A sensitive method for the determination of methamphetamine(MA) and amphetamine(AM) in hair was developed by gas chromatography/mass spectrometry using stable isotope-labeled internal standards, amphetamine-d$_{5}$ and methamphetamine-d$_{5}$. Hair sample was washed with MeOH, incubated with MeOH(I% HCI) overnight at $37^{\circ}C$ while stirring and extracted using solid phase extraction column on a vacuum manifold. The extract obtained was pentafluoropropionated, and applied to GC/MS. The calibration curves of MA and AM were linear from 2.5 to 250 ng (r>0.99 for both). The limit of detection was 0.1 ng/mg in hair and cut-off level was set at 0.25 ng/mg for both. Hair samples of 27 MA abusers showed positive results in the range 0.7 to 106.8 ng/mg. AM, its metabolite, was detected in 20 out of 27 samples. The ratio of MA versus AM was 4.6~38.3 in specimens. Hair analysis for methamphetamine by GC/MS is an effective method for identifying long-term drug abusers.
A current interest in chemistry concerns traceability of analytical measurements to the International System of Units (SI) and the proper estimation of their uncertainties in accordance with the internationally agreed guide provided by the International Organization for Standardization (ISO). Isotope dilution mass spectrometry (IDMS) is regarded as a primary method, which make the measurement results traceable to SI units without significant empirical correction factors. Our laboratory, as the national standards institute of Korea, participated in an intercomparison of environmental analysis, pp'-DDE in corn oil, which was organized by the CCQM under supervision of the CIPM to test feasibility of IDMS as a primary method for the trace analysis of organic compounds. In this report, we provide basic equations used for the calculation of the concentration of the analyte in a sample and a precise description of the processes for the evaluation of the uncertainties of the measurement results. Also, we report the experimental conditions adopted to improve the accuracy of the IDMS measurement. The principles contained in ??Guide to the Expression of Uncertainty in Measurement'' provided by ISO are followed for the uncertainty evaluation.
As the air pollution caused by diesel vehicles goes worse, so non-road vehicles exhaust gas standards are more strict in an foreign countries. There is growing evidence that diesel vehicles could play the important role in determining health effects. Most of the particle number emitted by diesel engines is in the nanopaticle range, D$\_$p/ < 50nm, while most of the mass is in the accumulation mode, 50nm < D$\_$p/ < 1000nm range. The aim of this work was to investigate pollutants in the exhaust of railroad diesel rolling stock under load tests.
Calcium isotopic ratios are precisely measured by removing isobaric interferences originated from water in the plasma. Liquid Ar cryogenic trap combined with membrane desolvator could eliminate backgrounds at m/z 42 and 44. Slow drift of ICP-MS is corrected by the frequent running of the standards. It is found necessary to separate Ca from the sample matrix using Ca oxalate precipitation technique. Currently, the RSD is 0.5-1.0% for 2 minutes of measurement but is expected to be improved if the measurement time is increased. The technique was applied to 42Ca enriched baby fecal samples and successfully determined 42Ca/44Ca ratio changes.
A variant peak was detected in the analysis of RP-HPLC of rHu-EPO, which has about 7% relative content. Fractions of the main and the variant peaks were pooled separately and further analyzed to identify the molecular structure of the variant peak. Total mass analysis for each peak fraction using ESI-TOF MS shows differences in molecular mass. The fraction of the main peak tends to result in higher molecular masses than the fraction of the variant. The detected masses for the variant are about 600-1000 Da smaller than those for the main peak. Peptide mapping analysis for each peak fraction using Asp-N and Glu-C shows differences in O-glycopeptide profiles at Ser126. The O-glycopeptides were not detected in the fraction of the variant. It is concluded that the variant peak is non-O-glycosylated rHu-EPO and the main peak is fully O-glycosylated rHu-EPO at Ser126.
Traceability establishment in chemical measurements is a like a linkage established through an unbroken chain from the measured results to the international system (SI) of units. The primary process for traceability establishment is the purity assignment of a target material to be measured. In this study, we studied the purity assignment of 17α-hydroxyprogesterone (17-OHP). The presence of 17-OHP is indicative of congenital adrenal hyperplasia (CAH) and it builds up due to the deficiency of 21-hydroxylase and 11β-hydroxylase enzyme in the human blood. The purity assignment of 17-OHP was performed by the mass balance method, in which the impurities are categorized into four classes: total related structural impurities, water, residual organic solvents, and nonvolatiles/inorganics. The total related structural impurities were characterized by HPLC-UV; water content was determined by Karl-Fisher coulometer; and the total residual solvents and nonvolatiles/inorganics were determined by TGA. The purity of 17-OHP from a commercial manufacturer was calculated as 993.30 mg/g, and the expanded uncertainty was 0.58 mg/g. The proposed method was validated by uncertainty evaluation and comparing with the actual value of purity.
The importance of separation comes from demands on study for exact effect of synthetic drugs and the reactivity of enantiomer in biological system. Racemization rate was measured under the influence of heat, acid, UV-light, enzyme(trypsin) and 6N-HCl at $105^{\circ}C$ on alanine, threonine, isoleucine, lecuine, aspartic acid, methionine, glutamic acid, tyrosine. The method for the identification of overlapped amino acids with GC was developed from the close study of fragmentation pattern with mass spectrometry. With cyclodextrin bonded phase by HPLC, the separation of dansyl amino acid was tested for compartison.
This study examined differences in body mass index (BMI), self-esteem, sociocultural attitude toward appearance, and diet awareness between genders as well as the effect of BMI, self-esteem, and sociocultural attitude toward appearance on diet awareness. This study surveyed adults between the ages of 20 and 29 who lived in Daegu and Uijeongbu from November $17^{th}$ to December $14^{th}$ 2014. Data collected from 258 respondents were analyzed using descriptive statistics, t-test, regression analysis, and ${\chi}^2$ test. The study results showed that the respondents could be divided into three groups (underweight group, standard weight group, and overweight group) by BMI and a large percentage of respondents were in the standard weight group. The percentage of standard weight females was higher than standard weight males. There were significant differences in BMI, sociocultural attitude toward appearance (such as self-awareness), and diet awareness between genders; however, there was no difference in self-esteem. Men also had a higher BMI than women; however, women were more self-aware of sociocultural standards for appearance, desired a slim body and wanted educational information on diet. BMI and sociocultural attitude toward appearance had positive effects on diet awareness. Self-awareness was a strong predictor of diet awareness and self-esteem had no significant effect on diet awareness.

A numerical program has been developed for the simulation of automotive engine cooling system. The program determines the mass flow rate of engine coolant circulating the engine cooling system and radiator cooling air when the engine speed is adopted by appropriate empirical correlation. The program used the method of thermal balance at individual element through the model for radiator component in radiator analysis. This study has developed the program that predicts the coolant mass flow rate, inlet and outlet temperatures of each component in the engine cooling system (engine, transmission, radiator and oil cooler) in its state of thermal equilibrium. This study also combined the individual programs and united into the total performance analysis program of the engine cooling system operating at a constant vehicle speed. An air conditioner system is also included in this engine cooling system so that the condenser of the air conditioner faces the radiator. The effect of air conditioner to the cooling performance, e.g., radiator inlet temperature, of the radiator and engine system was examined. This study could make standards of design of radiator capacity using heat rejection with respect to the mass flow rate of cooling air. This study is intended to predict the performance of each component at design step or to simulate the system when specification of the component is modified, and to analyze the performance of the total vehicle engine cooling system.
Migration levels of plasticizers, di-n-butyl phthalate (DBP), benzyl-butyl phthalate (BBP), di-n-octyl phthalate(DNOP), di-iso-decyl phthalate (DIDP) and di-iso-nonyl phthalate (DINP), di-(2-ethylhexyl) adipate (DEHA), from 46 poly(vinyl chloride) (PVC) wrap films and 54 PVC gaskets into food simulants were determined using gas chromatography/mass spectrometry (GC/MS). The method was validated with limit of detection (LOD) of $0.01{\sim}0.02\;{\mu}g/mL$ for DBP, BBP, DNOP and DEHA, and $2\;{\mu}g/mL$ for DIDP and DINP. The linearity were found to be > 0.99 for all the compounds in concentration range of $0.1{\sim}81.4\;{\mu}g/mL$, and overall recoveries were ranged from 90.4 ~ 99.6%. DBP, BBP, DNOP, DEHA, DIDP and DINP were not detected in food simulants, except 1 wrap sample from which 0.28 and $0.99\;{\mu}g/mL$ of DEHA were detected respectively when tested with 20% ethanol and n-heptane as food simulants. These values were below the regulatory limitation in European Union (EU).
