Effects of Concentration of Inhibitor on the Production of $\alpha$-amylase and Growth of Bacillus amyloliquefaciens (Bacillus amyloliquefaciens 성장과 $\alpha$-amylase 생합성에 미치는 저해제의 영향)

  • In this study, Bucillus amyloliquefaciens was adopted as bacterial source to investigate the concentration of carbon source by adding inhibitors in the batch culture. By adding acetic acid at $10g/\ell$ of initial glucose concentration, maximum dry cell density was obtained with the highest value of /$\3.9gell$ at $1.0g/\ell$ of initial acetic acid concentration. By adding acetic acid al 10g/$\ell$ of initial glucose concentration, maximum ${\alpha}$-amylase production was obtained with 331.55unit/m1 at $2.0g/\ell$ of initial acetic acid concentration. ${\alpha}$-Amylase production was decreased with the increase of initial acetic acrid concentration. By adding acetic acid to the medium, cell growth and ${\alpha}$-amylase production was higher in glucose than in maltose. By adding lactic acid to the medium, cell growth was decreased.

BIOFILTRATION OF GASEOUS TOLUENE USING ADSORBENT CONTAINING POLYURETHANE FOAM MEDIA

  • In this study, conventional biofilters packed with flexible synthetic polyurethane (PU) foam carriers were operated to remove toluene from a contaminated air stream. PU foams containing various adsorbents (e.g., zeolite, sepiolite, dolomite and barite) were synthesized for the biofilter media and their adsorption characteristics of toluene were determined. Adsorption capacity of PU-adsorbent foam was in the order of PU-dolomite ${\approx}$ PU-zeolite > PU-sepiolite > PU-barite. During the biofiltration experiment, influent toluene concentration was in the range of 0-160 ppm and EBRT (i.e., empty bed residence time) was 45 seconds. Pressure drop of the biofilter bed was 4-5 mm $H_2O/m$ column height. The maximum removal capacity was in the order of PU-dolomite > PU-zeolite > PU-sepiolite > PU-barite, while the complete removal capacity was in the order of PU-dolomite > PU-sepiolite > PU-zeolite > PU-barite. The better biofiltration performance in PU-dolomite foam was because PU-dolomite foam had lower density and higher porosity than the others providing favorable conditions for microbial growth. The results of biodegradation kinetic analysis showed that PU-dolomite foam had higher maximum removal rate ($V_m\;=\;11.04\;g$ toluene/kg dry material/day) and saturation constant ($K_s\;=\;26.57\;ppm$) than the other PU foams. This supports that PU-dolomite foam was better than the others for biofilteration of toluene.

  • In order to investigate the strength characteristics of weathered granite soils in unsaturated state, the five physically different weathered granite soils and the common soil (sandy loam) were examined. The disturbed and the undisturbed material were prepared for triaxial compression test. The following conclusions were drawn from the study; 1. Dry density of the undisturbed soil samples was lower than maximum dry density determined from the compaction test and it showed the higher value at the well graded soil. 2. The failure strength of the samples decreased with the increase of moisture content of the soil and these results were highly pronounced at the common soil sample having a good cohesive property. 3. On weathered granite soils, the cohesion was lower measured and the internal friction angle highly, the decrease rate at internal friction angle with increase of moisture content of the soil was more significant than that of cohesion 4. The modulus of deformation of the samples decreased with increase of moisture content of the soil and these phenomena were highly pronounced at the weathered granite soils than common soil. 5. The failure strength of the samples increased with in crease of confining pressure and effect of confining pressure on failure strength was highly significant at the lower moisture content of the soil.

  • The paper describes the comparison between observed and predicted stress-strain characteristics of marine silty clay in Dangjin district. For prediction, the hyperbolic model which is applied the parameters acquiring by physical and triaxial compression test was adopted, and the obtained results were summarized as follows: 1. The Young's modulus were increased with decreasing of moisture contents and increasing of dry density. 2. The most affective factor to hyperbolic model is lateral stress and dry density. and than cohesion and internal friction angle. 3. The comparision between the statistical and hyperbolic values of maximum deviator stress have few accordance. and the statisticals is lower than the hyperbolics. 4. Without. much labor and tiresome procedures, effective computer program was made and applied, but technical procedure for prevents test errors of parameter calculation is importants.

Natural Sand in Korea - Quality Evaluation - (한국의 모래 -품질평가-)

  • This study was conducted for evaluation the geological, physical, and chemical properties of domestic sand by analyzing about 4,800 quality data of natural sand from river and land area surveyed until 2023 through the aggregate resource survey conducted by the Ministry of Land, Infrastructure and Transport. The average depth of the Quaternary unconsolidated sedimentary layer in Korea, which includes a sand layer, is about 10m (maximum depth 66m). The thickness of the sand layer within the sedimentary layer is most dominant in the range of 0.5m to 4.0m. This accounts for about 70% of the entire sand layer. In the sand layer, the ratio of sand, gravel, and clay is 60:20:10. Regardless of the provenance or geology, the sand is mainly composed of quartz, plagioclase, and K-feldspar, and the minor minerals are muscovite, biotite, chlorite, magnetite, epidote. The sand includes in 45~75% of quartz, 5~20% of plagioclase and K-feldspar, each other. And other minor minerals are included in 10%. The average grain size of sand is 0.5mm to 1.0mm, which accounts for 44% of sand samples. The water absorption rate and soundness are estimated to be suitable for aggregate quality standard in almost all sand, and the absolute dry density is suitable for 66%.

Construction of Correlation between Basic Soil Properties and Deformation Modulus of Trackbed Soils Based on Laboratory and Field Mechanical Tests (역학적 실내외 시험에 의한 철도궤도 상부노반용 흙재료의 기본물성과 변형계수 상관성 평가)

  • The soils used as trackbed in Korea are selected using USCS utilizing basic soil properties such as Grain Size Distribution(GSD), % passing of #200 sieve ($P_{200}$), % passing of #4 sieve ($P_4$), Coefficient of uniformity ($C_u$), and Coefficient of curvature ($C_c$). Degree of compaction of the soils adapted in the code by KR should be evaluated by maximum dry density (${\gamma}_{d-max}$) and deformation modulus $E_{v2}$. The most important influencing factor that is critical to stability and deformation of the compacted soils used as trackbed is stiffness. Thus, it is necessary to construct a correlation between the modulus and the basic soil properties of trackbed soil in order to redefine a new soil classification system adaptable only to railway construction. To construct the relationship, basic soil test data is collected as a database, including GSD, maximum dry unit weight (${\gamma}_{d-max}$), OMC, $P_{200}$, $P_4$, $C_u$, $C_c$, etc.; deformation modulus $E_{v2}$ and $E_{vd}$ are obtained independently by performing a Repeated Plated Bearing Test (RPBT) and Light Weight Deflectometer Test (LWDT) for ten different railway construction sites. A linear regression analysis is performed using SPSS to obtain the relationship between the basic soil properties and the deformation modulus $E_{v2}$ and $E_v$. Based on the constructed relationship and the various obtained mechanical test data, a new soil classification system will be proposed later as a guideline for the design and construction of trackbed foundation in Korea.

Evaluating the Characteristics of Growth and Seedling Quality of Tetradium daniellii (Benn.) T. G. Hartley using Five Different Container Types (용기 종류에 따른 쉬나무 용기묘의 생장 및 묘목품질 특성)

  • There is an increasing demand for Tetradium daniellii seedlings due to their uses as alternative energy, for ecological restoration, and as a honey plant. This study was conducted to determine the optimum container for superior seedling production of T. daniellii. Experiments were performed using five plastic container types (500, 350, 320, 300, and 250 ml) for forestry facility cultivation. The height and root collar diameter growth of T. daniellii seedlings were significantly high in the 350-ml container. High growth appeared primarily in the container with a larger cavity volume and lower growing density. Root development was most active in full sunlight. The maximum dry matter production was observed in the 350-ml container, which was similar to the results of height and root collar diameter growth. QI, an index showing the quality of a seedling, was maximum at 0.97 in the 350-ml container. In conclusion, the 350-ml container is optimum for superior seedling production of T. daniellii.

Unconfined Compressive Strength Characteristics of Eco-Friendly Stabilizers and Carbon Fiber Reinforced Soil (친환경고화재와 탄소섬유 보강토의 일축압축강도 특성)

  • In this study, to reinforce the surface layer of weathered soil slopes where erosion and collapse of surface layer occur, compression strength tests were conducted by mixing carbon fiber and eco-friendly stabilizer (E.S.B.) To determine the optimal mixing ratio of E.S.B. and carbon fiber, E.S.B. was set at conditions of 10%, 20%, and 30%, and carbon fiber at 0.3%, 0.6%, 0.9%, and 1.2%. Additionally, to analyze the changes in compressive strength according to dry density and curing period, 85% and 95% of the maximum dry unit weight were applied, and curing periods were set to 3 days, 7 days, and 28 days. The standard strength for surface layer reinforcement of slopes is proposed as 4 MPa at 7 days and 6 MPa at 28 days according to ACI 230.1R-09 (2009). The compression test results showed that the unconfined compressive strength of E.S.B. reinforced soil met the standard strength at an E.S.B. mixing ratio of 10% or more for 95% compaction. Moreover, when carbon fiber was mixed with E.S.B. reinforced soil, a ductile fracture pattern was observed after the yield point due to compressive strength, indicating that the mixture could compensate for post-yield failure. It was analyzed that the maximum strength is exhibited at a carbon fiber mixing ratio of 0.6%. The unconfined compressive strength of carbon fiber reinforced soil increases by approximately 54-70% compared to the condition without carbon fiber.

Nitrogen fixation, and growth characteristics of Three Legume cover crops in no-tillage paddy field

  • A field experiment was conducted to investigate the performance of three legume species in a zero-tillage, non-fertilized rice field in a temperate zone. Before the experiment for 5 years, from 1995 to 1999, plant growth patterns of three legume species grown as over-wintering (October-May) cover crops on a paddy field were maintained to study N balance and $\textrm{N}_2$ fixation. Decrease in plant density accelerated from after winter to flowering from 1,090, 320, and 5 to 732, 232, and 6 plants $\textrm{m}^{-2}$ in Chinese milk vetch (CMV), white clover (WC), and hairy vetch (HV), respectively. Total dry weights of plants above-ground level were 0.05, 0.11, and 2.43 g $\textrm{plant}^{-1}$. in WC, CMV and HV respectively but steeply increased at ripening up to 0.77, 2.33, and 26 g $\textrm{plant}^{-1}$. The root dry weight of HV and CMV rapidly increased while in WC, root dry weight increased slightly towards flowering. The highest nodule numbers were recorded in CMV to April thereafter WC produced the highest. Nodule size was distributed within 7mm in CMV but it was larger in HV varying from 1 to 10mm. Shoot N (g $\textrm{m}^{-2}$) greatly increased from over-wintering to flowering in CMV, HV and WC and it ranged from 1.66, 0.5 and 1.92 to 12.6, 3.1 and 13.02 g $\textrm{m}^{-2}$, respectively. After wintering, the initial shoot N content (%) was more in CMV. Root N content (%) was constant or slightly decreased in HV and WC. Soil total N in the control plot (clean fallow) was the highest on Mar. 2 then decreased rapidly to flowering. Soil N content was constant in HV plots whereas it was low in WC plots for the entire growth period except just after winter. Maximum nitrogenase activities were 9, 37.8, and 131 mol $\textrm{C}_2\textrm{H}_4$ $\textrm{plant}^{-1}$ $\textrm{hour}^{-1}$. in CMV, HV, and WC, respectively. Nitrogenase activity showed a direct correlation with nodule number, size and fresh weight. As a cover crop preceding a rice crop, CMV is more suited to colder regions due to its earlier ripening characteristics. Hairy vetch and WC are recommended for regions with a mild winter and a long summer owing to their late ripening and great N fixation activity.

Mass Production of Resting Eggs by the Freshwater Rotifer, Brachionus calyciflorus Pallas with Exchange of Culture Water and Supplemental Aeration (환수 및 공기공급에 따른 담수산 rotifer, Brachionus calyciflorus의 내구란 대량생산)

  • This study was performed to investigate the formation and mass production of resting eggs by freshwater rotifer, Brachionus calyciflorus as influenced by different aeration supplements and exchange intervals of culture water in 15-L culture vessels and 500 L culture tanks. The maximum densities and mixis rates of the rotifers were not different between experimental group exposed to air or oxygen supplements. However, the fertilization rate and formation of resting eggs of the rotifers in the air-supplemented group were significantly higher than those in the oxygen supplemented group. In the experiment concerning exchange interval of culture water, the maximum density of the rotifers and formation of resting eggs in the batch culture were significantly higher than those in the semi-continuous culture with exchange of water every day. The formation of resting eggs per Chlorella dry weight was highest in the semi-continuous culture with exchange of water every day. The resting eggs of rotifers were produced at a density of $51.8{\sim}57.9{\times}10^6$ eggs in 500-L culture tanks. In this study, the batch culture with air is an effective method for mass production of resting eggs by the freshwater rotifer, B. calyciflorus, and the efficiency of mass production of resting eggs by this rotifer was similar to that of the seawater rotifers, B. plicatilis and B. rotundiformis.
