Identification of Two Novel Amalgaviruses in the Common Eelgrass (Zostera marina) and in Silico Analysis of the Amalgavirus +1 Programmed Ribosomal Frameshifting Sites

  • The genome sequences of two novel monopartite RNA viruses were identified in a common eelgrass (Zostera marina) transcriptome dataset. Sequence comparison and phylogenetic analyses revealed that these two novel viruses belong to the genus Amalgavirus in the family Amalgaviridae. They were named Zostera marina amalgavirus 1 (ZmAV1) and Zostera marina amalgavirus 2 (ZmAV2). Genomes of both ZmAV1 and ZmAV2 contain two overlapping open reading frames (ORFs). ORF1 encodes a putative replication factory matrix-like protein, while ORF2 encodes a RNA-dependent RNA polymerase (RdRp) domain. The fusion protein (ORF1+2) of ORF1 and ORF2, which mediates RNA replication, was produced using the +1 programmed ribosomal frameshifting (PRF) mechanism. The +1 PRF motif sequence, UUU_CGN, which is highly conserved among known amalgaviruses, was also found in ZmAV1 and ZmAV2. Multiple sequence alignment of the ORF1+2 fusion proteins from 24 amalgaviruses revealed that +1 PRF occurred only at three different positions within the 13-amino acid-long segment, which was surrounded by highly conserved regions on both sides. This suggested that the +1 PRF may be constrained by the structure of fusion proteins. Genome sequences of ZmAV1 and ZmAV2, which are the first viruses to be identified in common eelgrass, will serve as useful resources for studying evolution and diversity of amalgaviruses.

  • Bayesian personalized ranking (BPR) is a state-of-the-art recommendation system techniques for implicit feedback data. Unfortunately, there might be a loss of information because the BPR model considers only the binary transformation of implicit feedback that is non-binary data in most cases. We propose a modified BPR method using a level of confidence based on the size or strength of implicit feedback to overcome this limitation. The proposed method is useful because it still has a structure of interpretable models for underlying personalized ranking i.e., personal pairwise preferences as in the BPR and that it is capable to reflect a numerical size or the strength of implicit feedback. We propose a computation algorithm based on stochastic gradient descent for the numerical implementation of our proposal. Furthermore, we also show the usefulness of our proposed method compared to ordinary BPR via an analysis of steam video games data.

  • Since concrete is an inhomogeneous material consisting of larger aggregates and sand embedded in a cement paste matrix, it relatively shows a complex failure mechanism. In order to assure the reliability of concrete structure. microscopic fracture behavior and internal damage progress of concrete under the loading should be fully understood. In this study, an acoustic emission(AE) technique has been used to clarify microscopic failure mechanism and their corresponding AE signal characteristics of concrete under three-point bending test. In addition 2-dimensional AE source location has been performed to monitor the progress of an internal damage and the successive crack growth behavior during the loading. The relationship between AE signal characteristics and microscopic fracture mechanism is discussed.

Optimization of soaking in aqueous ammonia pretreatment of canola residues for sugar production (당 생산을 위한 카놀라 부산물의 암모니아 침지 전처리 공정의 최적화)

  • Bioenergy production from lignocellulosic biomass and agriculture wastes have been attracted because of its sustainable and non-edible source. Especially, canola is considered as one of the best feedstock for renewable fuel production. Oil extracted canola and its agriculture residues are reuseable for bioethanol production. However, a pretreatment step is required before enzymatic hydrolysis to disrupt recalcitrant lignocellulosic matrix. To increase the sugar conversion, more efficient pretreatment process was necessary for removal of saccharification barriers such as lignin. Alkaline pretreatment makes the lignocellulose swollen through solvation and induces more porous structure for enzyme access. In our previous work, aqueous ammonia (1~20%) was utilized for alkaline reagent to increase the crystallinity of canola residues pretreatment. In this study, significant factors for efficient soaking in aqueous ammonia pretreatment on canola residues was optimized by using the response surface method (RSM). Based on the fundamental experiments, the real values of factors at the center (0) were determined as follows; $70^{\circ}C$ of temperature, 17.5% of ammonia concentration and 18 h of reaction time in the experiment design using central composition design (CCD). A statistical model predicted that the highest removal yield of lignin was 54% at the following optimized reaction conditions: $72.68^{\circ}C$ of temperature, 18.30% of ammonia concentration and 18.30 h of reaction time. Finally, maximum theoretical yields of soaking in aqueous ammonia pretreatment were 42.23% of glucose and 22.68% of xylose.

Effects of Al-doping on IZO Thin Film for Transparent TFT

  • Amorphous transparent oxide semiconductors (a-TOS) have been widely studied for many optoelectronic devices such as AM-OLED (active-matrix organic light emitting diodes). Recently, Nomura et al. demonstrated high performance amorphous IGZO (In-Ga-Zn-O) TFTs.1 Despite the amorphous structure, due to the conduction band minimum (CBM) that made of spherically extended s-orbitals of the constituent metals, an a-IGZO TFT shows high mobility.2,3 But IGZO films contain high cost rare metals. Therefore, we need to investigate the alternatives. Because Aluminum has a high bond enthalpy with oxygen atom and Alumina has a high lattice energy, we try to replace Gallium with Aluminum that is high reserve low cost material. In this study, we focused on the electrical properties of IZO:Al thin films as a channel layer of TFTs. IZO:Al were deposited on unheated non-alkali glass substrates (5 cm ${\times}$ 5 cm) by magnetron co-sputtering system with two cathodes equipped with IZO target and Al target, respectively. The sintered ceramic IZO disc (3 inch ${\phi}$, 5 mm t) and metal Al target (3 inch ${\phi}$, 5 mm t) are used for deposition. The O2 gas was used as the reactive gas to control carrier concentration and mobility. Deposition was carried out under various sputtering conditions to investigate the effect of sputtering process on the characteristics of IZO:Al thin films. Correlation between sputtering factors and electronic properties of the film will be discussed in detail.

Advanced Low-k Materials for Cu/Low-k Chips

  • As the critical dimensions of integrated circuits are scaled down, the line width and spacing between the metal interconnects are made smaller. The dielectric film used as insulation between the metal lines contributes to the resistance-capacitance (RC) time constant that governs the device speed. If the RC time delay, cross talk and lowering the power dissipation are to be reduced, the intermetal dielectric (IMD) films should have a low dielectric constant. The introduction of Cu and low-k dielectrics has incrementally improved the situation as compared to the conventional $Al/SiO_2$ technology by reducing both the resistivity and the capacitance between interconnects. Some of the potential candidate materials to be used as an ILD are organic and inorganic precursors such as hydrogensilsequioxane (HSQ), silsesquioxane (SSQ), methylsilsisequioxane (MSQ) and carbon doped silicon oxide (SiOCH), It has been shown that organic functional groups can dramatically decrease dielectric constant by increasing the free volume of films. Recently, various inorganic precursors have been used to prepare the SiOCH films. The k value of the material depends on the number of $CH_3$ groups built into the structure since they lower both polarity and density of the material by steric hindrance, which the replacement of Si-O bonds with Si-$CH_3$ (methyl group) bonds causes bulk porosity due to the formation of nano-sized voids within the silicon oxide matrix. In this talk, we will be introduce some properties of SiOC(-H) thin films deposited with the dimethyldimethoxysilane (DMDMS: $C_4H_{12}O_2Si$) and oxygen as precursors by using plasma-enhanced chemical vapor deposition with and without ultraviolet (UV) irradiation.

A Study of Operating the Engineering Accreditation System to Contribute to Improving Rate of Employment (취업률 향상에 기여하는 공학인증 시스템 운영에 관한 연구)

  • The employment rate of a college has long been one of the most important elements in college survival strategies. Employment rates may not be key to colleges' survival in other countries, but they are fundamental to colleges in Korea as seen in the Korean government-led National Competency Standard (NCS), which has been underway since its introduction to colleges in Korea. Against this backdrop, if the engineering accreditation system can contribute to improving employment rate in this context, it can take root more quickly. The process conducive to improving employment rates can be classified as both direct and indirect methods. With dwindling school-age population having impacts on colleges and universities, the environment and the education directions of colleges have rapidly been shifting since the introduction of the NCS. Therefore, various survival strategies for colleges need to be developed. An indicator of employment rate of college graduates is among the most important things during this transition process. The Department of Mechanical Engineering at Koje College has consistently shown good records during the past 5 years by matching the accreditation schemes and employment strategies. The department will help put in place the engineering technology education accreditation system at the college through its various innovative and effective programs. To formulate a better employment strategy, it is needed to reflect and consolidate the engineering accreditation scheme into the existing employment policy of college. In short, this paper examines both the engineering accreditation scheme and the employment policy in ways that helps produce a coupling matrix structure. This study looks into the measures for improving employment rates by better adjusting the engineering education methods under the accreditation scheme.

Mechanical Properties and Morphology of Polyamide 6/Maleated Polypropylene Blends (폴리아미드6/반응성 폴리프로필렌(PA6/PP-g-MA) 블렌드의 기계적 특성과 모폴로지)

  • Melt blends of polymide 6(PA6) and polypropylene grafted maleic anhydride(PP-g-MA) were prepared to study the influence of chemical reaction between the two polymer components. The tensile, flexural, izod impact, dynamic mechanical properties and phase structure were investigated for this blend system. Tensile strength and modulus of the blends showed synergetic effect upon blending of two polymer components. Flexural properties maintained the value of numerical mean calculated from the weight ratio of two components. Also, notched izod impact strengths showed maximum in th PA6/PP-g-MA 50/50 wt % blend. From the change of tan ${\delta}$ observed, we confirmed the increase of miscibility in this blend system by chemical reaction between PA6 and PP-g-MA. Blends of good impact resistance could be obtained when the PP-g-MA particles of $2{\mu}m$ was dispersed in the PA6 matrix.

Extraction of Ground Points from LiDAR Data using Quadtree and Region Growing Method (Quadtree와 영역확장법에 의한 LiDAR 데이터의 지면점 추출)

  • Processing of the raw LiDAR data requires the high-end processor, because data form is a vector. In contrast, if LiDAR data is converted into a regular grid pattern by filltering, that has advantage of being in a low-cost equipment, because of the simple structure and faster processing speed. Especially, by using grid data classification, such as Quadtree, some of trees and cars are removed, so it has advantage of modeling. Therefore, this study presents the algorithm for automatic extraction of ground points using Quadtree and refion growing method from LiDAR data. In addition, Error analysis was performed based on the 1:5000 digital map of sample area to analyze the classification of ground points. In a result, the ground classification accuracy is over 98%. So it has the advantage of extracting the ground points. In addition, non-ground points, such as cars and tree, are effectively removed as using Quadtree and region growing method.

Molecular Cloning and Identification of Novel Genes, Gomsin, Characteristically Expressed in Snailfish, Liparis tanakae (꼼치에서 특징적으로 발현되는 새로운 유전자 곰신의 분리 및 동정)

  • In order to obtain the specific genes of snailfish a subtracted cDNA library was constructed, and analysed by sequencing and GenBank search. Among them C90-171 clone was turned out to be genes showing low homology and nonredundant genes. This novel clone was named Gomsin(C90-171). Gomsin was shown to be intensely expressed in the epithelial cells, some mesenchymal cells, and sheaths of muscle bundles in the result of immunohistochemistry. In the cross reaction assay of Gomsin antibody against various human tissues, the Gomsin was strongly expressed in the ductal and acinar cells of salivary glands, which was similar to the expression patterns of proline-rich proteins(PRPs) of human. The antibody raised against the Gomsin was clearly cross-reacted with human salivary PRPs and also recombinant proteins of human PRPs in the Western blot and immunoprecipitation analysis. Contrast to the salivary PRPs, the Gomsin was not easily degraded in the mixed saliva, but rapidly attacked on the cultured keratocytes in vitro. The simulated protein structure of Gomsin was similar to the whorled pattern of PRPs, even though the amino acid sequence of Gomsin was quite different from those of PRPs. These data suggest that the Gomsin is a characteristic matrix protein in the skin and body of snailfish, which is also utilized for the tissue protection in the similar way to the PRPs of human muco-secretory organs.
