Identification of triacylglycerols in coix seed extract by preparative thin layer chromatography and liquid chromatography atmospheric pressure chemical ionization tandem mass spectrometry

  • Here we reported a methodology for identification of triacylglycerols (TAGs) and diacylglycerols (DAGs) in coix seed by preparative thin layer chromatography (prep-TLC) and non-aqueous reversed-phase liquid chromatography (NARP LC)-atmospheric pressure chemical ionization (APCI) tandem mass spectrometry (MS/MS). Lipid components were extracted from coix seed by reflux extraction using n-hexane for 3 hr. TAGs and DAGs in coix seed extract were effectively purified and isolated from matrix interferences by prep-TLC and then analyzed by LC-APCI-MS and MS/MS for identification. TAGs were effectively identified taking into consideration of their LC retention behavior, APCI-MS spectra patterns, and MS/MS spectra of $[DAG]^+$ ions. In MS/MS spectra of TAGs, diacylglycerol-like fragment $[DAG]^+$ ions were useful to identify TAGs with isobaric fragment ions. Based on an established method, 27 TAGs and 8 DAGs were identified in coix seed extract. Among them, 15 TAGs and 8 DAGs were for the first time observed in coix seed. Interestingly, some of TAGs isolated by prep-TLC were partly converted into DAGs through probably photolysis process during storing in room temperature. Thus, degradation phenomenon of TAGs should be considered in the quality evaluation and nutritional property of coix seed. LC-APCI-MS/MS combined with prep-TLC will be practical method for precise TAG and DAG analysis of other herbal plants.

Fully Printed Chipless RFID Tags Using Dipole Array Structures with Enhanced Reading Ranges

  • This article proposes a design of chipless RFID tag with dipole array structure that is fully printable using conductive ink. The proposed tags encode data based on spectral signature modulations. The reading range is considerably increased (2 m) while maintaining low transmission power (1 mW). Several prototype chipless RFID tags were fabricated and measured in the SHF and UHF bands. The proposed dipole array structure enhances the antenna gain of the passive tags and contributes to overcoming the low conductivity of conductive ink. In order to verify the utility of our proposal, the tags are manufactured on paper, using conductive ink, for the purpose of economic mass production.

  • The mass spectrometry (MS) provides the mass-to-charge ratios of atoms, molecules, stable/metastable complexes, and their fragments. I have taken a long journey with MS to address outstanding issues and problems by experiments and theory and gain insights into underlying principles in chemistry. By looking through the mass-to-charge ratio, I have studied thermochemical problems in silicon chemistry, the infrared multiphoton dissociation spectroscopy of organometallic intermediates, unimolecular dissociations of halotoluene radical cations, and the kinetics of association/dissociation of alkali halide triple ions with Lewis bases. Various MS platforms have been used to characterize non-covalent interactions between porphyrins and fullerenes and those between the group IIB ions and trioctylchalcogenides, and to examine the binding of the group IA, IIA and porphyrin ions to G-quadruplex DNA. Recently, I have focused on mass-balanced H/D isotope dipeptide tags for MS-based quantitative proteomics, a simple chemical modification method for MS-based lipase assay, and the kinetics and dynamics of energy-variable collision-induced dissociation of chemically modified peptides. Now, I see an important role of MS in global issues in the post-COVID era, as the society demands high standards for indoor air quality to contain the airborne-pathogen transmission as well as in-situ monitoring and tracking of carbon emissions to reduce global warming.

  • New proteomic techniques have been pioneered extensively in recent years, enabling the high-throughput and systematic analyses of cellular proteins in combination with bioinformatic tools. Furthermore, the development of such novel proteomic techniques facilitates the elucidation of the functions of proteins under stress or disease conditions, resulting in the discovery of biomarkers for responses to environmental stimuli. The ultimate objective of proteomics is targeted toward the entire proteome of life, subcellular localization biochemical activities, and the regulation thereof. Comprehensive analysis strategies of proteomics can be classified into three categories: (i) protein separation via 2-dimensional gel electrophoresis (2-DE) or liquid chromatography (LC), (ii) protein identification via either Edman sequencing or mass spectrometry (MS), and (iii) proteome quantitation. Currently, MS-based proteomics techniques have shifted from qualitative proteome analysis via 2-DE or 2D-LC coupled with off-line matrix assisted laser desorption ionization (MALDI) and on-line electrospray ionization (ESI) MS, respectively, toward quantitative proteome analysis. In vitro quantitative proteomic techniques include differential gel electrophoresis with fluorescence dyes. protein-labeling tagging with isotope-coded affinity tags, and peptide-labeling tagging with isobaric tags for relative and absolute quantitation. In addition, stable isotope-labeled amino acids can be in vivo labeled into live culture cells via metabolic incorporation. MS-based proteomics techniques extend to the detection of the phosphopeptide mapping of biologically crucial proteins, which ale associated with post-translational modification. These complementary proteomic techniques contribute to our current understanding of the manner in which life responds to differing environment.

Analysis of Lipids in Deciduous Teeth by Matrix-Assisted Laser Desorption/Ionization Mass Spectrometry (MALDI MS)

  • Recently, deciduous teeth have been proposed as a promising biomatrix for estimating internal and external chemical exposures of an individual from prenatal periods to early childhood. Therefore, detection of organic chemicals in teeth has received increasing attention. Organic materials in tooth matrix are mostly collagen type proteins, but lipids and other small organic chemicals are also present in the tooth matrix. In this study, matrix-assisted laser desorption/ionization (MALDI) mass spectrometry (MS) was employed to obtain lipid fingerprints from deciduous teeth. Phospholipids and triacylglcerols (TAGs) from deciduous teeth were successfully detected by MALDI MS with 2,5-dihydroxybenzoic acid (DHB) or gold nanoparticle (AuNP) as a matrix.

A TMT-based quantitative proteomic analysis provides insights into the protein changes in the seeds of high- and low- protein content soybean cultivars

  • The presence of high amounts of seed storage proteins (SSPs) improves the overall quality of soybean seeds. However, these SSPs pose a major limitation due to their high abundance in soybean seeds. Although various technical advancements including mass-spectrometry and bioinformatics resources were reported, only limited information has been derived to date on soybean seeds at proteome level. Here, we applied a tandem mass tags (TMT)-based quantitative proteomic analysis to identify the significantly modulated proteins in the seeds of two soybean cultivars showing varying protein contents. This approach led to the identification of 5,678 proteins of which 13 and 1,133 proteins showed significant changes in Daewon (low-protein content cultivar) and Saedanbaek (high-protein content cultivar) respectively. Functional annotation revealed that proteins with increased abundance in Saedanbaek were mainly associated with the amino acid and protein metabolism involved in protein synthesis, folding, targeting, and degradation. Taken together, the results presented here provide a pipeline for soybean seed proteome analysis and contribute a better understanding of proteomic changes that may lead to alteration in the protein contents in soybean seeds.

  • Recently the RFID system, which can recognize multiple tags simultaneously through wireless communication, is emerging as a new technology that can replace the barcode system. Furthermore, related industries are carrying out active research on tags and authentication protocols with guaranteed security that are widely applicable to logistics, distribution, etc. The present study proposes a protocol with enhanced security by introducing the concept of RBAC to the authentication protocol, and a method with lower security for effective mass authentication. The proposed method is advantageous in that it guarantees security against spoofing attack, traffic analysis, replay attack, etc. based on hash function.

  • The speech synthesis technology is widely used and its application area is also being broadened to an automatic response service, a learning system for handicapped person, etc. However, the sound quality of the speech synthesizer has not yet reached to the satisfactory level of users. To make a synthesized speech, the existing synthesizer generates rhythms only by the interval information such as space and comma or by several punctuation marks such as a question mark and an exclamation mark so that it is not easy to generate natural rhythms of people even though it is based on mass speech database. To make up for the problem, there is a way to select rhythms after processing language from a higher level information. This paper proposes a method for generating tags for controling rhythms by analyzing the meaning of sentence with speech situation information. We use the Systemic Functional Grammar (SFG) [4] which analyzes the meaning of sentence with speech situation information considering the sentence prior to the given one, the situation of a conversation, the relationship among people in the conversation, etc. In this study, we generate Semantic Speech Control Tag (SSCT) by the result of SFG's meaning analysis and the voice wave analysis.

  • 한국 재래 닭의 유전적 특성 규명 및 기능 유전체 연구의 기초재료를 확보하고자 대량 EST 염기서열 결정 및 생물정보학 분석을 실시하였다. 대량 EST 염기서열 분석을 위한 첫 번째 단계로 재래 닭의 뇌, 비장, 정소, 배아 생식기를 이용하여 cDNA library를 구축하였다. 각각의 library로부터 총 15,121개의 클론을 선정하여 염기서열을 결정하였다. 생물정보학 분석결과 15,121개의 염기서열은 총 10,353개의 contig로 정리되었다. 이들 염기서열을 기존 데이터베이스를 대상으로 tBlastX(http://www.ncbi.nlm.nih.gov/BLAST) 분석을 실시한 결과, 염기서열 중 56 %가 기존 데이터베이스에 존재하는 유전자와의 상동성을 보였다. 상동성을 보이는 유전자들은 유전자의 구조 및 기능 분석에 이용될 것이고, 상동성을 보이지 않는 유전자들은 microarray와 같은 대량 유전자 발현분석 시스템을 이용하여 선별한 후 기능분석이 실시될 것이다.
