Assessment of prognostic factors in dogs with mammary gland tumors: 60 cases (2014-2020)

  • Canine mammary gland tumors are the most common neoplasms in intact female dogs. Approximately half of all mammary tumors are malignant, and there is a risk of metastasis, which is associated with a poor prognosis. This study was to evaluate the prognostic factors of canine mammary gland tumors and the risk factors associated with the development of malignant tumors. From 2014 to 2020, 60 dogs with mammary gland tumors that underwent surgical treatment were evaluated in this retrospective study. Tumor size, TNM stage, and histopathological results were prognostic factors for 2-year survival after surgery. Every 10 mm increase in tumor size, increased the risk of death within 2 years after surgery 1.213 times. Dogs with TNM stage IV or V had 8.667 fold risk of death within 2 years after surgery. The 2-year survival rate for dogs with benign tumors was 90.2% and for malignant tumors was 67.3%. Tumor size is the most important prognostic factor for canine mammary gland tumors. As tumor size increased by 10 mm, the risk for development of malignant tumors increased by 1.487 times. Tumors larger than 30 mm are highly likely to be malignant, and metastatic evaluation and wide resection should be considered.

Distribution of mast cell on the development of DMBA-induced mammary carcinoma)

  • In order to observe the distribution of mast cell on the stages of the mammary carcinogenesis, the numerical changes of mast cells in the mammary tumor development in rats treated with DMBA and compound 48/80 have been investigated by the light microscope. The results observed were summarized as follows: The appearance of tumor were not observed during the whole experimental period in the rats of the control group received injection of sterile saline, but tumors appeared in 100% of the animals, the tumor induction time that represented the number of days elapsing between the 3rd DMBA administration until a first tumor became $10{\times}10mm$ in diameter was $42.5{\pm}4.7$ days and the mean number of tumor masses per rat was $3.4{\pm}1.2$ in the DMBA treated group. And the majority of the DMBA-induced mammary neoplasms were appeared cervical mammary gland and thoracic mammary gland. The histological findings of mammary carcinoma were recognized adenocarcinoma in the DMBA treated group. Mast cells were distributed within the adipose tissues and the interglandular connective tissue in the control, but found to be randomly dispersed within the tumor cell masses, in the connective tissues adjacent to the periphery of the tumor, the adipose tissues and the subcutaneous tissues contiguous to the region of tumor development in the DMBA treated group. Numerical alterations of mast cells were observed in the mammary tumors that separated into three major classes of tumors: hyperplasia, atypical hyperplasia and carcinoma. The number of mast cells were distributed in the connective tissues adjacent to the mammary gland was $45.3{\pm}3.4$ cells in the control group, but was $50.2{\pm}4.9$ cells, $126.7{\pm}10.5$ cells and $340.3{\pm}19.2$ cells according to each stages of mammary tumorigenesis in the DMBA treated group.

생쥐의 유선발달에 있어서 Elvax 40P Implant를 이용한 서방형 Adenosine의 역할 (Effect of Sustained-Release of Adenosine by Elvax 40P Implant on Mouse Mammary Development)

  • To examine local response of adenosie(purine nucleoside) on the developing mammary gland, Elvax 40P implants containing adenosine were surgically implanted into mammary fat pad of the five week old female ICR mice. Inguinal(the 4th) mammary glands of anesthetized mice were exposed andplaced the implants for 12 days. One gland was treated with an adenosine implant, while the contralateral gland received a blank implant as control. For whole-mount preparations, glands were stained with alum carmine, and for histological observation, micro-selected mammary glands were stained with hematoxylin and eosin Y. Implantation with Elvax 40P did not affect on the damage of neighboring mammary tissue. Adenosie 25 or 250$\mu\textrm{g}$ per slow-release implant stimulated local mammary end bud formation of ovariectomized mice such as end bud size and numbers of end bud per gland in a dose dependent manner(P<0.05), and lower concentration of adenosie(2.5 or 25$\mu\textrm{g}$/implant) increased numbers of end bud(P<0.05) and end bud size(P<0.1) of intact mice. Adenosine treatment and intact ovarian function had moderate interation effects on the stimulation of end bud formation at 2.5$\mu\textrm{g}$ adenosine/implant(P<0.1). In histological observation, adenosine implants increased numbers of mammary epithelial type of cells at mammary duct in the presence or absece ofovary. These results indicate that adenosine should be one of regulators in mouse mammary ductal growth.

Mammary Gland Indices at the End of Lactation in the Superovulated Javanese Thin-Tail Ewes

  • Thirty lactating Javanese thin-tail ewes (12 ewes had been injected, prior to mating, with 700 IU pregnant mare serum gonadotropin, and 18 ewes with saline as a control) were used to evaluate the effect of superovulation on milk production during lactation and mammary chemical indices at the end of lactation. Thirteen ewes (9 control and 4 superovulated ewes) were fed at low and the other 17 ewes (9 control and 8 superovulated ewes) were fed at high quality ration. Superovulated ewes, either fed at low or high quality ration, had dramatically higher milk yields (57%). At the end of lactation, superovulated ewes had higher mammary dry fat-free tissue, mammary DNA concentration, total mammary DNA and RNA contents than nonsuperovulated ewes. Superovulation did not affect mammary RNA and collagen concentrations, and total collagen content. Ration quality did not significantly increase milk production during lactation and mammary chemical indices at the end of lactation. The observed increase in milk production in the superovulated ewes was probably due to the increased mammary secretory cell number and their synthetic activities during lactation as a result of the increased endogenous hormonal stimulation of mammary growth and development during pregnancy.

Genome-wide identification of long noncoding RNA genes and their potential association with mammary gland development in water buffalo

  • Objective: Water buffalo, an important domestic animal in tropical and subtropical regions, play an important role in agricultural economy. It is an important source for milk, meat, horns, skin, and draft power, especially its rich milk that is the great source of cream, butter, yogurt, and many cheeses. In recent years, long noncoding RNAs (lncRNAs) have been reported to play pivotal roles in many biological processes. Previous studies for the mammary gland development of water buffalo mainly focus on protein coding genes. However, lncRNAs of water buffalo remain poorly understood, and the regulation relationship between mammary gland development/milk production traits and lncRNA expression is also unclear. Methods: Here, we sequenced 22 samples of the milk somatic cells from three lactation stages and integrated the current annotation and identified 7,962 lncRNA genes. Results: By comparing the lncRNA genes of the water buffalo in the early, peak, and late different lactation stages, we found that lncRNA gene lnc-bbug14207 displayed significantly different expression between early and late lactation stages. And lnc-bbug14207 may regulate neighboring milk fat globule-EGF factor 8 (MFG-E8) and hyaluronan and proteoglycan link protein 3 (HAPLN3) protein coding genes, which are vital for mammary gland development. Conclusion: This study provides the first genome-wide identification of water buffalo lncRNAs and unveils the potential lncRNAs that impact mammary gland development.

Nutrition-induced Changes of Growth from Birth to First Calving and Its Impact on Mammary Development and First-lactation Milk Yield in Dairy Heifers: A Review

  • This review focuses on the nutritional effects from birth until age at first calving on growth, mammary developmental changes, and first-lactation milk yield in heifer calves. The advancement in the genetic potential and the nutritional requirements of the animals has hastened the growth rate. Genetic selection for high milk yield has suggested higher growth capacity and hence increasing nutritional inputs are required. Rapid rearing by feeding high energy or high concentrate diets not only reduces the age of sexual maturity but also lowers the time period of attaining the age of first calving. However, high energy diets may cause undesirable fat deposition thereby affecting future milk yield potential. Discrepancies exist whether overfed or overweight heifers at puberty can influence the mammary development and future milk yield potential and performance. The data on post-pubertal nutritional management suggested that body weight at calving and post-pubertal growth rate is important in first lactation milk yield. There is a continuous research need for strategic feeding that accelerates growth of dairy heifers without reduction in subsequent production. Nutritional management from birth, across puberty and during pregnancy is critical for mammary growth and for producing a successful cow. This review will mostly highlight studies carried out on dairy breeds and possible available opportunities to manipulate nutritional status from birth until age at first calving.

Relaxin Hormone의 직접젇인 작용에 의한 돼지 유선상피세포의 발달과 세포외간 기질의 분해개조 촉진 (Direct Action of Relaxin on the Pig Mammary Glands to Promote Development of the Alveoli and Remodeling of the Extracellular Matrix)

  • 돼지에 있어서 약 115일간의 임신 기간 중 마지막 40일 동안에 유선 내에서 유액도관(duct), 분비소엽(lobules), 분비낭포(alveoli) 등의 parenchyma 조직이 현저하게 발달하게 되는데, 이는 단백질 호르몬 relaxin과 estrogen의 공동작용에 기인하여 촉진되는 것으로 알려져 있다. 최근에는 면역조직화학적 실험방법을 통해 relaxin hormone이 유선조직 세포(mammary gland parenchyma cells)에 직접적으로 작용하여 lobuloalveolar의 발달을 촉진할 수 있는 가능성을 시사하였다. 즉 Relaxin에 특이적이고 포화 가능한 Relaxin의 결합부위가 lobulo-llveolar 구조 및 유액도관(lactiferous duct)의 상피세포(epithelial cells)뿐만 아니라 혈관세포에서도 존재함을 관찰하였다. 본 연구의 목적은 유선조직 세포에 존재하는 relaxin 결합부위가 기능적으로 활성을 지닌 relaxin 수용체(functional relaxin receptor)인지를 규명하고 동시에 relaxin hormone이 유선조직 세포에 직접적으로 작용하여 lobulo-alveolar의 발달을 촉진하는 지에 대한 직접적인 증거를 제공하는 데 있다. 정상적인 생리주기를 가진 3마리의 생후 7개월 된 잡종재지에 대하여 양쪽 난소를 모두 제거하는 양측난소 절제시술(Bilateral ovariectomy)을 실시하였다.(day 0). 난소 절제시술 후 15일부터(day 15) 29일(day 29)까지 각 개체별로 estradiol benzoate(1 mg/2 ml /Com oil)를 12시간 간격으로 근육주사 하였다. 시술 후 22일째부터(day 22) 순수 정제된 relaxin을 miniature osmotic pump를 통하여 몸의 anterior end로부터 왼쪽부위의 4번째 유선에 일정한 속도(1 ug/hr)로 주입하기 시작하였으며, 동일한 개체의 오른쪽 부위의 4번째 유선에는 동일한 방업으로 생리적 식염수(대조군)을 주입하였다. 난소 절제수술 후 29일째(day 29) 각 개체를 희생하여 젖꼭지의 base로부터 1cm 떨어진 지점에서 osmotic pump의 위치로부터 가까운 parenchyma 조직 시료를 채취하여 약 5-mm cube으로 자른 다음 조직학적 분석을 위한 일반적 조직시료 처리과정을 시행하였다. 조직절편을 Weigert's iron hematoxylin과 함께 염색한 다음 Olympus AH-2 광학현미경을 이용하여 lobulo-alveolar의 발달과 세포외간 섬유사인 collagen의 밀도와 정렬정도를 조사하였다. 생리식염수로 처리된 유선조직에 있어서는 lobulo-alveolar의 발달이 거의 일어나지 못한 반면, relaxin을 처리한 유선조직은 lobulo-alveolar의 현저한 발달을 보였을 뿐만 아니라 분비낭포(alveoli) 내에서는 두드러진 분비물이 존재하였다. 또한 식염수로 처리된 유선조직에서는 비교적 조밀하고 매우 잘 발달된 collagen 섬유사 묶음을 가진 세포의 간 기질의 특징을 보였으나, relaxin 호르몬이 처리된 유선조직에서는 collagen 섬유사가 비교적 분산되어 있고 느슨하며 덜 조밀한 구조적 특징을 나타내었다. 결론적으로, 본 연구의 결과는 유선조직 내의 특이적 relaxin 결합세포가 기능적으로 활성을 지닌 relaxin 수용체를 함유하고 있으며, relaxin hormone이 유선조직 세포에 직접적으로 작용함으로써 lobulo-alveolar의 발달을 촉진함을 보여준다.

홍삼 사포닌에 의한 유선상피 및 유선암세포의 분화 유도 효과 연구 (Effects of Ginseng Saponins on the Induction of Differentiation in Mammary Epithelial Cells and Mammary tumor Cells)

  • 정상 흰쥐 유선상피세포 및 DMBA로 유도안 흰쥐 유선암세포를 in vitro 상태에서 배양시키며 이들의 성장과 증식에 대한 홍삼 사포닌(조사포닌, 총사포닌, Rbl, Rb2, Rc, Rd, Re, Rhl, Rtl2)의 영향을 관찰하였다. 홍삼 사포닌들의 세포 배양중 발생하는 4종의 세포집락의 형태(cobble stone, spindle, honey comb, senescence)에 영향을 미쳤다. 배양한 세포집락에 lucifer yellow용액을 이용하여 SLDT결과 홍삼 사포닌을 첨가하고 배양한 세포군에서 배양 2주째 세포 간극을 통한 세포간 정보전달체계가 양성을 나타내었다. 이것으로 홍삼사포닌이 분화가 덜된 상피 간세포의 분화를 촉진시켜 분화의 지표 중 하나인 세포체포신호전달을 유도한 것으로 사료된다. 그리고 정상 유선 및 유선암 미세절편들을 Matrigel에 배양한 결과 분화의 종 지표인 수종의 다세포구조물이 생성됨을 확인하였는데 정상 조직과 유선암세포로부터 생성된 구조물 ductal, webbed, stellate, squamous colony들of 생성되었는데 유선암세포로부터는 alveloar unit, foamy alveolar nit, squmaous, lobule-ductal, stellate, webbed colony들이 생성되어 다소 상이함을 확인하였다. 그러나 홍삼사포닌을 첨가하여 배양하였을 때 수종의 다세포 구조물 발생 빈도가 변하는 것으로 미루어 보아 홍삼 사포닌이 이들 다세포 구조물의 발생에도 영향을 미치는 것으로 생각된다. 편평상피암의 전암병소라 일컬어지는 편평상피화생의 경우홍삼 사포닌의 존재 하에 이들의 생성이 억제되고 분화가된 ductal colony들의 수가 증가된 것으로 보아 암예방 효과의 기전을 이해하는데 중요한 단서를 제공할 것으로 사료된다.

한우와 홀스타인 암송아지의 성장, 유선발달, 혈청 단백질과 혈청 중 Growth Hormone, Prolactin 및 Estradiol의 농도 비교 (Comparisons of Growth, Mammary Development, Serum Proteins and Serum Concentrations of Estradiol, Growth Hormone and Prolactin between Korean Native and Holstein Heifers)

  • Growth, mammary development, serum concentrations of growth hormone, prolactin and estradiol which affect the growth and mammary development, were investigated in Korean Native heifers and Holstein heifers from 5 months old to 15 months old. From 5 months old to 15 months old, body weights in Korean Native heifers averaged 37%, lower than those in Holstein heifers of the same age (P<.01). Teat volumes in Korean Native heifers are average 67% smaller than those in Holstein heifers. Analysis of serum protein by secondary electrophoresis showed differences in protein pattern between the breeds and between the individuals. From 5 months old to 15 months old, serum concentration of growth hormone in Korean Native heifers was averge 47% lower than that in Holstein (P<.01), yet prolactin concentrations in serum were about the same between the two breeds. However serum concentration of estradiol in Korean Native heifers was average 20% higher than that in Holstein of same age (P<.01). This result showed evidently that the decrease of the growth hormone concentration in serum, while growing up, causes to retard the growth and mammary development in Korean Native heifers compared to Holstein heifers.

Rat mammary carcinoma의 발육(發育)에 있어서 비만세포(肥滿細胞)의 영향(影響)에 관한 병리조직학적(病理組織學的) 연구(硏究) 2. 종양발육(腫瘍發育)에 있어서 mast cell mediator의 영향 (Histopathological Studies on the Influence of Mast Cell in the Growth of Rat Mammary Carcinoma 2. Effect of Mast Cell Mediator on the Development of Rat Mammary Carcinoma)

  • In order to know the influence of mast cells on the mammary tumor development, the growth of the mammary carcinoma, the numerical changes and the morphological findings of mast cells appeared in the tumor were microscopically observed in the rat treated with DMBA and each chemical of histamine, heparin, pyrilamine or cimetidine. The results observed were summarized as follows: The tumor induction time that represented the number of days elapsing between the 3rd DMBA administration until a first tumor became $10{\times}10mm$ in diameter was $42.5{\pm}4.7$ days, and the mean number of tumor mass per rat was $3.4{\pm}1.2$ in the DMBA-treated group. No significant difference was apparent in the tumor induction time of the histamine-treated group, heparin-treated group or pyrilamine-treated group compared with the control group, but in the cimetidine-treated group the tumor induction time was $61.8{\pm}10.6$ days (p<0.005). The mean number of tumors per rat was $2.1{\pm}0.9$ in the cimetidine-treated group in contrast to $3.4{\pm}1.3$ in the control group (p<0.005). Numerical changes of mast cells were observed according to the development of DMBA induced mammary tumors that were separated into three major classes of tumors. The numbers of mast cells in all the experimental group were inclined to increase significantly according to the mammary tumor development (p<0.005), and the histamine-treated group, heparin-treated group, or pyrilamine-treated group were nearly similar to the control group. But the mast cells in the each stage of tumor development were more numerous in the cimetidine-treated group than in the control group (p<0.005). There were not significant in the numerical changes of mast cells among the experimental groups on each stage of carcinomas separated by early stage, middle stage and late stage. In the morphological characteristics of mast cells, the degranulation was not detectable from the hyperplasia stages to the early stage of carcinoma, but its degranulation was observed at the middle stage of carcinoma. Most mast cells were nearly degranulated at the late stage of carcinoma. The histamine treated group, pyrilamine-treated group and cimetidine treated group did not differ from the control group in morphological changes of mast cells, but the degranulation was shown mild in the heparin-treated group. And the degranulation gave rise to the depletion of intercellular matrix via exocytosis all the experimental group. From above results, it is supposed that mast cells inhibit the tumor development and that the inhibition is not caused by a single-factor, but by a complex activities of mast cell mediators.
