Analysis of Off-flavor Generated from a Polyethylene Terephthalate Water Bottles and Caps by Using an Electronic Nose (MS-전자코를 사용한 페트(polyethylene terephthalate) 생수병과 뚜껑의 이취 이행 분석)

  • The objective of the study was to investigate the off-flavor generated from PET water bottles and their caps by using a mass spectrometry-based electronic nose. The ion fragment data obtained from the electronic nose were used for discriminant function analysis (DFA). In the case of increased concentrations of the contamination of water, the off-flavor pattern depended on the discriminant function second score instead of the discriminant function first score. To identify the cause of off-flavor in polyethylene terephthalate (PET) bottled water, the PET bottle and its cap were analyzed by DFA. The results showed that the cap generated more volatile compounds than the bottle or mineral water did. The substances causing the off-flavor were predicted to be 2,4-di-tert-butylphenol (2,4-DTBP), nonanal, and decanal when the main peak of the mass spectrum was compared with the major ion fragments of the electronic nose. Thus, using this method, we could determine whether the PET water bottle was contaminated and whether the off-flavor resulted from contamination of the bottle cap.

Changes in Nutrient Levels of Aqueous Extracts from Radish (Raphanus sativus L.) Root during Liquefaction by Heat and Non-heat Processing

  • The amount of cellular components including soluble sugars, amino acids, organic acids and glucosinolates (GLS) was investigated during radish root processing to develop a radish beverage. The radish root was divided into two parts, white and green tissue, and processed separately by extracting the juice from the fresh tissue and from the boiled tissue to compare differences in the components content among the preparations. The overall palatability of both the fresh and boiled extracts from the green part of the radish was higher than that of the same extracts from the white part. The sweetness of extract by boiling increased and its pungency decreased, thereby the palatability increased by being compared to the fresh radish extract. The sweetness was affected by sucrose not by glucose or fructose of monosaccharides by showing different sucrose contents according to treatment comparing palatability. Malic acid was identified as primary organic acid, and the content was higher in both the fresh and boiled extracts from the white part than in the extracts from the green part of the radish. The fresh extract from the green part of the radish contained more essential amino acids, such as threonine and valine, and more hydrophilic amino acids including glutamic acid, aspartic acid, and arginine than those of the fresh extract from the white part, suggesting the green fresh part is more palatable than the white fresh part. The main sulfur compound was ethylthiocyanate in radish, and others were butyl isothiocyanate, dimethyl-disulfide, and 4-methylthio-3-butylisothiocyanate. The four GLS were detected much more in the fresh green and fresh white parts of the radish because they evaporated during boiling. The contents of the four sulfur compounds were higher in the white fresh part than in the green fresh part, which is likely the reason the pungency was higher and the palatability was lower in the white fresh part than in the green fresh part of the radish. The ascorbic acid content was higher in the fresh extract compared to the boiled extracts from both the green and white parts. Taken together, these findings indicate that fresh radish extract is superior to obtain in terms of retaining desirable nutritional and functional components for health.

Quality Characteristics of Yakju as Affected by Rice Nuruk with Different Degrees of Milling (도정도별 쌀누룩 약주의 품질특성)

  • This study aimed to investigate the effect of rice nuruk prepared from rice with different degrees of milling (0, 10, 30, 50, and 70%) on physicochemical qualities of yakju. The results indicated significant differences in the qualities of yakju prepared using rice nuruk from rice with different degrees of milling. Specially, pH, amino acidity, brownness, b value, and color difference were significantly reduced as the degrees of milling increased, whereas contents of alcohol and volatile acid, reducing sugar, and L value were increased. Malic acid, succinic acid, and lactic acid were the main organic acids in yakju, and there were significant differences among the treatments in malic acid, lactic acid. However, there was no tendency according to different degrees of milling. In addition, contents of total free amino acids and nitrogen compounds showed no tendency according to degrees of milling, whereas the content of aspartic acid showed a tendency to decrease significantly. The content of 2-phenyl alcohol in the volatile flavor components showed a tendency to increase significantly with higher degree of milling of rice nuruk, whereas contents of n-propanol and iso-amyl alcohol tended to decrease significantly.

Determination of the volatile flavor components of orange and grapefruit by simultaneous distillation-extraction (연속수증기증류추출법에 의한 오렌지와 자몽의 휘발성 유기화합물 확인)

  • The volatile flavor components of the fruit pulp and peel of orange (Citrus sinensis) and grapefruit (Citrus paradisi) were extracted by simultaneous distillation-extraction (SDE) using a solvent mixture of n-pentane and diethyl ether (1:1, v/v) and analyzed by gas chromatography-mass spectrometry (GC-MS). The total volatile flavor contents in the pulp and peel of orange were 120.55 and 4,510.81 mg/kg, respectively, while those in the pulp and peel of grapefruit were 195.60 and 4,223.68 mg/kg, respectively. The monoterpene limonene was identified as the major voltile flavor compound in both orange and grapefruit, exhibiting contents of 65.32 and 3,008.10 mg/kg in the pulp and peel of orange, respectively, and 105.00 and 1,870.24 mg/kg in the pulp and peel of grapefruit, respectively. Limonene, sabinene, ${\alpha}$-pinene, ${\beta}$-myrcene, linalool, (Z)-limonene oxide, and (E)-limonene oxide were the main volatile flavor components of both orange and grapefruit. The distinctive component of orange was valencene, while grapefruit contained (E)-caryophyllene and nootkatone. $\delta$-3-Carene, ${\alpha}$-terpinolene, borneol, citronellyl acetate, piperitone, and ${\beta}$-copaene were detected in orange but not in grapefruit. Conversely, grapefruit contained ${\beta}$-pinene, ${\alpha}$-terpinyl acetate, bicyclogermacrene, nootkatol, ${\beta}$-cubebene, and sesquisabinene, while orange did not. Phenylacetaldehyde, camphor, limona ketone and (Z)-caryophyllene were identified in the pulp of both fruits, while ${\alpha}$-thujene, citronellal, citronellol, ${\alpha}$-sinensal, ${\gamma}$-muurolene and germacrene D were detected in the peel of both fresh fruit samples.

Alternaria mycotoxins and its incidence in fruits and vegetables

  • Alternaria is a ubiquitous fungal genus, widely distributed in the environment and a range of different habitats. It includes both plant pathogenic and saprophytic species, which can affect crops in the field or cause post-harvest spoilage of plant fruits and kernels. Numerous Alternaria species cause damage to agricultural products including cereal grains, fruits and vegetables, and are responsible for severe economic losses worldwide. Most Alternaria species have the ability to produce a variety of secondary metabolites, which may play important roles in plant pathology as well as food quality and safety. Alternariol (AOH), alternariol monomethyl ether (AME), tenuazonic acid (TeA), tentoxin (TEN) and altenuene (ALT) are considered the main Alternaria compounds thought to pose a risk to human health. However, food-borne Alternaria species are able to produce many additional metabolites, whose toxicity has been tested incompletely or not tested at all. Both alternariols are mutagenic and their presence in cereal grain has been associated with high levels of human esophageal cancer in China. TeA exerts cytotoxic and phytotoxic properties, and is acutely toxic in different animal species, causing hemorrhages in several organs. The possible involvement of TA in the etiology of onyalai, a human hematological disorder occurring in Africa, has been suggested. Altertoxins (ALXs) have been found to be more potent mutagens and acutely toxic to mice than AOH and AME. Other metabolites, such as TEN, are reported to be phytotoxins, and their toxicity on animals has not been demonstrated up to now. Vegetable foods infected by Alternaria rot are obviously not suitable for consumption. Thus, whole fresh fruits are not believed to contribute significantly with Alternaria toxins to human exposure. However, processed vegetable products may introduce considerable amounts of these toxins to the human diet if decayed or moldy fruit is not removed before processing. The taxonomy of the genus is not well defined yet, which makes it difficult to establish an accurate relationship between the contaminant species and their associated mycotoxins. Great efforts have been made to organize taxa into subgeneric taxonomic levels, especially for the small-spored, food associated species, which are closely related and constitute the most relevant food pathogens from this genus. Several crops of agricultural value are susceptible to infection by different Alternaria species and can contribute to the entry of Alternaria mycotoxins in the food chain. The distribution of Alternaria species was studied in different commodities grown in Argentina. These food populations were characterized through a polyphasic approach, with special interest in their secondary metabolite profiles, to understand their full chemical potential. Alternaria species associated with tomato, bell pepper, blueberry, apples and wheat cultivated in Argentina showed a surprisingly high metabolomic and mycotoxigenic potential. The natural occurrence of Alternaria toxins in these foods was also investigated. The results here presented will provide background for discussion on regulations for Alternaria toxins in foods.

Removal Characteristics of Natural Organic Matter and Taste and Odor by Advanced Water Treatment Process around the Han River Water Supply System (한강수계 고도정수처리 공정에서의 유기물과 맛·냄새의 제거특성)

  • The water treatment plants in Seoul Metropolitan Area, which are under Korea Water Resources Corporation(KOWACO)'s management, take water from Paldang Reservoir in Han River System for drinking water supply. There are taste and odor (T&O) problems in the finished water because the conventional treatment processes do not effectively remove the T&O compounds. As part of countermeasures for taste and odor control, KOWACO is planning to introduce advanced water treatment process such as ozone and GAC in near future. This study evaluated the removal characteristics of T&O and dissolved organic matter (DOM) to find design and operation parameters of advanced water treatment processes in a pilot-scale treatment plant. The GAC adsorption capacity for DOC in the two GAC system (GAC and $O_3$-GAC) at an EBCT of 14min was mostly exhausted after 9months. The differency of the removal efficiency of DOC between $O_3$-GAC and GAC increased with increasing operation time because the bioactivity in $O_3$-GAC process was enhanced by post-ozone process. Removal by conventional treatment was unable to reach the target TON(threshold odor number) of 3 but GAC systems at an EBCT(empty bed contact time) of 14 min were able to archive the target with few exception. During the high T&O episodes, PAC as a pretreatment together with GAC could be useful option for T&O control. However, substantial TON removal continued for more than two year (> 90,000 bed volumes). At the spiking of less concentration 26 to 61 ng/L in the influent of GAC systems, GAC absorber and $O_3$-GAC processes could meet the treatment target. The better spike control after 12 and 19 months of operation compared to that after 7 months of operation is a strong indication of biological control. The results presented in this study had shown that $O_3$-GAC process was found to be more effective for T&O control than GAC process. And the main removal mechanism in GAC systems were adsorption capacity and biodegradation.

Effects of Garlic Juice on Toxicity of Mercury in Rat (마늘즙 투여가 흰쥐의 수은 독성에 미치는 영향)

  • It was attempted to observe the antidotic effects of garlic on mercury intoxicated rat model in vivo. The experimental model group consists of garlic juice treated group (G), garlic juice-mercury treated group(MG), mercury treated group (M), and control group (basal diet), The garlic juice of 2% to the daily diet by weight was administered orally to G and MG groups, The single dose of 2.5mgHG/Kg per week was given orally to M and MG groups. the study was carried out for 4 weeks. The results of experiment were as follows. For the group of Mg, and the weight increasing rate was improved to about 30% compared to that of group M. Furthermore a general tonic efficay of garlic was observed in G group in term of increased weight gain rate (bout 15%) than control. In the biochemical studies of rat blood garlic showed effects on lowering the abnormally elevated GPT, GOT, uric acid creatinine value, and especially in lowering the BUN value of Hg treated rat that was selectively elevated in the case of impared renal function such as acute gromerulonephritis caused by Hg intoxication. In the analytical studies blood and renal Hg contents. HG group showed lower value (0.3, 0.33ppm) than that of M group (0.46, 0.51 ppm) Significant difference in reducing Hg level due to the antidotic effect of garlic was observed. In conclusion, it was revealed out from this research, the main principles of garlic, nonprotein sulfur amino acid (alliin) and sulfur compounds (allicin and diallyl disulfides) seem to almost certainly have an antidotic effect on mercury intoxication of rat in vivo.

  • Two major flavonoid compounds having inhibition activity of xanthine oxidase from onion skin were separated, isolated and identified by ODS chromatography, Sephadex LH-20 chromatography, UV/visible absorption spectroscopy and FAB Mass. Spectral analyses indicated that $F_1$ was a flavonol having 3,5,7,3'-OH and 4'-glucoside (quercetin 4'-glucoside), and $F_2$ was a flavonol having 3(5),7,3',4'-OH (quercetin). FAB-Mass of fraction $F_1\;and\;F_2$ in positive-ion-mode produced a spectra containing main ions at m/z 465, corresponding to the $(M+H)^+$ ion of quercetin 4'-glucoside, and m/z 303, corresponding to the $(M+H)^+$ ion of quercetin. The inhibition mechanisms of $F_1\;and\;F_2$ were a mixture of the uncompetative and non-competative modes, with respect to xanthine as a substrate.

  • Pectin and tannin analysis were carried out to inverstigate any available components from Pinus densiflora, P. koraiensis, P. thunbergii and P. rigida. To analyze terpenoid components, the essential oils were extracted with steam distillation method from four kinds of pine needles. The essential oil was analyzed by GC and GC-MS spectroscopy. The results were summarized as follows: Pectin content was highest in P. koraiensis with 0.40%, and tannin content was highest in P. koraiensis with 1.05. Major components of P. densiflora needles were ${\alpha}$-pinene, ${\beta}$-caryophyllene, ${\Delta}^3$-carene and phytol. ${\alpha}$-Pinene, ${\Delta}^3$-carene, ${\beta}$-caryophyllene, germacrene D, and camphene were found major components in P. koraiensis. Major components of P. thunbergii needles were ${\beta}$-pinene, ${\alpha}$-pinene, ${\beta}$-caryophyllene and germacrene D. ${\beta}$-Pinene, ${\alpha}$-piene, humulene oxide and ${\alpha}$-elemene were major components in P. rigida. Sabinene and citronellol were infrequent components in P. koraiensis, and ${\alpha}$-pinene oxide was present only in P. rigida. ${\alpha}$-Pinene, limonene, and bornylacetate well known as the main components of green air bath were found in P. densiflora and P. koraiensis.

Carbonation Characteristics of Alkali Activated Blast-Furnace Slag Mortar (알칼리활성 고로슬래그 모르타르의 탄산화 특성)

  • Alkali-activated slag (AAS) is the most obvious alternative materials that can replace OPC. But, AAS industrial usage as a structural material should be evaluated for its durability. Carbonation resistance is one of the most important factors in durability evaluation. Test results for 18 slag-based mortars activated by sodium silicate and 6 OPC mortars were obtained in this study to verify the carbonation property. Main variables considered in the study were flow, compressive strength before and after carbonation, and carbonation depth. Mineralogical and micro-structural analysis of OPC and AAS specimens prior to and after carbonation was conducted using XRD, TGA, FTIR FE-SEM. Test results showed that CHS was major hydration products of AAS and, unlike OPC, no other hydration products were found. After carbonation, CSH of hydration product in AAS turned into an amorphous silica gel, and alumina compounds was not detected. From the analysis of the results, it was estimated that the micro-structures of CSH in AAS easily collapsed during carbonation. Also, the results showed that this collapse of chemical chain of CSH lowered the compressive strength of concrete after carbonation. By increasing the dosage of activators, carbonation resistance and compressive strength were effectively improved.
