SEM, X선 마이크로 분석기, TEM에 의한 코디에라이트의 특성 연구 (Characterization of Cordierite by SEM, Microanalysis X and TEM)

  • The cordierite (MgO, SiO2, Al2O3) is of great interest for microelectronic packaging of integrated circuits. Its main advantages are low dielectric constant and low thermal expansion. The cordierite precursor is obtained by sol-gel synthesis using organic and inorganic compounds. The obtained cordierite precursor is an amorphous state at about 900\ulcorner. Green and fired cordierite samples were studied by SEM. Microanalysis X and TEM for microscopic properties. The fired cordierite shows forte diminution of Mg in comparison with its value at volume and the deficit of Mg compensates by sugmentation of Al and Si \ulcornercordierite and \ulcornercordierite are present near the surface (< 100) and small quantities of magnesium aluminate (MgAl2O4)is presented spinnel phase.

Antiviral Activity Enhancement through the SATE Prodrug of a 2'-Modified 5'-Norcarbocyclic Adenine Analogue

  • We synthesized and tested the anti-HIV activity of the SATE prodrug of a 2'-methyl 5'-norcarbocyclic adenine analogue. The introduction of a methyl group in the 2'-position was performed by the addition of a carbonyl using isopropenyl magnesium bromide. The adenine base was efficiently coupled using the Mitsunobu reaction. The chemical stability study of the bis(SATE) derivative 18 was measured at neutral (pH 7.2) and slightly acid (milli-Q water, pH 5.5) pH, and compounds 16 and 18 were evaluated as potential anti-HIV-1 agents.

  • Hot-rolled and Extruded plates of AZ type magnesium alloys were successfully joined by friction stir welding(FSW). AZ31B-H24 and AZ61 plates with the thickness of 4mm were used, and the microstructural development in the stir zone were investigated using optical and scanning electron microscopes. The grain size of base metal and stir zone were investigated using the line-intersecter method. Hardness of the stir zone was remarkably increased due to very fine recrystallized grain structure both in AZ3l and AZ6l alloys. Tensile strengths of the FS welded Mg alloys AZ31 and AZ61 were strongly affected by formation of the intermetallic compounds, ${\beta}$-Al$\sub$12/Mg$\sub$17/.

FSW를 이용한 AZ31B Mg합금의 접합성 평가 (Evaluation of Joint Properties of Friction Stir Welded AZ31B Mg Alloy)

  • Friction stir weldability of AZ31B Mg alloy was studied using microstructural observation and mechanical tests. Defect free joints was obtained under the condition of 2000rpm-100mm/min. In TMAZ, a lot of twin deformation were observed due to the mechanical effect of the FSW tool and thus relatively high hardness was obtained. In SZ, the twin deformation was disappeared by recovery and the hardness decreased because the. grain structure was coarsened by dynamic recrystallization and grain growth. The Al-Mn precipitates were observed throughout the joint regions. On the other hand, $$\beta$-Mg_{17}Al_{12}$ intermetallic compounds were not observed in either of the zone. The joint efficiency was about 80% and the impact value of the joint was almost equal to that of base metal.

Study on Property Change with a Fire Retardant Content in the Manufacture of Polymer Composites for Cable Sheath

  • Four different polymer compounds were manufactured to make cable sheaths for the shipping industry. Two kinds of ethylene vinyl acetate (EVA) as the main matrix polymers and EVA-grafted maleic anhydride (EVA-g-MAH) as the coupling agent were selected for compounding with fire retardant, crosslinking agent, filler, plasticizer, and other additives. The properties of the four compounded materials were investigated with different contents of the fire retardant, silanecoated magnesium dihydroxide (S-MDH). In the rheology evaluation, the $t_{60}$ and ${\Delta}T$ values increased with increasing S-MDH contents. On the other hand, the tensile strength decreased with increasing S-MDH content due to a relative decrease in binder polymers. With increasing S-MDH content, fire resistance increased, but cold resistance showed no obvious enhancement due to the polar effect of vinyl acetate in EVA.

Sesquiterpenoids Bioconversion Analysis by Wood Rot Fungi

  • Sesquiterpenoids are defined as $C_{15}$ compounds derived from farnesyl pyrophosphate (FPP), and their complex structures are found in the tissue of many diverse plants (Degenhardt et al. 2009). FPP's long chain length and additional double bond enables its conversion to a huge range of mono-, di-, and tri-cyclic structures. A number of cyclic sesquiterpenes with alcohol, aldehyde, and ketone derivatives have key biological and medicinal properties (Fraga 1999). Fungi, such as the wood-rotting Polyporus brumalis, are excellent sources of pharmaceutically interesting natural products such as sesquiterpenoids. In this study, we investigated the biosynthesis of P. brumalis sesquiterpenoids on modified medium. Fungal suspensions of 11 white rot species were inoculated in modified medium containing $C_6H_{12}O_6$, $C_4H_{12}N_2O_6$, $KH_2PO_4$, $MgSO_4$, and $CaCl_2$ for 20 days. Cultivation was stopped by solvent extraction via separation of the mycelium. The metabolites were identified as follows: propionic acid (1), mevalonic acid lactone (2), ${\beta}$-eudesmane (3), and ${\beta}$-eudesmol (4), respectively (Figure 1). The main peaks of ${\beta}$-eudesmane and ${\beta}$-eudesmol, which were indicative of sesquiterpene structures, were consistently detected for 5, 7, 12, and 15 days These results demonstrated the existence of terpene metabolism in the mycelium of P. brumalis. Polyporus spp. are known to generate flavor components such as methyl 2,4-dihydroxy-3,6-dimethyl benzoate; 2-hydroxy-4-methoxy-6-methyl benzoic acid; 3-hydroxy-5-methyl phenol; and 3-methoxy-2,5-dimethyl phenol in submerged cultures (Hoffmann and Esser 1978). Drimanes of sesquiterpenes were reported as metabolites from P. arcularius and shown to exhibit antimicrobial activity against Gram-positive bacteria such as Staphylococcus aureus (Fleck et al. 1996). The main metabolites of P. brumalis, ${\beta}$-Eudesmol and ${\beta}$-eudesmane, were categorized as eudesmane-type sesquiterpene structures. The eudesmane skeleton could be biosynthesized from FPP-derived IPP, and approximately 1,000 structures have been identified in plants as essential oils. The biosynthesis of eudesmol from P. brumalis may thus be an important tool for the production of useful natural compounds as presumed from its identified potent bioactivity in plants. Essential oils comprising eudesmane-type sesquiterpenoids have been previously and extensively researched (Wu et al. 2006). ${\beta}$-Eudesmol is a well-known and important eudesmane alcohol with an anticholinergic effect in the vascular endothelium (Tsuneki et al. 2005). Additionally, recent studies demonstrated that ${\beta}$-eudesmol acts as a channel blocker for nicotinic acetylcholine receptors at the neuromuscular junction, and it can inhibit angiogenesis in vitro and in vivo by blocking the mitogen-activated protein kinase (MAPK) signaling pathway (Seo et al. 2011). Variation of nutrients was conducted to determine an optimum condition for the biosynthesis of sesquiterpenes by P. brumalis. Genes encoding terpene synthases, which are crucial to the terpene synthesis pathway, generally respond to environmental factors such as pH, temperature, and available nutrients (Hoffmeister and Keller 2007, Yu and Keller 2005). Calvo et al. described the effect of major nutrients, carbon and nitrogen, on the synthesis of secondary metabolites (Calvo et al. 2002). P. brumalis did not prefer to synthesize sesquiterpenes under all growth conditions. Results of differences in metabolites observed in P. brumalis grown in PDB and modified medium highlighted the potential effect inorganic sources such as $C_4H_{12}N_2O_6$, $KH_2PO_4$, $MgSO_4$, and $CaCl_2$ on sesquiterpene synthesis. ${\beta}$-eudesmol was apparent during cultivation except for when P. brumalis was grown on $MgSO_4$-free medium. These results demonstrated that $MgSO_4$ can specifically control the biosynthesis of ${\beta}$-eudesmol. Magnesium has been reported as a cofactor that binds to sesquiterpene synthase (Agger et al. 2008). Specifically, the $Mg^{2+}$ ions bind to two conserved metal-binding motifs. These metal ions complex to the substrate pyrophosphate, thereby promoting the ionization of the leaving groups of FPP and resulting in the generation of a highly reactive allylic cation. Effect of magnesium source on the sesquiterpene biosynthesis was also identified via analysis of the concentration of total carbohydrates. Our current study offered further insight that fungal sesquiterpene biosynthesis can be controlled by nutrients. To profile the metabolites of P. brumalis, the cultures were extracted based on the growth curve. Despite metabolites produced during mycelia growth, there was difficulty in detecting significant changes in metabolite production, especially those at low concentrations. These compounds may be of interest in understanding their synthetic mechanisms in P. brumalis. The synthesis of terpene compounds began during the growth phase at day 9. Sesquiterpene synthesis occurred after growth was complete. At day 9, drimenol, farnesol, and mevalonic lactone (or mevalonic acid lactone) were identified. Mevalonic acid lactone is the precursor of the mevalonic pathway, and particularly, it is a precursor for a number of biologically important lipids, including cholesterol hormones (Buckley et al. 2002). Farnesol is the precursor of sesquiterpenoids. Drimenol compounds, bi-cyclic-sesquiterpene alcohols, can be synthesized from trans-trans farnesol via cyclization and rearrangement (Polovinka et al. 1994). They have also been identified in the basidiomycota Lentinus lepideus as secondary metabolites. After 12 days in the growth phase, ${\beta}$-elemene caryophyllene, ${\delta}$-cadiene, and eudesmane were detected with ${\beta}$-eudesmol. The data showed the synthesis of sesquiterpene hydrocarbons with bi-cyclic structures. These compounds can be synthesized from FPP by cyclization. Cyclic terpenoids are synthesized through the formation of a carbon skeleton from linear precursors by terpene cyclase, which is followed by chemical modification by oxidation, reduction, methylation, etc. Sesquiterpene cyclase is a key branch-point enzyme that catalyzes the complex intermolecular cyclization of the linear prenyl diphosphate into cyclic hydrocarbons (Toyomasu et al. 2007). After 20 days in stationary phase, the oxygenated structures eudesmol, elemol, and caryophyllene oxide were detected. Thus, after growth, sesquiterpenes were identified. Per these results, we showed that terpene metabolism in wood-rotting fungi occurs in the stationary phase. We also showed that such metabolism can be controlled by magnesium supplementation in the growth medium. In conclusion, we identified P. brumalis as a wood-rotting fungus that can produce sesquiterpenes. To mechanistically understand eudesmane-type sesquiterpene biosynthesis in P. brumalis, further research into the genes regulating the dynamics of such biosynthesis is warranted.

Effects of thinning intensity on nutrient concentration and enzyme activity in Larix kaempferi forest soils

  • Background: As the decomposition of lignocellulosic compounds is a rate-limiting stage in the nutrient mineralization from organic matters, elucidation of the changes in soil enzyme activity can provide insight into the nutrient dynamics and ecosystem functioning. The current study aimed to assess the effect of thinning intensities on soil conditions. Un-thinned control, 20 % thinning, and 30 % thinning treatments were applied to a Larix kaempferi forest, and total carbon and nitrogen, total carbon to total nitrogen ratio, extractable nutrients (inorganic nitrogen, phosphorus, calcium, magnesium, potassium), and enzyme activities (acid phosphatase, ${\beta}$-glucosidase, ${\beta}$-xylosidase, ${\beta}$-glucosaminidase) were investigated. Results: Total carbon and nitrogen concentrations were significantly increased in the 30 % thinning treatment, whereas both the 20 and 30 % thinning treatments did not change total carbon to total nitrogen ratio. Inorganic nitrogen and extractable calcium and magnesium concentrations were significantly increased in the 20 % thinning treatment; however, no significant changes were found for extractable phosphorus and potassium concentrations either in the 20 or the 30 % thinning treatment. However, the applied thinning intensities had no significant influences on acid phosphatase, ${\beta}$-glucosidase, ${\beta}$-xylosidase, and ${\beta}$-glucosaminidase activities. Conclusions: These results indicated that thinning can elevate soil organic matter quantity and nutrient availability, and different thinning intensities may affect extractable soil nutrients inconsistently. The results also demonstrated that such inconsistent patterns in extractable nutrient concentrations after thinning might not be fully explained by the shifts in the enzyme-mediated nutrient mineralization.

  • 토양과 토양용액의 경계면에서 토양내 흡착부위에 대해 경쟁적 이온이 존재할 때 카드늄의 흡착 현상을 연구하기 위하여, 토양의 Bt층으로부터 토양시료를 채취하여 토양의 물리화학적 특성을 분석하였다. 한편 Ethy-lone glycol monoethyl ether(EGME)와 질소가스를 가지고 각각의 토양의 특정표면적을 조사하였다. 토양층 내에서 토양입자와 반응성을 지닌 물질의 동태를 연구하는 일반적 지침으로, 물리화학적 특성이 완전히 다른 토양을 가지고 일반적인 등온흡착곡선을 취하였다. 단일 등온흡착곡선에서 보여 주었던 것처럼 물질간의 경쟁적흡착을 포함한 여러가지의 물리 화학적 요인들이 흡착에 영향을 미칠 것으로 추정된다. 한편 각각의 토양입자에 의한 카드늄 흡착의 정도는 사용된 토양과 용액의 회석 비율뿐만 아니라 각각의 토양의 표면적에 의존한다는 것을 알 수 있었다. 이러한 결과는 서로 다른 토양에서의 카드늄이 마그네시움과의 경쟁적 흡착에서도 적용되는 것을 알 수 있었다. 이러한 등온흡착곡선은 유해한 무기성분의 흡착과 동태 해석 뿐만 아니라 표준 배취흡착과정을 연구개발하는데 복합 요인으로 작용한다는 것을 증명하기 위한 예로서 사용할 수 있다.

Effect of Different Cations on Pidan Composition and Flavor in Comparison to the Fresh Duck Egg

  • The effects of different cations on its composition and flavor characteristics of pidan white and yolk produced with duck egg in comparison to its fresh egg were investigated. Mineral content such as calcium, magnesium, sodium and potassium were significantly increased in pidan yolk irrespective of its cations in pickle solution in comparison to the fresh yolk (P<0.05). It confirmed the migration of minerals from the pickling solution to the egg. However, calcium and magnesium was found lower in 0.2% $PbO_2$ treated pidan. Less pidan flavor compounds were generated in pidan white produced with the aid of 0.2% $PbO_2$. It confirmed that binding of lead prevent the maillard reaction in the pidan treated with $PbO_2$. Benzaldehyde, ketones, alcohol and acid found in the pidan white treated with 0.2% $ZnCl_2$ reveals that volatiles are generated most likely from maillard reaction. However, pidan yolk of both 0.2% $PbO_2$ and 0.2% $ZnCl_2$ showed higher generation of volatiles more likely from yolk lipids. Butanal, pentanal, hexanal, and heptanal are of those aldehydes found in 0.2% $ZnCl_2$ treated pidan yolk whereas hexanal is the only aldehyde detected in 0.2% $PbO_2$ treated pidan yolk. Thus, cations in the pickling solution affect the flavor characteristics of pidan white and yolk.

블랜칭 및 용질 처리에 따른 참취(Aster scaber Thunb.)의 phytochemical 성분 및 품질특성 (Phytochemical compounds and quality characteristics of Aster scaber Thunb. in response to blanching conditions and treatment with solutes)

  • 블랜칭 처리조건 확립을 위하여 시간(1, 3, 5 min) 및 용질의 종류[0.1% soluble Ca(Sol-Ca), 0.1% soium bicarbonate($NaHCO_3$), 0.1% magnesium sulfate($MgSO_4$)]를 달리한 참취의 품질특성을 비교하였다. 블랜칭 시간이 경과함에 따라 녹색도와 chroma 값은 감소되고 색차는 증가하였으며, polyphenol 함량은 유의적으로 감소하였고 블랜칭 처리시간이 3분 이상 경과될 시 급격히 저하되었다. Carotenoid 함량의 경우 적정 블랜칭 처리 시 함량은 증가하는 반면 ascorbic acid 함량은 유의적으로 감소하는 결과를 나타내어 열적 손실이 동반되는 것으로 관찰되었다. 경도 및 절단력은 처리 시간이 길어짐에 따라 유의적으로 감소하였으며, pectinesterase와 polygalacturonase 저해활성은 3분 이후부터는 불활성화 수치가 증가하여 효소활성을 저해시키는 것으로 나타났다. 총균수 및 대장균군수의 변화를 측정한 결과, 대조구의 경우 5 log CFU/g 수준으로 검출되었으나, 블랜칭 처리한 처리구에서는 2~4 log CFU/g 수준으로 미생물이 감소하였으며, 대장균군수는 검출되지 않았다. Chlorophyll 함량은 증가하는 반면 chlorophyll a 및 b의 비율(Ca/Cb)은 유의적으로 감소하였으며, 관능적 기호특성에서는 외관 및 색상의 경우 블랜칭 처리 시간이 길어짐에 따라 감소하는 경향을 나타낸 반면 향미 및 전반적인 기호도에서는 블랜칭 처리에 의하여 기호특성이 증가하였다가 점차 감소하였으며, 전반적으로 3분간 블랜칭 처리 시 높은 선호도를 나타내었다.
