IgE-dependent histamine-releasing factor (HRF) is found extracellularly to regulate the degranulation process of histamine in mast cells and basophils and known to play a predominant role in the pathogenesis of chronic allergic disease. HRF has been also identified in the intracellular region of the cell. Previously, we reported that HRF interacts with the 3rd cytoplasmic domain of the alpha subunit of Na,K ATPase and inhibits Na,K-ATPase activity. The predicated phosphorylation site in HRF by PKC was mapped to one serine residues (S98) by the computer analysis. In this study, we identified that S98 residue of HRF was phosphorylated using anti-HRFpS98 antibody which specifically recognizes the phosphorylated serine residue of HRF and HRFS98A mutant construct. We also performed $^{86}Rb^{+}-uptake$ assay to understand the role of HRF wild-type and HRFS98A mutants on the regulation of Na,K-ATPase activity. Dephosphorylation of HRF at serine 98 residue recovers slightly the inhibitory function of HRF, suggesting that phosphorylated HRF at serine 98 may not suppress the Na,K-hfpase activity.
Cortex Mori (Moros alba L.), the root bark of mulberry tree has been used as an autiphlogistic, diuretic and expectorant in herval medicine. Recently, a few papers reported that phenolic extract of Cortex Mori had the hypotensive, hypoglycemic, antiviral and anticancer effects, and hot water extract of Cortex Mori(CM) had inhibitory effect on the degranulation and histamine release from activated mast cells. These previous studies suggest a possibility that CM has an antidotal activity against inflammation which was mediated mainly by macrophage-secreting inflammatory factors. This study was performed to evaluate the influences of CM on carrageenan-induced edema in vivo and release of inflammatory mediators such as NO, TNF and IL-1 by macrophages stimulated with LPS or $IFN-{\gamma}$ in vitro. Subcutaneous injections of carrageenan into the mouse paw rapidly induced local edema by increasing vascular permeability, but single intraperitoneal injection of CM extract at 30 minutes before carrageenan suppressed the development of edema. NO and TNF production from macrophage stimulated by LPS or $IFN-{\gamma}$ were significantly suppressed, especially TNF secretion by up to 3-4 folds. LPS stimulated IL-1 production was also inhibited, but not significantly. Cell viability assay verified that the inhibition was not due to general cell toxicity. These results suggest that reduction of NO, TNF and IL-1 production may be one of the means by which CM prevent inflammation associated diseases.
Objectives Seunggal-tang is one of the prescriptions of oriental herbal medicine, which has been applied to several allergic diseases such as atopic dermatitis. This study was planned to compare differences of anti-allergic effects based on different form of Seunggal-tang by manufacturing differently. Methods Two types herb medicine products were used; aqueous extract (SG-T, Seunggal-Tang) and powder (SG-S, Seunggal-San) which were made from the same mixed formula of Seunggal-tang. To investigate in vitro anti-allergic activities, rat basophilic leukemia (RBL-2H3) cells were treated with SG-T and SG-S for 1 hour, and then stimulated with the phorbol 12-myristate 13-acetate (PMA) plus calcium ionophore A23187. We examined the release of beta-hexosaminidase, as a marker of degranulation, and the releases of tumor necrosis factor (TNF)-alpha and interleukin (IL)-4, as proinflammatory cytokines. Results SG-T and SG-S didn't have effects on cell viabilities in concentrations under 2㎎/㎖. In additionto that, SG-T more inhibited releasing ${\beta}$-hexosaminidase, TNF-${\alpha}$ and IL-4 than SG-S. Conclusions These results indicate that SG-T is more effective against mast cell-mediated allergic reactions than SG-S.

This experimental research has been done to study the effects of Palmul-tang on the anti-allergic reaction. We found the several important results from the research which has been performed by two experiments toward immediately type and delayed type in order to study the effects of Palmul-tang on hypersensitivity response to mice. The results obtained from our research are as following: The survival rate of one group to which we injected only the compound 48/80 is almost 0% according to its density and timing test. In the other hand. the survival rates of the other group to which we injected both of the compound 48/80 and Palmul-tang are 20%, 0%. 40%, 10%, 20% and 50% according to 0.025, 0.05, 0.1, 0.25 0.5 and 1 (mg/g) of compound 48/80. Time dependency test also shows the 30% and 20% survival rates in 5 and 10 minutes. Palmul-tang revealed the significantly inhibitory effect on Compound 48/80 induced Mast cell degranulation. Palmul-tang showed the significantly inhibitory effect in the delayed type hypersensitivity response to picry1 chloride. Palmul-tang showed the significantly inhibitory effect in the delayed type hypersensitivity response to sheep red blood cell. Our research provides the important evidence that Palmul-tang is benificial to the prevention and treatment of allergy related aiseases.

We found the several important results from the research which has been performed by two experiments toward immediately type and delayed type in order to study the effects of BJIKT on hypersensitivity response to mice. The results obtained from our research are as following: 1. The survival rate of one group to which we injected only the compound 48/80 is almost 0% according to its density and timing test. In the other hand, the survival rates of the other group to which we injected both of the compound 48/80 and BJIKT are 20%, 10%, 30%, 10%, 40%, and 70% according to 0.025, 0.05, 0.1, 0.25 0.5 and 1(mg/g) of compound 48/80. Time dependency test also shows the 10% and 0% survival rates in 5 and 10 minutes. 2. BJIKT revealed the significantly inhibitory effect on Compound 48/80 induced Mast cell degranulation. 3. BJIKT showed the significantly inhibitory effect in the delayed type hypersensitivity response to picryl chloride. 4. BJIKT showed the significantly inhibitory effect in the delayed type hypersensitivity response to sheep red blood tell. Our research provides the important evidence that BJIKT is benificial to the prevention and treatment of allergy related diseases.
Some flavonoids have spasmolytic activities in various smooth muscles, but structure-activity relationships on their spasmolytic activity and its mechanism are unclear. In this study, effects of flavones (flavone and apigenin) and flavonols (quercetin and rutin) on the rat ileal smooth muscle contraction were studied in vitro and in vitro. In the electric stimulation-induced contraction, all of four flavonoids inhibited concentration-dependently the rat ileal smooth muscle contraction induced by electric stimulation (10 mV, 0.1 cps, 0.1 msec duration), IC$_{50}$ of quercetin, apigenin, flavone and rutin were 0.98${\times}$10$^{-5}$, 1.20${\times}$10$^{-5}$, 1.55${\times}$10$^{-5}$ and 1.85${\times}$10$^{-5}$ M, respectively. Flavonoids at a concentration of 2${\times}$10$^{-5}$ M also significantly inhibited the anaphylactic contraction and decreased concentration-dependently the mast cell degranulation by anaphylactic reaction, IC$_{50}$ of quercetin, apigenin, flavone and rutin were 4.0${\times}$10$^{-5}$, 7.5${\times}$10$^{-5}$, 8.0${\times}$10$^{-5}$ and 9.5${\times}$10$^{-5}$ M, respectively. These results indicated that flavones and flavonols inhibited the rat ileal smooth muscle contraction induced by electric stimulation because of their antagonism against acetylcholine and have spasmolytic activities on anaphylactic contraction which may be due to their mast cell-stabilizing activities. Furthermore, double bond of C$_{2,3}$ in benzene ring of flavonoids may be important in the their antispasmodic activities on the rat ileal smooth muscle contraction induced by electric stimulation and anaphylactic reaction.
Purpose : To assess the histomorphologic changes in the rat lung injury induced by radiation, to determine whether captopril reduces the rat lung injury and to evaluate change in TNF-${\alpha}$ and TGF-${\beta}$ in rat lung damage by radiation and captopril Methods and material : Right lungs in male Sprague-Dawley rats were divided irradiation alone (10, 20, 30 Gy) or radiation (same dose with radiation alone group) with captopril (500 mg/L). Radiation alone group were sacrificed at twelve hours and eleven weeks after radiation and radiation with captopril group (captopril group) were sacrificed at eleven weeks after radiation with captopril. We examined the light microscope and electron microscopic features in the groups. Results : In radiation alone group, there were patch parenchymal collapse and consolidation at twelve hours after radiation. The increase of radiation dose shows more prominent the severity and broader the affected areas. Eleven weeks after radiation, the severity and areas of fibrosis had increased in proportion to radiation dose given in the radiation alone group. There was notable decrease of lung fibrosis in captopril group than in radiation alone group. The number of mast cells rapidly increased with increase of radiation dose in radiation alone group and the degree of increase of mast cell number and severity of collagen accumulation more decreased in captopril group than in radiation alone group. In radiation alone group, expression of TNF-${\alpha}$ and TGF-${\beta}$ increased according to increase of radiation dose at twelve hours after radiation in both group. At eleven weeks after radiation, expression of TGF-${\beta}$ increased according to increase of radiation dose in radiation group but somewhat decreased in captopril group. In the captopril group the collagen deposition increased but less dense than those of radiation alone group. The severity of perivascular thickening, capillary change, the number and degranulation of mast cells more decreased in the captopril group than in the radiation alone group. Conclusion : It is concluded that the effect of captopril in the rat lungs after radiation was considered to be due to its effect on inhibition of mast cells and reduction of collagen deposition, and captopril may be protect in lung damage after radiation. We observed expression of TNF-${\alpha}$ and TGF-${\beta}$ increased at the early phase after radiation and expression of TGF-${\beta}$ increased in proportion to increase of radiation dose at the chronic phase after radiation. This results will contribute to future investigation in reduction mechanism of captopril in lung damage after radiation.

Optimal conditions for extraction of bakkenolide B from Petasites japonicus leaves were determined by using response surface methodology. A second-order Box-Behnken design representing three extraction temperatures (80, 100, $120^{\circ}C$), three extraction times (30, 45, 60 min), and three solvent pH's (5, 7, 9) was executed. The efficiency of the extraction conditions was defined using the ${\beta}$-hexosamidase assay by comparing both the bakkenolide B content and its anti-allergic activity expressed as extract inhibition on degranulation. The response surface plot described for the bakkenolide B content showed that the maximum content was predicted as 121.6 ${\mu}g/g$ with extraction conditions of $127.1^{\circ}C$, 46.6 min, and pH 7.7. Extraction temperature and time were important factors in determining bakkenolide B content. Using regression analysis, correlation between the inhibition effect of mast cell degranulation and bakkenolide B content was found to be low.
Objectives : In this report, we investigated the effect of ethanol extract of Syzygium aromaticum(L.) Merr. & Perry.(SAE) on the RBL-2H3 cell-mediated allergic response and studied its possible mechanisms of action. Methods : Cytotoxicity on RBL-2H3 cell was evaluated by MTT assay. Anti-allergic activity of SAE was assessed by ${\beta}$-Hexosaminidase and Histamine secretion, ${\beta}$- Hexosaminidase and Histamine secretion were measured by ELISA assay. Evaluate the mechanisms of effect of SAE on the secretion of degranulate mediators, we examined the effect of SAE on the activation of mitogen-activated protein kinases using western blot analysis. Results : SAE had no cytotoxicity on rat basophilic leukemia cell(RBL-2H3). Moreover SAE dose-dependently inhibited RBL-2H3 cell degranulation and histamine release. SAE specifically blocked the IgE-induced p38 mitogen-activated protein kinase activation. Conclusions : Our findings provide evidence that Syzygium aromaticum ethanol extract inhibits mast cell derived allergic reaction, and also demonstrate the involvement of p38 MAPK phosphorylation.
Objectives: This study investigated the administration of Jeungmiyijin-tang (JYT) to rats with reflux esophagitis (RE) induced by pylorus and forestomach ligation operations. Methods: Twenty laboratory rats were divided into three groups with 5~7 rats in each group. The control group consisted of rats with no inflammation (CON). The RE group had rats with gastroesophageal reflux elicited by pylorus and forestomach ligation operations. The JYT group had rats that were orally administered Jeungmiyijin-tang (1.5 ml/day/300 g) once a day for 14 days before reflux esophagitis was induced by the pylorus and forestomach ligation operations. Six hours after the operations, the rats were sacrificed, morphological changes were observed, and histological examinations were done in the stomach and esophagus lesion areas. If apoptosis was observed, the apoptotic cells in the esophagus lesion areas were counted. Results: The morphological and histochemical changes consisted of various injuries from hemorrhagic erosion in the RE group, while there were significantly fewer in the JYT group. The RE group marked increases of gastric mucosa erosion and infiltration of inflammatory cells in the submucosa, as well as cell division in the epithelial layer, the proliferation and degranulation of mast cells, and increases in the IL-$1{\beta}$, TNF-${\alpha}$, and MMP-9 expressions in the esophagus of the rats. The JYT group was inhibited above expression compared with the RE group. Apoptosis was statistically significantly decreased in the JYT group compared with the RE group. Conclusions: According to the above results, it appears that Jeungmiyijin-tang inhibits the expression of pro-inflammatory cytokines (TNF-${\alpha}$, IL-$1{\beta}$, and MMP-9) and apoptosis in the esophagus mucosa, thereby preventing esophageal mucosal damage from esophageal reflux.
