Disodium Kromoglycate와 Ketotifen의 사람정장 유도 비만세포 활성화 억제작용 (Inhibitory Effect of Disosium Cromoglycate and Ketotifen on Human Seminal Plasma-Induced Mast Cell Activation)

  • Background: Human seminal plasma (HSP)-induced hypersensitivity is one of the serious complications with sexual intercourse. The clinical manifestations of HSP-induced hypersensitivity may be related to the release of vasoactive mediators from mast cell induced by HSP. It has recently been reported that HSP modulates immune systems and induces mast cell degranulation and histamine release from rat peritoneal mast cells (RPMC). Ketotifen and disodium cromoglycate (DSCG), anti-asthmatic and anti-allergic drugs, have a role of mast cell stabilization and inhibit mast cell-induced leukocyte rolling and adhesion. But the inhibitory agents of HSP-induced mast cell activation are unknown. This study was performed to investigate the effects of DSCG and ketotifen on the HSP-induced mast cell activation. Methods: For this, influences of DSCG and ketotifen on the human seminal plasma-induced degranulation, histamine release and morphological changes of RPMC were observed. Results: The mast cell degranulation and histamine release of RPMC by HSP were induced in a dose-dependent fashion. The HSP-induced cytomorphological changes such as swelling, intracellular vacoules, and interrupted cell boundary were significantly inhibited by pretreatment with DSCG or ketotifen. DSCG and Ketotifen inhibited the HSP-induced degranulation and histamine release from RPMC. Conclusion: From the above results, it is suggested that DSCG and ketotifen have a inhibitory effect of the HSP-induced mast cell activation. DSCG and ketotifen may be used for treatment of HSP-induced hypersensitivity.

Mast Cell Concentrations in Peripheral and Central Giant Cell Granulomas: Is there any Angiogenetic Role?

  • Background: In the maxillofacial region, giant cell granulomas occur in 2 clinical forms, central and peripheral. Despite histopathological similarity between these 2 forms totally different clinical behaviors have been reported. The present study was undertaken to compare mast cell and vascular concentrations in these pathologic lesions. Materials and Methods: In this cross-sectional descriptive study, 20 pathological samples of central giant cell granuloma (CGCG) and 20 samples of peripheral giant cell granuloma (PGCG) were selected and examined through toluidine blue staining for mast cell assessment and immunohistochemical staining by VEGEF antibody for comparing the number of mast cells. T-test, chi-squared test and backward multivariate linear regression were used for statistical analysis using SPSS 20. Statistical significance was set at P<0.05. Results: This study showed significantly greater VEGF expression and mast cell concentrations in CGCG compared to PGCG cases. Also there was a significant correlation between VEGF expression and the concentration of mast cells. No relation was found between age, sex and site of the lesion and concentration of mast cells or VEGF expression. Conclusions: It is feasible that higher concentrations of mast cells in CGCG versus PGCG samples might lead to more aggressive clinical behavior via vascular proliferation and angiogenesis. However, other biologic mechanisms should be considered in this situation.

경혈에서의 비만세포 분포특성에 대한 연구 고찰 (The Association between Mast Cell Distribution and Acupoint Specificity: A Comprehensive Review)

  • Objectives : Mast cells, the multifunctional immune cells which can be activated by various stimulations, have been in the limelight recently at the fields of acupuncture research. This study aimed to investigate the association between the distribution of mast cell and acupoint specificity. We focused on the characteristics of mast cell distribution of acupoints in normal, pathologically sensitized, and acupoint-stimulated state. Methods : Literature searches were performed on PubMed and EMBASE for dates ranging to February 2019, by using terms "acupuncture", "moxibustion", "acupoint", "mast cell". Finally, 18 papers were collected by inclusion and exclusion criteria. To assess the quality of included studies, modified SYRCLE's risk of bias tool for animal studies was used. Results : Overall, the studies showed that the number of the mast cells was higher in acupoints than non-acupoints. After pathological sensitization, increasing tendency towards the mast cell number was reported in acupoints. The increase of mast cells was also observed after acupuncture or moxibustion. However, when the acupoint stimulation was repeated for several days in pathological models, the results did not show consistent tendency. In quality assessments, most of the studies showed unclear risk of bias. Conclusions : The studies showed a consistent trend about the association with mast cell distribution and acupoint specificity. However, we could not certainly affirm the relationship due to insufficient qualities of included studies. Not only the distribution but also the functions should be considered in further researches to identify the relationship between mast cells and acupuncture effect.

포유류 조직비만세포의 주사전자현미경적 연구 (Scanning Electron Microscopic Study on the Tissue Mast Cells of Mammals)

  • A Scanning electron microscope which can obtain additional information not readily available with either the light or transmission electron microscope was used to study the mast cell shape and its granules in normal mammal tissue(rat mesentery, stomach and mouse stomach) by fretting cut using liquid nitrogen. The results showed that rat mesentery and mouse stomach mast cell surfaces had no ridges and microvilli, but revealed several microvilli projecting into the surrounding connective tissue in the rat stomach mast cell. The shape of the mast cell varied from discoid(in the rat mesenteric mast cell) to ellipsoid (rat and mouse stomach), ranging from 7.5 to $10{\mu}m$ in diameter. The shape of the nucleus was ellipsoid and nucleic membrane was adherent to the outer surface of the granules. The granules, approximately 0.2 to $0.9{\mu}m$ in diameter, were various shapes. Frequently, rounded protrusions of cytoplasmic granules could be discerned under the cell membrane. Many small granules were seen in the cytoplasm.

Aloe 성분 NY945의 항알러지 작용 (The Inhibitory Mechanism of Aloe Component (NY945) on the Mediator Releases evoked with Mast Cell Activation)

  • By using guinea pig lung mast cells, this study aimed to examine the effects of Aloe component(NY945) on the mediator releases caused by mast cell activation, and also aimed to assess the effects of NY945 on the mechanism of mediator releases in the mast cell activation. We partially purified mast cells from guinea pig lung tissues by using the enzyme digestion, the rough and the discontinuous density percoll gradient method. Mast cells were sensitized with $IgG_1$ (anti-OA) and challenged with ovalbumin. Histamine was assayed by fluorometric analyzer, leukotrienes by radioimmunoassay The phospholipase D activity was assessed more directly by the production of labeled phosphatidylethanol or phosphatidylbutanol which was produced by phospholipase D-mediated transphosphatidylation in the presence of ethanol or butanol. The amount of mass 1,2-diacylglycerol was measured by the [$^3H$]1,2-diacylgycerol produced when prelabeled with [$^3H$]myristic acid. In the mast cells prelabeled with L-[$^3H$]methyl methionine the phospholipid methylation was assessed by measuring the incorporation of the [$^3H$]methyl moiety into phospholipids. Pretreatment of NY945(10$\mu$g) significantly decreased histamine and leukotrienes releases during mast cell activation. The decrease of histamine release was stronger than that of leukotrienes during mast cell activation. The phospholipase D activity increased by the mast cell activation was decreased by the dose-dependent manner in the pretreatment of NY945. The amount of mass 1,2-diacylglycerol produced by activation of mast cells were decreased in the pretreatment of NY945. NY945 pretreatment strongly inhibited the incorporation of the [$^3H$]methyl moiety into phospholipids. The data suggest that NY945 purified from Aloe inhibits in part an increase of 1,2-diacylglycerol which is produced by activating mast cells with antigen-antibody complexes which is mediated via phosphatidylcholine-phospholipise D and phosphatidylinositole-phospholipise C systems, and then followed by the inhibition of histamine release. Furthermore, NY945 reduces the phosphatidylcholine production by inhibiting the methyltransfsrase I and II, which decrease the conversion of phosphatidylcholine into arachidonic acid and inhibits the production of leukotrines.

가미옥병풍산(加味玉屛風散)의 비만세포에서의 알레르기 반응에 대한 효과 연구 (Effect of modified-Okbyungpoongsan on mast cell-mediated allergic responses in RBL-2H3 mast cells)

  • Objectives : In this study, we investigated the effect of modified-Okbyungpoongsan (mOP) on mast cell-mediated allergic response in basophilic leukemia cell line, RBL-2H3 mast cells. Methods : Cells were stimulated with anti-DNP-IgE after the treatment of DNP-HSA (AI/D), and then incubated with different concentrations of mOP (0.1, 0.2, 0.5 and 1 mg/$m{\ell}$) in RBL-2H3 cells. Cell toxicity was determined by WST-1 assay. The degranulation of mast cells was observed by microscope with toluidine blue staining and also the levels of beta-hexosaminidase, histamine and TNF-alpha were measured in culture supernatants by enzyme-linked immunosorbant assay. Results : mOP inhibited anti-DNP-IgE-imduced degranulation of mast cells in RBL-2H3 cells. mOP also significantly decreased the levels of histamine and inflammatory cytokine, TNF-alpha in RBL-2H3 cells, but slightly decreased the level of beta-hexosaminidase. Conclusions : These results indicate that mOP, an oriental prescription could be inhibit the allergic response through suppressing the mast cell activation.

Helicobacter pylori 감염 위염에서의 비만세포 증가와 Stem Cell Factor Receptor (c-kit)의 발현 (Mast Cell Increase and Stem Cell Factor Receptor (c-kit) Expression in Helicobacter pylori-infected Gastritis)

  • It is known that mast cells (MCs) are increased in H. pylori-infected gastritis and its increase is mediated by stem cell factor (c-kit ligand). To determine the mechanism of mast cell recruitment and activation by stem cell factor, weinvestigated the expression of stem cell factor receptor (c-kit) in H. pylori-positive and -negative gastric mucosa. Biopsy specimens from 16 H. pylori-negative and 20 positive subjects were examined. H. pylori infection in gastric mucosa was examined by the Warthin-Starry method. MC and c-kit were identified by immunohistochemisty, using a monoclonal antihuman MC tryptase antibody and a polyclonal anti-human c-kit antibody. Densities of MC and c-kit positive cell were measured by a computerized image analysis system. MCs were detected in the lamina propria of both H. pylori-positive and -negative gastric mucosa. Densities of MC and c-kit positive cell were significantly greater in H. pylori-positive than -negative subjects. c-kit was located on the surface of MCs. These results indicate that stem cell factors may be one of the factors involved in mast cell increase and that they activate mast cells by binding with c-kit.

백서 복직근피판의 허혈-재관류 손상에 대한 히스타민 차단제의 효과 (The Effects of Anti-Histamine and Mast Cell Stabilizer against Ischemia-Reperfusion Injury to TRAM Flap in Rat)

  • Purpose: The purpose of this study was to evaluate the role of mast cell and histamine as typical product of mast cell in ischemia-reperfusion injury of muscle flap using H2 receptor blocker and mast cell stabilizer. Methods: Thirty-five Sprague-Dawley rats weighing 250-300 gm were divided into four groups; Group I: Control group without ischemia, Group II: Normal saline injection group with ischemia, Group III: Cimetidine injection group with ischemia, Group IV: Sodium cromoglycate injection group with ischemia. Well established single pedicled transverse rectus abdominis musculocutaneous(TRAM) flap was designed in all rats and were rendered ischemia by clamping the artery for 150 minutes. All injections were applied intramuscular around gluteal area 30 minutes before reperfusion. The flap survival was evaluated at 7 days after operation. Neutrophil counts and mast cell counts were evaluated 24 hours after reperfusion. Results: The difference of skin flap survival between control group and cimetidine injection group was not significant. In the normal saline injection group flap survival was markedly decreased compared to that of control group. The muscle flap survival was similar to the results of skin flap survival. The neutrophil counts were significantly decreased in control group and sodium cromoglycate injection group than normal saline injection group. The mast cell counts were significantly decreased in cimetidine injection group and control group than both normal saline injection and sodium cromoglycate injection groups. The protective effect of sodium cromoglycate was not seen in the skin flap, but the muscle flaps showed protective effects of sodium cromoglycate compared to normal saline injection group. Conclusions: It is suggests that commonly used antihistamine(H2 receptor blocker) has protective effect against ischemia-reperfusion injury to skin and muscle flaps by reducing neutrophil and mast cell. The mast cell stabilizer was not effective for skin flap but, possibly, for muscle flap.

가미소요산(加味逍遙散)의 항산화 효과와 serotonin 대사과정에 미치는 영향 (The Effect of Gamisoyo-san(Jiaweixiaoyaosan) on Serotonin Metabolism)

  • Objectives : This experiment was designed to investigate the effect of Gamisoyo-san(Jiaweixiaoyaosan, SYS) on serotonin activity of P815 Mast Cell. Methods : The effects of SYS on activation of TPH-1 mRNA and AAADC mRNA in P815 mast cell were investigated. The effect of SYS on content of serotonin in P815 mast cell was investigated. The effects of SYS on activation of DPPH and SOD in P815 mast cell were investigated. Results : 1. The SYS increased the activation of SOD and DPPH in P815 mast cell. 2. The SYS decreased the manifestation TPH-1 mRNA in P815 mast cell. 3. The SYS decreased the manifestation AAADC mRNA in P815 mast cell. Conclusions : This experiment shows that Gamisoyo-san might not be effective for treating depression in terms of biogenic amine theory. However, Gamisoyo-san showed significant anti-oxidative effect and it can not yet be ruled out for treating depression. Therefore, pathogenesis of depression and clinical research of Gamisoyo-san is suggested for future research.

Effects of IL-3 and SCF on Histamine Production Kinetics and Cell Phenotype in Rat Bone Marrow-derived Mast Cells

  • Background: Rat mast cells were regarded as a good model for mast cell function in immune response. Methods: Rat bone marrow mast cells (BMMC) were prepared both by recombinant rat IL-3 (rrIL-3) and by recombinant mouse stem cell factor (rmSCF), and investigated for both proliferation and differentiation in time course. Rat BMMC was induced by culture of rat bone marrow cells (BMCs) in the presence of both rrIL-3 (5 ng/ml) and rmSCF (5 ng/ml). Culture media were changed 2 times per week with the cell number condition of $5{\times}10^4/ml$ in 6 well plate. Proliferation was analyzed by cell number and cell counting kit-8 (CCK-8) and differentiation was by rat mast cell protease (RMCP) II and histamine. Results: Cell proliferation rates reached a maximum at 8 or 11 days of culture and decreased thereafter. However, both RMCP II production and histamine synthesis peaked after 11 days of culture. By real time RT-PCR, the level of histidine decarboxylase mRNA was more than 500 times higher on culture day 11 than on culture day 5. By transmission electron microscopy, the cells were heterogeneous in size and contained cytoplasmic granules. Using gated flow cytometry, we showed that cultured BMCs expressed high levels of $Fc{\varepsilon}RI$ and the mast cell antigen, ganglioside, on culture day 11. Conclusion: These results indicate that rat BMMCs were generated by culturing BMCs in the presence of rrII-3 and rmSCF and that the BMMCs have the characteristics of mucosal mast cells.
