Investigation of Cytoplasmic Male Sterility and Development of Maintainer and Restorer Lines in Rye (Secale cereale L.)

  • Rye has been a major winter forage crop in Korea. Varietal improvement of rye has been practiced either by hybrid or population breeding systems. Hybrid breeding offers important advantages over population breeding since it is normally a cross-pollinated crop. The hybrid breeding in rye has been possible since cytoplasmically inherited forms of male sterility (CMS) and corresponding nuclear restorer genes were found. The objectives of this research were to develop the maintainer and restorer lines of Korean inbred lines and to estimate the effect of 'Pampa' type of CMS cytoplasm on yield and its related characteristics. For easy discrimination of male-sterile status of plants, anther scoring and the restore index system in which seed-setting and pollen quantity of viability were taken into account were established. High significant correlation between pollen quantity and pollen viability was found. For "Pampa" cytoplasm, four of 14 Korean inbred lines tested turned out to be a maintainer but no restorer was found. But for "235b" CMS cytoplasm, seven inbred lines acted as complete restorers. The Korean inbred rye lines acted mainly as maintainers in "Pampa" cytoplasm but acted mainly as restorer in "235b" cytoplasm. The 'Pampa' cytoplasm inducing male sterility reduced cohn length and plant height and increased the number of tiller, so forage yield and grain yield were enhanced.   • Grain Sorghum varieties grown in Korea have low productivity and are constrained by the fact that farmers' choice of improved varieties is limited. Hybrid cultivars have been demonstrated to be more productive and food security than pure line varieties. However, There's no available hybrid cultivars and never planted before in Korea. This study was, therefore, conducted to determine combining ability of Korean landrace varieties and cultivar, the level of heterosis of experimental hybrids depending on environments. Two cytoplasmic male-sterile lines were crossed with six male-fertile lines in accordance with North Carolina II mating scheme to generate 12 experimental hybrids. The hybrids were evaluated in replicated row-column alpha designs at two sites in Texas and South Korea under on-season production conditions. For each trait, general combining ability (GCA) and specific combining ability (SCA) effects were estimated using the line-tester method of analysis. Results indicated significant differences among genotypes for both grain yield potential and secondary traits. Hybrids were predominant for grain yield, and displayed up to 127% heterosis of A03017 ${\times}$ Sodamchal in Korea. Positive GCA for yield was observed for Donganme, Hwanggeumchal and Jungmo4002 in Korea. Among them Jungmo4002 and Donganme had highly positive for yield (227.90 and 84.90 kg ha-1), while it showed negative GCA for yield in College station (-40.90, -189.60 kg ha-1). ATx630 ${\times}$ Sodamchal had the greatest SCA for yield (468.10 kg/ha) in College station but at South Korea its SCA effect was -302.40 kg/ha. A similar trend was observed, followed by A03017 ${\times}$ Donganme (SCA effect = 215 kg ha-1 at College station vs. -44.20 kg ha-1 at South Korea). At South Korea the greatest positive SCA effect for grain was observed in ATx630 ${\times}$ Jungmo4002 as 322.20 kg/ha. Both GCA and SCA effects significantly interacted with site effects demonstrating the need for region adaptation of potential cultivars and test for developing Korea suitable cultivar.

New Mid-late Maturing F1 Hybrid Onion Cultivar, "Yeongpunghwang" (양파 추파형 중·만생 다수성 신품종 "영풍황")

  • New onion hybrid cultivar, "Yeongpunghwang" was developed from cross between "MOS8", male sterile line and "Mokpo 11", pollen line by Mokpo Sub-Station, National Institute of Horticultural & Herbal Science, RDA in 2008. The crossing line was performed yield test in 2003. IT was evaluated as "Mokpo 11" in the regional yield trial test(RYT) from 2006 to 2008 during three years. This line is semi-erect plant growth type and has green leaf colour. It has broad ovate bulb shape and yellow skin colour in bulb characteristics. Plant height, leaf number, and bolting rate was 78cm, 7.9, and 3.9%, respectively. Lodging date as harvesting indicator was on 26 May. The yield potential of "Yeongpunghwang" was out-yielding 6% compare to check variety ("Sunpower") by with 8,344 kg/10a in RYT. It showed moderated cold hardness in all regional location which was conducting RYT, including Muan, Goheung, Changyeong and Andong. So this variety was recommend as a leading variety at southern area of korea.

Some Management Practices Affecting Outcrossing and Seed Production in Burley Tobacco (Nicotiana tabacum L.) (연초 버어리종의 자연교잡율과 종자생산에 관련된 몇가지 요인)

  • In this study effects of isolation distance, transplanting time of maternal plants, and bagging of flower head with the gauze-cloth bag on the outcrossing of burley tobacco (Nicotiana tabacum L.) were investigated. Also the effect of fertilizer level and control of the number of capsules per plant on seed production and quality were examined. A male sterile line. produced 0.3 to 3.8 capsules Per plant when it was Planted with normally flowering tobacco with the average outcrossing of 7.2 plants, ranging from 2 to 18 out of 20 plants. With the farther the isolation distance between maternal plants and pollen donor plant, the lower the outcrossing occurred. Outcrossing occurred even at the isolation distance of 312 m. When the maternal plants were transplanted 35 days after transplanting the pollen donor ones, the outcrossed plants were not decreased significantly. The bagging of the flower head with the gauze-cloth bas (#0.9∼ 1.0 mm) decreased the outcrossed plants significantly, but couldn't prevent the outcrossing completely. The seed amount per plant was higher in the highly fertilized cultivation. The number of seed capsules per plant affected significantly on seed yield and quality. When the seed capsules was controlled by 30 or 50 capsules per plant, the weight of 1,000 seeds and germination rate were higher than those with 70 or 90 capsules per plant. A FA Iinterspecific Hybrid Lily 'Green Star' with Unspotted Greenish Yellow Petals

  • An interspecific hybrid lily cultivar 'Green Star' was bred in 2005 at the National Institute of Horticultural and Herbal Science (NIHHS), Rural Development Administration (RDA), Korea. The crossing and in vitro embryo rescue was conducted between Lilium FA97-2 (L. ${\times}$ formolongi 'Silky White' ${\times}$ L. Asiatic 'Sunray') and L. Asiatic 'Bomi (Byeongga ${\times}$ Connecticut King)' by cut style pollination method (CSM) at Suwon in 2000. The first selection was done and was tentatively named as 'FA03-5' in 2003. After in vitro multiplication and bulbing production of 'FA03-5' line, growth and flowering characteristic tests were conducted from 2003 to 2005. The evaluation of characteristics and consumer preferences were surveyed at a lily flower show of NIHHS in 2005. 'Green Star' flowered in the middle of June and grew more than 120 cm stem in length. Flowers bloomed facing upward, unspotted in petals and greenish yellow (RHS, Y6D). 'Green Star' was male sterile. Year-round flowering can be done by storing the bulb under $-1.5^{\circ}C$ conditions. It was needed to control the Botrytis disease in wet season.

Prospect on the Fixation of $F_1$ Hybrid Seed by Means of 2n Apomixis (2n性 單爲생殖 이용에 의한 固定 $F_1$種子 생산과 그 展望)

  • Plants belonging to the category of 2n apomixis or agamospermy form embryos and seeds without the processes of normal meiosis and syngamy. Seeds produced in this way have identical genotype of their maternal parent. Three different types of agamospermy are recognized: diplospory, apospory, and adventitious (adventive) embryony. $F_1$ hybrid cultivars cannot be used as seed sources in the next ($F_2$) generation because this generation would be extremely variable as a result of genetic segregation. Hybrid vigor is also reduced in the $F_2$ generation. Therefore, parental stocks for hybrid seed production need to be maintained and cross must be continuously repeated. Agamospermic 2n apomixis would make it possible to fix the genotype of a superior variety so that clonal seeds faithfully representing that genotype could be continuously and cheaply produced independent of pollination. That is, $F_1$ hybrid seeds could be produced for many generations without loss of vigor or genotype alteration. Production of apomictic $F_1$ hybrid seed would be simplified because line isolation would not be necessary to produce seed or to maintain parental lines, and the use of male-sterile lines could be avoided. Overall, apomixis would enable a significant reduction in hybrid seed production costs. Additionally, the production of clonal seed is not only important for seed propagated crops, but also for the propagation of heterozygous fruit trees and timbers. Clonal seed would help avoid costly and time-consuming vegetative propagating methods that are currently used to ensure the large-scale production of these plants. Apomixis is scattered throughout the plant kingdom, but few important agricultural crops possess this trait Therefore, most research to date has centered on introgressing the trait of apomixis into agricultural crops such as wheat, maize, and some forage grasses from wild distant relatives by traditional cross breeding. The classical breeding approach, however is slow and often impeded by many breeding barriers. These problems could be surmounted by taking mutagenesis or molecular approach. Arabidopsis thaliana is a tiny sexually reproducing plant and is convenient in constructing and screening in molecular researches. Male-sterile mutants of Arabidopsis are particularly suitable genetic background for mutagenesis and screening for apomictic mutants. Molecular approaches towards isolating the genes controlling the apomictic process are feasible. Direct isolation of genes conferring apomixis development would greatly facilitate the transfer of this trait to wide variety of crops. Such studies are now in progress.

Allelism and Molecular Marker Tests for Genic Male Sterility in Paprika Cultivars (파프리카 시판 품종에 대한 유전자적 웅성불임성의 대립성 및 분자표지의 유용성 검정)

  • Paprika (Capsicum annuum L.), a colored bell-type sweet pepper, is one of the most important money making vegetable crops in Korea. The cultivation area, total production, and exports of paprika are gradually getting increased, but the paprika cultivars used in Korea are all imported. It was well-known that the genic male sterility (GMS) is the main way to produce paprika hybrid seeds. However, it is little known that how many and what kinds of ms genes are used for breeding of paprika $F_1$ varieties. In this study, eight paprika cultivars ('Special', 'Debla', 'Plenty', 'Fiero', 'Boogie', 'Fiesta', 'Derby', and 'Minibell'), popularly cultivated in Korea and three different genic male sterile lines ('GMSP', 'GMS3', and 'GMSK') were used. For allelism test among the $F_1$ cultivars, half diallel crosses were performed. The result demonstrated that the most of the GMS in paprika cultivars except for 'Minibell' were same allele. To identify which GMS gene(s) were used for paprika $F_1$ cultivars, top crosses between previously known GMS lines and the $F_1$ cultivars were performed. As a result, we found that the $ms_k$ and the $ms_p$ genes were alleles for the GMS of 'Minibell' and for the other cultivars, respectively. We also confirmed that the GMS gene identification using GMSK-CAPS marker linked to the $ms_k$ gene and the PmsM1-CAPS marker linked to the $ms_p$ gene in $F_2$ progenies of 'Minibell' and 'Fiesta' and 'Derby' cultivars, respectively. In addition, we developed the PmsM2-CAPS marker for 'Plenty', 'Fiero', and 'Boogie' cultivars. We expect that these markers will be very useful for breeding new maternal (male sterile) line of paprika.

A New F1 Hybrid Variety of Rapeseed 'Suan' with Early Maturing and High Oleic Acid (조숙 올레인산 고함유 1대잡종 유채 신품종 '수안')

  • 'Suan' is a new $F_1$ hybrid variety of rapeseed (Brassica napus L.) with early maturing and high oleic acid. This hybrid variety was bred by the cross between Mokpo-CGMS (male sterile line) and 8516-B-5-6-5-3 (restore line) for the production of bio-diesel and edible oil in 2006. 'Suan' has green and parted leaf, yellowish flower, and black seed coat. 'Suan' is more tolerant to lodging and stem rot compared to 'Sunmang', check variety. The ripening date of 'Suan' is June 2nd which is 6 days earlier than 'Sunmang'. Yield trials were conducted from 2006 to 2007 and regional adaptation trials were examined at five locations each in 2008 and 2009. The average seed yield of regional adaptation trials was 381 kg/10a that was 4% higher than that of 'Sunmang'. Total oil content of 'Suan' was 44.3%. Oleic acid content was 68.3%, which is 5.2% higher than 'Sunmang' but, erucic acid was not detected. Total glucosinolate content was 2.31 mg/g. Therefore, this variety is recommended as a leading variety at southwestern area including Jeonnam, Jeonbuk and Kyongnam provinces of South Korea.

A FA Intersectional Hybrid Lily 'Golden Center' with Light Yellow Petals (연노랑색 FA 종간잡종 나리 신품종 '골든센터' 육성)

Flower blooms facing upward, with light yellow petals (RHS, GW157C). The pollen of 'Golden Center' is sterile. Year-round flowering can be done by storing the bulb under $-1.5^{\circ}C$ conditions. It is needed to control Botrytis disease in wet season.

A New F1 Hybrid Variety of Rapeseed (Brassica napus L.), 'Jomang' with Early Maturation, High Yield and High Oleic Acid (조숙, 다수성, 올레인산 고함유 유채 신품종 '조망')

  • 'Jomang' is a new $F_1$ hybrid variety of rapeseed (Brassica napus L.) with early maturation, high yield and high oleic acid. This hybrid variety was developed from the cross between Mokpo-CGMS (male sterile line) and 8516-B-5-6-5-3 (restore line) for the production of edible oil and biodiesel in 2006. Yield trials were conducted from 2007 to 2008, and regional adaptation trials were examined at four locations in 2008. 'Jomang' has green and parted leaf, yellowish flower and black seed coat. The ripening date of 'Jomang' is 1~4 days earlier than 'Sunmang' and 'Tammiyuchae'. The average seed yield of the regional adaptation trials was 406 kg/10a, which was 7% and 42% higher than those of 'Sunmang' and 'Tammiyuchae,' respectively. Total oil content of 'Jomang' was 44.5%. 'Jomang' has no erucic acid in fatty acid composition. However, oleic acid content is 68.2%, which is 3.4% and 5.1% higher than 'Tammiyuchae' and 'Sunmang,' respectively. Total glucosinolate content was 2.31 mg/g. Therefore, this variety is recommended as a leading variety at southwestern area including Jeonnam, Jeonbuk and Kyongnam provinces and Cheju island of Korea.
