  • 음식 알러지는 만성적인 장애를 일으킬 수 있는 즉각적이며 잠재적으로 생명을 위협하는 반응이다. 이 중 계란은 알러지 위험성이 높은 물질로, 아이들에게 음식 알러지를 유발하는 흔한 물질로 알려져 있다. 계란 알러젠 중에서 계란 흰자 단백질인 OVA이 주요한 알러젠으로 알려져 있다. 우리는 이번 연구에서 OVA이 염증 및 면역 반응에 중요한 역할을 한다고 알려져 있는 $NF-{\kappa}B$ 활성화와 COX-2와 iNOS 발현에 어떠한 영향을 미치는지를 알아보았다. OVA은 $NF-{\kappa}B$ 활성화와 COX-2와 iNOS의 발현을 증가시켰다. 이러한 연구는 앞으로 계란 알러젠에 의한 알러지 작용기전 규명 및 알러지 치료제 개발에 중요한 역할을 할 것으로 기대한다.

임상병리학, 검사의학, 의료생물학 관련 지원보건직 명칭에 대한 제안 (Job Title Recommendations for Allied Health Professionals Related to Clinical Pathology, Laboratory Medicine, and Medical Biology)

  • 본 연구의 목적은 임상병리사의 전공과 직업에 정체성을 부여할 수 있는 용어를 찾는 것이다. 임상병리학이라는 용어는 병리학의 모든 분야, 즉 해부병리학, 화학병리학, 혈액학, 미생물학 및 이들의 모든 하위 전문 분야를 포함하도록 의도되었다. 불행히도, 많은 나라에서 임상병리학이라는 용어는 해부병리학을 배제하는 것으로 생각되었는데, 이는 이름이 영어 이외의 언어로 번역되었을 때 문제가 복잡해졌다. 임상병리학(미국, 영국)은 의학의 한 전문 분야이다. 유사한 용어로는 검사의학(독일, 폴란드), 의료/임상생물학(프랑스, 네덜란드) 또는 임상분석학(스페인)이 있다. 의료기술은 개인, 회사 및 기관과 같이 질문하는 사람에 따라 약간 다르게 정의된다. 물론, 질문하는 언어에 따라 다르다. 의료기술은 임상검사기술학, 지원보건과학, 의료기기, 의생명공학, 보건기술로도 번역될 수 있다. 지원보건과학에서는 '임상병리기술학·병리기술학'이라는 용어를 사용하지 않는다. '보건계열 임상병리학 및 임상병리사'의 명칭은 '의생명검험과학·의생명검험사' 또는 '임상검사분석학·임상검사분석사'로 대체할 수 있다. 본 연구에서는 학문과 자격의 명칭을 '의생명'이 결합된 용어로 '의료생물학·의료생물사'로 변경할 것을 제안해 본다.

Helicobacter pylori Chaperone-Like Protein CagT Plays an Essential Role in the Translocation of CagA into Host Cells

  • Most of the Helicobacter pylori strains containing the cag pathogenicity island (PAI) have been associated with more severe gastric disease in infected humans. The cag PAI is composed of 27 proteins, and some of the components are required for CagA translocation into host cells as well as induction of proinflammatory cytokines, such as interleukin-8 (IL-8); however, the exact function of most of the components remains unknown or poorly characterized. In this study, we demonstrated that CagT (HP0532), which is an essential structural component of the cag PAI apparatus, plays an important role in the translocation of CagA into host epithelial cells. In addition to being located on the bacterial surface, CagT is also partially localized in the inner membrane, where it acts as a chaperone-like protein and promotes CagA translocation. However, CagT secretion was not detected by immunoprecipitation analysis of cell culture supernatants. Meanwhile, CagT was related to the introduction of IL-8 of the host cell. These results suggest that CagT is expressed on both the inner and outer bacterial membranes, where it serves as a unique type IV secretion system component that is involved in CagA secretion and cag PAI apparatus assembly.

Translocation and Phosphorylation of Calcyclin Binding Protein during Retinoic Acid-induced Neuronal Differentiation of Neuroblastoma SH-SY5Y Cells

  • For better understanding of functions of the Calcyclin Binding Protein (CacyBP) and exploring its possible roles in neuronal differentiation, the subcellular localization of human CacyBP was examined in retinoic acid(RA)-induced and uninduced neuroblastoma SH-SY5Y cells. Immunostaining indicated that CacyBP was present in the cytoplasm of uninduced SH-SY5Y cells, in which the resting $Ca^{2+}$ concentration was relatively lower than that of RA-induced cells. After the RA induction, immunostaining was seen in both the nucleus and cytoplasm. In the RA-induced differentiated SH-SY5Y cells, CacyBP was phosphorylated on serine residue(s), while it existed in a dephosphorylated form in normal (uninduced) cells. Thus, the phosphorylation of CacyBP occurs when it is translocated to the nuclear region. The translocation of CacyBP during the RA-induced differentiation of SH-SY5Y cells suggested that this protein might play a role in neuronal differentiation.

  • Intraoperative Neurophysiological Monitoring (INM) is very useful in monitoring the motorsensory pathway and vascular circulation system during intraspinal, or intracranial neurosurgery. Brainstem Auditory Evoked Potentials (BAEPs) are for detecting the problems along the auditory pathways including, the eighth cranial nerve and brainstem. Motor Evoked Potentials (MEPs) is a useful adjunct to conventional monitoring of Somato-sensory Evoked Potentials (SEPs) during surgery. Visual Evoked Potentials (VEPs) has been regarded as having limited significance for the preservation of visual function during neurosurgical procedures. In this paper, we propose that the most appropriate averaging of the number of inspections in the inspection of each used in the operative field, is good and efficient, functionally.

Cardamonin Inhibits the Expression of Inducible Nitric Oxide Synthase Induced by TLR2, 4, and 6 Agonists

  • Toll-like receptors (TLRs) play an important role for host defense against invading pathogens. The activation of TLRs signaling leads to the activation of $NF-{\kappa}B$ and the expression of pro-inflammatory gene products such as cytokines and inducible nitric oxide synthase (iNOS). To evaluate the therapeutic potential of cardamonin, which is a naturally occurring chalcone from Alpinia species (zingiberaceous plant species), $NF-{\kappa}B$ activation and iNOS expression induced by MALP-2 (TLR2 and TLR6 agonist) or LPS (TLR4 agonist) were examined. Cardamonin inhibited the activation of $NF-{\kappa}B$ induced by MALP-2 or LPS. Cardamonin also suppressed the iNOS expression induced by MALP-2 or LPS. These results suggest that cardamonin has the specific mechanism for anti-inflammatory responses by regulating of TLRs signaling pathway.

Removal of the Glycosylation of Prion Protein Provokes Apoptosis in SF126

  • Although the function of cellular prion protein (PrP$^C$) and the pathogenesis of prion diseases have been widely described, the mechanisms are not fully clarified. In this study, increases of the portion of non-glycosylated prion protein deposited in the hamster brains infected with scrapie strain 263K were described. To elucidate the pathological role of glycosylation profile of PrP, wild type human PrP (HuPrP) and two genetic engineering generated non-glycosylated PrP mutants (N181Q/N197Q and T183A/T199A) were transiently expressed in human astrocytoma cell line SF126. The results revealed that expressions of non-glycosylated PrP induced significantly more apoptosis cells than that of wild type PrP. It illustrated that Bcl-2 proteins might be involved in the apoptosis pathway of non-glycosylated PrPs. Our data highlights that removal of glycosylation of prion protein provokes cells apoptosis.

Effect of Scutellariae Radix as a Novel Antibacterial Herb on the ppk(Polyphosphate Kinase) Mutant of Salmonella typhimurium

  • The antibacterial effects of water extracts of Scutellariate Radix (a dried root of Scutellaria baicalensis GEORGI) and its major flavonoid components, Baicalin and Baicalein, on Salmonella typhimurium, a representative enteric pathogen, were studied. Through a Kriby-Bauer disc analysis, the growth-inhibition activity of Scutellariae Radix against. S. typhimurium was found to be compatible with commercial antibiotics, such as ampicillin, chloramphenicol, and streptomycin. In contrast, the growth of a nonpathogenic E. coli strain was unaffercted by Scutellariae Radix. To examine the effect of polyphosphate kinase (ppk), a putative virulence factor, on the antibacterial activity of Scutellariae Radix, the growth profile of a ppk mutant of S. typhimurium was investigated in a tryptic soy broth containing different concentrations of water extracts of Scutellariae Radix. The ppk mutant was able to grow in 6 mg/ml of water extracts of Scutellariae Radix, whereas in 6 mg/ml of water extracts of Scutellariae Radix, whereas the wild-type could not, implying that the inactivation of ppk made S. typhimurium more resistant to the antibacterial activity of Scutellariae Radix. No enhanced resistance was observed in a ppk mutant of S. typhimurium complemented with a ppk expression vector. The attenuation of the virulence by ppk inactivation was also observed in a virulence assay using BLAB/c mice. Neither Baicalin nor Baicalein exhibited any growth-inhibition activity against S. typhimurium. The water extracts of Scutellariae Radix stimulated the transcription of ppk, especially in the early growth-stage of S. typhimurium.
