Identification of Trichiurus (Pisces: Trichiuridae) Eggs and Larvae from Korea, with a Taxonomic Note

  • The hairtail (currently recognized as Trichiurus lepturus in Korea) is one of the most important commercial fish species in Korea, Japan, China, and Taiwan. Because the amount of catches has been steadily declining, we must determine the early life stages of the hairtail from the viewpoint of resource management. Furthermore, the taxonomic status of the hairtail is unclear among ichthyologists, potentially creating management difficulties. Therefore, the purpose of this study was to compare morphological and molecular information on eggs, larvae, and adults of hairtail from Korea with that of T. lepturus from the Atlantic Ocean, and to review the taxonomic status of the hairtail. A total of 510 base pairs of the mitochondrial DNA cytochrome oxidase subunit I sequences of 12 eggs, 2 larvae, and 11 adults of the hairtail from the Korean waters clearly matched those of Trichiurus japonicus adults (d = 0.000-0.014) from the East China Sea rather than those of T. lepturus (d = 0.100-0.110) from the Atlantic Ocean. Our results also showed that larvae of the Korean hairtail are different than those in the Atlantic Ocean in having no melanophores along the ventral edge of the lower jaw. Therefore, our findings suggest that the hairtail in the Korean waters may not be T. lepturus, but T. japonicus.

Sexual Maturity and Early Life History of the Mudskipper Scartelaos gigas (Pisces, Gobiidae): Implications for Conservation

  • Scartelaos gigas is an amphibious mudskipper species that inhabits mud flats in Korea, China, and Taiwan. This fish is at risk of extinction because of its very restricted habitat and overexploitation. Information about this fish's reproductive characteristics is needed for species conservation. The sexual maturity and early life history of S. gigas were investigated through histological methods and direct observation of eggs in the wild, respectively. In total, 560 individuals of S. gigas were collected with the aid of fishermen from March 2003 to October 2003 at Jung-do Island, southwest Korea. Through microscopic observations of gonadal development, it was determined that S. gigas of both sexes were immature in April, but began to reach maturity in May, and were then fully mature by June, which was maintained until July. In August, some female fish developed early oocytes, but by September oocytes were observed to have degenerated and had been absorbed. Spawned eggs were elliptical and had an average size of 1.37 mm (long axis) by 0.69 mm (short axis). The newly hatched larvae (3.03 mm total length, TL) had an open mouth and anus, two melanophores near the anus, and one large melanophore between the 18th and 19th myomeres. The larvae (3.18 mm TL) showed absorption of the yolk and oil globule within 5 days after hatching and became prelarvae. This species should be considered vulnerable or conservation-dependent, and thus parental fish need to be protected from fishermen during the main spawning season (June).

Genetic identification of anisakid nematodes isolated from largehead hairtail (Trichiurus japonicus) in Korea

  • Background: The nematode species belonging to genus Anisakis occur at their third larval stage in numerous marine teleost fish species worldwide and known to cause accidental human infection through the ingestion of raw or undercooked fish or squids. They may also draw the attention of consumers because of the visual impact of both alive and dead worms. Therefore, the information on their geographical distribution and clear species identification is important for epidemiological survey and further prevention of human infection. Results: For identification of anisakid nematodes species isolated from largehead hairtail (Trichiurus japonicus), polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) analysis of internal transcribed spacers of ribosomal DNA were conducted. Mitochondrial cytochrome c oxidase subunit 2 gene was also sequenced, and phylogenetic analysis was conducted. From the largehead hairtail (n = 9), 1259 nematodes were isolated in total. Most of the nematodes were found encapsulated throughout the viscera (56.2 %, 708/1259) or moving freely in the body cavity (41.5 %, 523/1259), and only 0.3 % (4/1259) was found in the muscles. By PCR-RFLP, three different nematode species were identified. Anisakis pegreffii was the most dominantly found (98.7 %, 1243/1259) from the largehead hairtail, occupying 98.7 % (699/708) of the nematodes in the mesenteries and 98.1 % (513/523) in the body cavity. Hybrid genotype (Anisakis simplex ${\times}$ A. pegreffii) occupied 0.5 %, and Hysterothylacium sp. occupied 0.2 % of the nematodes isolated in this study. Conclusions: The largehead hairtail may not significantly contribute accidental human infection of anisakid nematode third stage larvae because most of the nematodes were found from the viscera or body cavity, which are not consumed raw. But, a high prevalence of anisakid nematode larvae in the largehead hairtail is still in concern because they may raise food safety problems to consumers. Immediate evisceration or freezing of fish after catch will be necessary before consumption.

순천 상내리 갯골에 출현하는 민물두줄망둑(Tridentiger bifasciatus)의 식성 (Feeding Habits of Tridentiger bifasciatus in the Tidal Creek at Sangnae-ri, Suncheon, Korea)

  • The feeding habits of Tridentiger bifasciatus were studied using 733 individuals collected in the tidal creek at Sangnae-ri, Suncheon, Korea, using a push net, monthly, from April 2011 to March 2012. The standard length (SL) of the specimens ranged from 1.5 to 8.2 cm. The stomach content analysis indicated that T. bifasciatus consumed mainly amphipods, such as Corophium sp., while copepods, polychaetes, and shrimps were other important prey species. In addition T. bifasciatus ate small quantities of crab larvae, fish, gastropod larvae, and insects. T. bifasciatus is a generalist that feeds on various zoobenthos, including fish. T. bifasciatus underwent an ontogenetic dietary shift. Smaller individuals (1-2 cm SL) fed mainly on amphipods. The proportion of amphipods decreased as body size increased, whereas the consumption of polychaetes increased gradually. The seasonal variation in the diet composition of T. bifasciatus was significant. Insects were common prey during summer, while the amounts of other prey, including polychaetes, were similar during spring, autumn, and winter.

Egg Development and Larvae and Juveniles Morphology of Carp, Cyprinus carpio in Korean

  • This study was conducted to observe egg and larvae morphological development of carp to obtain basic data for resource conservation and taxonomic research. Brood carp used in the research (total length 67.3-75.5 cm, average $71.0{\pm}3.45cm$) were bred in a circular rearing aquarium ($600{\times}300{\times}100cm$) using a running water system from January to July, 2015. Breeding water temperature was maintained at $23.0-25.0^{\circ}C$(average $24.0^{\circ}C$). Fertilized carp eggs were translucent and globular, and their size was 1.75-1.89 mm (average $1.82{\pm}0.06mm$). Blastoderms formed 10 min after fertilization and reached the two-cell stage 30 min after fertilization. Then, the embryo turned dark and exhibited melanophores, and blood started flowing from the heart across the egg yolk at 42 hrs and 50 min after fertilization. Hatching began 70 hrs and 26 min after fertilization larvae emerged through the egg membrane, starting from the head. The length of prelarvae immediately after hatching was 5.23-5.38 mm (average $5.31{\pm}0.11mm$) the mouth and anus were closed, and the pectoral fin was formed. Postlarvae at 18 days after hatching had a total length of 11.9-13.9 mm (average $12.9{\pm}1.40mm$), separate anal fin and back membranes, and fin ray. Juveniles fish at 35 days after hatching had a total length of 29.9-30.2 mm (average $30.1{\pm}0.13mm$), with the body covered with scales, and the same number of fin rays, color, and shape as their broodstork.

봄-여름 남해 중부 연안 자어 군집구조 (Community Structure of Larval Fish Assemblage in the Coastal Waters of South-central Korea during Spring and Summer)

  • 본 연구는 남해 중부 연안역에서 2016년 4~8월까지 자어군집구조의 월 변화를 조사하였다. 조사기간 동안 총 42종의 자어가 출현하였다. 우점종 중 멸치(E. japonicus)는 여름보다는 봄철에 우점하였고, 용서대(C. abbreviates)는 봄철에만 출현하였으며 수조기(N. albiflora)는 여름철에만 출현하였다. 자어 군집은 계절별로 4~5월 (봄)과 6~8월 (여름)의 그룹으로 유의하게 나눌 수 있었고, 여름철은 또다시 6~7월과 8월의 두개의 유의한 그룹으로 나눌 수 있었다. 2016년 8월 조사해역에서는 이상 고수온 현상이 발생하여 표면수온이 $28^{\circ}C$까지 상승하였고, 멸치와 수조기 자어의 출현량은 산란 적정수온을 넘은 수온으로 인해 급감하였다.

3종 양식어류의 전기자어기에 미치는 원유 수용성 획분(WSF) 의 영향 (Effects of the Water Soluble Fraction from Kuwait Crude Oil on the Early Larval Stages of Three Cultured Fish Species)

  • 원유중 수용성 획분(WSF)이 양식 대상종인 감성돔 (Acanthopagrus schlegeli), 참돔(Pagrus major) 및 넙치 (Paralichthys olicaceus)의 전기자어에 미치는 영향을 파악하였다. 실험구는 0%(대조구), 25%, 50%, 75% 및 100% WSF로 하였고, 이에 따른 자어의 난황 흡수속도, 유영활성, 서장도, 기형률 및 생존율을 조사하였다. 수정란으로부터 부화후 자어까지 지속적으로 WSF에 노출시켰을 때, 전기자어의 난황 흡수 속도는 3어종 모두 대조구에 비해 고농도의 WSF에서 느렸다. 유영빈도와 유영속도는 WSF 농도가 높아질수록 낮아지는 경향이었고, 성장은 대조구에 비해 고농도의WSF에서 느린 것으로 나타났다. 고농도의 WSF(50%, 75% 및 100%) 노출은 대조구에 비해 높은 기형률을 나타내었고, 3어종중 참돔의 자어에서 가장 높았다. 전기자어의 생존율은 3어종 모두에서 WSf 농도가 높을수록 낮았다. 부화후의 자어를 WSF에 노출시켰을 때, 유영활성은 3어종 모두에서 SF 농도가 높아질수록 낮아지는 경향이었다. 기형률은 고농도의WSF (50%, 75% 및 100%)에서 높았으며, 3종중 참돔에서 가장 높았다. WSF 농도가 높아짐에 따라 생존율도 유의하게 낮아졌다.

겨울철 산란하는 대구, 꼼치, 노래미 전기 자어의 발달 비교 (Comparative Early Developments in Winter Spawned Three Pre-larval Fishes(Gadus macrocephalus, Liparis tanakae, Hexagrammos agrammus))

  • 겨울철 환경에 대한 초기 적응을 조사하기 위해 대구(Gadus macrocephalus), 꼼치(Liparis tanakae), 노래미(Hexagrammos agrammus) 자어의 부화 직후와 첫 섭식 시 외부형태, 소화기관, 유영능력의 발달을 확인하였으며, 다른 계절에 부화하는 어종과 비교하였다. 평균 전장(${\pm}SD$)은 대구 $4.35{\pm}0.11mm$, 꼼치 $5.26{\pm}0.08mm$, 노래미 $7.48{\pm}0.35mm$로 부화 시 다른 계절 부화자어에 비하여 컸으며, 세 어종 모두 초기소화기관을 갖춘 후 부화하여 다른 계절 부화자어에 비해 초기소화기관 확립시기가 빨랐다. 대구, 꼼치, 노래미의 첫 섭식 시기가 부화 후 3일, 2일, 0일이었고, 난황을 갖고 있는 기간이 5일 이상으로 다른 계절에 부화하는 자어에 비해 혼합섭이기간이 비교적 길었다. 이 3종 자어의 유영능력은 다른 계절 부화자어에 비해 순항유영속도와 돌진유영속도 모두 부화 직후부터 빨랐다. 이러한 특징들은 자어가 생존하기 어려운 겨울철 환경에 대한 자연선택 및 환경적응의 결과로 생각된다.
