Research on the Production of CO2 Absorbent Using Railway Tie Concrete Waste (콘크리트 철도 침목 폐기물을 활용한 CO2 포집제 제조 연구)

  • In recent years, excessive emissions of carbon dioxide(CO2) have become the cause of global climate change. Consequently, there has been significant research activity aimed at both removing and utilizing CO2. This study assesses the potential utilization of railway tie concrete waste, generated from railway infrastructure, as a CO2 absorption material and investigates the physicochemical properties before and after CO2 absorption to understand the CO2 removal mechanisms. Railway tie concrete waste primarily consists of Si(26.60 %) and contains 9.82 % of Ca. Compared to samples of Cement and Normal concrete waste, it demonstrated superior potential for use as a CO2 absorption material, with approximately 98 % of the Ca content participating in CO2 absorption reactions. Through Thermogravimetric Analysis(TGA) and X-ray Diffraction(XRD) analysis, it was confirmed that the carbonate reaction, where the Ca in railway tie concrete waste converts into CaCO3 through reaction with CO2 gas, is the primary mechanism for CO2 removal. Furthermore, Scanning Electron Microscopy(SEM) analysis revealed the formation of numerous CaCO3 particles with sizes less than 0.1 ㎛ after the CO2 absorption reaction. This transformation of large internal voids in the CO2 absorption material into mesopores resulted in an increase in the specific surface area of the material.

Influence of Vetiver Grass (Vetiveria zizanioides) on Rhizosphere Chemistry in Long-term Contaminated Soils (중금속으로 오염된 토양에서 근권부의 화학적 특성에 미치는 vetiver grass (Vetiveria zizianioides)의 영향)

  • A detailed understanding and appreciation of the important mechanisms operating at the soil:root interface, commonly identified as the rhizosphere, is critical for evaluating the potential for particular plant species to be successfully used as part of a phytoremediation technique. For specific plants, mechanisms may exist to overcome the inherit limitation of the phytoremediation technique when poorly mobile soil metals are of interest. In the present study, the influence of root exudates on the rhizosphere chemistry of soil and consequential metal uptake were investigated following culture of vetiver grass (Vetiveria zizanioides), recognized as a promising plant for land stabilization, in three different long-term contaminated soils and one non-contaminated control soil. The soil solution pH increased (0.3-1.1 units) following vetiver grass culture and dissolved organic carbon (DOC) also significantly increased in all soils with the highest increase in PP02 (23 to $173mg\;L^{-1}$). Chemical changes are contributed to root exudation by vetiver grass when exposed to high concentration of heavy metals. Chemical changes, consequently, influenced metal (Cd, Cu, Pb, and Zn) solubility and speciation in the rhizosphere. The highest solubility was observed for soil Ko01 (eg. 2091 and $318{\mu}g\;L^{-1}$ for Cd and Pb, respectively). Initial heavy metal solubility in soils varied with soil and either increased or decreased following vetiver grass culture depending on the soil type. An increase in pH following plant culture generally resulted in a decrease in metal solubility, while elevated DOC due to root exudation resulted in an increase in metal solubility via the formation of metal-DOC complexes. Donnan speciation demonstrated a significant decrease in free Cd and Zn in the rhizosphere and the concentration of Cd, Pb, and Zn in vetiver grass shoot was highly correlated with soluble concentration rather than total soil metal concentration.

Effect of Trace Metal on Accumulation and Physiological Response of the Polychaete, Perinereis nuntia (미량금속 노출에 따른 갯지렁이(Perinereis nuntia)의 미량금속 축적 및 생리학적인 반응)

  • Metal exposure experiments using polychaete (Perinereis nuntia) as a bio-indicator of trace metals contamination were conducted to evaluate the bioaccumulation and the biomarkers responses such as metallothionein-like protein (MTLPs) and glutathione S-transferase (GST) which was simultaneously exposed to Cadmium (Cd) and Copper (Cu). Cu and Cd concentrations in polychaete were enhanced with increasing exposure time and their concentrations of aqueous medium. Initial accumulation of Cd was higher than that of Cu. Our results showed that the bioaccumulation of Cu and Cd were prohibited, especially at higher Cu levels, suggesting the different cellular uptake mechanisms when Cu and Cd are co-exist. Net accumulation rate of Cu was declined with exposure time but it did not show any significant change for Cd. Although the highest MTLPs concentration was observed at 6 hr of exposure time, it did not show any significant change related to exposure times and metals concentrations. An increase of GST activity tended to increase as a function of exposure time and metals concentrations. And GST activities in P. nuntia have similar tendency with bioconcentration factors in high concentration of Cu (treatment group IV) at post 24 h of exposure. Our results provide new information of the bioaccumulation and biomarker responses to understand the effects of co-existing contaminants (Cu and Cd) using polychaete. Further studies are required to elucidate the bioaccumulation and biomarkers responses for various contaminants.

Postprandial hypoglycemic effects of mulberry twig and root bark in vivo and in vitro (In vivo와 in vitro에서 상지 및 상백피 에탄올추출물이 식후 혈당 상승 억제 조절에 미치는 영향)

  • Purpose: Our previous study demonstrated the hypoglycemic effects of mulberry (Morus alba L.) leaf and the underlying mechanisms. Here we explored the potency of mulberry twigs (TW) and root barks (RB) in postprandial hypoglycemic effects in vitro and in vivo. Methods: The major components of TW and RB were determined by high performance liquid chromatography (HPLC). Alpha-glucosidase inhibition and glucose/fructose uptake inhibition in Caco-2 cells were determined for TW, RB, and their major components, followed by an oral sugar tolerance test (OSTT) in streptozotocin-induced diabetic rats. Male Wistar rats were fed a high-fat diet for 2 weeks and then a single dose of streptozotocin (35 mg/kg B.W) was administered by intraperitoneal injection. Rats with fasting blood glucose levels above 126 mg/dL were randomly divided into 5 groups (n = 8/group) for the following treatments by gavage for 4 weeks: vehicle (normal control and diabetic control), 200 mg/kg B.W of TW or RB or 100 mg/kg B.W of oxyresveratrol (OXY). Results: OXY and mulberroside A were identified as the major components of TW and OXY, mongolicin, and kuwanon H for RB. A significant inhibitory activity on ${\alpha}-glucosidase$ was found for TW, RB, and OXY (p = 0.0099). There was a dose-dependent inhibition of TW and RB on the intestinal sugar uptakes in Caco-2 cells, showing a greater impact on fructose compared to glucose. The OSTT showed that TW and RB significantly delayed time to maximal concentration (p = 0.0088) and decreased maximal concentration (p = 0.0043) compared to the control group. Conclusion: These results suggest that TW and RB may have a postprandial hypoglycemic effect, particularly in the case of high fructose or sucrose intake. OXY was suggested as a contributor to the hypoglycemic effect of TW and RB. Further studies are needed for the systemic effect of TW and RB in circulation.

Expressions of Norepinephrine Transporter in Pre-eclamptic Placenta (자간전증 태반에서의 Norepinephrine Transporter(NET) 발현)

  • Placenta has been shown to be a site of expression of several of the monoamine membrane uptake transporters. However, the correlation between the expressions of norepinephrine transporter (NET) and placental development including gynecological diseases is still unknown. To investigate the expression and functions of NET in placenta, we conducted to compare NET expression in normal and preeclamptic placenta and analyzed the function of NET in HTR8-SV/neo trophoblast cells after NET gene transfection. The expression of NET was analyzed in placental tissues from the following groups of patients (none underwent labor): 1) term normal placenta (n=15); 2) term with preeclamptic placeneta (n=15); and 3) pre-term with preeclamptic placenta (n=11) using semi-quantitative RT-PCR, immunohistochemistry, and Western blot. In order to evaluate the function of NET, NET gene plasmid and NET gene-specific siRNA were trnasfected into HTR-8/SVneo trophoblast cells for 24 hours. NET had low expression in the pre-eclamptic placenta compare with normal placenta but no difference in western blot data. NET was expressed in the trophoblasts, and the up-regulation of NET gene stimulated the invasion of HTR-8/SVneo trophoblast cells by 2.5 fold (p<0.05), whereas the NET-siRNA treatment reduced invasion rates. Also, we observed that the expression of NET induces to expression and activity of MMP-9 in HTR-8/SVneo trophoblast cells in zymography. The results suggest that the expression of NET were reduced in pre-eclampsia and should be inhibited invasion activity of trophoblasts. Therefore, these findings provide useful guidelines for the mechanisms of trophoblast invasion as well as for the basic understanding of gynecological diseases including pre-eclampsia.

Effects of Mifepristone and Tamoxifen on Calcium Modulation in DU-145 Prostate Cancer Cells (DU-145 전립선 암세포에 있어서 mifepristone과 tamoxifen이 칼슘조절에 미치는 영향)

  • Mifepristone (MIF) and Tamoxifen (TAM) have been used in the treatment of prostate cancer and breast cancer for more than a decade. MIF can induce apoptosis in both AR-positive and negative prostate cancer cells. Because of its pleiotropic ligand-receptor properties, TAM exerts cytotoxic activity in estrogen (ER)-positive and various ER.negative cancer cells. However, the molecular mechanisms of these two substances are not yet clear. In the present work, we report that the cytotoxic effects of MIF and TAM are due to the modulation of intracellular $Ca^{2+}$ level in DU-145, androgen-insensitive cells. When the cells were treated with micromolar concentrations of either MIF or TAM, the growth and viability were significantly decreased in a dose- and time-dependent manner. The apoptosis induced by MIF or TAM was further proved and analyzed by confocal laser scanning microscopy (CLSM) and fluorescence-activated cell sorting (FACS). In the cells cultivated in a normal 1.5 mM $Ca^{2+}$ medium, both MIF and TAM also induced an increase of the intracellular $Ca^{2+}$ level in a dose-dependent fashion. Since a change in calcium level could not be found in cells of the $Ca^{2+}$-free medium, the increase of intracellular $Ca^{2+}$ level might be due to an increase in extracellular calcium uptake. Our results show that the apoptotic effect was more prominent in TAM treatment compared to MIF treatment in DU-145 cells. The above findings might be due to the difference in the uppermost pathways of apoptosis induced by either MIF or TAM. When we checked the level of procaspase-8 activation, TAM showed minor level of activation, as opposed to MIF, which exerted strong activation. In both treatments, the levels of anti-apoptotic protein Bcl-2 decreased, and pro-apoptotic protein Bax level increased more than 2-fold. The activation of caspase-3, a key protease enzyme in the downstream pathway of apoptosis, was much higher in the cells treated with TAM, compared to the MIF treatment. The overall apoptotic activity shown in the present work was closely related to intracellular $Ca^{2+}$ concentration levels. Therefore, the cytotoxic activity induced by MIF and TAM might have been due to intracellular calcium modulation.

Effect of McN-A-343, Clonidine and Cocaine on the Contractile Response of Rat and Rabbit Vas Deferens to Field Stimulation (McN-A-343, Clonidine 및 Cocaine의 백서(白鼠)와 가토정관(家兎精管)의 자극효과(刺戟效果)에 대(對)한 영향(影響))

  • 1) Effects of McN-A-343, clonidine and cocaine on the contractile response of the rat and rabbit vas deferens to field stimulation were investigated. The action mechanisms of these drugs have shown to be associated with endogenous norepinephrine(NE). 2) The contractile response to the stimulation of 5-10 Hz was markedly inhibited by the cumulative doses of McN-A-343 (0.003, 0.03, $0.3{\mu}g/ml$), clonidine (0.003, $0.3{\mu}g/ml$) and cocaine $(0.03\;{\mu}g/ml)$. The inhibition was antagonized by yohimbine and pjperoxan. 3) The inhibitory effect of McN-A-343 $(0.03{\mu}g/ml)$ and cocaine $(0.03{\mu}g/ml)$ was markedly enhanced by the same dose of cocaine and McN-A-343, respectively. This enhanced inhibition was also antagonized by yohimbine. 4) The contractile response to the stimulation of 0.01 Hz and 5-10 Hz was markedly potentiated by comparatively large doses of McN-A-343 $(30\;{\mu}g/ml)$ and $(3\;{\mu}g/ml)$. This potentiation was not observed in the presence of thymoxamine. The potentiation by McN-A-343 also did not appear in the presence of atropine. 5) The contractile response to the above stimulation was potentiated by muscarine and the potentiation was markedly attenuated in the presence of thymoxamine and atropine.

Change of Nutrients and Behaviour of Total Coliforms in the Natural Treatment of Wastewater by Subsurface Flow Wetland System (인공습지를 이용한 자연정화 오수처리시설에서 영양물질의 변화와 대장균군의 행동)

  • The constructed wetland system which is applicable to rural wastewater treatment was examined by pilot plant experiment. Removal rates of nutrients including nitrogen and phosphorus and total coliform were evaluated. The $NH_4\;^+$ concentration of the influent was in the range of 91.57 to 275.88mg/l and the effluent concentration was about 40% lower than the influent. The decreasing of the $NH_4\;^+$ concentration might be due to volatilization, plant uptake, adsorption onto soil particles, and mainly nitrification. However, generally concentrations of $NO_2\;^-$ and $NO_3\;^-$ were increased in the effluents compared to the influent concentrations, which implies that nitrogen components in the system were nitrified. Overall, the average removal rate of the nitrogen was about 5% which seems inadequate as a wastewater treatment system, and this system needs improvement on nitrogen removal mechamism. The removal rate of the phosphorus was quite high and effluent concentration was very low. Reason for high removal rate of the phosphorus might be mainly strong adsorption characteristic onto soil particles. The average removal rate of the total coliforms was about 83%, and main removal mechanisms are thought to be adsorption onto soil and inability to compete against the established soil microflora. From the results of the study, the constructed wetland system needs to be improved in nitrogen removal mechanism for field application.

Mapping the Research Landscape of Wastewater Treatment Wetlands: A Bibliometric Analysis and Comprehensive Review (폐수 처리 위한 습지의 연구 환경 매핑: 서지학적 분석 및 종합 검토)

  • Constructed wetlands (CWs) are effective technologies for urban wastewater management, utilizing natural physico-chemical and biological processes to remove pollutants. This study employed a bibliometric analysis approach to investigate the progress and future research trends in the field of CWs. A comprehensive review of 100 most-recently published and open-access articles was performed to analyze the performance of CWs in treating wastewater. Spain, China, Italy, and the United States were among the most productive countries in terms of the number of published papers. The most frequently used keywords in publications include water quality (n=19), phytoremediation (n=13), stormwater (n=11), and phosphorus (n=11), suggesting that the efficiency of CWs in improving water quality and removal of nutrients were widely investigated. Among the different types of CWs reviewed, hybrid CWs exhibited the highest removal efficiencies for BOD (88.67%) and TSS (95.67%), whereas VSSF, and HSSF systems also showed high TSS removal efficiencies (83.25%, and 78.83% respectively). VSSF wetland displayed the highest COD removal efficiency (71.82%). Generally, physical processes (e.g., sedimentation, filtration, adsorption) and biological mechanisms (i.e., biodegradation) contributed to the high removal efficiency of TSS, BOD, and COD in CW systems. The hybrid CW system demonstrated highest TN removal efficiency (60.78%) by integrating multiple treatment processes, including aerobic and anaerobic conditions, various vegetation types, and different media configurations, which enhanced microbial activity and allowed for comprehensive nitrogen compound removal. The FWS system showed the highest TP removal efficiency (54.50%) due to combined process of settling sediment-bound phosphorus and plant uptake. Phragmites, Cyperus, Iris, and Typha were commonly used in CWs due to their superior phytoremediation capabilities. The study emphasized the potential of CWs as sustainable alternatives for wastewater management, particularly in urban areas.
