  • Mitochondrial DNA (mtDNA), a multicopy genome found in mitochondria, is crucial for oxidative phosphorylation. Mutations in mtDNA can lead to severe mitochondrial dysfunction in tissues and organs with high energy demand. MtDNA mutations are closely associated with mitochondrial and age-related disease. To better understand the functional role of mtDNA and work toward developing therapeutics, it is essential to advance technology that is capable of manipulating the mitochondrial genome. This review discusses ongoing efforts in mitochondrial genome editing with mtDNA nucleases and base editors, including the tools, delivery strategies, and applications. Future advances in mitochondrial genome editing to address challenges regarding their efficiency and specificity can achieve the promise of therapeutic genome editing.

  • This study was conducted to suggest the optimal periods for planting and felling Larix kaempferi tree by monitoring its cambial activity period. In addition, the heat summation to induce the cambial activity of Larix kaempferi was investigated. The study sites were at Mt. Worak and Mt. Midong. After selecting 5 trees at Mt. Worak and Mt. Midong, the cambium samples were collected using a mini trephor with 2 mm diameter from April 7 to October 6, 2017 at Mt. Worak, and from April 7 to September 29, 2017 at Mt. Midong. After the collected cambium samples were embedded in PEG2000, transverse thin sections with a thickness of 10 to 15 ㎛ were prepared using a microtome. One Larix kaempferi sample from each site, Mt. Worak and Mt. Midong, in which the accurate monitoring of cambial activity was impossible due to the formation of traumatic resin canal, was excluded from the study. The observation of the initiation date of cambial activity under a light microscopy revealed that 2 specimens from Mt. Worak and 3 from Mt. Midong showed the initiation on April 28. The remaining 2 specimens of Mt. Worak and 1 specimen of Mt. Midong were initiated on May 4, which was a week later than the others. The heat summation that induced the initiation of cambial activity was 196.4-271.8 at Mt. Worak and 204.7-277.3 at Mt. Midong, which was similar. The termination of cambial activity occurred between August 4 and 25 at Mt. Worak, and between August 4 and September 1 at Mt. Midong. Based on the above results, it was found that the optimal planting period for Larix kaempferi in Mt. Worak and Mt. Midong was before April, about a month before the cambium activity, and the felling period was from October when the cambial activity was completely terminated.

  • We isolated a rice (Oryza sativa L.) WRKY gene which is highly upregulated in senescent leaves, denoted OsWRKY42. Analysis of OsWRKY42-GFP expression and its effects on transcriptional activation in maize protoplasts suggested that the OsWRKY42 protein functions as a nuclear transcriptional repressor. OsWRKY42-overexpressing (OsWR KY42OX) transgenic rice plants exhibited an early leaf senescence phenotype with accumulation of the reactive oxygen species (ROS) hydrogen peroxide and a reduced chlorophyll content. Expression analysis of ROS producing and scavenging genes revealed that the metallothionein genes clustered on chromosome 12, especially OsMT1d, were strongly repressed in OsWRKY42OX plants. An OsMT1d promoter:LUC construct was found to be repressed by OsWRKY42 overexpression in rice protoplasts. Finally, chromatin immunoprecipitation analysis demonstrated that OsWRKY42 binds to the W-box of the OsMT1d promoter. Our results thus suggest that OsWRKY42 represses OsMT1d-mediated ROS scavenging and thereby promotes leaf senescence in rice.

Sampling was conducted two times in May.June and October.November, 2000. Main two areas are Mt. Anpyong (470.0m) and Mt. Daedun (877.0m) area. Total taxa of benthic macroinvertebrates were 111 species, 47 families, 15 orders, 6 classes in 4 phyla. Aquatic insects were composed of 29 species in ephemeroptera, 10 species in odonata, 7 species in plecoptera, 2 species in hemiptera, 2 species in megaloptera, 23 species trichuptera, 6 species in coleoptera and 17 species in diptera. Other non-insects were composed of 1 species in platyhelminthes, 8 species in gastropoda, 2 species in oligochaeta, 3 species in hirudinea and 1 species in crustacea. Ephemeroptera, trichoptera and plecoptera as indicators in clean water were very abundantly. Occurrence species at each surrey area was 77 species at Mt. Anpyong area and 89 species at Mt. Daedun area, respectively. Mean values of species diversity indices were 3.04 and 2.69, respectively. According to the saprobic system based on the species diversity indices of benthic macroinvertebrates, Mt. Anpyong region are determined as polysaprobic area or ${\beta}-mesosaprobic$ area and Mt. Daedun region are determined as polysaprobic area, $\beta-mesosaprobic$ area or ${\alpha}-mesosaprobic$ area.

  • Restriction endonucleases were used to search for intraspecific variation at 32 cleavage sites in mitochondrial DNA(mtDNA) purified from fourteen strains of Drosophila melanogaster helonging to different localities of the world. mtDNA of D. melanogaster was displayed site variation(Hpall, Haelll and Seal endonucleases) and length variation(maxirnum 550bp). Six genotypes, Ml, M2, M3, M4, M6 and M7, could be distinguished based on ihe site types witti a low average of intraspecific substitution rate (1.88%),but M5 type of Ogasawara strain in Japan was not detected in this study. A possible explanation for the low divergence was that mtDNA variation of fourteen strains in D. melanogaster could not he accumulated sufficiently owing to recent divergence of few individuals, and that sequence divergence was prevented by frequent migration in spite of the geographical isolation.

  • Although there are several methods for the preparation of mitochondrial DNA (mtDNA) from mammalian tissues, most are relatively long ultracentrifugation or manipulations by a small-scale method. We escribed a rapid method for large-scale extraction of mtDNA from human placental and horse liver tissues. The method is based on the preparation and homogenization of tissues, urification of crude mitochondria by differential centrifugations and isolation of mtDNA by alkaline Iysis. It was improved from Pre-existing methods by replacing some steps with simpler ones and discarding many others. This method gives a high yield of pure mtDNA(approximately 1-5mg from one placenta; ca. 400-600 g wet weight), depending on its sources (fresh tissue gave better results than frozen one). The resulting mtDNA indicated that this method can yield mtDNA in sufficient purity and quantity to identify the direct restriction analysis on agarose gel, random-primed labeling as a probe, and end labeling. Therefore, the method is ideal for obtaining good mtDNA samples to conduct routine restriction fragment length polymorphism (RFLP) analyses of natural populations for genetic studies.

  • In this study, the development and production of electric vehicles and hydrogen vehicles are presented as a method for realizing carbon-neutral. Accordingly, the demand and need for development of underground metal mineral resources such as copper and nickel has increased. The research was carried out using MT survey, which is very useful for deep exploration such as mineral resources and oil exploration because of it's low cost and explorable depth. In Korea, there are very few cases of MT exploration in terms of mineral development, so the study was conducted based on the MT exploration conducted previously in AusLAMP, Australia. Through comparative analysis of the MT exploration data conducted to identify the ore body in the deep area of the Olympic Dam in Australia, with the data directly calculated in 2D inversion, it was confirmed that it can have a positive effect on the possibility of resource development and carbon neutrality using MT exploration in Korea.

  • To tolerate environmentally adverse conditions such as cold, drought, and salinity, plants often synthesize and accumulate proline in cells as compatible osmolytes. ${\Delta}^1$-Pyrroline-5-carboxylate synthetase(P5CS) catalyzes the rate-limiting step of proline biosynthesis from glutamate. Two complete genes, MtP5CS1 and MtP5CS2, were isolated from the model legume Medicago truncatula by cDNA cloning and bacterial artificial chromosome library screening. Nucleotide sequence analysis showed that both genes consisted of 20 exons and 19 introns. Alignment of the predicted amino acid sequences revealed high similarities with P5CS proteins from other plant species. The two MtP5CS genes were expressed in response to high salt and low temperature treatments. Semi-quantitative reverse transcription-polymerase chain reaction showed that MtP5CS1 was expressed earlier than MtP5CS2, indicating differential regulation of the two genes. To evaluate the reverse genetic effects of nucleotide changes on MtP5CS function, a Targeting Induced Local Lesions in Genomes approach was taken. Three mutants each were isolated for MtP5CS1 and MtP5CS2, of which a P5CS2 nonsense mutant carrying a codon change from arginine to stop was expected to bring translation to premature termination. These provide a valuable genetic resource with which to determine the function of the P5CS genes in environmental stress responses of legume crops.

  • In order to investigate the effects of Mangumtang(MT) and Gamimangumtang(GMT) experimental studies were performed in Ex.1(treated with cholesterol and administration of sample at a time) and Ex.2(administration of sample after the treatment of cholesterol). The results were summerized as follows : 1. The content of Total cholesterol in the serum was not significantly decreased in MT group, whereas in GMT group significantly decreased compared with control group in Ex.1 and all significantly decreased in MT group and GMT group in Ex.2. 2. The content of Triglyseride in the serum was all significantly decreased in MT group and GMT group in all case. 3. The content of Phospholipid in the serum was not significantly decreased in MT group, whereas in GMT group was significantly decreased compared with control group in Ex.1 or Ex.2. 4. The content of Free Fatty Acid in the serum was all significantly decreased in MT group and GMT group in all case. 5. The content of HDL-Cholesterol in the serum was tended to decreased compared with control group but did not show a significance in Ex.1 or Ex.2. 6. The content of LDL-Cholesterol in the serum was not significantly decreased in MT group and GMT group in Ex.1, whereas in Ex.2 not significantly decreased in MT group but significantly decreased in GMT group. According to the above results, it is assumed that Mangumtang and Gamimangumtang have a valid effect on Hyperlipidemia, especially Gamimangumtang have a more valid effect on prevention and cure of Hyperlipidemia.
