Quantitative Analysis and Enantiomeric Separation of Ephedra Alkaloids in Ma Huang Related Products by HPLC-DAD and UPLC-MS/MS

  • Ephedra is a genus of the Ephedraceae family and is found in temperate regions, such as Central Asia and Europe. Among the various ephedra species, Ma Huang (Ephedra herb) is derived from the aerial parts of Ephedra sinica S tapf, Ephedra equisetina Bunge, and Ephedra intermedia Schrenk & C.A. Mey. Ma Huang contains various ephedra alkaloids, including (-)-ephedrine, (+)-pseudoephedrine, (-)-norephedrine, (+)-norpseudoephedrine, (-)-methylephedrine, and (+)-methylpseudoephedrine, which are found naturally as single enantiomers, although they can be prepared as racemates. Although the use of Ma Huang in foods is prohibited in Korea, products containing Ma Huang can be imported, and so it is necessary to develop a suitable analytical technique for the detection of Ma Huang in foods. Herein, we report the development of analytical methods for the detection of ephedra alkaloids in products containing Ma Huang. Following sample purification by solid phase extraction, quantitative analysis was performed using ultra-performance liquid chromatography-triple quadrupole mass spectrometry (UPLC-MS/MS). Additionally, the enantiomers were successfully separated using HPLC-DAD. We successfully analyzed various food samples, where the ephedra alkaloids were qualitatively and quantitatively determined, and the enantiomers were separated. It is expected that these methods may contribute toward preventing the distribution of illegal products containing Ma Huang.

Chemical Analysis of Fly Ashes from Municipal Solid Waste Incinerators (생활폐기물 처리시설 배출 비산재의 조성분석)

  • Analysis of fly ashes from the MSW incinerators was carried out using XRF, ICP-MS and ICP-AES. It was found that the major elements of fly ash were Ca, K, Na, Si, Al, S, Cl and O by the XRF analysis. The XRD spectra showed that the fly ashes were mainly consisted with the chlorides, hydroxides, carbonates and also oxides of former elements. For the determination of minor elements such as Zn, Pb, Cu, Cr, and Cd, we used ICP-AES and ICP-MS after microwave digestion and the results were compared with the result of XRF.

Analysis of animal glue by pyrolysis/GC/MS (열분해/GC/MS에 의한 아교의 분석)

  • Animal glue is a traditional material that was used widely as an adhesive in cultural artifacts, such as Buddhist paintings, dancheong (traditional multicolored paintwork on wooden buildings), mother-of-pearl inlay, and so on. Spectroscopic methods, such as infrared spectroscopy, have been used for the analysis of animal glue. However, such methods do not yield sufficient information about the constituents of the glue. Because pyrolysis/GC/MS analysis is able to estimate the components of a material through the examination of its pyrolyzed products, it is useful for the analysis of polymeric material. In this study, pyrolysis/GC/MS analysis was used to determine the chemical composition of animal glue. The appropriate conditions for analyzing animal glue were established by examining pyrolysis temperature, quantity of the sample, and the repeatability of the method. Some products of pyrolysis were identified. We also analyzed commercial gelatin and animal glue using the established method and found almost no differences in chromatograms among the samples. The results will be used as supporting data to confirm the use of animal glue in cultural artifacts and to replace traditional animal glue by commercial gelatin.

  • We reported that amorphous silicon (a-Si) thin film provide sample plate exhibiting a multimodality to measure biomolecules by secondary ion mass spectrometry (SIMS) and laser desorption/ionization mass spectrometry (LDI-MS). Kim et al.1 reported that a-Si thin film were suitable to detect small molecules such as drugs and peptides by SIMS and LDI-MS. Recently, bacterial identification has been required in many fields such as food analysis, veterinary science, ecology, agriculture, and so on.2 Mass spectrometry is emerging for identifying and profiling microbiology samples from its advantageous characters of label-free and shot-time analysis. Five species of bacteria - S. aureus, G. glutamicum, B. kurstaki, B. sphaericus, and B. licheniformis - were sampled for MS analysis without lipid extraction in sample preparation steps. The samples were loaded onto the a-Si thin film with a thickness of 100 nm which did not only considered laser-beam penetration but also surface homogeneity. Mass spectra were recorded in both positive and negative ionization modes for more analytical information. High reproducibility and sensitivity of mass spectra were demonstrated in a mass range up to mass-to-charge ratio(m/z) 1200 by applying the a-Si thin film in mentioned above MS. Principle component analysis (PCA) - a popular statistical analysis widely used in data processing was employed to differentiate between five bacterial species. The PCA results verified that each bacterial species were readily distinguished and differentiated effectively from our MS approach. It shows a new opportunity to rapid bacterial profiling and identification in clinical microbiology. More details will be discussed in the presentation.

GC-MS Analysis of Chemical Constituents from Various Agarwood (GC-MS를 이용한 침향류의 성분 비교 연구)

  • Objectives : The purpose of this study is to evaluate the quality of four different kinds of agarwood and analysis chemical constituents by GC-MS. Methods : Four source plants were two of Vietnamese agarwood, Indonesian agarwood and Myanmar agarwood. These plant materials were named sample No. 1-4. The chemical constituents of each agarwood were analysed by GC-MS. Results : GC-MS analysis showed that ${\beta}$-selinene was found in all the samples. The Vietnamese agarwood, Sample 1, and 4 has contained 6.861% and 7.497% of ${\beta}$-selinene and reached the highest level at the retention times 46.245 and 46.274 (min). Sample 2, the Myanmar agarwood has contained 1.608%, the lowest level of ${\beta}$-selinene, and it also has contained about 20% of ${\alpha}-,{\beta}-,{\gamma}$-eudesmol and 4.076% of valerianol. Sample 3, the Indonesian agarwood has contained 9.59% of ${\beta}$-selinene, 9.493% of ${\beta}$-selinene and shown its highest level (more than 19%) at the retention time 43.998 (min). Conclusion : It suggested that ${\beta}$-selinene could be a standard marker to evaluate agarwood by GC-MS analysis.

Field Application and Evaluation of Health Status Assessment Tool based on Dietary Patterns for Middle-Aged Women (중년 여성의 식생활 중심 건강상태 판정 도구의 현장 적용 및 평가)

  • Objectives: This study was performed to verify the validity and judgment criteria setting of a health status assessment tool based on dietary patterns for middle-aged women. Methods: A total of 474 middle-aged women who visited the Comprehensive Medical Examination Center at Hanmaeum Hospital in Changwon were enrolled (IRB 2013-0005). The validity was verified using clinical indicators for the diagnosis of metabolic syndrome (MS), and it was used to set the criteria for the tool. A logistic regression analysis was performed for validation. The area under-receiver operation (AUC), sensitivity, specificity, and Youden Index were calculated through ROC curve analysis. Statistical analysis was performed by SPSS 21, and p value <0.05 was considered to be statistically significant. Results: The mean score of the group with no MS (73.3 points) was significantly higher compared to the group with MS (65.7 points) (p<0.001). An analysis of the association between the tool scores and risk of MS showed a 0.15-fold reduction in the risk of MS every time the tool's score increased by one point. As the result of the ROC curve analysis, the assessment reference point was set to 71 points, indicating 77.0% sensitivity and 61.0% specificity. Risk of MS was significantly higher in the group with a score of less than 71.0 than a group with more than 71 points (OR=5.28, p<0.001). Conclusions: This study was the first attempt to develop a health status assessment tool based on the dietary patterns for middle-aged women, and this tool has proven its usefulness as an MS assessment tool through the application of middle-aged women in the field of health screening.

Semiquantitative Dynamic Headspace GC-MS Analysis for Organic Compounds Outgassed from FAB Materials of Air Shower (에어샤워부품의 용출 가스 중 유기화합물의 반 정량적 Headspace GC-MS 분석)

  • The polymeric FAB materials of air shower used in clean room of wafer industry have been outgassed with the dynamic headspace (ca.$100^{\circ}C$) for half an hour, and analyzed using GC-MS. The air in the clean room running air shower was sampled using sorbent tube method, and the organic compounds adsorbed in the sorbent tube were extracted using Soxhlet extraction method, and analyzed using GC-MS. The analytical results from FAB materials of air shower (electric over current relay, acryl plate. polycarbonate window, filter, fan housing, steel galvanized cold plate and canvas buffer) indicated that most of chemicals were originated from polymer fragments of FAB materials. Their analytical results have been compared with those from the air of clean room running air shower. These comparative results could lead to identify whether the sources of trace organic contaminants in clean room air are originated from the polymeric FAB material of air shower.

Establishment of a library of fragments for the rapid and reliable determination of anabolic steroids by liquid chromatography-quadrupole time of flight-mass spectrometry

  • Anabolic steroids have similar structures to testosterone, both of which promote the growth of muscle mass and increase strength. However, the side effects of anabolic steroid use may lead to heart attacks or strokes. Additionally, the excessive use of steroids inhibits the production of the sex hormones in the body via a negative feedback loop, which results in testicular atrophy in males and amenorrhea in females. Currently, the method of choice used to test for the presence of anabolic steroids is GC-MS. However, GC-MS methods require chemical derivatization of the steroid sample to ensure compatibility with the analytical method; therefore, analysis of many different samples is difficult and time consuming. Unlike GC-MS, the liquid chromatography-quadrupole-time of flight mass spectrometry (LC-Q-TOF-MS) method is suitable for many samples. Twenty-two different anabolic steroids were analyzed by LC-Q-TOF-MS with various collision energies (CE). Accurate mass spectral data were obtained using a Q-TOF-MS equipped with an electro-spray ionization source and operated in the positive MS/MS mode for several classes of steroids that are often the targets of testing. Based on the collected data, fragmentation pathways were carefully elucidated. The high selectivity and sensitivity of the LC-Q-TOF-MS instrument combined with these fragmentation pathways offers a new approach for the rapid and accurate screening of anabolic steroids. The obtained data from the 22 different anabolic steroids will be shared with the scientific community in order to establish a library to aid in the screening of illegal anabolic steroids.

A Study on the dimensional model of the meridian system (경락시스템의 층차적 모형에 관한 고찰)

  • Objective : It is limited to verify existence and a part of characteristic of MS(Meridian system) in modern MS hypothesis. Because it is that an object of scientific approach is to prove a structure of MS. Then according to DMMS, we will research on a subject of MSs rules. This is a paper on the investigation of DMMS in aspects of propriety and supplement. Results : DMMS is composed of organizational anatomy system, MS, signal system. This means that the contents of classic MS theory divide into three dimensions. It includes classic MS theory and explains modern MS hypothesis with DMMS. But is has two problems that one is the difference between the right side and the left in the same meridian and the other is a lack of dynamic idea(動態觀念). After apply this ti analysis system, it will be reasonable to DMMS. It indicates to use various ways of a science, especially, mathematics, system theory, information theory, control theory to supplement the DMMS. Conclusions : Although the scientific study of MS is stagnant, the approach of this DMMS will provide us with a new result of MS.

Analysis and Detection of Malicious Data Hidden in Slack Space on OOXML-based Corrupted MS-Office Digital Files

  • OOXML-based MS-Office digital files are extensively utilized by businesses and organizations worldwide. However, OOXML-based MS-Office digital files are vulnerable to forgery and corruption attack by including hidden suspicious information, which can lead to activating malware or shell code being hidden in the file. Such malicious code can cause a computer system to malfunction or become infected with ransomware. To prevent such attacks, it is necessary to analyze and detect the corruption of OOXML-based MS-Office files. In this paper, we examine the weaknesses of the existing OOXML-based MS-Office file structure and analyzes how concealment and forgery are performed on MS-Office digital files. As a result, we propose a system to detect hidden data effectively and proactively respond to ransomware attacks exploiting MS-Office security vulnerabilities. Proposed system is designed to provide reliable and efficient detection of hidden data in OOXML-based MS-Office files, which can help organizations protect against potential security threats.
