$^{13}C\;and\;^{51}V $ Nuclear Magnetic Resonance Studies of Vanadium (Ⅴ)-$\alpha$-Hydroxycarboxylate Complexes

  • The interaction of vanadium(V) with various a-hydroxycarboxylate ligands in aqueous solution at pH 3.2 have been studied by $^{51}V$ and $^{13}C$ NMR spectroscopies. From the results it is supposed that vanadates mainly form the octahedral complexes with lactate, 2-hydroxybutyrate, glycerate, and malate. While, vanadates form the trigonal-bipyramidal complexes with glycolate, tartarate, and 2-hydroxy-3-methylbutyrate, and tetrahedral complexes with pyruvate(diol), 2-hydroxyisobutyrate, and 2-hydroxy-3-methylbutyrate. The bipyramidal products are formed as monomeric compounds. The octahedral products are formed as dimeric compounds with no evidence for a significant proportion of the monomeric derivatives. The complexes are mainly formed through the coordination at the carboxylate and the 2-hydroxyl groups of the ligands.

Effect of Light on Development of Microbody Functions in the Cotyledons of Rape (Brassica napus L.) Seedlings (유채 종자의 Microbody 기능 발달에 미치는 빛의 영향)

  • The changes in activities of glyoxysomal and peroxisomal enzymes during the transition from fat degradation to photosynthesis were investigated with the cotyledns of rape (Brassica napus L.) seedlings. The development and disappearance of glyoxysomal enzyme (isocitrate lyase, EC 4.1.3.1; malate dehydrogenase, EC 1.1.1.37; catalase, EC 1.11.1.6) activities took place independently of light. It is concluded that the mobilization of storage fat is independent of photomorphogenesis. During early periods of development in the dark of light (days 1 through 3), the glyoxysomal enzyme activities were relatively high and the enzyme activities rose to a peak at 3rd day after sowing. Thereafter, the activities decreased gradually. While glyoxysomal enzyme activities were dropping, the peroxisomal enzyme (glycolate oxidase, EC 1.1.3.1) activities were increasing rapidly during the transition period in the light. Moreover, the changes of enzyme activities of the common microbody marker, catalase, indicated both functional patterns. The enzyme patterns in rape cotyledons indicate that the glyoxysomal function of microbodies is replaced by the peroxisomal function of these organelles during the transition from fat degradation to photosynthesis.

Electrophoresis of Reptilian Blood Proteins (파충류 혈액 단백질의 전기영동)

  • The blood proteins of ten reptilian species were studied by cellulose acetate elctrophoresis. Three members examined of the genus Agkistrodon have unusually similar patterns in plasma protein, hemoglobin, lactate dehydrogenase and malate dehydrogenase. On the basis of their electrophoretic patterns, it was concluded that Agkistrodon blomhoffii brevicaudus was closely related to the others. In general the plasma protein patterns reflect species specificity. Under the conditions employed, all snakes had a single hemoglobin band except Dinodon rufozonatum rufozonatum which showing two component patterns. Two members of the Chelonia showed four bands of hemoglobin. The zymograms indicated a distinct divergence in blood proteins of the Squamata from those of the Chelonia. The results reflected superficially the established phylogenies of these groups.

Choline supplementation improves the lipid metabolism of intrauterine-growth-restricted pigs

  • Objective: The objective of this study was to investigate the effects of dietary choline supplementation on hepatic lipid metabolism and gene expression in finishing pigs with intrauterine growth retardation (IUGR). Methods: Using a $2{\times}2$ factorial design, eight normal birth weight (NBW) and eight IUGR weaned pigs were fed either a basal diet (NBW pigs fed a basal diet, NC; IUGR pigs fed a basal diet, IC) or a diet supplemented with two times more choline than the basal diet (NBW pigs fed a high-choline diet, NH; IUGR pigs fed a high-choline diet, IH) until 200 d of age. Results: The results showed that the IUGR pigs had reduced body weight compared with the NBW pigs (p<0.05 from birth to d 120; p = 0.07 from d 120 to 200). Increased (p<0.05) free fatty acid (FFA) and triglyceride levels were observed in the IUGR pigs compared with the NBW pigs. Choline supplementation decreased (p<0.05) the levels of FFAs and triglycerides in the serum of the pigs. The activities of malate dehydrogenase and glucose 6-phosphate dehydrogenase were both increased (p<0.05) in the livers of the IUGR pigs. Choline supplementation decreased (p<0.05) malate dehydrogenase activity in the liver of the pigs. Gene expression of fatty acid synthase (FAS) was higher (p<0.05) in the IC group than in the other groups, and choline supplementation decreased (p<0.05) FAS and acetyl-CoA carboxylase ${\alpha}$ expression in the livers of the IUGR pigs. The expression of carnitine palmitoyl transferase 1A (CPT1A) was lower (p<0.05) in the IC group than in the other groups, and choline supplementation increased (p<0.05) the expression of CPT1A in the liver of the IUGR pigs and decreased (p<0.01) the expression of hormone-sensitive lipase in both types of pigs. The gene expression of phosphatidylethanolamine N-methyltransferase (PEMT) was higher (p<0.05) in the IC group than in the other groups, and choline supplementation significantly reduced (p<0.05) PEMT expression in the liver of the IUGR pigs. Conclusion: In conclusion, the lipid metabolism was abnormal in IUGR pigs, but the IUGR pigs consuming twice the normal level of choline had improved circulating lipid parameters, which could be related to the decreased activity of nicotinamide adenine dinucleotide phosphate-generating enzymes or the altered expressions of lipid metabolism-related genes.

Fractionation of the Cells of Staphylococcus aureus (Staphylococcus aureus의 균체분획(菌體分劃))

  • The growing cells of S. aureus were fractionated along the Schmidt-Thannhauser-Schneider's technique into several fractions such as TCA(trichloroacetic acid)-soluble, lipid, nucleic acid, protein and residue fraction. They were also fractionated according to their cellular structure into Sonic-supernatant, SDS(sodium lauryl sulfate)-soluble, Formamide-soluble and Residue fraction. Fractionation was carried out by orderly treatment of the Sonic pellet with 1.0% SDS and hot$(150^{\circ}C)$ formamide, and the pellet was prepared by centrifugation of the cells sonic osillated for 20 minutes at 150 watt. Sonic-supernatant fraction contained a 91.3% of total DNA while other fractions contained less than 9.5%. SDS-soluble fraction showed a high activity of malate dehydrogenase(13.67 unit/mg protein) and which was higher 22.3 times than the activity found from unsoluble fraction. Formamide-soluble fraction prepared from SDS-undoluble pellet by using the hot formamide exhibited a clear action of reducing sugars against the Anthronesulfate, while it exhibited no clear action against the ninhydrin. However, contrastly, the residue remainnning after extraction with formamide exhibited a clear action against ninhydrin and glucosamine was detected form the hydrolysate of residue by paper chromatography. From these results it is considered that the Sonic-supernatant fraction is mainly consisted of plasmic component of the cells. Other fractions, SDS-soluble, Formamide-soluble and Residue, should be consisted of plasma membrane, lipoplysaccharide and peptidoglycan of the cell, respectively.

  • In order to remove the high content of malic acid (0.48%) in freshly fermented apple wine by applying malo-alcoholic fermentation(MAF) using malate-decomposing yeasts, Schizosaccharomyces japonicus var. japonicus or Schizosaccharomyces pombe, some factors which influenced malic acid decomposition in apple wine by malate-decomposing yeasts were investigated. By incubating the apple wine with these yeasts, 80% of initial malic acid was decomposed in 13 days at $20^{\circ}C$, whereas only 32% reduction in malic acid was observed in 40 days when MAF was not induced. For malic acid decomposition, Schizosaccharomyces pombe was superior to Schizosaccharomyces japonicus var. japonicus in natural medium such as apple wine, whereas Schizosaccharomyces japonicus var. japonicus was superior to Schizosaccharomyces pombe in synthetic medium. The optimum temperature for the MAF using these yeasts was between $20\;to\;30^{\circ}C$ and no adverse effect was detected by the addition of $SO_2$ up to 200ppm. Additions of $Mg^{2+}$ or certain amino acids such as alanine, aspartic acid to the wine could enhance the composition of malic acid.

Isolation of Pseudoxanthomonas sp. W12 and WD32 Producing Extracellular Protease (단백질분해효소를 생산하는 Pseudoxanthomonas sp. WD12와 WD32의 분리)

  • Proteases catalyze hydrolytic cleavage of a peptide bond between amino acids and occupy pivotal positions in application in physiological and commercial fields. During the screening for novel bacteria producing extracellular protease, two bacterial strains, WD12 and WD32, were isolated from rotten trees and they made clear zone on LB plates supplemented with 1% skim milk. The similarities of 16S rRNA gene sequence of either WD12 or WD32 to GenBank database showed the highest to Pseuoxanthomonas mexicana as 97.8 and 99.8%, respectively. Phylogenetic analysis showed that both isolated was located within the cluster comprising P. mexicana and P. japonesis. WD12 and WD32 were catalase- and oxidase-positive, Gram-negative rod strains. In case of WD12, it could assimilate malate, but could not assimilate D-mannose, which were different characteristics from P. mexicana. Both Pseuoxanthomonas sp. WD12 and WD32 optimally produced extracellular protease at $35-37^{\circ}C$, and maximal activity showed as 656 unit/ml and 267 unit/ml, respectively.

Cell Biological Studies on Brain Formation at the Early Stage of Chick Embryogenesis (초기계배의 뇌 형성에 관한 세포 생물학적 연구)

  • The effect of tryptophan on brain formation at the early stage of chick embryo has been investigated morphologically using electron microscope. The electron micrographs of cerebral cortex cells of $5\\sim10$ day old chick embryo, which received 1.0mg of tryptophan showed that the irregularity, evagination and disruption of nuclear membrane and nuclear chromatin condensation, nucleolar chromatin margination and segragation. Hypertrophy of stalks, vesicles, and vacuoles can be seen and dilation and vesiculation of rough endoplamic reticulum and polysome disaggregation occured. Protein and RNA levels and the activity of several enzymes such as lactate dehydrogenase, succinate dehydrogenase, malate dehydrogenase and glucose 6-phosphate dehydrogenase of tryptophan administered group were significantly lower than those of control group suggesting that the tryptophan administration depressed protein biosynthesis resulting in the decrease of enzyme activity. It was found that serotonin content of egg yolk which has been incubated for 10 days were as much as three times that of control egg yolk. It is not clear whether the increase of serotonin content might inhibit intracellular yolk granule degradation which might result in malformation of chick embryo, but it is likely that tryptophan administration might depress protein biosynthesis, consequently, the enzyme biosynthesis would be impaired. This might give rise to improper development of chick embryo.

Interorder Protoplast Fusion between Pleurotus ostreatus and Ganoderma applanatum (느타리버섯과 잔나비걸상버섯과의 이목간(異目間) 원형질체(原形質體) 융합(融合))

  • Interorder heterokaryons were obtained by polyethylene glycol induced fusion of protoplasts from auxotrophic mutants of Pleurotus ostreatus in agaricales and Ganoderma applanaturm in aphyllophorales. When transferred to MMM plates, all fusion colonies exhibited an extremely growth rate. During three times subcultivation on MCM the growth rate of fusants showed faster little by little. Seventy-five % fusion products of potoplasts showed mixed morphologies between those of P. ostreatus and G. applannatum in the first subcultivation on MCM and MGM. The phenotype of these fusants changed similar those of P. osteatus type after three times subcultivation on MCM. However, phenotype of 25% stable strains did not change on subcultivation. Hyphae of all fusion products did not form true clamp connection. All these types did not produce primordia. A comparrison of interorder somatic hybrids was made using isozyme analysis of esterase, malate dehydrogenase and peroxidase. In most cases the enzyme patterns of G. applanatum were not distinct, however, fusant showed non-parental bands.

Morphological and Biochemical Characteristics in the Korean Landraces of Mulberry Tree, Morus (한국 재래뽕의 생태 및 품종간 특이성에 관한 연구)

  • This study was conducted to characterize mulberry varieties by the analysis of morphological characters as well as biochemical and molecular biological markers. As for the budding stage Geumsang, Chosang i and Yeongbyeonchuwoo were early, but Dangsang 6, Hwangchuwoo were late. The lowest varietiy in rate of death atop was Dangsang 8(0.0%). Suncheonppong was the highest leaf yields in spring and autumn rearing season. In biochemical isozyme analysis, peroxidase gave good zymogram patterns in isoelectric focusing electrophoresis. There were high variations in RAPD analysis among the mulberry trees. From the obtained peroxidase and RAPD variations, cluster phylogenetic analysis was carried out using NT-SYS PC program. There were no clear grouping patterns between native varieties and leading varieties. The highest similarity was observed between Suwonsang 1 and Suwonsang 2 at about 90% similarity level.
