Effects of various medium on mass propagation of in vitro cultured Platycodon grandiflorum with yellow green petals

  • Propagation by crown division in Platycodon grandiflorum is too slow for producing many plants in a short time although the plants are uniform. This study was performed to enhance the mass propagation for Platycodon grandiflorum containing yellow green petals via various medium compositions and the growth regulators. The nodes containing yellow green petals were used as materials to execute the study with a variety of MS medium concentrations. The 1/4MS medium showed the best development of adventitious root, while the 1/2MS medium exhibited the potential growth. The higher the concentration of sucrose showed the better development and growth of both shoots and adventitious roots. Many adventitious roots were developed at the controlled culture medium at pH 4.8 with a tendency of suppression with higher levels of pH. Also, the cultivated node and leaf explants with the treatments of simple and combined applications with auxin and cytokinin at the 1/4MS culture medium with adding 5% of sucrose were used to identify the influences of growth regulators. The regeneration of plantlets at the 234single application showed a good result with the addition of BA $1mg{\cdot}L^{-1}$ and the development and growth of adventitious roots appeared to be good at the addition of NAA $1mg{\cdot}L^{-1}$. For the combined applications, the regeneration of plantlets and the development of adventitious roots were prosperous at the combined applications with BA $0.1mg{\cdot}L^{-1}$ and IAA $0.5mg{\cdot}L^{-1}$. The addition of IAA for the leaf explants induced a number of plantlets that showed the potential regeneration, and the highest results was obtained from the combined applications of both BA $1mg{\cdot}L^{-1}$ and IAA $2.5mg{\cdot}L^{-1}$. In addition, the development of adventitious roots showed the satisfactory results at the combined application of both BA $1mg{\cdot}L^{-1}$ and IAA $0.5mg{\cdot}L^{-1}$.

생장조절제가 하월시아 만상(Haworthia maughanii)의 기내 대량증식에 미치는 영향 (Effect of growth regulators on In Vitro mass propagation of Haworthia maughanii)

  • 하월시아(Haworthia maughanii)의 기내 식물체 대량생산을 위하여 캘러스 유도에 적합한 부위와 신초 및 발근에 영향을 미치는 생장조절제 종류 및 농도를 구명하고자 본 시험을 수행하였다. 캘러스 유도에 적합한 식물 부위를 구명 하기 위해 잎, 화뢰, 화경을 이용하여 NAA와 TDZ의 농도를 달리하여 MS배지에 치상한 결과, 잎은 $NAA\;1mgL^{-1}$$TDZ\;1mgL^{-1}$, 화뢰는 $NAA\;2mgL^{-1}$$TDZ\;2mgL^{-1}$, 화경는 $NAA\;1mgL^{-1}$이상의 단용처리에서 100% 캘러스 형성율을 보였고, 모든 부위에서 캘러스 형성율은 높았으나 가장 많은 배양절편 확보가 가능한 잎이 가장 효율적으로 판단되었다. $NAA\;0.1mgL^{-1}$$BA\;0.1mgL^{-1}$ 혼용처리구에서 다른 처리에 비해서 신초가 22.0개로 신초 발생에 가장 효과적이었다. 또한 신초의 대량증식은 $NAA\;0.1mgL^{-1}$$BA\;0.1mgL^{-1}$처리구에서 신초가 16.3개로 가장 많이 형성되었다. 이러한 결과는 최적의 기내 배양 조건을 통해 하월시아 만상의 대량생산 가능성을 보여주었다.

  • We, Tong Yang Moolsan Co. Ltd. (TYM) set up the mass-production system for virus-free seed garlic via tissue culture technique. TYM's tissue culture technique is called as 'Multiple shoot propagation technique'. This technique can lead mass propagation of genetically homogeneous seed garlic in a short period because of its highly proliferation rate of in vitro shoots ($15^{10}$ /year). TYM researchers applied the technique to some selected garlic cultivars with superior characteristics and carried out field test of productivity in the inside and outside of the country for several years. According to the yearly results of field test with virus-free seed garlic, we ascertained that virus-free seed garlic can produce the highly yield increase (max. above $50\%$) and also can enhance the product quality. Consequently, we estimated that TYM's seed garlic will contribute to farmers with increase of income and can elevate the national position of garlic market in the world for its competitive power of technical and production cost.

폐기물 소각로 베드에서의 연소현상 관찰을 위한 실험적 연구 (An Experimental Study on the Bed Combustion Phenomena in MSW(Municipal Solid Waste) Incinerator)

  • Experimental studies have been performed to observe the basic phenomena of waste bed combustion in MSW incinerator. A reduced scale apparatus was utilized to simulate the combustion behavior in real plant with 1-dimensional transient behavior at the experimental setup, which uses wet cubic wood with ash content as simulated waste. LHV (lower heating value) of solid fuel, fuel particle size and flow rate of combustion air were taken as important parameters of the bed combustion. For the quantitative analysis, FPR (flame propagation rate), TBT (total burn-out time) and PBT (particle burn-out time) was defined. LHV represent the capability of heat release of the fuel, so that a higher LHV results in faster reaction rate of the fuel bed, which is shown by higher FPR. Fuel particle size is related with surface area per unit mass as well as heat and mass transfer coefficient. As the particle size increases the FPR decreases owing to decreasing specific surface area. Air injection supplies oxygen to the reaction zone. However oversupply of combustion air increases convection cooling of the bed and possibly extinguishes the flame.

열적 메커니즘에 의한 펄스레이저 어블레이션 현상의 수치계산 (Numerical computation of pulsed laser ablation phenomena by thermal mechanisms)

  • High-power pulsed laser ablation under atmospheric pressure is studied utilizing numerical and experimental methods with emphasis on recondensation ratio, and the dynamics of the laser induced vapor flow. In the numerical calculation, the temperature pressure, density and vaporization flux on a solid substrate are first obtained by a heat-transfer computation code based on the enthalpy method, and then the plume dynamics is calculated by using a commercial CFD package. To confirm the computation results, the probe beam deflection technique was utilized for measuring the propagation of a laser induced shock wave. Discontinuities of properties and velocity over the Knudsen layer were investigated. Related with the analysis of the jump condition, the effect of the recondesation ratio on the plume dynamics was examined by comparing the pressure, density, and mass fraction of ablated aluminum vapor. To consider the effect of mass transfer between the ablation plume and air, unlike the most previous investigations, the equation of species conservation is simultaneously solved with the Euler equations. Therefore the numerical model computes not only the propagation of the shock front but also the distribution of the aluminum vapor. To our knowledge, this is the first work that employed a commercial CFD code in the calculation of pulsed ablation phenomena.

모시풀 조직배양에서 소독방법 및 생장조절제의 Multiple Shoot 유기 효과 (Effect of Growth Regulator and Sterilization Method on Multiple Shoot Induction through Sucker and Stem Node Culture in Ramie(Boehmeria nivea Hoooker et Arnot))

  • 모시풀의 경편과 흡지의 배양을 통하여 완전한 식물체를 대량증식하기 위하여 소독방법과 생장조절제 처리 효과에 대한 기내배양을 실시하여 다음과 같은 결과를 얻었다. 1. 모시풀 경편 배양시 소독은 초음파세체기를 이용한 2% NaClO를 20분 동안 하였을때 오염률이 3.3%로 가장 낮았으며, 식물체도 79%가 생존하였고, 건실한 묘를 생산할 수 있었다. 2. 생장조절제 처리효과에서는 NAA(0.02mg/$\ell$)+ BA(1.5mg/$\ell$) + GA3(0.1mg/$\ell$) 혼합처리가 캘러스 형성이 안되고, 식물체 형성률이 96%였으며, 건실한 묘를 생산할 수 있었다. 3. 치상부위별로는 흡지보다 경편배양이, 품종별로는 개량종인 서방종보다 재래종인 보성종이 증식효률이 높았다. 4. 순화 효율은 상토 배합과 호르몬 처리에 있어서 버미큐라이트 : 모래 :황토 =1 : 1 : 1의 배합과 NAA 1000ppm을 30분간 담근 후 이식한 순화율이 99%로서 건실하였으며, 식물체는 대부분 정상이었다.
