In this study, lotus leaf was added to a Takju (Korean milky rice wine) preparation to evaluate its effects on physicochemical and fermentation properties. For rapid saccharification Takju was stored at 25$^{\circ}C$ for 2 days and then temperature was switched to 18$^{\circ}C$ to mature the wine for 21 days. From the start of fermentation to 2-days, the pH of Takju decreased rapidly and acidity increased. Maltose, a byproduct of starch saccharification, increased for 2 days then rapidly decreased. Glucose fluctuated for 7 days and of the 21-days fermentation. The ethanol production rate was highest during the first 7 days, then slowed. Total viable yeast and lactic acid bacterial counts increased rapidly for 2 days and then decreased gradually thereafter. Leuconostoc spp. grew rapidly for 1 day and sharply disappeared with decreasing pH. DPPH radical scavenging activity was significantly higher for Takju prepared with lotus leaf than without. The overall acceptance of Takju tended to increase when it was prepared with lotus leaf.
The epidermal structure and stomatal types in vegetative and reproductive organs of three species of Bryophyllum(B. crenatum, B. diagremontion, B. tubiflorum) were described. The epidermal cells were polygonal, isodiametric, and rectangular in the leaves and stems, and elongated cells in the stamens, styles, and ovaries. These cells were commonly thick, and arched or sinuous in the leaves, epiphylous bunds, petals and ovaries. They were straight in the stems, petioles, pedicels, and peduncles. In both vegetative and reproductive organs, the subsidiary cell walls were commonly thin and mostly arched in all the organs. The great majority of the mature stomata in all the organs were helicocytic type with a helix of four to six subsidiary cells. The mature stomata varied from organ to organ with regard to the number and arangement of subsidiary cells. The ontogenetic type of stomata in all the organs was mostly helico-eumesogenous type. This type was subdivided into three subtypes such as parahelico-eumesogenous, anomohelico-eumesogenous, and diahelico-eumesogenous stomata on the basis of the division angle fo the guard mother cell. Sometimes, the anisoeumesogenous type was found in various organs. This type was subdivided into three subtypes such as paraniso-eumesogenous, anomoaniso-eumesogenous, and dianisoeummesogenous stomata. The tetra-eumesogenous and duplotetra-eumesogenous types were rarely found; the former in the leaf of B. crenatum and the latter in the leaf of B. diagremontiana. Anomometric patterns in the mesogenous categorry of stomatal types was observed in a few organs of all the materials. A new stomatal type with tetra-eumesogenous stoma within a girdle of three subsidiary cells fo aniso-eumesogenous in the leaf of B. diagremontiana was firstly observed in the vascular plants. This stoma was termed the cotetra-aniso-eumesogenous type. Anormal stomata such as aborted stomata, single guard cells, stoma with a constricted part in the middle of large guard cells, and arrested stomata were found in the various organs of all the materials.
Field experiment was conducted to get the basic information about the difference in color and plastid pigments at the different stage of harvest of flue-cured tobacco cultivated in paddy field, and compared to upland ones. All tested crops were primed in lots of 3 to 4 at each priming, beginning at the bottom of the plant. Leaf pigments and chromatic values of tobacco leaves were determined in samples collected at weekly intervals, and according to analyses obtained from 4 stalk positions. By the degree of maturity, leaves harvested were separated with visual characters into four classes of immature, mature, ripe and mellow. regardless of stalk position, chlorophyll contents was higher in the order of immature > mature > ripe > mellow, respectively. In fresh leaves, chlorophyll levels was higher with ascending stalk position. chlorophyll a and b in cured leaves were less than 10% of the amount present at harvest, and it was estimated that maturity condition also influenced the rates of degradation of plastid pigments. Fully mature leaves of upland diverted from paddy field showed slightly higher in chlorophyll contents than in those of upland tobacco ones but somewhat lowered in $L^*$ and $b^*$ values, and there was no difference in $a^*$ value.
Defensins are small cysteine rich peptides with a molecular mass of 5-10 kDa and some of them exhibit potent antifungal activity. We have cloned the coding region of a cDNA of 225 bp cysteine rich defensin, named as Tfgd1, from the legume Trigonella foenum-graecum. The amino acid sequence deduced from the coding region comprised 74 amino acids, of which the N-terminal 27 amino acids constituted the signal peptide and the mature peptide comprised 47 amino acids. The protein is characterized by the presence of eight cysteine resisdues, conserved in the various plant defensins forming four disulphide bridges, which stabilize the mature peptide. The recombinant protein expressed in E coli exhibited antifungal activity against the broad host range fungus, Rhizoctonia solani and the peanut leaf spot fungus, Phaeoisariopsis personata.
In soil-to-plant transfer of heavy metals, the amount absorbed and accumulated varies depending on the environment conditions. The absorption rate of cadmium (Cd) in plants differs considerably depending on the bioavailability of Cd in the soil, while usage by various organic matters is also reported to affect absorption patterns. Therefore, this study aimed to identify the difference in the transfer of essential metal ions and Cd to various plant parts when rice straw compost was used to cultivate soybean (Glycine max L. cv. Daepung). In the two-leaf stage of soybean cultivated in a greenhouse, Cd was mixed in the soil, after which the Cd and essential metal ions contents, and physiological changes of soybean seedlings were studied on the 15th and 25th day. The Cd toxicity in the plant was reduced with the use of rice straw compost. Further, the Cd content varied with the plant part, and was higher in young leaves (3rd and 4th leaf) than in the stem. When analyzed by leaf age, the Cd transfer was highest in young leaves (3rd and 4th leaf), followed by mature leaves (1st and 2nd leaf). While there was no significant difference between plant tissues in the absorption rate of copper (Cu) and zinc (Zn) when rice straw compost was used against Cd toxicity, the absorption rate of manganese (Mn) and iron (Fe) showed a significant decline in both the control and rice straw compost treatment conditions, as well as a significant difference between leaf ages. Therefore, these results confirm that the use of rice straw compost against Cd toxicity is effective, and implies that the rate of Cd transfer in the soybean plant varies significantly with leaf age.

The nutritive value and utilization of whole crop rice silage (WCRS), Hamasari, at yellow mature stage was determined by three studies. In first study, chemical composition, in vivo digestibility and metabolizable energy (ME) content of WCRS was determined by Holstein steers. WCRS contains 6.23% CP, its digestibility is 48.4% and estimated TDN is 56.4%. Its ME content was 1.91 Mcal/kg DM. Gross energy (GE) retention (% of GE intake) in steers is only 22.7% most of which was lost through feces (44.7% of GE intake). It takes 81 minutes to chew a kg of WCRS by steers. In another study, the effect of Hamasari at yellow mature stage at three stages of lactation (early, mid and late lactation) and two levels of concentrate (40 or 60%) on voluntary intake, ME content and ME intake, milk yield and composition using lactating Holstein dairy cows were investigated. Total intake increased with the concentrate level in early and mid lactation, but was similar irrespective of concentrate level in late lactation. WCRS intake was higher with 40% concentrate level than with 60% concentrate. ME intake by cows increased with the concentrate level and WCRS in early lactating cows with 40% concentrate can support only 90% of the ME requirement. Milk production in accordance with ME intake increased with the increase in concentrate level in early and mid lactating cows but was similar in late lactating cows irrespective of concentrate level. Fat and protein percent of milk in mid and late lactating cows were higher with for 60% concentrate than 40%, but reverse was in early lactating cows. Solids-not-fat was higher with for 60% concentrate than 40% concentrate. Finally in situ degradability of botanical fractions such as leaf, stem, head and whole WCRS, Hamasari at yellow mature stage was incubated from 0 to 96 h in Holstein steers to determine DM and N degradability characteristics of botanical fractions and whole WCRS. Both DM and N solubility, rate of degradation and effective degradability of leaf of silage was lower, but slowly degradable fraction was higher compared to stem and head. Solubility of DM and N of stem was higher than other fractions. The 48 h degradability, effective degradability and rate of degradation of leaf were always lower than stem or head. In conclusion, voluntary intake of silage ranged from 5 to 12 kg/d and was higher with low levels of concentrate, but milk yield was higher with high levels of concentrate. Fat corrected milk yield ranged from 19 to 37 kg per day. For consistency of milk, early lactating cows should not be allowed more than 40% whole crop rice silage in the diet, but late lactating cows may be allowed 60% whole crop rice silage.
IL-18. formerly known as IGIF(interferon -gamma inducing factor), is structurally IL-l related but functionally IL-12 related pro-inflammatory cytokine. The human IL -18(hIL-lS), like IL-$1{\beta}$, is synthesized as a biologically inactive precursor of 24kDa lacking a signal peptide, and then cleaved into an active mature form by cystein protease IL-$1{\beta}$ converting enzyme (ICE: caspase- 1), We tested if the mature hIL -18 can be expressed and secreted into culture medium by transforming the forming gene construct consisting of a mature hIL-18 gene fused to signal peptide of rice amylase lA. Secondly, we were tested if the pro- IL-18 could be processed into a biologically active form by caspase-l like protease in plant. Cell suspension culture was established from the leaf-derived calli of transgenic tobacco plant. Southern and Northern blot analysis indicated the expression of both pro-hIL-18 and mature hIL-18 plant cells. Western blot analysis introduced the protein products of pro- hIL -18 and mhIL -18 were observed in transigenic cell lines. In addition, the molecular size of recombinant pro-hILl-18 and mhIL-18 were estimated to be 24kDa and 18kDa, respectively. ELISA revealed that the amount of pro- hIL -18 was 1.3ug per gram of fresh weight calli. Moreover, the presence of mhIL-18 was detected in the culture medium and it appeared to be 25ug/L.
This study was conducted to explain the characters of the structure of biomass production in the thrifty-mature Quercus mongolica stands and investigate the relationships between the leaf weight or leaf area and the sapwood area in the bole. Also we intended to identify the allocation ratio of stem, branches, and leaves or heartwood, sapwood and bark in trees and the characters of productive structure of stem and leaf biomass by the tree height. The results obtained were as follows : 1. The allocation ratio of biomass based on dry weight was 70-84% in stem, 11-25% in branches, and 3-6% in leaves. 2. In the bole, the ratios of composition of heartwood, sapwood, and bark were showed 37-43%, 38-46%, and 16-19%, respectively. 3. The volume of sapmood was exceeded more than that of heartwood in dominant and intermediate trees, while it was reversely appeared in suppressed trees. 4. The weight and area of leases significantly correlated with the sectional area of sapwood in bole (r>0.9. 1% significant level). 5. The ratio of leaf area($m^2$) to sapwood areal($cm^2$), k varied 0.35 to 2.05. 6. The basal diameter and the cross sectional area of a branch significantly correlated with the leaf weight r>0.9. 1% significant level. 7. The leaf weight in a tree is showed a normal distribution curve and the accumulative volume of bole is showed a tapering type.

The expression of a chimeric transgene (nos-npt II) has been examined in 9 years old transgenic poplars (Populus koreana x P. nigra) growing in a nursery. The expression of the gene in twenty six independentely transformed plants were examined by 1) enzyme (NPT II) assay, 2) RT-PCR, and 3) resistance to kanamycin. High NPT II activities in young leaves of all the transformed plants were found even without a selection pressure for antibiotics for 9 years. However, the activity varied with the positions of leaves in the stem in that young leaves showed higher activity than did mature tissues. When leaf segments were cultured in the presence of 150 mg/l kanamycin, only those from young leaves produced vigorously growing callus. However, as in the case of NPTII assay, the leaf segments from mature leaves did not form callus well on the media. RT-PCR with nptII specific primers also showed that amplification products were observed only when RNAs from young tissues were used. The total RNA gel showed that while RNA in young leaves are relatively stable and in a large quantity, those in old leaves were mostly degraded. All the above results suggest that the gene is transcriptionally active only in young tissue even though it is attached to a constituitive promoter. Therefore, the expression of foreign gene in poplar plants seemed to be affected by the metabolic state of the cells and thus vary greatly with the developmental stages and the age of tissue.
