Parentage Testing for the Offspring Produced by Embryo Transfer with Frozen Embryo in the Dog (개에서 동경 수정란 이식 후 생산된 산자의 친자감별)

  • The dornor, 2 years old, 20kg and mixed breed, was bred naturally on day 1 and day 3 of estrus and eight gastrulae were collected by flushing the uterus of the donor after laparatomy on day 13 after the second mating. The embryos were frozen by programmable freezer and preserved for about 3 months in liquid nitrogen. Another bitch in natural estrus, 2 years old, 30kg, mixed breed, was selected as the recipient and the frozen embryos(8 gastrulae) were thawed and each 4 embryos were transferred into upper partr of left and right uterine horn, respectively, on day 13 after the proper mating day determined by vaginal smear. The ecipient delivered 6 offspring 48 days after embryo transfer. Of 6 puppies, one was still birth and two puppies died one month after birth. Parentage test was performed by DNA analysis using microsatellite sequences for 3 puppiers, the recipient, the donor, the male dog bred with the donor, and the male dog raised near to the recipient. The markers selected for the test were CXX 873(133-157 base pair) and CXX 894(141-165 base pair). Using primers manufactured according to the markers, the blood samples were processed for polymerase chain reaction and the PCR products were treated for electrophoresis. The three puppies showed identical band to that of recipient, consequently, it was concluded that the puppies were offspring of the recipient mated naturally by the male dog, not the offspring by embryo transfer.

  • In this study, the stiffness of an LMU (Leg Mating Unit), which is a device required for installing the top side part of an offshore structure, was examined through structural analysis. This unit is mounted on the supporting point of the structure and is used to absorb the shock at installation. It is a cylindrical structure with an empty center. To support the vertical load, elastomeric bearings (EBs) and iron plates are laminated in layers. The stiffness of the EBs is basically influenced by the size of the bearings, but it varies with the number of laminated sheets inside the same sized structure. The relationship between the stiffener and the compressive stiffness is investigated, and its design is suggested. The stiffness of the EBs is analyzed by calculating the reaction force, while controlling the displacement. First, the relationship between the size of the reinforcing plate and the compressive stiffness is considered. Next, the relationship between the number of stacked reinforcing plates and the compression stiffness is considered. Different loads are required for each installed point. The goal is to design the compression stiffness in such a way that the same deformation occurs at each point in the analysis. In this study, ANSYS is used to perform the FE analysis.

Population Structure of the Gibberella fujikuroi Species Complex Associated with Rice and Corn in Korea

  • Several species belonging to the Gibberella fujikuroi species (Gf ) complex are commonly associated with rice and corn, not only causing serious diseases, but also producing fumonisins, a group of mycotoxins harmful to animals and humans. To characterize the population structure of the putative fumonisin-producing Gf complex in Korea, we obtained 276 candidate isolates from rice and corn harvested in 2009 and 2010 by diagnostic polymerase chain reaction with several specific primer sets. Phylogenetic trees were constructed using multilocus sequences (combined RPB2 and EF1A, totaling 1.6 kb) from these isolates. Among the 135 isolates from rice, F. fujikuroi (teleomorph: G. fujikuroi; 59.3%) and F. proliferatum (G. intermedia; 13.3%) were predominant, followed by F. concentricum (5.9%). Additionally, twenty-five (18.5%) rice isolates belonged in a distinct subclade of F. commune, a non-member of the Gf complex. In contrast, F. verticillioides was the most predominant species (38.3%) among the 141 corn isolates, and followed by F. fujikuroi (27.7%), F. proliferatum (14.9%), F. subglutinans (7.1%), and F. concentricum (2.8%). A single mating type (MAT1-1) was found predominantly among the Gf complex isolates examined. Possible distinct subclades were detected within the populations of F. fujikuroi and F. proliferatum; however, this needs further confirmation. This is the first reported population-level characterization of putative fumonisin-producing Gf complex associated with rice and corn in Korea.

  • Ceramic on ceramic total hip prostheses are developed to apply to young patients because lifetime of polyethylene joint prostheses is limited by loosening due to biological response. As mating faces of all-ceramic joint must be highly conformed to reduce stress concentration, wear properties of flat surfaces are investigated in this study. Through wear tests at 2 MPa of contact pressure and 36 mm/s of sliding velocity, alumina and silicon carbide keep low wear rate, high hardness and smooth surface. Soft surface film was detected after the test in bovine serum. This suggests that boundary lubrication is effective to reduce wear in all-ceramic joint.

Development of Genetic Markers for Triploid Verification of the Pacific Oyster, Crassostrea gigas

  • The triploid Pacific oyster, which is produced by mating tetraploid and diploid oysters, is favored by the aquaculture industry because of its better flavor and firmer texture, particularly during the summer. However, tetraploid oyster production is not feasible in all oysters; the development of tetraploid oysters is ongoing in some oyster species. Thus, a method for ploidy verification is necessary for this endeavor, in addition to ploidy verification in aquaculture farms and in the natural environment. In this study, a method for ploidy verification of triploid and diploid oysters was developed using multiplex polymerase chain reaction (PCR) panels containing primers for molecular microsatellite markers. Two microsatellite multiplex PCR panels consisting of three markers each were developed using previously developed microsatellite markers that were optimized for performance. Both panels were able to verify the ploidy levels of 30 triploid oysters with 100% accuracy, illustrating the utility of microsatellite markers as a tool for verifying the ploidy of individual oysters.

Prevalence of Feline Blood Types in Seoul and Kangwon Area of Korea (서울 및 강원지역 고양이의 혈액형 빈도 조사)

  • To determine the distribution of feline blood types and then to estimate the risk of neonatal isoerythrolysis (NI) in non-pedigree cats, we typed blood of 482 cats of both genders and various breeds (336 domestic shorthair cat and 146 pedigree) from August 2005 through July 2007. Blood samples from Seoul and Kangwon province were typed within 5 days after collection by the simple tube method. High-titer anti-A antiserum and anti-B reagent, prepared with Triticum vulgaris lectin, were used to determine type A and type B blood, respectively. The majority of cats were type A (n = 465, 96.5%) and only 3.5% (n = 17) were type B. No type AB blood were detected. Blood type distributions among the non-pedigree and pedigree cats were similar: for non-pedigree cats, 96.4% were type A and 3.6% were type B, whereas for pedigree cats, 96.6% were type A and 3.4% were type B. All type B cats had a very strong agglutination reaction to anti-A antiserum: 8 sample for 3+ and 9 for 4+. Assuming 19% of estimated frequency for the type-B allele in domestic cats, the calculated proportion of random mating from this population at risk for developing NI was 3.4%. Based on this finding, it is strongly recommended that blood typing be performed prior to any blood transfusion or breeding to minimize blood type incompatibilities. Further comprehensive studies on the titer of naturally occurring antibodies in cat populations in Korea and the prevalence of possible NI in practice are clearly required.

Assessment of the Ability of Extracellular Enzyme Production in Hybrid Strains of Lentinula edodes by Chromogenic Reaction-based Plate Assay (발색반응 분석법을 이용한 표고 교배균주의 세포외효소 분비 능력 평가)

  • Shiitake breeding requires the procedures of mating of two different parental strains and selection of hybrid strains that have good traits for the mushroom production. In this study, we tested the possibility of the use of chromogenic plate-based assay for extracellular enzyme production in order to assess and find good biochemical properties-possessed hybrid strains that were generated from genetic cross of the monokaryotic strains derived from two different parental strains of Lentinula edodes Sanjo-101ho and Sanjo-108ho. We observed that there was difference in the ability of producing ${\beta}$-glucosidase, avicelase, CM-cellulase, amylase, pectinase, xylanase, and protease among the monokaryotic strains. We could also comparatively assess that the ability of the seven extracellular enzymes production in the hybrid strains depended on the mating combination of the monokaryotic strains. Our results demonstrate that the assessment method for extracellular enzyme production using chromogenic plate assay could be usefully applied to the assessment of the hybrid strains derived from the breeding procedure of L. edodes.

Expression of in vitro-tested ribozyme against cucumber mosaic virus RNA in tobacco plant (시험관내에서 합성한 오이모자이크 바이러스 RNA단편을 성공적으로 절단한 ribozyme의 식물체내의 발현)

  • In vitro-tested ribozyme against synthesized cucumber mosaic virus (CMV) RNA (Agric. Chem. & Biotech. 37:56-63(1994)) was expressed in tobacco plant to develop virus resistant plants. The ribozyme sequence was linked to cauliflower mosaic virus 35S promoter and nopaline synthase(nos) terminator and this chimeric 35S-ribozyme-nos gene was sequenced. The sequenced chimeric gene was transferred to Agrobacterium tumefaciens LBA4404 using tri-parental mating system. The E. coli HB101 containing chimeric gene was incubated with E. coli HB101(pRK2073) as a helper and Agrobacterium tumefaciens LBA4404. Then Agrobacterium cells containing the ribozyme construct was cocultivated with tobacco leaf pieces. Ten different plants were regenerated from kanamycin containing MS medium. The presence of the ribozyme construct in the transgenic tobacco plants was confirmed by polymerase chain reaction (PCR). Seven different transgenic plants in ten different kanamycin resistant plants showed the expected size (570 base pairs) of 35S-ribozyme-nos gene fragment. Total RNAs were isolated from four different transgenic plants and separated on a 1% agarose gel containing formamide. Northern hybridization with 35S-ribozyme-nos gene fragment as a probe indicated that ribozyme transcripts may be degraded tv nuclease. Therefore, nuclease-resistant ribozymes are needed for the development of virus-resistant transgenic plants using ribozymes.

  • In the assembling process by industrial robots, many difficulties stem from the fact that the assembly operation is impossible or the parts to be assembled can be damaged by reaction forces due to even little misalignment in part mating. In this paper a flexible and sensible robot wrist is developed to make possible the precision insertion operation. The flexibility of the developed wrist were evaluated both analytically and experimentally in actual insertion process. The results show that without the use of feedback control the wrist is capable of doing insertion operations with a small clearance at a low inserting force. For smaller clearance the assembly process was devised involving insertion force feedback and a control algorithm for this active accommodation was developed. The simulation results show that if the active feedback control is used the insertion action can be performed with much less force, as compared with a passive accommodation method.

A Study on Corrosive Wear Characteristics and the Mechanism of Austempered Low-Alloy Ductile Iron (오스템퍼링 한 저합금구상흑연주철의 부식마멸특성 및 그 기구에 관한 연구)

  • This study was undertaken to investigate the corrosive wear charateristics upon various transformation condition of austempered low-alloy ductile cast iron in corrosive environments against mating specimen made of the hardened SM45C. The corrosive wear test was carried out by rubbing the annular surface of two test pieces in distilled water and aqueous solution at constant sliding speed of 0.5m/s. In severe wear region, the corrosive wear rate Wc increased hastily with NaCl concentration owing to intermetallic adhesion but Wc went down slowly in mild wear region due to lubricating effect of the corrosion product. The critical sliding distance decreased with increasing NaCl concentration due to increased generation rate of the corrosion product and the specific corrosive wear rate has maximum in 1% NaCl aqueous solution at mild wear region. With the variation of matrix, the corrosive wear resistance of the fine acicular bainite was higher than that of coarse upper bainite because of reducing the local cell reaction by carbides. A growth in volume fraction of retained austenite in matrix increased the Wc due to soften surface, but has a declining tendency of Wc in mild wear region.
