Optimization of Regeneration Condition Under Agrobacterium-mediated Transformation in in vitro Cultured Korean Soybean

  • Soybean is the important crop in Asian countries as protein source, oil production and animal feed. Improving soybean using genetic transformation is the principal tool in nowadays. Developing herbicide resistant transgenic soybean plants through Agrobacterium-mediated transformation has been worked in many previous studied. However, the transformation efficiency is still low. Many attempts try to find the optimum media condition for plant regeneration after infection. After transformation, the plant regeneration is very important condition to promote growth of transgenic plant. In this study, we optimized a regeneration condition for two Korean soybean cultivar, Dawonkong and Pungsannamulkong using cotyledon, cotyledonary nodes and hypocotyl as explant. The results showed that shoot regeneration of cotyledonary nodes on B5 medium containing 2 mg/L 6-benzylaminopurine showed the highest percentage of regeneration in Dawonkong (75.8%) while Pungsannamulkong presented high number of shoots 2.12 shoots per explant. For transformation condition, co-cultivation in 7 days showed a high number of GUS positive expression. Most of explants can survived under media including 5 mg/L of glufocinate which refers phosphinotricin for 2-week selection. Washing with 400 mg/L of cefotaxime in several times and selection in plant regeneration media with 400 mg/L of cefotaxime can prevent bacteria growth, effectively.

  • Today, the fashion business environment of the 4.0 generation is changing based on fashion technology combined with advanced digital technologies such as AI (Artificial Intelligence), big data and IoT (Internet of Things). "Digital Transformation" means a fundamental change and innovation in a digital paradigm including corporate strategy, organization, communication, and business model, based on the utilization of digital technology. Thus, this study examines digital transformation strategies through the fashion brand Burberry. The study contents are as follows. First, it examines the theoretical concept of digital transformation and its utilization status. Second, it analyzes the characteristics of Burberry's digital transformation based on its strategies. For the research methodology, a literature review was performed on books and papers, aligning with case studies through websites, social media, and news articles. The result showed that first, Burberry has reset their main target to Millennials who actively use mobile and social media, and continues to communicate with them by utilizing digital strategy in the entire management. Second, Burberry is quickly delivering consistent brand identity to consumers by internally creating and providing social media-friendly content. Third, they have started real-time product sales and services by using IT to enhance access to brands and to lead consumers towards more active participation. In this study, Burberry's case shows that digital transformation can contribute to increased brand value and sales, keeping up with the changes in the digital paradigm. Therefore, the study suggests that digital transformation will serve as an important business strategy for fashion brands in the future.

  • The advent of the "un-tact 2.0 era," described as a "new normal non-face-to-face social relationship," accelerates the transformation of the living paradigm as a fully digital mediated social relationship. The emergence of these new forms of digital behaviors and mediated relationships significantly influences the industry prospect and consumers' individual lives. Advertising has played a decisive role in moving the formation forces of society, creating a dynamic flow of the capitalist system across races and geographical boundaries. Its role will become more fundamental in the physical contactless environment due to the COVID-19 outbreak. Accordingly, advertising has changed and modified its shape and meaning throughout history to adapt to the dynamic external environment. In this article, we illustrated four primary stages of the evolution in the advertising industry from simple advertising to convergence of advertising. Finally, we also identified the challenges of the present advertising industry and the paradigm transformation of "un-tact 2.0" with various related examples.

Organogenesis and Production of Some Transgenic Legume Plants by Agrobacterium tumefaciens-mediated Herbicide Resistance Gene Transformation

  • Development of herbicide resistant transgenic legume plants through Agrobacterium-mediated transformation has been worked in many previous studied. Plant regeneration after infection is the important step to obtain successful transgenic plants. Many attempts try to find the optimum media condition for plant regeneration after infection. However, the transformation efficiency of legume plants is still low. In this study, regeneration of some Korean legume species including two soybean cultivars (Dawon and Pungsan) and pea have been done with organogenesis which is used various kind of explants such as cotyledonary-nodes in soybean and bud-containing tissue in pea. We developed the optimum media condition for plant regeneration regulators under Agrobacterium-mediated transformation using different kind and various concentration of plant growth. As the results, B5 medium containing 2 mg/L of 6-benzylaminopurine was selected in this study for the optimum plant regeneration media. The segments were inoculated with Agrobacterium suspension harbored an IG2 vector containing bar gene which confers resistance to phosphinotricin (PPT) in 3, 5 and 7 days. The transformation efficiency was achieved in Dawon 3.03 % and pea 1.46 % with co-cultivation period of 7 days which is showed a high number of GUS positive expression period.

  • Recombinant chimeric plasmid constructed with Xba I digested pUBl10 and -pE194 was transformed by polyethylene glycol induced protoplast transformation system into Bacillus subtilis BR 151 on the mannitol regeneration media, and two genes of antibiotics resistance were expressed simultaneously in the transfromant. Transformation frequency of the recombinant plasmid was 6.5 $\times$ 10$^{-5}$ on the mannitol regeneration agar plate containing neomycin and erythromycin. The replication of recombinant plasmid in the recipient cells was confirmed by the alkaline extraction method and agarose gel electrophoresis.

  • In this paper, we propose an adaptive transformation engine that provides smart devices with optimal user interfaces and media contents to their characteristics so as to conveniently control home networks remotely using smart devices in the wired and wireless home networks which various kinds of smart devices and home network devices are connected. The proposed engine dynamically transforms UI contents and transfers them using device profiles and UI transformation policies to make it suitable for smart devices. Moreover, the proposed engine offers suitable media contents for smart devices through capabilities of real-time media transcoding and streaming considering device and media profiles.

Improvement of Transformation Efficiencies using Agrobacterium-Mediated Transformation of Korean Rice

  • A reproducible transformation system via optimized regeneration media for Korean rice cultivars was established using Agrobacterium tumefeciens LBA4404 (pSBM-PPGN; gusA and bar). Although japonica rice genotypes were easier to produce transgenic plants compared to Tongil type cultivars, transformation efficiencies were not always correlated with regeneration efficiencies of non-transgenic callus on the control medium. Regeneration efficiencies of Donganbyeo, Ilmibyeo, and Manchubyeo were over 50% in non-transgenic control, however, transformation efficiencies were significantly low when only sucrose was added to the media as a carbon source. However, the medium, MSRK5SS-Pr (or MSRK5SM-Pr), that contains $5\textrm{mgL}^{-1}$ kinetin, $0.5\textrm{mgL}^{-1}$ NAA, 2 % sucrose (or maltose), 3% sorbitol, and $500\textrm{mgL}^{-1}$ proline, was the most efficient not only for regeneration of non-transgenic callus but also for regeneration of transgenic callus in the presence of L-phosphinotricin (PPT). Average transformation efficiencies of 16 Korean rice cultivars were significantly enhanced by using the optimized medium from 1.5% to 5.8% in independent callus lines and from 2.9% to 19.4% in tromsgenic plants obained. Approximately 98.9% (876 out of 885) transgenic plants obtained on optimized media showed basta resistance. Stable integration, inheritance and expression of gusA and bar genes were continued by GUS assay and PCR and Southern analysis of the bar gene. With Pst1 digestion of genomic DNA of transgenic plants, one to five copies of T-DNA segment were observed; however, 76% (19 out of 25 transgenic plants) has low copy number of T-DNA. The transformants obtained from one callus line showed the same copy numbers with the same fractionized band patterns.

Fast Prediction Mode Decision in HEVC Using a Pseudo Rate-Distortion Based on Separated Encoding Structure

  • A novel fast algorithm is suggested for a coding unit (CU) mode decision using pseudo rate-distortion based on a separated encoding structure in High Efficiency Video Coding (HEVC). A conventional HEVC encoder requires a large computational time for a CU mode prediction because prediction and transformation procedures are applied to obtain a rate-distortion cost. Hence, for the practical application of HEVC encoding, it is necessary to significantly reduce the computational time of CU mode prediction. As described in this paper, under the proposed separated encoder structure, it is possible to decide the CU prediction mode without a full processing of the prediction and transformation to obtain a rate-distortion cost based on a suitable condition. Furthermore, to construct a suitable condition to improve the encoding speed, we employ a pseudo rate-distortion estimation based on a Hadamard transformation and a simple quantization. The experimental results show that the proposed method achieves a 38.68% reduction in the total encoding time with a similar coding performance to that of the HEVC reference model.

The Transformation of Alternative Public Sphere and its Motive in Korea (한국 대안적 공론장의 변화과정과 추동 요인에 대한 고찰)

  • The concept of alternative sphere is terminologically various; resistance media, counter media, minjung media, grassroots media, underground media, alternative media, civic media, etc. Each terminology reflect the feature of times and the emphasis of advocator. This article explore the transformation of alternative public sphere and its motive in Korea. In a world, Korea's alternative public sphere has changed as following: resistance media and liberty media in 70', counter media and minjung media in 80', alternative media in 90', and alternative media and civic media in 2000'. The motive of transformation is basically the expansion of civil society and extension of social movement in Korea. As social movement's character has changed, the feature of alternative public sphere has also changed. Minjung movement played key role of social movement in 1980', the alternative public sphere characterized as minjung media or counter media which was based on working class consciousness. After this, According as civil movement have initiative of social movement, the character of alternative public sphere changed by alternative media and civic media. Besides, this article argue that the alternative public sphere of Korea has changed dialectically with social movement.

Identifying Research Trends in Big data-driven Digital Transformation Using Text Mining (텍스트마이닝을 활용한 빅데이터 기반의 디지털 트랜스포메이션 연구동향 파악)

  • A big data-driven digital transformation is defined as a process that aims to innovate companies by triggering significant changes to their capabilities and designs through the use of big data and various technologies. For a successful big data-driven digital transformation, reviewing related literature, which enhances the understanding of research statuses and the identification of key research topics and relationships among key topics, is necessary. However, understanding and describing literature is challenging, considering its volume and variety. Establishing a common ground for central concepts is essential for science. To clarify key research topics on the big data-driven digital transformation, we carry out a comprehensive literature review by performing text mining of 439 articles. Text mining is applied to learn and identify specific topics, and the suggested key references are manually reviewed to develop a state-of-the-art overview. A total of 10 key research topics and relationships among the topics are identified. This study contributes to clarifying a systematized view of dispersed studies on big data-driven digital transformation across multiple disciplines and encourages further academic discussions and industrial transformation.
