG extracts and pure ginsenosides were administered to rats to study the pharmacokinetics and tissue distribution characteristics of the following ginsenosides-DRg1, Re, Rb1, and Rd. The concentrations of the ginsenosides in the plasma and tissues were determined using UPLC-MS/MS. Results: The rate and extent of absorption of Rg1, Re, Rb1, and Rd appeared to be affected by the different methods used in processing the ginseng samples. The areas under the plasma drug concentration-time curves (AUCs) of Rg1, Re, Rb1, and Rd were significantly higher than those of the pure ginsenosides. In addition, the AUCs of Rg1, Re, Rb1, and Rd were different for WG, FG, and RG. The amounts of Rg1, Re, Rd, and Rb1 were significantly (p < 0.05) higher in the tissues than those of the pure ginsenosides. The amounts of Re, Rb1, and Rd from the RG extract were significantly higher than those from the WG and FG extracts in the heart, lungs, and kidneys of the rats. Conclusion: Our results show that the use of different methods to process ginseng might affect the pharmacokinetics and oral bioavailability of ginseng as well as the tissue concentrations of Rg1, Re, Rd, and Rb1.
