  • This manuscript describes the relationship between climate change and forest growth, forest species, carbon stocks, and tree mortality. 1) In the aspect of forest growth, the growth of major coniferous species, including Pinus densiflora, had a negative correlation with temperature. On the other hand, major deciduous oak species, including Quercus variabilis and Quercus mongolica, had a positive correlation with temperature. 2) When considered in the aspect of the forest species distribution, various models commonly showed a decrease in the distribution of coniferous species and an increase in oak species due to climate change in the medium to long term. 3) From the carbon stock perspective, there was a difference in the estimation according to the status of forest management. Most of Korean forests will mature to become over-matured forest after year 2030 and are estimated to produce approximately 410 million ton forest biomass until 2090 with the current cutting regulations for sustainable forest management announced by the Korean Forest Service. 4) In the forest mortality, the mortality rate of the major coniferous species showed a clear tendency to increase higher temperatures while it decreased for the oak species with no verification of statistical significance. Moreover, the mortality of the subalpine coniferous species was projected to progress rapidly. considering the overall impacts described above, there should be a management strategy for coniferous species that are relatively vulnerable to climate change. Moreover, a sustainable forest plan in the aspect of ecosystem services, carbon sequestration and storage, which is linked to global issues such as Sustainable Development Goals, ecosystem services and negative emission.

  • To investigate the forest structure of Mt. Bukhan. ranging from Seoul to Kyongkido, twenty plots were set up by the vegetation physiognomy and vegetation analysis was carried out. According to the leading dominant tree species in canopy stratum, forest communities were classified into three large groups of natural forest communities, semi-natural forest communities and artificial forest communities, and each of them covered 82.64, 7.03, and 5.71% of Mt. Bukhan area, respectively. Pure or mixed natural forest communities of Pinus densiflora and Quercus mongolica were major forest communities and covered 70.8% of Mt. Bukhan area. The important planted tree species were Robinia pseudoacacia, Pinus rigida, and Alnus birsuta and they were mainly planted at the southern slope and roadside. The degree of human disturbance of vegetation of 8, 7, and 6 area covered 82.64, 0, and 12.74%, respectively. According to forest dimensions, most of forest communities were young aged forests of which mean DBH was 20cm and canopy height below 10m. However, a few mature forest communities of Pinus densiflora or Quercus mongolica were found in the small area. The range of Shannon's species diversity of major natural forest communities, pure or mixed forest communities of Pinus densiflora and Quercus mongolica was 1.085~1.242. According to stand dynamic analysis by DBH class distribution, the present Quercus mongolica communities arid Robinia pseudoacacia communities may last long their present forest structure and most of other communities may be succeeded to Quercus mongolica communities, however, a few communities invaded by Robinia pseudoacacia and Quercus aliena-Quercus acutissima communities may be succeeded to Robinia pseudoacacia communities and Quercus aliena communities, respectively. DCA was the most effective method of this study. DCA ordination were showed that successional trends of tree species seem to be from Pinus densiflora through Quercus serrata. Prunus sargrntii. Sorbus alnifolia to Q. mongolica. Fraxinus mandsburica, F. rhynchophylla in the upper layer and from Zanthoxylum schinifolium, Lespedeza crytobotrya trough Rhus trichocarpa. Rh. verniciflua. Rhododendron mucronulatum. Rh. schlippenbachii to Acer pseudo-sieboldianus. Magnolia sieboldii, Euonymus sieboldianus.

  • Off-white, friable embryogenic calluses were formed on the internal integument of mature seeds of yuzu (Citrus junos) cultured on Murashige and Skoog's basal medium at a frequency of 1.2%. Embryogenic calluses were proliferated when cultured on medium with 1 mg/L 2,4-D. Upon transfer to medium with 0.1 mg/L kinetin, embryogenic calluses produced numerous somatic embryos. Embryogenic suspension cultures were established by placing embryogenic calluses into liquid medium with 1 mg/L 2,4-D. When plated onto medium with 0.5 mg/L ABA, embryogenic cells developed into somatic embryos at a high frequency, and then regenerated into plantlets. Plantlets were successfully transplanted to potting soil and grown in a greenhouse.

  • Introgressive hybridization of Pinus densiflora in Korea was investigated by examining the position of resin ducts in 4,992 trees from 67 populations. Judging that P. densiflora has only external resin ducts, P. thunbergii only medial resin ducts, and any tree which has even one needle with external and medial resin ducts is hybrid, hybrids are found at all populations investigated. As variation in position of resin ducts was found among needles on a tree, the number of needles investigated from a tree affects judgment of the tree's hybridity. Therefore, hybridity seems to be judged by many traits rather than by the single trait, position of resin ducts. Seed production by artificial pollination between P. densiflora and P. thunbergii was very poor, i.e. one full seed from five female flowers or from two mature cones. This poor seed production, in addition to the low hybrid vigor, if any, seems to make the artificial pollination between the two species for mass-production of seeds for planting impractical.

  • Parts of damaged trees are being transplanted in accordance with the Environmental Impact Assessment Manual. Problems such as death or poor growth are constantly being addressed in the process of transplanting trees from the forest they originally inhabited to temporary and final transplant sites. The purpose of this study is to analyze the differences in soil properties in the surrounding forest, the temporary transplant site, and the construction site and to suggest methods for improving the soil to make it suitable for the growth of transplanted trees. For 10 development projects, 2 soil samples were sampled from the surrounding forest, temporary transplant site, and construction site. A total of 60 soil samples were analyzed for physical and chemical properties. Among the physical properties such as coefficient of permeability, available moisture, and hardness, and chemical properties such as acidity, organic matter content, total nitrogen, and available P showed significant differences among groups. The soil of the construction site is harder than the surrounding forest because of construction equipments, the coefficient of permeability is higherthan the surrounding forest because of high sand content, and the available moisture was low. It does not retain the moisture necessary for plants in the soil and drains immediately. It is necessary to implement tillage to improve the physical properties and structure of the soil. In addition, it is necessary to cover the surface with wood chips or fallen leaves after adding mature organic matter to improve the physical and chemical properties of the soil together.

  • In order to optimize tissue culture conditions for genetic transformation of orchardgrass, the effects of culture medium supplements on tissue culture responses were investigated with mature seeds of 3 cultivars, 'Frode'. 'Roughrider' and 'Frontier' as explant tissues. Callus induction medium containing 3mg/L 2,4-D or 3mg/L dicamba each with 0.1mg/L BA was optimal fer embryogenic callus formation from mature seed and had a strong effect on successive plant regeneration. The regeneration frequency from embryogenic callus among cultivars were descending order of Roughrider > Frode > Frontier. Supplementation of the regeneration media with 10mg/L $AgNO_3$ and 40mg/L cysteine enhanced frequency of plant regeneration. Efficient regeneration system established in this study will be useful fur molecular breeding of orchardgrass through genetic transformation.

  • Morphogenetic responses were investigated by culturing embryo and leaf explants of Korean wild type tea plant, Camellia sinensis (L.) O. Kuntze. Induction of direct somatic embryogenesis as well as adventitious and/or axillary shoots was obtained from mature zygotic embryo cultures on Murashige and Skoog (MS) basal medium having 5 to $20\mu\textrm{M}$cytokinin a lone. Morphogenetic response was decreased dramatically by the addition of auxins tested. One hundred percent of induced and isolated shoots formed roots after four weeks of culture on half-strength MS or quarter-strength Schenk and Hildebrandt (SH) media supplemented with $10\mu\textrm{M}$indole-3-butyric acid (IBA). Immature zygotic embryos were shown to be a suitable explant for embryogenic callus formation in the presence of 2, 4-dichlorophenoxyacetic acid(2, 4-D) in basal medium. Mature zygotic embryo originated leaves were used to test their ability for mophogenesis by incorporating plant growth regulators such as IBA, naphthyl-1-acetic acid (NAA), and 6-benzylaminopurine (BAP). Apparently, the morphogenetic responses of the cultured explant sources on the types and/or levels of plant growth regulators tested were observed visually.

  • Excised mature embryos of P. koraiensis were plated on a series of G.D. basic media supplemented with growth regulators as follow; $0.1mg/{\ell}$ NAA(M-1); $0.1mg/{\ell}$ NAA and 2,4-D (M-2); $0.1mg/{\ell}$ 2,4-D and BAP (M-3); $0.1mg/{\ell}$ 2,4-D and kinetin (M-4). Cytological study of the callus showed that thercentage of occurrance of diploid cells was observed 52% in M-3 and 36% in M-4, while that was revealed 29% in M-1 and 17% in M-1. The frequency of diploid cells was increased on the M-3 and M-4 media. The stable chromosome state is a crucial factor for organogenesis. Therefore, it can be inferred that the callus tissues cultured on media supplemented with both auxin and cytokinin have the greatest possibility of organogenesis.

  • Multiple shoot induction and plant regeneration using apical bud and nodal explants of 100 year old tree of Anthocephalus cadamba, an important sacred and medicinal tree in India was achieved for the first time. Aseptic explants cultured in Murashige and Skoog (MS) medium augmented with different concentrations of BAP (0.1, 0.5, 1.0, 2.5, 5.0 and 10 mg/l), when maintained for 60 days, healthy shoots were induced in presence of BAP (1 mg/l). Lower concentrations of BAP (0.1 - 0.5 mg/l) induced only one shoot per explant. Increase in number of shoots per explant was observed in presence of higher concentrations of BAP (2.5, 5.0 and 10 mg/l). However, elongation of shoots was completely inhibited. Bud break and shoot regeneration was largely associated with seasonal factors. Apical buds cultured during June to August exhibited early bud break within two weeks of initial culture. In rest of the months, bud break and shoot regeneration was very slow irrespective of the various concentrations of BAP used in the medium. Explants sourced from three different maturity levels of shoots indicated that actively growing shoots from the mother plant with 1 - 2 nodal segments was more suitable for culture initiation than the explants collected from mature shoots at dormant stage. Regenerated shoots with 2 - 3 pairs of leaves when transferred to half strength MS medium fortified with IBA (1 mg/l), 60% of the shoots induced healthy roots, indicating the possibility of large scale micropropagation.

  • Present experiment has been carried out in order to make clear the abnormalities of the female gametophyte formation and its relation to fertility, using the short-style of F. koreana, the results of which are summarized as follows : (1) Anatropous ovule has single integument with thick cell-layer and tiny nucellus consisting of nucella-epidermis and megaspore mother cell. (2) Meiotic division of megaspore mother cell takes place around middle or latter part of March, while that of microspore mother cell occurs from the end of September to the beginning of October. (3) Megaspore mother cell stage is long, and ranges from October to March next year. (4) Formation of mature embryo sac is not completed until the beginning of May, approximately one month after blooming. (5) Normal embryo sac is rare, most of the nucellus being devoid of embryo sac.
