Spermatogenesis and Sexual Maturation in Male Mactra chinensis (Bivalvia: Mactridae) of Korea

  • Spermatogenesis, the reproductive cycle, and the size at first sexual maturity in male Mactra chinensis were investigated by cytological and histological observations. The spermatozoon exhibits a primitive type morphology and is similar to those of other bivalves in that it contains a short midpiece with four mitochondria surrounding the centrioles. The morphologies of the sperm nucleus type and the acrosome shape of this species are cylindrical and modified cap-like, respectively. The spermatozoon is approximately $40-45\;{\mu}m$ in length including the sperm nucleus (about $1.46\;{\mu}m$), acrosome (about $1.20\;{\mu}m$) and tail flagellum. The axoneme of the sperm tail flagellum consists of nine pairs of microtubules at the periphery and a pair at the center. The axoneme of the sperm tail shows a 9+2 structure. The spawning period of this species lasts from June to September, and the main spawning occurs in July and August, when the seawater temperature is greater than $20^{\circ}C$. The percentage of individual male clams at first sexual maturity was 56.5% for those whose shell lengths were 35.1-40.0 mm, and 100% for over 45.1 mm. Accordingly, harvesting clams <35.1 mm in shell length could potentially cause a drastic reduction in recruitment, and a measure indicating a prohibitory fishing size should be taken for adequate fisheries management.

Maturity and Spawning of Sebastes thompsoni in the Coastal Waters off Ulleungdo, Korea (울릉도 연안에 서식하는 불볼락(Sebastes thompsoni)의 성숙과 산란)

  • We investigated the maturation and spawning of Sebastes thompsoni in the coastal waters off Ulleungdo, based on samples collected by gill net from March 2013 to February 2014. We analyzed monthly changes in maturity stage, gonadosomatic index (GSI), egg diameter, fecundity, and total length (TL) at 50% group maturity. The average TLs of female and male S. thompsoni were 22.6 cm and 22.5 cm, respectively. The main parturition period of females was during March and April. The GSI of females began to increase in January, and reached a maximum in March. The GSI of male reached a maximum between January and February. The spawning period of males was shorter than that of females. The egg diameter during gestation stage was 1.4±2.1 mm. The TL of females at 50% group maturity was 22.3 cm.

Induced Ovulation and Histological Changes of the Oocytes according to HCG and Trout Pituitary Extract in the Korean Loach, Misgurnus anguilicaudatus (한국산 미꾸라지에 있어서 HCG나 송어의 뇌하수체에 의한 배란유기와 난모세포의 조직학적 변화)

  • This study was undertaken to determine the effects of HCG and trout pitutary on sexual maturation and ovulation in the Korean loach, Misgurnus anguillicaudatus. Sexual maturation was observed in the fish treated with 250+125 IU HCG within 2-4 days following injections. Total ovary weight and GSI were significantly greater in the hormone-treated fish than in the control fish. Furthermore, in the hormone-treated fish body weight increased by about 4-9 percent in 2 days. The water content of the ovulated eggs of the fish treated with 250+125 IU HCG was also greater than that in the control. Changes in both macroscopic & microscopic appearnce of Korean loach ovaries are describled, as well as changes in the gonadosomatic index. Oocytes pass through seven cytologically changes in the relative stages. By examining these stages, it was found that oogenesis occures in two broad phases. The previtellogenic phasebegins as a new oogonia continues to arise following spawning. These rapidly develop into early perinucleolus oocytes, which in turn develop into resting stage oocytes. Most oocytes remain in this stage when they develop into late perinucleolus oocytes. The vitellogenic phase begins as these late perinucleolus oocytes become transformed into early maturing oocytes through the accumulation of yolk. The cytoplasm completely fills with yolk as oocytes reach the late maturing stage. Shortly before spawning the final hyaline stage of developing is reached. Changes in the microscopic appearances of the ovaries were well correlated with changes in both gonadosomatic index and macroscopic appearance.

  • We investigated maturation and spawning of Miichthys miiuy in the south-western water of Korea, based on samples collected by stow net from August 2015 to December 2016. We analysed monthly changes in maturity stage, gonadosomatic index (GSI), total length (TL) at 50% group maturity. The spawning period was August to October. We estimated the TL at 50% group maturity as 54.8 cm for females and 49.0 cm for males.

  • This stduy was carried out to obtain basic information available on ovulation, spawning, fertilization rate, hatching rate, and deformity rate after hCG injection in cyprinid loach, Misgurnus mizolepis. The results obtained in these experiments were as follows: 1. Ovulation and spawning occurred simultaneously and spawing was completed within 1 hour after ovulation. 2. More than 80% of fertilization rates appeared within 12 hours at 21$^{\circ}C$, 8 hours at $25^{\circ}C$, and 4 hours at 29$^{\circ}C$, respectively, following the onset of spawing. Afterwards, the fertilization rates of released eggs sharply decreased in three different water temperatures. 3. More than 70% of hatching rates appeared within 8 hours at 21$^{\circ}C$, 6 hours at $25^{\circ}C$, and 2 hours at 29$^{\circ}C$, respectively, following the onset of spawing. Afterwards, hatching rates of spawned eggs abruptly decreased in three different water temperatures. 4. The deformity rates of hatched larvae were high at $25^{\circ}C$, 8 hours following the onset of spawing. 5. Based on the developmental ability of oocytes, the optimum time of fertilization was 4 hours (stage 5) following the onset of spawing.

  • On maturation of abalone KIKUCHI'S concept of accumulative temperature expressed ${\Sigma}(t_i-{\theta})$ have been apply importantly for an anniversary spawning of the animals. In case of Haliotis discus hannai he was formulated ${\Sigma}(t_i-7.6)>1300$. This expression is able to modify by UKI'S the relationship between the daily feeding rate ($\%$) (Y) ana temperature ($^{\circ}C$) (t) expressed Y= 0.4291 t-0.1501. The modified expression is as the formula ${\Sigma}(Y-3.05)>570$. And the concept its meaning involves is replaced with accumulative ingestion instead of accumulative temperature. This modified concept is understood that on maturation of abalone temperature decide on satiation level, food ingestion decide on gonad maturity rate.

Reproductive Cycle and Induced Sexual Maturation of the Pacific Oyster, Crassostrea gigas (참굴, Crassostrea gigas의 생식주기와 성 성숙 유도)

  • Reproductive cycle, the condition index, sex ratio of the Pacific oysters, Crassostrea gigas were investigated by histological and morphometric data. The specimens were collected in the two oyster farms of Geoje and Namhae, Gyeongsangnam-do, Korea, from November 1995 to October 1996. Growth of shell length in two regions was similar, but growth of total weight of the oyster in Namhae was faster than that in Geoje oyster farm. The spawning periods in female and male clams were from July to October in Geoje and from June to October in Namhae oyster farm. Ripe oocytes were approximately 50 m in diameter. The reproductive cycle of in females and males in Geoje and Namhae oyster farms can be divided into five successive stages: early developing, late developing, ripe, partially spawned and spent/inactive. Monthly changes in gonad developmental phases showed somewhat different patterns between female and male clams except for the spawning period. On the whole, however, monthly changes in the gonad developmental phases showed a similar pattern in the same sex. The sex ratios of females to males in Geoje and Namhae oyster farms were not significantly different from a 1:1 sex ratio ($x^2$ = 0.55 (p > 0.05) in Geoje and $x^2$ = 0.27 ( p > 0.05) in Namhae). Artificial induction of maturation by heating of adult oysters (two-year-old) was investigated from 17 January to 18 March in 1996. Maturity at the fixed water temperature group of $20^{\circ}C$ was 80%, it showed the highest maturity of experimental groups cultured for five weeks. The survival (%) of Crassostrea gigas in the raised water temperature experimental groups (15, 20, $25^{\circ}C$) were over 98.5%, as similar to the control group (100%). But, the survival of C. gigas in the fixed water temperature experimental groups (15, 20, 25, $30^{\circ}C$) were decreased with the increase of the water temperatures. In the fixed water temperature experimental group of $30^{\circ}C$, the survival was 51.1%. Base on these results, the fixed water temperature of $20^{\circ}C$ was the best condition for artificial induction of sexual maturation.

Plasma Sex Steroid Hormone and Vitellogenin Profiles during Ovarian Development of the Wild Marbled Sole (Limanda yokohamae) (자연산 문치가자미 (Limanda yokohamae)의 난소 발달에 따른 혈중 성호르몬과 난황단백전구체 농도 변동)

  • This study correlated changes in estradiol-l7$\beta$ ($E_2$), testosterone (T), 17$\alpha$,20-dihydroxy-4-pregnen-3-one (DHP), and vitellogenin (VTG) levels with changes in the gonadosomatic index (GSI) and ovarian histology during the annual reproductive cycle of the wild marbled sole, Limanda yokohamae. Synchronous oocyte development occurs in this fish. Ovary maturity was classified into four periods, based on histological observations: the spawning (December to February), post-spawning (February to April), recovery (May to August), and vitellogenic (September to November) periods. Seasonal changes in the GSI were inversely correlated with water temperatures and reflected the degree of ovarian maturity. Plasma VTG levels were correlated with changes in the GSI, which increased from September to a peak in January, and levels remained comparatively high until February. Estradiol-17$\beta$ was at baseline levels (<0.11 ng/mL) during the spring and summer, and peaked rapidly (1.55$\pm$0.445 ng/mL) from October to January. Plasma T and DHP levels had a similar profile; they rose markedly during the spawning period and remained low (or were not detectable) from spring through autumn. These data indicate that changes in plsama steroid hormones and VTG levels are correlated with the annual ovarian activity of the marbled sole. Based on these results and published reports, it appears that in this species DHP is the most important maturation-inducing steroid and that T is also related to final maturation.

Gonad Maturation and Spawning of the Blue Crab, Portunus trituberculatus (Miers, 1876) from the West Sea of Korea (한국 서해안 꽃게, Portunus trituberculatus (Miers, 1876)의 난 성숙과 산란)

  • The gonad maturation and spawning of blue crab, Portunus trituberculatus (Miers, 1876) using samples caught by trammer net in the western coast of Korea from January to December. The gonadosomatic index (GSI) showed the highest value of 15.0 in June and lowest value of 1.29 in August. Hepatosomatic index (HSI) showed the lowest in the spawning season from June to July. Thereafter, from August the value started to increase to 5.77 in December, showing an adverse trend against GSI. In the histological studies of the female reproductive organ, the gonad filled with primary oocytes from January to February and secondary oocytes in May. The female gonads mature from June, and mature and immature groups could be distinguished from December to February or March. In August, female developed another cycle of gonad development, showing new primary oocytes in the gonads again. Some of female crabs had receptaculum filled with already active sperm cells in it from March to May. In the time of August to September, almost all the females had the sperm sacs in the receptaculum. From the result, the coupling period estimated from August to September. Total RNA and DNA values of the crabs were highest in May (2.51, $0.57{\mu}g/mg$). The DNA value remained unchanged except for the value in May ($0.09-0.13{\mu}g/mg$). while RNA value was lowest in January ($1.08{\mu}g/mg$). The RAN/DNA ratio was the lowest (6.23%) in May and highest (18.05%) in July. The value of correlations coefficient between body weight and fecundity of the crabs was higher that of between carapace width and fecundity.

  • Gonadosomatic index, reproductive cycle, spermatogenesis and first sexual maturity of Chlamys farreri were investigated by cytological and histological observations, from January 1998 to December 1999. The gonadosomatic index (GSI) rapidly increased in April and reached a maximum in May when seawater temperature rapidly increase. Then the GSI gradually decreased from June to August when spawning occur. Accordingly, monthly changes in the GSI in males coincide with the reproductive cycle. The spermatozoon of Chlamys farreri is the primitive type found in external fertilization species. The head of the spermatozoon is approximately $2.75{\mu}m$ in length including the acrosome measuring about $0.50{\mu}m$ in length, and its tail was approximately $20{\mu}m$, the axoneme of the tail flagellum consists of nine pairs of microtubules at the periphery and a pair at the center. Five spherical mitochondria around the centriole (the satellite body) appear in the middle piece of the sperm. The spawning period was from June to August and the main spawning occurs from July to August when seawater temperatures are greater than $20^{\circ}C$ The reproductive cycle of this species can be categorized into five successive stages; early active stage (January to March), late active stage (March to April), ripe stage (April to August), partially spawned stage (June to August), and spent/inactive stage (August to January). Over 50% of male scallops attained first sexual maturity between 50.0 and 60.0 mm in shell height, and 100% of those over 60.0 mm in shell height achieved maturity. Accordingly, we assume that male individuals begin reproduction at three years of age.
