  • This experiment was carried out to investigate the effects of liming, manure application, and inoculation with Rhizobium on the early growth, nodulation, seedling establishment, winter survival, and forage yield and quality of alfalfa(Medicago sativa L.) at the Experimental Livestock Farm, College of Agriculture, Seoul National University, Suweon in 1985 and 1986. The results obtained are as follows: 1. Seedling vigor and nodule formation of alfalfa at early stage were the best in the treatments of liminginoculation( L1) and liming-manure-inoculation(LM1). 2. Seedling establishment was not significantly affected by the treatments of liming, manure application, nodulation and the combination of these. 3. The LI and LMI treatments showed the highest winter survival of 94.3 and 83.5%, respectively, but the lowest winter su.rvival of 55.9 and 58.5% was found in no treatment(None) and inoculation( I ), respectively. 4. No differences in crude protein(CP1, acid detergent fiber(ADF) and neutral detergent fiber(NDF) contents were found among the treatments. 5. There were significant differences in the total dry matter yield of alfalfa among the treatments. The total dry matter yields of 10,618, 11,830, 11,855 and 13,456 kg/ha from the treatments of L, LM, LI and LMI, respectively, were significantly higher than those of None, M and I treatments. 6. Based on the results of the experiment, it appears that the dry matter yield of alfalfa could be enhanced by the treatments of liming with Rhizobium inoculation and also liming, manure application and Rhizobium inoculation as a practical method.

Callus Formation from Alfalfa (Medicago sativa L.) Seed and Plant Regeneration from Alfalfa Calli (알팔파 종자로부터 캘러스 유도 및 식물체 재분화)

  • The conditions for callus formation and plant regeneration were confirmed in four varieties of alfalfa(Medicago sativa L.). Among four varieties of alfalfa, "Vernal" expressed the highest rate for both of callus formation and plant regeneration. Otherwise, among SH(Schenk and Hildebrandt), MS(Murachige and Skoog) and N6 medium (Chu), SH medium was highest degree of efficiencies respectively in callus formatio and plant regeneration. In this study, we determined volume of hormones and other compounds appended in media. For callus formation, only $3mg/{\ell}$ of 2,4-D (2,4-dichlorophenoxy acetic acid) was appended in their media. For plant regeneration, the three kinds of media were used; the medium appended $5mg/{\ell}$ of NAA (1-naphtalene acetic acid) and $2mg/{\ell}$ of kinetin (6-furfurylaminopurine), the medium appended $11mg/{\ell}$ of 2,4-D and $1mg/{\ell}$ of kinetin, and the medium appended $1.6g/{\ell}$ of ammonium sulfate and $5.75g/{\ell}$ of proline. We obtained alfalfa plants from callus by regeneration, about sixty five days later transfer calli to regeneration media.

Correlations among Shearing Force, Morphological Characteristic, Chemical Composition, and In situ Digestibility of Alfalfa (Medicago sativa L) Stem

  • Alfalfa (Medicago sativa L) is a high-quality forage for ruminants and the main stem is the dominant morphological component contributing to the forage nutritive value in mature alfalfa forage. Shearing force, a fracturing property of plant stem, is an important indictor of forage value. The objectives of this study were to investigate the effects of morphological characteristic on shearing force, the relationship between shearing force and chemical composition, and the relationship between shearing force and in situ digestibility of alfalfa stem. The results showed that linear density (weight per unit length of stem) was more important than chemical composition in affecting shearing force. There was a positive relationship between lignin content and shearing force (r = 0.78). Correlations were not found between shearing force and other chemical components such as neutral detergent fiber (NDF), acid detergent fiber (ADF), cellulose and hemicelluloses. In situ digestibility (of dry matter and NDF) was related to shearing force. A negative correlation was found between shearing force and dry matter (DM) digestibility (r = -0.70), and there was also a negative correlation between shearing force and NDF digestibility (r = -0.87). When shearing force was standardized for stem diameter or stem linear density, the relationship between shearing force and digestibility was consistent regardless of stem diameter and stem linear density. Shearing force was significantly correlated with lignin content and in situ digestibility (of DM and NDF), and was a more direct indicator for estimating forage nutritive value related to animal performance, so it can be used to predict the forage value of alfalfa.

Effect of Phos phatic Fertilization Level on Growth , Crude Potein Content and Nitrohen Fixation Ability of Alfalfa ( Medicago sativa L. ) at Sowing Year (인산시비수준이 알팔파 ( Medicago sativa L. ) 의 파종초년도 생육 , 조단백질함량 및 질소고정능에 미치는 영향)

  • The experiment was carried out to investigate the effects of phosphate fertilizer level on growth, crude protein content and nitrogen fixation ability of alfalfa(Medicugo sariva L.) at sowing year. The experimental field was conducted using a randomized block design with three replications according to the phosphate application level (200 and 400kfia) in farm of Kongju National University. Research samples were collected at intervals of 7 days from October 7 to December 9 and nitrogen fixation activity of nodule was measured by using the acetylene reduction method (Hardy et al. 1968). The results obtained are summarized as follows : 1. Number of branches, root length and dry matter weight at phosphate fertilizer level of 400kg in sowing year were more increase than those of 200kfia. The number of nodules was not significantly difference between phosphate fertilizer level, because of low temperature to be enough to nodulation of alfalfa. 2. The crude protein content of leaf, stem and root were not significantly affected by the phosphate fertilizer level. but that of leaf was increased at 400kgha on 72 days after sowing. The crude protein content of each part according to growing stages was increased at 44 days and 72 days after sowing. 3. The dry weight of root was not significantly difference between phosphate fertilizer level from 30 days to 51 days after sowing, but that was increased at phosphate fertilizer level of 400kgha from 51 days afier sowing. 4. The amount of ethylene in the samples taken 2 hours and calculated amount of nitrogen fixation were increased at phosphate fertilizer level of 400kfia from 30 days to 65 days after sowing. but there was vice versa from 58 days after sowing.

알팔파 종자로부터 캘러스유도 및 재분화 조건

  • 4품종의 알팔파 (Medicago sativa L.) 종자로부터 직접 캘러스를 유도하고, 형성된 캘러스로부터 식물체를 재분화하는 조건을 확립하였다. 공시품종중 "Vernal" 품종이 캘러스 유도 및 재분화에서 가장 우수한 것으로 판명되었으며, SH(Schenk and Hildebrandt), MS (Murashige and Skoog), N6 (Chu) 배지중에서 SH 배지가 캘러스 유도와 재분화시에 모두 유리한 것으로 나타났다. 캘러스 유도 및 증식시에는 2,4-D를 3mg/$\ell$ 농도로 첨가한 SH-3 배지에서 가장 효율이 좋았으며, kinetin을 첨가하거나 kinetin의 농도가 높을수록 캘러스 형성 정도는 급격히 저하되었다.(중략)저하되었다.(중략)

Remobilization of Nitrogen Reserves and Analysis of Xylem Exudate during Regrowth of Alfalfa (Medicago sativa) (알팔파의 예취 후 재생시 저장질소의 재이동과 목부 삼출액의 분석)

  • Experiments with non-nodulated alfalfa (Medicago sativa L. cv, Europe) plants grown in hydroponic cuiture, were carried out to estimate the remobiJization of nitrogen (N) reserves and to investigate the transported forms of reduced soluble-N in xylem during regrowth following shoot removal. Endogenous N remobilization were estimated by $^{15}N$ labelling and amino acids in xylem sap were analysed. The, $^{15}N$ contents of ~egr9wing leaves and stems increased as a result of remobilization of N reserves mainly from root ,system, Regrowing leaves were a stronger sink than regrowing stems, with about tWo-thirds of remobilized 15N being recoved in leaves. Endogenous N in lateral roots accounted for about 46% of the total N reserves used for regrowth, while tap roots accounted for 23%, About 72% of total endogenous N remobilized to regrowing shoot, occurred during the first 10 days of regrowth, The outflow of reduced soluble-N (mainly amino acids) was greater than that of protein-N, while the latter was the largest storage pool in tap root and lateral roots. It is suggested that amino acids-N was the most readily avaiable form of N reserves. Asparagine, which repre5ented about 75% of amino acids-N in xylem sap, was the main transported form of reduced N. Its relative contents, during the first 10 days of regrowth, decreased from 75% to 59%. This decline was accompanied by compensatory increase in the relative contents of asparatate and glutamine.

Non-traditional Straws: Alternate Feedstuffs for Ruminants

  • The nutritive value of 4 straws, obtained after thrashing of seeds from fodder crops, was assessed as complete feed for ruminants. Sixteen male Murrah buffaloes (liveweight 365.8${\pm}$19.5 kg), were divided into 4 equal groups and offered ad lib. straw of either Trifolium resupinatum, Trifolium alexandrium, Medicago sativa or Lolium perenne, supplemented with minerals and vitamin A, for 40 days in a completely randomized design. Simultaneously, each straw was offered to 3 rumen fistulated male buffaloes in order to assess the biochemical changes in the rumen. Compared to other straws M. sativa straw had higher (p<0.05) organic matter (OM), crude protein (CP), acid-detergent fiber (ADF) and cellulose content. L .perenne had the highest (p<0.05) hemicellulose and lowest (p<0.05) CP and acid-detergent lignin (ADL) content. T. resupinatum had the lowest concentration of cell wall constituents (CWC). The digestibility of nutrients of T. resupinatum and L. perenne straw was similar, but higher (p<0.05) than that of other straws. M.sativa straw showed highest (p<0.05) digestibility of CP. The highest OM digestibility of T. resupinatum and CP digestibility of M. sativa were responsible for highest (p<0.05) total volatile fatty acids and trichloroacetic acid precipitable nitrogen in the strained rumen liquor. The digestible crude protein (DCP) was highest (p<0.05) in M. sativa followed by that in T. alexandrium. The total purine derivatives excreted in urine varied from 0.22-0.32 mmol/kg $W^{.75}/d$. The efficiency of microbial protein synthesis indicated that OM of straws of M. sativa and that of T. alexandrium was used more (p<0.05) efficiently. The microbial protein synthesized was highest in T. resupinatum, but statistically similar to other groups. The values for N-retention and apparent biological value were highest for L. perenne, though comparable with that of M. sativa and T. alexandrium. The available metabolizable energy (ME) was highest (p<0.05) in T. resupinatum followed by that in L. perenne and lowest in M. sativa. It was concluded that all the straws, supplemented with minerals and vitamin A, could be fed exclusively to adult ruminants with no adverse affect, as animals were able to maintain body weight (372${\pm}$20.1 kg).
