Effects of Dietary Thiazolidinedione Supplementation on Growth Performance, Intramuscular Fat and Related Genes mRNA Abundance in the Longissimus Dorsi Muscle of Finishing Pigs

  • The objective of this study was to investigate the effect of dietary supplementation with thiazolidinedione (TZD) on growth performance and meat quality of finishing pigs. In Experiment 1, 80 castrated finishing pigs (Large White${\times}$Landrace, BW = 54.34 kg) were randomly assigned to 2 treatments with 5 replicates of 8 pigs each. The experimental pigs in the 2 groups were respectively fed with a diet with or without a TZD supplementation (15 mg/kg). In Experiment 2, 80 castrated finishing pigs (Large White${\times}$Landrace, BW = 71.46 kg) were divided into 2 treatments as designed in Experiment 1, moreover, carcass evaluations were performed. The results from Experiment 1 showed that TZD supplementation could significantly decreased the average daily feed intake (ADFI) (p<0.05) during 0 to 28 d, without impairing the average daily gain (ADG) (p>0.05). In Experiment 2, the ADG was significantly increased by TZD supplementation during 14 to 28 d and 0 to 28 d (p<0.05) and the feed:gain ratio (F:G) was significantly decreased by TZD supplementation during 0 to 28 d (p<0.05). Compared with the control group, TZD group had significantly higher serum triglyceride (TG) concentration at 28h and serum high-density lipoprotein (HDL) levels at 14 d (p<0.05). Moreover, there was an apparent improvement in the marbling score (p<0.10) and intramuscular fat (IMF) content (p<0.10) of the longissimus dorsi muscle in pigs treated by TZD supplementation. Real-time RT-PCR analyses demonstrated that pigs of TZD group had higher mRNA abundance of $PPAR{\gamma}$ coactivator 1 (PGC-1) (p<0.05) and fatty acid-binding protein 3 (FABP3) (p<0.05) than pigs of control group. Taken together, these results suggested that dietary TZD supplementation could improve growth performance and increase the IMF content of finishing pigs through regulating the serum parameters and genes mRNA abundance involved in fat metabolism.

경제적인 조피볼락용 배합사료 설계 및 평가 (Evaluation of Economical Feed Formulations for Korean Rockfish (Sebastes schlegeli))

  • 여러 가지 종류의 단백질 원료를 혼합하여 성장이 좋고, 가격이 싼 조피볼락용 배합사료를 개발하기 위해 설계한 사료의 효과를 사육실험을 통해 조사하였다. 북양어분 간접식 $29\%$와 갈색 어분 $29\%$ 첨가구를 대조구로 정하고 원료의 필수아미노산 조성을 고려하여 대두박, 콘글루텐 밀, 육분, 혈분, 우모분, 소맥분을 각각 다른 비율로 첨가하여 어분 단백질원인 북양어분과 갈색어분을 $40\%$에서 $65\%$까지 대체한 사료 6종류와 RMP(생사료+분말사료)구를 선정하여 총 8종류의 실험사료를 제조하였다. 33 g 전후의 조피볼락을 실험사료마다 각각 3반복으로 15주간 사육 실험한 결과, 증체율은 사료 2, 3, 4구와 RMP 사료가 대조구와 유의차없이 좋은 결과를 보였다(P>0.05). 반면에 사료효율은 RMP구가 가장 높았고, 사료 7이 가장 낮았다. 어체 1 kg 증육시키는데 소요되는 사료원가는 RMP구가 1,332원으로 가장 높았고, 대조구가 539원으로 나타났으며, 그 외 사료들은 $452\~498$원으로 비슷하였다. 위와 같이 사료 $5\~7$을 제외한 사료들은 성장효과가 대조구와 유의차가 없어 30 g 이상의 어체에서는 식물성 및 동물성 원료를 적절히 혼합 첨가하면 사료중의 어분을 $55\%$까지 대체 가능하였으며, 양식 생산비를 절감할 수 있을 것으로 나타났다.

  • 동물성 식품의 생산을 증가시키기 위하여 다양한 종류의 ehdafn성 약물이 사용되고 있고 공업의 발달과 더불어 각종의 공해물질이 수질과 토양을 오염시키고 있다. 따라서 이와 같은 화학물질이 직접 혹은 간접적으로 동물성 식품을 오염시키거나 식품에 잔류할 가능성이 높아지고 있다. 이에 따라 농림수산부와 보건 사회부에서는 약제 잔류를 방지하기 위하여 잔류 허용기준을 따로이 마련하였고 독자적으로 잔류에 대한 조사연구에 착수하였다. 잔류 약제에 대한 규제는 국민건강을 지킨다는 의미에서 착실히 추진되어야 할 것이나 생산기반이 흔들리지 않는 범위 내에서 점진적으로 진행되어야 할 것이고, 국내에서의 동물성 식품의 안전성 확보를 통하여 수출을 확대하고 무분별한 수입을 억제할 수 있도록 이원화되어 있는 업무를 일원화되어야 한다. 검사를 통하여 잔류를 방지하고자 하는 정책 보다는 생산 단계에서 안전휴약기간을 지키도록 적극 교육, 홍보, 지도하여야 한다. 이 심포지엄 연제에서는 미국과 한국 농림수산부에서는 잔류검사기법에 대한 개요와 검사 계획, 국내에서의 잔류물질에 대한 조사연구 개황, 그리고 허용기준 설정방법 등에 대하여 논의한다.

Effects of Amino Acid-enriched Ruminally Protected Fatty Acids on Plasma Metabolites, Growth Performance and Carcass Characteristics of Hanwoo Steers

  • This study was conducted to determine the effects of amino acid-enriched ruminally protected fatty acid (AARPFA) on plasma fatty acids and amino acids, growth performance and carcass characteristics of Korean native steers (Hanwoo) by simultaneous supply of fatty acids and limiting amino acids (methionine and lysine). Eighteen finishing Hanwoo steers, 18 months of age and weighing an average of $459.0{\pm}38.9\;kg$, were used for studies of the metabolism of plasma fatty acids and amino acids during supplementation of AARPFA. Also, 45 finishing Hanwoo steers, 16 months of age and weighing an average of $408.6{\pm}26.5\;kg$, were used for growth performance and carcass characteristics during supplemention of AARPFA. There were three treatments which comprised a basal diet supplemented with AARPFA at 0 g (T1), 50 g (T2) or 100 g (T3), respectively. Concentrations of saturated, unsaturated and total fatty acids in plasma were increased in T3 compared with other treatments (p<0.05). Concentrations of methionine and lysine in plasma were linearly increased with increasing levels of AARPFA (p<0.01). Average daily gain, dry matter intake and feed conversion ratio were not different among the treatments. Marbling score measured by ultra-sound scanning was higher in T3 than in T1 at 24 months of age (p<0.05). Rib eye area, back fat thickness, yield index and yield grade score were similar across the treatments. Marbling score and quality grade score were higher in T3 compared with other treatments (p<0.01). Thus, plasma fatty acids, methionine and lysine metabolism were affected by supplementing with 100 g of AARPFA which also had positive effects on marbling score and meat quality grade of finishing Hanwoo steers.

Replacement value of cassava for maize in broiler chicken diets supplemented with enzymes

  • Objective: Pellet durability, particle size distribution, growth response, tibia bone characteristics and energy retention were measured to evaluate cassava as an alternative energy source to replace maize in broiler diets with or without Ronozyme (A+VP) enzyme composites. Methods: A total of 480 one-day broiler chicks were randomly assigned to 8 treatments in a 4×2 factorial arrangement. Four levels of cassava: (0%, 25%, 50%, 75%) and 2 levels of enzymes (0 and 500 g/tonne) were used. Each treatment was replicated six times, with ten birds per replicate. Results: The particle size distribution in the diets showed an increasing trend of small particles with increase in cassava level. Pellet durability decreased (p<0.05) with cassava inclusion. Feed intake was highest in birds fed diets with medium cassava level at 1 to 24 d and 1 to 35 d of age. The body weight gain of birds reduced (p<0.037) as cassava level increased, but it increased (p<0.017 when enzymes were added. The feed conversion ratio was high (p<0.05) when cassava level was increased, but it reduced (p<0.05) when enzymes were added. The dressing percentage (DP), and weight of drumsticks reduced (p<0.05) with increasing cassava level. Enzyme supplementation increased (p<0.05) DP, and weight of breast, thighs and drumsticks. Ash content, weight, length, width, and bone strength decreased (p<0.05) when cassava level was increased, however, they were increased with enzyme addition. The contents of Ca, K, and Zn were raised (p<0.001) with increasing cassava level. Enzyme inclusion increased (p<0.001) all mineral contents in tibia bones. Body fat and energy retained as fat decreased (p<0.001) as cassava level increased. Enzyme inclusion increased (p<0.05) body protein content and energy retained as protein. Conclusion: Although broiler performance was depressed by high levels of cassava inclusion, it was not affected by low levels, which further improved by enzyme supplementation.

  • Development process of agricultural technology has been studied with a case study of Korean agriculture. Technological is considered as a transformer of inputs into outputs and hence technological appropriateness, an important aspect of agricultural development strategies, is considered as a dynamic concepts. Considering the concept of agricultural system as a delivery system for providing essential materials and services to producers and consumers, it has been divided into two major groups of dimensions vis. external challenge dimensions and internal response dimensions. Market, investment and agro-ecosystem constitute the external challenge dimensions : whereas trade , technology as well as production and resources allocation constitute internal response dimensions. The system manager is responsible for maintaining equilibrium in the mentioned six sub-systems. Two kinds of alternatives paths of technological development viz. land saving technology and labour saving technolog have been studied. Technology is considered as a combination of four basic components viz. facilities, abilities, facts and frameworks. Adoption of innovation in agriculture depends on profitability, awareness, risk aversion, financial capacity, institutional infrastructure, availability of physical inputs and adaptability to the local conditions. For a cast study of Korea, changes in the agricultural system through external challenge dimensions are investigated. The impacts of industrialization on agro-ecosystem reported are shift of labour from the agricultural sector to non-agricultural sectors and continuously increasing demand of farm the agricultural sector to non-agricultural sectors accompanied by increase in land prices. The impacts on the commodity market discussed are shift in demand from rice, barley and other cereals to meat , dairy products and vegetables : and increasing in supply capacity of agricultural inputs. The process of agricultural development from 1962 to 19 1 9 (i.e. from start of the first to the end of the sixth five year plan) are also discussed in details with several policy measures taken. The trend of agricultural income and productivity are also analyzed. The main cause of increase in the agricultural income is considered as increase in labour productivity. The study revealed that during the span of 1965-88, holding size has not changed significantly, but both the land and labour productivity increased and so did the agricultural income. R&D activities in Korea have changed over time in three stages vix. import of improved technology, localization by adaptive research and technological mastery. For the new technology to be made affordable to farmers, policy measures like fertilizer and food grain exchange system, dual price system in rice and barely and loan for machinery were strengthened.

Genetic diversity of Halla horses using microsatellite markers

  • Background: Currently about 26,000 horses are breeding in Korea and 57.2% (14,776 horses) of them are breeding in Jeju island. According to the statistics published in 2010, the horses breeding in Jeju island are subdivided into Jeju horse (6.1%), Thoroughbred (18.8%) and Halla horse (75.1%). Halla horses are defined as a crossbreed between Jeju and Thoroughbred horses and are used for horse racing, horse riding and horse meat production. However, little research has been conducted on Halla horses because of the perception of crossbreed and people's weighted interest toward Jeju horses. Method: Using 17 Microsatellite (MS) Markers recommended by International Society for Animal Genetics (ISAG), genomic DNAs were extracted from the hair roots of 3,880 Halla horses breeding in Korea and genetic diversity was identified by genotyping after PCR was performed. Results and conclusion: In average, 10.41 alleles (from 6 alleles in HTG7 to 17 alleles in ASB17) were identified after the analysis using 17 MS Markers. The mean value of $H_{obs}$ was 0.749 with a range from 0.612(HMS1) to 0. 857(ASB2). Also, it was found that $H_{\exp}$ and PIC values were lowest in HMS1 (0.607 and 0.548, respectively), and highest in LEX3(0.859 and 0.843, respectively), and the mean value of $H_{\exp}$ was 0.760 and that of PIC was 0.728. 17 MS markers used in this studies were considered as appropriate markers for the polymorphism analysis of Halla horses. The frequency for the appearance of identical individuals was $5.90{\times}10^{-20}$ when assumed as random mating population and when assumed as half-sib and full-sib population, frequencies were $4.08{\times}10^{-15}$ and $3.56{\times}10^{-8}$, respectively. Based on these results, the 17 MS markers can be used adequately for the Individual Identification and Parentage Verification of Halla horses. Remarkably, allele M and Q of ASB23 marker, G of HMS2 marker, H and L of HTG6 marker, L of HTG7 marker, E of LEX3 marker were the specific alleles unique to Halla horses.

1978년 진해만 적조와 양식굴의 대량폐사 (MASS MORTALITYS OF OYSTER DUE TO RED TIDE IN JINHAE BAY IN 1978)

  • 1978년 진해만에서 있었던 적조현상과 양식 굴의 대량폐사에 관한 조사결과를 요약하면 아래와 같다. 1. 진해만에서 1978넌 9월중 양식 굴의 대량폐사가 있었다. 폐사량은 $96\%$인 5,687M/T으로 피해액은 22억 7천 5백만원이었다. 2. 상기의 대량폐사는 지속된 고온, 대규모의 적조와 그로 인한 저층수의 빈산소 내지 무산소화로 인한 질식이 주 원인의 일보였다고 추측된다. 3. 적조의 원인종은 와편모조의 일종인 Ceratium fusus로서 각장 $340{\mu}m$, 각폭 $20{\mu}m$ 정도의 대형 식물플랑크톤이었다. 클로로필-a량은 최고 $50mg/m^3$ 이었다.4. 춘기의 가뭄과 6월의 다우로 성층이 형성된데다 지속된 고온과 장일조 그리고 해상의 평온함이 대규모의 적조를 유발시킨 원인의 일부라 생각된다.

넙치(Paralichthys olivaceus) Ball 제품의 제조 및 품질 특성 (Processing and Quality Properties of Olive Flounder Paralichthys olivaceus Balls Product)

  • Olive flounder Paralichthys olivaceus production has increased gradually in recent years, but prices have fallen. Thus, the development of a variety of processed foods incorporating olive flounder would help to increase the income of fishermen. This study was conducted to investigate the best method for olive flounder ball processing. Clean olive flounder were divided into five portions. Olive flounder meat (100 g with added egg white 39 g) was chopped and then mixed with 10 mL fresh cream and ingredients. The dough was molded into the shape of a ball. The olive flounder balls were then processed by two different methods. In the first method, the flounder ball was boiled in water for 3 min then vacuum-packed in polyethylene film and stored at $-20^{\circ}C$ for 7 days. After 7 days, the ball was thawed and heated in a microwave for 2 min (Sample-1). In the second method, the ball was vacuum-packed in polyethylene film without boiling and then stored at $-20^{\circ}C$ for 7 days before thawing and boiling in water for 3 min (Sample-2). After heating, both types of olive flounder balls were evaluated. Various factors (including the viable bacterial count, chemical composition, pH, hardness, thiobarbituric acid level, salinity, and free amino acid content) were measured, and a sensory evaluation was conducted. Based on the results of the sensory and hardness evaluations, Sample-1 was deemed to be superior to Sample-2.

Combined Effects of High Pressure and Heat on Shear Value and Histological Characteristics of Bovine Skeletal Muscle

  • Changes in shear force value, transverse sections, myofibrils and intramuscular connective tissue of bovine skeletal muscle exposed to the combination of high-pressure up to 400 MPa and heat (30 and $60^{\circ}C$) were studied. The shear force value decreased by pressure-heat treatment up to 200 MPa at 30 and $60^{\circ}C$, and then slightly increased over 200 MPa at $30^{\circ}C$. Shear force values of treated muscles were lower than those of untreated ones. Gaps between muscle fibers in the untreated muscle were a little clear, and then they became very clear in the treated muscles up to 200 MPa at 30 and $60^{\circ}C$. However, the gaps reduced significantly over 200 MPa at $30^{\circ}C$. The remarkable rupture of I-band and loss of M-line materials progressed in the myofibrils with increasing pressure applied. However, degradation and loss of the Z-line in myofibrils observed in the muscle treated at $60^{\circ}C$ was not apparent in the muscle treated at $30^{\circ}C$. The length of the sarcomere initially contracted by pressure-heat treatment of 100 MPa at $30^{\circ}C$ seemed to have recovered with increase of the pressure up to 400 MPa. In the muscle treated at $60^{\circ}C$, the length of sarcomere gradually decreased with increase of the pressure up to 400 MPa. In the treated muscles, changes in the honeycomb-like structure of endomysium were observed and accelerated with increase of the pressure. A wavy appearance clearly observed at the inside surface of endomysium in the untreated muscles gradually decreased in the treated muscles with increase of the pressure. Tearing of the membrane was observed in the muscles treated over 150 MPa at $30^{\circ}C$, as observed in the sample pressurized at 100 MPa at $60^{\circ}C$. The roughening, disruption and fraying of the membrane were observed over 200 MPa at $60^{\circ}C$. From the results obtained, the combination of high-pressure and heat treatments seems to be effective to tenderize tough meat. The shear force value may have some relationship with deformation of intramuscular connective tissue and myofibrils.
