Characteristics of Thiamine Uptake by the BeWo Human Trophoblast Cell Line

  • Little is known concerning the mechanisms responsible for the transplacental transfer of thiamine. So, the aim of this work was to characterize the placental uptake of thiamine from the maternal circulation, by determining the characteristics of $^3H$-thiamine uptake by a human trophoblast cell line (BeWo). Uptake of $^3H$-thiamine (50-100 nM) by BeWo cells was: 1) temperature-dependent and energy-independent; 2) pH-dependent (uptake increased as the extracellular medium pH decreased); 3) $Na^+$-dependent and $Cl^-$-independent; 4) not inhibited by the thiamine structural analogs amprolium, oxythiamine and thiamine pyrophosphate; 5) inhibited by the unrelated organic cations guanidine, N-methylnicotinamide, tetraethylammonium, clonidine and cimetidine; 6) inhibited by the organic cation serotonin, and by two selective inhibitors of the serotonin plasmalemmal transporter (hSERT), fluoxetine and desipramine. We conclude that $^3H$-thiamine uptake by BeWo cells seems to occur through a process distinct from thiamine transporter-1 (hThTr-1) and thiamine transporter-2 (hThTr-2). Rather, it seems to involve hSERT. Moreover, chronic (48 h) exposure of cells to caffeine ($1\;{\mu}M$) stimulated and chronic exposure to xanthohumol and iso-xanthohumol (1 and $0.1\;{\mu}M$, respectively) inhibited $^3H$-thiamine uptake, these effects being not mediated through modulation of the expression levels of either hThTr-1 or hSERT mRNA.

Isolated 3-Methylcrotonyl CoA Carboxylase Deficiency Detected by Newborn Screening Program Using Tandem Mass Spectrometry (탠덤매스 신생아 스크리닝 검사로 발견된 Isolated 3-Methylcrotonyl CoA Carboxylase 결핍증 1례)

  • Isolated deficiency of 3-methylcrotonyl CoA carboxylase is a rare disorder of the catabolic pathway for leucine and many patients have mild symptoms or no symptom. However, the introduction of tandem mass spectrometry in newborn screening has revealed an unexpectedly high incidence of this disorder. We report an asymptomatic premature infant with isolated 3-methylcrotonyl CoA carboxylase deficiency detected by newborn screening program using tandem mass spectrometry. She was born at preterm, 36 weeks of gestation and her birth weight was 1,912 gm. She was delivered by Cesarian section due to maternal preeclampsia and oligohydramnios. An elevation of 3-hydroxyisovalerylcarnitine in a blood sample obtained at Seven days was detected by tandem mass screening. Massively elevated excretion of 3-hydroxyisovalerate and 3-methylcrotonylglycine was detected in the urine collected at 15 days. L-carnitine(100 mg/kg/day) was administrated orally to correct sencondary carnitine deficiency. Carnitine is conjugated with metabolites, to decrease the potential toxic effects. She is asymptomatic to date, and her growth and development are within normal limits.

Influence of Fatigue, Depression and Anxiety on Quality of Life in Pregnant Women with Preterm Labor (조기진통 임부의 피로, 우울과 불안이 삶의 질에 미치는 영향)

  • Purpose: The purpose of this study was to identify effects of fatigue, depression and anxiety on quality of life in pregnant women with preterm labor. Methods: With a survey design, data were collected from 138 mothers who were admitted at a hospital in Seoul, between June 2014 and September 2015. Instruments used to collect the data for the study were: Fatigue Continuum Form, Depression anxiety stress scale (DASS-21) and maternal postpartum quality of life (MAPP-QOL). Results: The mean fatigue score was 68.30 with 50.7% of women being depressed and 79.7% of the 138 women being anxious. The mean quality of life was 18.92 with quality of life being associated with fatigue, depression and anxiety. Depression and fatigue explained 26% of the variance in quality of life. Conclusion: Depression and fatigue adversely affected women's quality of life. It is important to address appropriate management of depression and fatigue in order to improve quality of life in pregnant women with preterm labor.

The Effects of Dietary Iodine Intake on the Postpartum Thyroiditis(PPT) Manifestation (산모의 요오드섭취가 산후 갑상선염 발현에 미치는 영향)

  • Iodine-rich seaweed soup has been traditionally supplied to postpartum women in Korea. This dietary habit might introduce over-intake of iodine above the recommended requirements, and might provoke postpartum thyroid dysfunction. Although the response to excess iodine intake is highly variable, goiter, hyperthyroidism, hypothyroidism, and thyroiditis could follow the daily intake of 1,500$\mu\textrm{g}$ of iodine. A few studies are available concerning iodine toxicity in Korea. The purpose of this study was to investigate the relationships between the dietary intake of iodine and thyroid function change as well as the incidence of postpartum thyroiditis. One hundred and thirty-seven postpartum women who had experienced normal deliveries were studied. Dietary intake of iodine and excretion concentration of iodine in breast milk and maternal urine were measured . Serum T$_3$, T$_4$, TSH, anti-thyroglobulin antibody, and anti-microsomal antibody were anlayzed 1 week before delivery and 1, 6, 12, and24 weeks after delivery. Iodine intake was analyzed by one-to-one interviews using 24-hr recall and a food frequncy questionnaire. The result showed that the intake of dietary iodine before delivery and 1 and24 weeks after delivery were 483$\mu\textrm{g}$/day, 3367$\mu\textrm{g}$/day, and 1069$\mu\textrm{g}$/day, respectively. The concentration of iodine in urine at the first week after delivery was 63$\mu\textrm{g}$/dL, and 23.9$\mu\textrm{g}$/dL in breast milk . The levels of serum T$_3$ and T$_4$ before delivery were 2.01ng/mL and 11.49$\mu\textrm{g}$U/dL, respectively, showing that the levels were gradually dropping to normal values after delivery. Positive serum anti-thyroglobulin antibody and anti-microsomal antibody appeared in 3 cases. After a 24 week follow-up period , 6 women(10.3%) experienced cases of postpartum thyroiditis, 5 of which were cases of hyperthyroidism and one of which was a case of hypothyroidism. These figures of postpartum thyroiditis are similar to those of other countries.

Changes in Maternal Blood Glucose and Plasma Non-Esterified Fatty Acid during Pregnancy and around Parturition in Twin and Single Fetus Bearing Crossbred Goats

  • The effects of fetal number (single or twin) on blood glucose and plasma NEFA during pregnancy and around parturition were studied on ten Alpine ${\times}$ Beetal crossbred goats in their first to third lactation. The animals were divided in-groups 1(carrying single fetus, n=4) and 2(twin fetus, n=6). The samples were drawn on day1 after estrus and then at 14 days interval (fortnight) for 10 fortnights. Around parturition the samples were taken on days -20, -15, -10, -5, -4, -3, -2, -1 prior to kidding and on day 0 and +1, +2, +3, +4, +5, +10, +15, +20 days post kidding. In twin bearing goats the blood glucose concentration continued to increase from 1st until 4th fortnight and thereafter gradually decline from 5th upto 8th fortnight. In single bearing goats there was increase in levels from 2nd upto 4th fortnight and thereafter it declined from 5th uptill 9th fortnight. The difference in sampling interval was highly significant (p<0.01) in both the groups. However the values were higher in single than in twin bearing goats. The plasma NEFA concentration was low in both the groups' upto 4th fortnight and thereafter it is continuously increased upto 9th fortnight. During prepartum period the blood glucose was higher in single than in twin bearing goats. The values were minimum on the day of kidding in both the groups. During postpartum period the values were significantly (p<0.01) higher in twin than in single fetus bearing goats. The plasma NEFA was significantly (p<0.05) higher in twin than in single fetus bearing goats. The blood glucose and plasma NEFA concentration can be used as index of nutritional status during pregnancy and around parturition in goats.

  • Data spread over 11 years (1986-1996) pertaining to a synthetic population developed by inter se mating of half-breds of Corriedale and Russian Merino with Nali maintained at CCS Haryana Agricultural University, Hisar were utilized for the estimation of genetic parameters for growth traits. The means for birth weight (BWT), weaning weight (WWT), six month body weight (SWT), yearling weight (YWT), preweaning daily gain (PRW) and post weaning daily gain (POW) were 3.35 kg, 10.79 kg, 13.28 kg, 18.96 kg, 82.6 gm and 27.6 gm, respectively. The effects of year and season of birth and sex of lamb were significant for all the traits under study except the effect of season of birth for WWT, SWT and PRW. No definite trend was observed over the years for the averages of body weight and gain. Lambs born during the spring season performed better for BWT, WWT and PRW while the performance of lambs born during autumn was better for the other traits included in the study. The male lambs were heavier than the females for body weight at all stages and gain in weight. The heritability estimates for WWT and PRW were low; for BWT and SWT were moderate and for YWT and POW were high. Birth weight had high heritability and high genetic correlations with subsequent body weights and gains but due to the presence of a maternal effect on BWT and WWT, a sequential selection procedure is recommended for the improvement of growth rate in sheep.

A Study on the Family Planning Practice of Some Urbanites (일부 도시지역 주민의 가족계획 실천에 관한 연구)

  • The Family Planning Project as a birthcontrol policy has been enthusiastically carried out by the government since 1962. But gradually it get less enthusiastic. Therefore, it is required to establish a more comprehensive and systematic plan and to carry out it thoroughly. And it is needless to say that people's knowledge about family planning, their attitudes and their practice should be concretly comprehended. Taking these things into consideration, this study surveys the general situation on family planning among 237 married women of less than 34 years in Guro 6-dong, one of target areas for Korea University Health Project from Sep. 17, 1984 to Oct. 13, 1984. The results are as follows; 1) Most of the subjects (62.8%) want 2 children and 13.6% want one child. But son-preference consciousness is remarkably revealed among them, which is thought to cause social inequality between man and woman. Therefore, it needs to change the traditional son-preference convention, for equality of all men regardless of sex. 2) The rate of induced abortion experience by person is 61.6% and the average frequency is 1.4. Almost all the induced abortion are carried out at clinics and hospitals during 2~3 months after conception. To prevent these unnecessary conception and induced abortion in view of maternal health, ethics and economics, proper contraceptive measures should be emphasized. 3) Temporary contraceptives should be sold more widely in the basis of free trade in order to practice the intensive and comprehensive family planning because the degree which Health Center has been utilized for buying temporary contraceptives is low. 4) There are serious problems such as the side effects and the lack of follow up care in permanent contraceptives. Those lower the practice rate of permanent contraception. Therefore it is necessary to improve the quality of permanent contraception practice and strengthen the follow-up care.

Influences of Parental Pairs on Progeny Sex Ratios of Nile Tilapia Oreochromis niloticus (틸라피아 Oreochromis niloticus의 성비 결정에 미치는 암수어미의 영향)

  • Sex of the Nile tilapia Oreochromis niloticus is mainly determined by an XX/XY system. However, accumulating evidences suggest the existence of additional sex modifying factors including environmental, autosomal and parental influences. In order to investigate the possibility of parental effects on sex ratios of tilapia progenies, in this study, a series of crosses was carried out using gynogenetic clonal fish, neomales, normal males and females, and YY fish. Crosses between clonal XX male and clonal female have yielded only female progenies and no parental influences were observed. However, in the crosses between clonal males and normal females, female parents were significantly associated with the progeny sex ratios ($X^2$=20.046, 7 d.f., p<0.01). Progeny sex ratios from the crosses between neomales and normal females ($X^2$=60.491, 5 d.f and $X^2$=28.072, 2 d.f.) also showed significant association with female parents (P<0.001). The stability of progeny sex ratios from repeated spawns were confirmed by using 6 different parental pairs. In 16 crosses between normal males and normal females, sex ratios of progenies showed clear maternal influences, and further analysis of the results revealed a negative correlation ($r^2$=0.7718, p<0.05) between the sex ratios of progenies from two different males, indicating a strong paternal influence. No statistically significant relationship between survival rates and sex ratios of progenies was observed in any genotypic groups. Taken together, the influence of parental pairs on progeny sex ratios in this species is evident although the cause of this influence is not clear.

Associations of Family Income and Ethnicity with Parents' Selection of Infant Care (가족소득 및 인종에 따른 영아 보육 선택)

  • In present study, using longitudinal data of 161 families from Durham Child and Development Study, we examine the characteristics of infant child care chosen by families of different income levels and ethnicity in the U.S. Time-related changes in child care selection for the first year of the child's life were also investigated. Infants from high-income families were more likely to be cared for by someone other than the mother, compared to infant from lower-income families. African-American families were more likely to use non-maternal care for their infant compared to European-American families. Although the effects of income on type of care were not significant, descriptively, more low-income families used relative care while high-income families used more formal arrangements such as daycare center. Infants from low-income families began care earlier and low-income families paid less for care than other families. The findings call for attention to the importance of child care policy for low-income families as well as further research examining the function of cultural differences in selection of infant child care.

Phosphorylation Status of RNA Polymerase II Carboxyl-terminal Domain in Porcine Oocytes and Early Embryos

  • Fertilization of the oocyte commences embryogenesis during which maternally inherited mRNAs are degraded and the embryonic genome is activated. Transcription of embryonic mRNA is initiated by embryonic genome activation (EGA). RNA polymerase II (RNA Pol II) is responsible for the synthesis of mRNAs and most small nuclear RNAs, and consists of 12 subunits, the largest of which characteristically harbors a unique C-terminal domain (CTD). Transcriptional activity of RNA Pol II is highly regulated, in particular, by phosphorylation of serine residues in the CTD. Here, we have shown the presence of RNA Pol II CTD phosphoisoforms in porcine oocytes and preimplantation embryos. The distribution pattern as well as phosphorylation dynamics in germinal vesicles and during embryogenesis differed in developmental stages with these isoforms, indicating a role of RNA Pol II CTD phosphorylation at the serine residue in transcriptional activation during both oocyte growth and embryonic genome activation. We additionally examined the effects of the RNA Pol II inhibitor, ${\alpha}$-amanitin, on embryo development. Our results show that inhibition of polymerase, even at very early stages and for a short period of time, dramatically impaired blastocyst formation. These findings collectively suggest that the functionality of maternal RNA Pol II, and consequently, expression of early genes regulated by this enzyme are essential for proper embryo development.
