Enhancement in the photocurrent of ZnO nanoparticles by thermal annealing

  • The optoelectrical characteristics of the ZnO nanoparticles (NPs) annealed in vacuum or oxygen condition from $200^{\circ}C$ to $600^{\circ}C$ were examined. Increased on-off ratio (or, the ratio of photocurrent to dark current) was observed when they were annealed at $300^{\circ}C$, $400^{\circ}C$ and $500^{\circ}C$ with the values enhanced about 4 orders compared to the as-prepared ZnO NPs in both annealing conditions, while the maximum efficiency was shown at the annealing temperature of $600^{\circ}C$ for the ZnO NPs annealed in vacuum with the value of 29.8 mA/W and at the temperature of $500^{\circ}C$ for those annealed in oxygen condition with the value of 40.3 mA/W. Photoresponse behavior of the ZnO NPs annealed in oxygen showed the sharp increase right after the ir exposure to the light followed by the slow decay and saturation during steady illumination, differing from the ZnO NPs annealed in vacuum which only exhibited the gradual increase. This difference occurred due to the curing effect of the oxygen vacancies. SEM images indicated no change in their morphologies with annealing, indicating the change in their internal structures by annealing, and most remarkably at $600^{\circ}C$. As for their photoluminescence(PL) spectra, the decrease of the deep-level(DL) emission was observed when they were annealed in oxygen at $400^{\circ}C$, and not at $200^{\circ}C$ and $600^{\circ}C$.

Kinetic Studies on Submerged Acetic Acid Fermentation of Acetobacter aceti (Acetobacter aceti균의 심부배양에 의한 초산발효의 동력학적 연구)

  • In order to determine the optimal conditions for the process of acetic acid fermentation, the kinetics of Acetobacter aceti fermentation in submerged batch cultures were studied at different agitation speeds and air flow rates. The maximum cell concentration was noted after about 48 hr fermentation and the time course of Acetobactey aceti fermentation showed a distinct feature of growth-associated product formation. At agitation speeds 700, 500, and 300 rpm fixed on air flow rate 1 v/v/M, specific grow rates were $3.97\times10^{-2},\;3.82\times10^{-2},\;and\;2.04\times10^{-2\} \;hr^{-1}$, saturation constants were 61.4, 64.6, and 69.4mg/ml. and volumetric oxygen transfer coefficients were 0.9337, 0.4468, and 0.1701 $min^{-1},$ respectively. At air flow rates 1.25, 1.00, and 0.75 v/v/M fixed on agitation speed 500 rpm, specific growth rates were $3.90\times10^{-2},\;3.82\times10^{-2},\;and\;2.37\times10^{-2}\;hr^{-1}$, saturation constants were 63.4, 64.6, and 64.9 mg/ml, and volumetric oxygen transfer coefficient were 0.4923, 0.4468, and 0.3509 $min^{-1},$ respectively.

Effect of seeding ratio on acidogenic biokinetics in high ammonia concentration

  • Anaerobic digestion is one of the well-known methods for biological treatment handling of concentrated organic matter such as swine $wastewater.^{1)} The anaerobic digestion can reduce organic loading but also hydrolyze non-biodegradable organic $matter.^{2)}$ The feces from the scrapper-type barn are usually collected to make compost and the urine is discarded with swine-slurry wastewater by ocean-dumping or treated by biological methods. The lagoon, aerobic digestion, anaerobic digestion, SBR, $A^{2}/O$, and UCT have been applied for treating swine $wastewater.^{3)} In this study, as a result of the analysis of swine wastewater, the total and soluble chemical oxygen demand was 130g/L and 60g/L, respectively. And the volatile fatty acid as chemical oxygen demand equivalent was 45g/L, which was 75% of soluble chemical oxygen demand. Before everything else, ammonia nitrogen concentration was 6.5 g/L. From biochemical acidogenic potential test, it was concluded that the enhanced acidification process to manage swine waste should be operated in the ammonia nitrogen concentration of less than 1.2 g/L. In the result of seeding ratio experiments with artificial $wastewater^{4)}, the lag period of acidogens was taken the long time because of the inhibition by the $ammonia^{5)}$, however no difference of period by the seeding ratio was not shown. The Haldane-based biokinetics were also evaluated using a method of fourth order Runge-Kutta $approximation.^{6,7)}$ The nonlinear least squares (NLLS) method with a 95% confidence interval was also used. The ranges of maximum microbial growth rate, ${/mu_{max}}$, and half saturation coefficient, $K_{s}$, for acidogenesis of various seeding ratio with artificial wastewater were 6.1 ~ 12.6 $d^{-1}$ and 45,000 ~ 53,500 mg glucose/L, respectively. Also, the methanogenic microbial yield coefficient, Y, and microbial decay rate coefficient, $k_{d}$, and inhibition substrate concentration, $K_{si}$, for the reactors were determined to be 0.32 ~ 0.465 ${/mu}g$/mg glucose; 0.42 ~ 1.01 $d^{-1}$ and 51,500 ~ 55,600 mg glucose/L, respectively.

Optimization of Induction Conditions for Bacillus-derived Esterase Production by High-cell Density Fermentation of Recombinant Escherichia coli (재조합 대장균의 고농도 배양과 유도조건 최적화를 통한 Bacillus 유래 esterase의 생산)

  • To increase the efficiency of esterase production by Bacillus, high cell-density culture of recombinant Escherichia coli through fed batch fermentation was tested. Cells were cultured to $OD_{600}$ of 76 (35.8 g/l DCW) with dissolved oxygen level controlled to least above 30% air saturation by supplying pure oxygen. Cells were cultured to an $OD_{600}$ of 90 (42.4 g/l DCW) with glucose feeding controlled to at least 1 g/l. However, the cells reached stationary phase at the late stage of culture, despite glucose being supplied. Cells were cultured to an $OD_{600}$ of 185 (87.3 g/l DCW) by supplying additional medium with fortified yeast extract. To increase the productivity of the recombinant protein, cell growth and esterase productivity based on induction time were evaluated. Late exponential phase induction for esterase production in fed batch fermentation resulted in maximum optical density $OD_{600}$ of 190 (89 g/l DCW) and maximum esterase activity of 1745 U/l, corresponding to a 5.8-fold enhancement in esterase production, compared to the early exponential phase induction. In this study, we established fermentation methods for achieving maximum production of Bacillus-derived esterase by optimizing IPTG induction time in high-cell density culture by supplying pure oxygen and a nitrogen source.

Seasonal Variations in Primary Productivity and Pigments of Downstream Water of the Han River (한강하류수역의 기초생산과 식물플랭크톤 색소량의 년변화)

  • This study was undertaken to assess the annual cycle of primary production and plant pigments in a downstream of the Han River. Measurements were carried out at three week intervals during April 1966 and March 1967, and ancillary data include water temperature, transparency, pH, dissolved oxygen and phytoplankton cell number. The seasonal cycle in water temperature profile shows the hihgest in the end of August with 27$^{\circ}C$, lowest in the middle of February with 0.2$^{\circ}C$. The transparency with Secchi disk reading varied from a maximum 4.0m in fall and a minimum 0.5m or less in early spring and flood season of summer. The pH of the river water varied from 6.5 to 7.3, averaged 6.91 in the surface water and 6.98 in the bottom water, showed little seasonalvariability. The dissolved oxygen in the surface water ranged from 5.93-9.64ml/L, while in the bottom water it ranged from 5.54-9.72 ml/L, and the oxygen saturation never fall below 94%. None thermal, the distribution of pH and content of oxygen, stratifications occurred. An apparent seasonal cycle of primary productivity was observed with remarkably high levels in the spring and fall, the lowest level in the winter. The range of net carbon assimilations showed 3.1-112.6 mgC/㎥/day or 15-427 mgC/㎡/day in spring, 37.0-271.2 mgC/㎥/day or 115-329 mgC/㎡/day in summer, 27.2-168.0 mgC/㎥ /day or 139-415 mgC/㎡/day in fall and 0.5-10.9 mgC/㎥/day or 5-19 mg/㎡/day in winter, respectively. Amount of chlorophyll ${\alpha}$ ranged from a minimum concentration of 0.2-0.3 mg/㎥ in the middle of February and a maximum 4.1-6.7 mg/㎥ in the middle of June. A general increase trend in chlorophyll ${\alpha}$ concentration was noted with increase of the river water temperature.

  • In order to predict the performance of biological wastewater treatment plant, the kinetic parameters and stoichiometric coefficient must be known. The theories and experimental procedures for determining the biological kinetic parameters were discussed in this study. Respirometric analysis in the batch reactor was carried out for the experimental assessment of kinetic parameters. A simple procedure to estimate kinetic parameters of heterotrophs and autotrophs under aerobic condition was presented. The difficulties in the interpretation of COD and VSS measurements encouraged the conversion of respirometric data to growth data. Maximum specific growth rate, yield coefficient, half saturation constant and decay rate of heterotrophic biomass were obtained from OUR(Oxygen Uptake Rate) data. Maximum specific growth rate of autotrophic biomass was obtained from the increase of nitrate concentration. The aim of this paper is to estimate the kinetic parameters of heterotrophic and autotrophic biomass by means of the respirometric analysis of activated sludge behavior in the batch reactors. These procedures may be used for the activated sludge modeling with complex kinetic parameters.

  • This research is to investigate the reaction kinetics by air-lift bioreactor using calcium hydroxide, the neutralization agent and immobilization media, for removing ethylene glycol remained after chemical pretreatment. It was found that the optimum hydraulic retention time was obtained as 24.2hours at the optimum F/M ratio of 1.32kg-$TCOD_{Mn}$/day.kg-MLVSS, and then, infiuent $TCOD_{Mn}$ and MLVSS concentration were 3,290mg/l and 2,472mg/l, respectively. During the steady state, the kinetics constants such as maximum specific substrate removal rate, half saturation velocity coefficient, yield coefficient and endogenous respiration coefficient were estimated in the base of $TCOD_{Mn}$ as substrate concentration. And they were 1.47day$^{-1}$, 3.95mg/l, 0.391 and 0.092day$^{-1}$, respectively. And also, the oxgen use coefficients for cell synthesis, a', and energy of maintenance, b', were obtained as 0.4kg-O$_{2}$/kg-$TCOD_{Mn}$ and 0.056day$^{-1}$, at the steady state by the experimental result of oxygen uptake rate.

  • Plasma polymerized thin films were manufactured inter-electrode coupled plasma polymerization apparatus. The deposition rate reached its maximum between 40[W] and 100[W]. In the ESCA analysis, peaks revealing -CH2, -CH, -C- were present at 285.4 and 285.5[eV] respectively. The C=O peak at 532.8[eV] and the C-O peak at 533.8[eV], which were grouped with an unignorable amount of oxygen were conformed. In ESR analysis, the curve revealing strong amplification was in saturation, which was affected by weak power. This is considered as a -CH-Ch=Ch- structure containing the Allyl group. The relative permittivity of the plasma polymerized thin films was about 3.5 at a frequency of 100[Hz]∼200[kHz]. The dissipation factor showed allow value of 0.008.

Intranasal administration of dexmedetomidine (DEX) as a premedication for pediatric patients undergoing general anesthesia for dental treatment

  • Background: The most important reason for pre-operative administration of medication is to reduce anxiety. Alleviation of fear and anxiety about surgery enables patients to remain comfortable during treatment. Dexmedetomidine (DEX) is a fast-acting drug that is used as a premedication in different circumstances because it has sedative and anti-anxiolytic effects, and stable hemodynamics. It also has the advantage of intranasal administration. The aim of this study was to investigate the effects and hemodynamic stability of DEX by retrospectively analyzing cases in which DEX was administered nasally as a premedication. Methods: Ten patients treated at Dankook University Dental Hospital, recruited between February and April 2015, received intranasal delivery of $2{\mu}g/kg$ DEX, 30 minutes prior to general anesthesia. Anesthesia records of anxiety, blood pressure, respiration, pulse, estimated arterial oxygen saturation ($SpO_2$), and partial pressure, or maximum concentration, of carbon dioxide ($ETCO_2$) were analyzed. Results: Administration of DEX prior to a general anesthetic effectively relieved anxiety. Respiratory depression, the most severe adverse effect of other sedatives, was not observed. Hemodynamic stability under general anesthesia was maintained during treatment and a reduction in emergence delirium was observed upon completion of treatment. Conclusions: Premedication administration of DEX is safe for pediatric patients undergoing dental treatment under general anesthesia.

MICROENVIRONMENT IN OYSTER FARM AREA 1. On the Eutrophication and Raft Density in Geoje Bay (굴 양식장의 미세환경에 관한 연구 1. 거제만의 양식장밀도 및 부영양화에 관하여)

  • After a great mortality owing to abnormal oceanographical condition and fungal disease in 1973 hanging cultch lines per raft has been reduced to 450 from 558, and oyster production per raft has also been decreased. It seems to be result of dense culture of oysters and its resulting accumulation of waste materials on the sea bottom in the farm area. The present study was carried out to investigate the effect of rearing density and the degree of eutrophication in Geoje Bay for 6 months from June through November in 1977. Total area of this bay is about $48.9\;km^2$, and the area of registered oyster farms as of November in 1977 is around $10.9\;km^2$, which is about $22.3\%$ of this bay. Water Quality during summer season was shown as transparency 5.5 m, COD 1.5ppm, degree of oxygen saturation $90.6\%$ in upper and middle layers and $82.2\%$ near bottom, chlorophyll-a 3.0 mg/m^3, and phytoplankton $8.7\times10^4\;cells/l$. In superficial mud in August COD was 35.4 mg/g, total sulphide 0.24 mg/g, and phaeophytin $43.7\;{\mu}g/g$dry mud. These values indicate that water qualify in this bay is so far excellent but quantities of chemical oxygen demend and sulphide in bottom mud show maximum level or a little over eutrophication standard.
