Incidence and Causes of Sub-Clinical Mastitis in Dairy Cows on Smallholder and Large Scale Farms in Tropical Areas of Tanzania

  • A study on the prevalence and causes of sub-clinical mastitis was conducted on ten smallholder and large-scale dairy farms in Morogoro urban and peri-urban areas. A total of 65 lactating cows were screened using the California Mastitis Test (CMT). Confirmatory tests used included; the direct microscopic somatic cell count (DMSCC), culture, bacteriological and biochemical tests. Structured questionnaires were used to collect information on management aspects. Results showed 62% and 4% cows as sub- clinical and clinical mastitis cases respectively. Levels of infection were higher on smallholder farms (75%) than on large-scale farms (25%). All tested cows had high cell counts (>500,000) per ml of milk. Incidences of mastitis were significantly (p<0.05) related to milking practices. The dominant bacterial isolates in the same order were Staphylococcus aureus, Streptococcus spp, and Escherichia coli. Other organisms isolated included Pseudomonas spp and Klebsiella spp. It was concluded that the high rates of sub-clinical mastitis in the research area were mainly due to poor management and unhygienic milking practices.

  • Mastitis is a common and serious infection of the mammary gland in dairy cattle and has a major economic impact on the production of milk and dairy products. Bacterial mastitis is caused by several pathogens and is most frequently associated with coagulase-negative staphylococci (CNS). Although CNS are typically associated with subclinical or mild mastitis, the importance of CNS has increased as these pathogens have emerged as predominant mastitis-related pathogens in many countries. CNS can cause persistent infections, resulting in increased milk somatic cell counts and thereby affecting milk quality and decreasing milk production. Globally, Staphylococcus chromogenes, S. epidermidis, and S. simulans are the predominant CNS species in dairy cattle mastitis. Antibacterial resistance of CNS varies with species, and most CNS are susceptible to vancomycin and resistant to penicillin and tetracycline. As the most frequently isolated CNS species, some strains of S. chromogenes exhibit phenotypic resistance to ampicillin, erythromycin, oxacillin, penicillin, and tetracycline. Some strains of S. epidermidis and S. haemolyticus are only susceptible to vancomycin and rifampicin. Therefore, more studies are needed to achieve the control and prevention of CNS as environmental pathogens.

  • Various kinds of antibiotic intramammary infusions are used for treatment of bovine mastitis. As antibiotic-resistant bacteria are increased, the therapeutic rate for bovine mastitis is decreased. The goal of this research is to detect significant synergic effects of combination of antibiotics with sulbactam, $\beta$-lactamase inhibitor, on methicilin-resistant Staphylococcus aureus (MRSA). We used 5 strains of MRSA isolated from bovine mastitis with clinical and subclinical signs. All of the bacteria isolated had resistance to oxacillin and showed multi-resistant patterns in the antimicrobial susceptibility tests. Minimal bactericidal concentrations of ampicillin, amoxicillin, cephalexin, ampicillin/sulbactam(2:1), amoxicillin/sulbactam (2:1), and cephalexin/sulbactam (1:1) were measured according to broth microdilution method suggested by National Committee for Clinical Laboratory Standards (NCCLS, M31-A2) to compare the synergic effects of sulbactam combination with each antibiotic alone. Ampicillin and amoxicillin showed synergic antibacterial activity to 4 and 3 respectively in 5 strains of MRSA in combination with sulbactam. This study demonstrates that ampicillin/sulbactam and amoxicillin/sulbactam can be therapeutic choices for mastitis associated with MRSA.

양측성 육아종성 유방염의 세침흡인 세포학적 소견 - 1예 보고 - (Fine Needle Aspiration Cytology of Bilateral Granulomatous Mastitis - A Case Report -)

  • Granulomatous mastitis is a rare inflammatory breast lesion, usually presented with rapidly enlarging palpable mass in young parous women and can simulate carcinoma. Unnecessary surgical procedure can be avoided if the nature of the lesion were defined by fine needle aspiratio(FNA) cytology. We experienced a case of bilateral granulomatous mastitis diagnosed by FNA cytology. The patient was a 31-year-old woman with one month history of a rapidly enlarging mass in the left breast, considered clinically to be malignant. After 4 years, she presented with a palpable mass in the right breast. The FNA cytology smears contained numerous aggregates of epithelioid histiocytes admixed with Langhans' and foreign body giant cells, lymphoytes, neutrophils and apoptotic debris leading to a diagnosis of granulomatous mastitis. The subsequent lumpectomy of the left breast confirmed the diagnosis of granulomatous mastitis. The FNA cytology smears from right breast showed identical cytologic findings.

  • A total of 2,053 quarter milk samples of 518 dairy cows from 46 herds were examined for mastitis and results obtained were as follows: 1. A total of 428 quarters(20.8%) of 271 dairy cows(52.3%) from 41 herds(89%) were found to be infected with mastitis. 2. It was found that 71 quarters(3.5%) of 41 cows(7.9%) from 21 herds(45.6%) were clinical mastitis and 357 quarters(17.3%) of 230 cows(44.4%) from 20 herds (43.4%) were subclinical mastitis. 3. The main causative organisms of the mastitis were Staphylococcus aureus(26%), Streptococcus uberis(16%), and Streptococcus agalactiae(10%). 4. The majority of causative organisms were very resistant to colistin(85.9%), penicillin(67%), and streptomycin(39%). Most of the other drugs were highly effective as inhibitor for the most causative organisms in vitro. However sulfisoxazole was moderatively effective against them.

전북지방(全北地方) 젖소의 이상유(異常乳) 발생상황(發生狀況)과 원인균(原因菌)에 관한 연구(硏究) (Studies on Incidence and Causative Agents of Abnormal Milk among Dairy Cattle in Jeonbug Area)

  • A total of 364 quarter milk samples of 91 dairy cattle from 4 area around Jeonju in Jeonbug area were examined for infection rate and causative abets of mastitis by the Laboratory Procedures used in the Connecticut Mastitis-Control Program. The results obtained were as follows: 1. One hundred and thirty quarters (35.7%) from 67 cows (73.6%) were found to be infected with mastitis. It was found that 5 (1.37%) of the infected quarters were clinical mastitis and all of the rest were subclinical mastitis (125 quarters). 2. The main causative agents were found to be Staphylococcus aureus (46 quarters), Streptococcus agalactiae (49 quarters), streptococcus disgalactiae (18 quarters), Steptococcus uberis (13 quarters). Corynebacteria, Coliform organisms, Pseudomonas were also occasionally found to be causative organisms. 3. Having examined the number of infection Quarter per head of mastitis dairy cattle samplings, major findings were that the average number of infection quarter to total dairy cattle examined was 1.4 quarters, and that average number of infection quarter to the infected cattle was 1.9 quarters.

유선염(乳腺炎) 의 한방(韓方) 외치법(外治法)에 대한 문헌적고찰(文獻的考察) (A bibliographic study on the external therapy of Acute Mastitis(乳癰Yu-ong))

  • Acute Mastitis(急性乳腺炎=乳癰) is an acute suppurative disease appearing in the breast. It is mostly caused by galactostasis, the stagnation of liver-qi(肝鬱) and stomach-heat(胃熱). It is mostly found in breast feeding women, mostly in primiparae. It usually appears in the third and the fourth weeks of postpartum. At the beginning in the chest there appear induration, distending pain, galactostasis, inversion to cold with fever followed by the enlargement of the masses, burning red, megalgia, not subduing of chills and fever, the accumulation of it to form pus. In the early phase it is advisable to follow the therapeutic principles of soothing the liver and clearing away heat, promoting lactation and subduing swelling. After the formation of pus it is advisable to cut radially and drain the pus. External Therapy(外治法) generally refers to all the methods to treat various diseases and symptoms with application of drugs and manipulation or together with proper instruments on the body surface except the method of taking medicine orally Acute Mastitis has been regarded as surgery or the method of taking medicine orally But, based on many bibliographies, acute mastitis was treated by external therapy. Thus through the historical bibliographic studies of external therapy about acute mastitis, this study was made to help the expanding of the methods of treating acute mastitis.

젖소의 유방염(乳房炎) 검진(檢診)을 위한 Modified Whiteside Test법(法)과 California Mastitis Test법(法)의 비교시험(比較試驗)에 관(關)하여 (Some Observations on the Comparison of the Modified Whiteside Test and the California Mastitis Test for the Detection of Bovine Mastitis)

  • The diagnostic effects of the Modified Whiteside Test(M.W.T.) and the California Mastitis Test(C.M.T.) for detection of bovine mastitis were studied using 372 quarter milk samples collected from six dairy herds in suburbs of Suwon city in order to compare the effectiveness of the two methods. The leucocyte count and bacteriological observation were also performed to support analysing the results. The results obtained were as follows; 1. The CMT showed more specificity than MWT for leucocytes in the abnormal milk. 2. When comparison was based on Identical ratings, agreement was obtained in 56.4% of the samples. The stronger reaction had the higher agreement and the CMT was found to be more specific than MWT in detection of the sub-clinical mastitis. 3. The CMT reaction was instantaneous and sufficiently clear-cut. 4. The results of the CMT leucocyte count and bacteriological observation were completely agreed in 69.9% of the samples. When degrees of reaction and the leucocyte count were considered, the CMT showed agreement in 93.5% of the samples.

Genome-wide DNA methylation pattern in a mouse model reveals two novel genes associated with Staphylococcus aureus mastitis

  • Objective: Staphylococcus aureus (S. aureus) is one of the major microorganisms responsible for subclinical mastitis in dairy cattle. The present study was designed with the aim to explore the DNA methylation patterns using the Fluorescence-labeled methylation-sensitive amplified polymorphism (F-MSAP) techniques in a S. aureus-infected mouse model. Methods: A total of 12 out-bred Institute of Cancer Research female mice ranging from 12 to 13 weeks-old were selected to construct a mastitis model. F-MSAP analysis was carried out to detect fluctuations of DNA methylation between control group and S. aureus mastitis group. Results: Visible changes were observed in white cell counts in milk, percentage of granulocytes, percentage of lymphocytes, CD4+/CD8+ ratio (CD4+/CD8+), and histopathology of mice pre- and post-challenge with S. aureus. These findings showed the suitability of the S. aureus-infected mouse model. A total of 369 fragments was amplified from udder tissue samples from the two groups (S. aureus-infected mastitis group and control group) using eight pairs of selective primers. Results indicated that the methylation level of mastitis mouse group was higher than that in the control group. In addition, NCK-associated protein 5 (Nckap5) and transposon MTD were identified to be differentially methylated through secondary polymerase chain reaction and sequencing in the mastitis group. These observations might play an important role in the development of S. aureus mastitis. Conclusion: Collectively, our study suggests that the methylation modification in Nckap5 and transposon MTD might be considered as epigenetic markers in resistance to S. aureus-infected mastitis and provided a new insight into S. aureus mastitis research in dairy industry and public health.

Mastitis Detection by Near-infrared Spectra of Cows Milk and SIMCA Classification Method

  • Mastitis is a major problem for the global dairy industry and causes substantial economic losses from decreasing milk production and considerable compositional changes in milk, reducing milk quality. The potential of near infrared (NIR) spectroscopy in the region from 1100 to 2500nm and chemometric method for classification to detect milk from mastitic cows was investigated. A total of 189 milk samples from 7 Holstein cows were collected for 27 days, consecutively, and analyzed for somatic cells (SCC). Three of the cows were healthy, and the rest had mastitis periods during the experiment. NIR transflectance milk spectra were obtained by the InfraAlyzer 500 spectrophotometer in the spectral range from 1100 to 2500nm. All samples were divided into calibration set and test set. Class variable was assigned for each sample as follow: healthy (class 1) and mastitic (class 2), based on milk SCC content. The classification of the samples was performed using soft independent modeling of class analogy (SIMCA) and different spectral data pretreatment. Two concentration of SCC - 200 000 cells/ml and 300 000 cells/ml, respectively, were used as thresholds fer separation of healthy and mastitis cows. The best detection accuracy was found for models, obtained using 200 000 cells/ml as threshold and smoothed absorbance data - 98.41% from samples in the calibration set and 87.30% from the samples in the independent test set were correctly classified. SIMCA results for classes, based on 300 000 cells/ml threshold, showed a little lower accuracy of classification. The analysis of changes in the loading of first PC factor for group of healthy milk and group of mastitic milk showed, that separation between classes was indirect and based on influence of mastitis on the milk components. The accuracy of mastitis detection by SIMCA method, based on NIR spectra of milk would allow health screening of cows and differentiation between healthy and mastitic milk samples. Having SIMCA models, mastitis detection would be possible by using only DIR spectra of milk, without any other analyses.
