Genetic Differentiation of Strains of Xanthomonas campestris pv. vesicatoria by Random Amplified Polymorphic DNA (RAPD) (Random Amplified Polymorphic DNA (RAPD)를 이용한 고추 더뎅이병균 균주의 유전적 분류)

  • Genetic diversity of forty-four strains of Xanthomonas campestris pv. vesicatoria from diverse geographic origins was investigated using random amplified polymorphic DNA (RAPD) of genomic DNA. One hundred and thirty-seven amplified fragments were produced by polymerase chain reaction with a set of 14 random primers, and the sizes of amplified DNA fragments ranged approximately from 0.3 to 3.2 kb. Cluster analysis of genetic similarity among the strains generated the dendrogram that clearly separated all strains from each other. The 44 strains of X. campestris pv. vesicatoria were classified into 4 major genomic DNA RAPD groups and 15 subgroups at the genetic similarity of 0.60 and 0.92, respectively. The strains from foreign countries formed discrete subgroups, but the United States strain 87-77 clustered closely with some of Korean strains together. Thirty-nine Korean strains were classified into 11 subgroups, and especially Masan strain Ms93-1 clustered distinctly far from the other Korean strains. RAPD polymorphism suggests strongly the occurrence of genetic differentiation of X. campestris pv. vesicatoria and the existence of genetically distinctive subgroups among the populations in Korea.

  • The shock tube is a useful device for investigating shock phenomena, spray combustion, unsteady gas dynamics, etc. Therefore, it is necessary to analyze exactly the flow phenomena in shock tube. In this study, the mechanics of its reflected shock zone has been investigated by using of the one-dimensional gas dynamic theory in order to estimate the transition from initial reflection of shock wave region. Calulation for four kinds of reflected shock tube temperature (i.e. (a) 1388 K (b) 1276 K (c) 1168 K (d) 1073 K) corresponding to the experimental conditions have been carried out sumarized as follows. (1) The qualitative tendency is almost the same as in that conditions in region of reflected wave region. (2) High temperature period (reflected shock wave temperature) $T_{5}$, exists 0-2.65 ms. (3) Transition period from temperature of reflection shock wave is far longer than the calculated one. This principally attributed to the fact that the contact surface is accelerated, also, due to the release of energy by viscoity effect. This apparatus can advance the ignition process of a spray in a ideal condition that involved neither atomization nor turbulent mixing process, where, using a shock tube, a column of droplets freely from atomizer was ignited behind a reflected shock.

HALL EFFECT AND RESISTIVITY OF AMORPHOUS $Fe_{83-x}Zr_{7}B_{10}Nb_{x}$ ALLOYS

  • The effect of small addition of Nb on the electrical resistivity and Hall coefficient of the amorphous $Fe_{83}Zr_{7}B_{10}$ alloy and annealed ones below the crystallization temperature were investigated, which has been considered to be suitable for high frequency core material. At room temperature, their resistivities $\rho$ and the spontaneous Hall coefficients $R_{s}$ are $~1.6\;{\mu}{\Omega}m$ and $~3{\times}10^{-8}m^{3}/As$, respectively. $R_{s}$ and $\rho$ are decreased with increasing temperature from 100 K to room temperature. Side-jump effect was adopted to analyze the effect of the small variation of concentration and annealing. The quantity of $R_{s}/{\rho}^{2}$ at room temperature, which is directly related to the electronic structure of the mother alloy, remained almost a constant except as quenched one as it can be predicted from the side-jump effect. We suggested the temperature dependence of $R_{s}/{\rho}^{2}$ can be compared to Ms{T}.

Implementation of Image Transmission Based on Vehicle-to-Vehicle Communication

  • Weak over-the-horizon perception and blind spot are the main problems in intelligent connected vehicles (ICVs). In this paper, a V2V image transmission-based road condition warning method is proposed to solve them. The encoded road emergency images which are collected by the ICV are transmitted to the on-board unit (OBU) through Ethernet. The OBU broadcasts the fragmented image information including location and clock of the vehicle to other OBUs. To satisfy the channel quality of the V2X communication in different times, the optimal fragment length is selected by the OBU to process the image information. Then, according to the position and clock information of the remote vehicles, OBU of the receiver selects valid messages to decode the image information which will help the receiver to extend the perceptual field. The experimental results show that our method has an average packet loss rate of 0.5%. The transmission delay is about 51.59 ms in low-speed driving scenarios, which can provide drivers with timely and reliable warnings of the road conditions.

  • This study was conducted to obtain basic informations on the lignan components from sesame seed. Two major lignans, sesamin and sesamolin, were isolated and identified by means of spectral methods, and quantitative analysis was by HPLC from sesame variety Danbaeggae. Separation was achieved by isocratic elution and reversed phase chromatography Develosil ODS column. The content of the major lignan components were about 0.42% and 0.30% for sesamin and sesamolin, respectively.

Petrogenesis and Metamorphism of Charnockite of Eastern Jirisan Area (지리산 동부 지역에 분포하는 차노카이트의 변성작용과 성인에 관한 연구)

  • Precambrian metamorphic rocks of southwest Sobaeksan massif consist of mainly granitic gneiss, porphyroblastic gneiss and quartzofeldspathic gneiss. The orthopyroxene-bearing rocks(charnockites) are found in the west of Hadong-Sancheong anorthosite complex. The charnockites are 3km wide, 12km long and divided into massive and foliated types based on their texture. The compositions of charnockites are comparable to granodiorite to adamellite and subalkaline. Variations in major and trace elemental abundances show typical magmatic differentiation trends. The geochemical data plotted on tectonic discrimination diagrams reveal that these charnockites were formed in the active tectonic environment. The massive and folidated charnockites are mainly composed of plagioclase, orthopyroxene, microcline, quartz and disseminated garnet. Camels generally show characteristic zonal textures with decreasing $X_{alm}$(0.74~0.83), $X_{Py}$ (0.07~0.12) and $X_{Mg}$ (0.12~0.08) and increasing $X_{grs}$(0.03~0.15) from core to rim. Metamorphic temperature and pressure of the charnockites estimated from orthopyroxene-garnet-plagioclase-quartz assemblages show wide range of variation of $600~900^{\circ}C$ and 2.5~7.5 kbar respectively. The results of P-T estimates indicate an anticlockwise P-T evolution path.

NecroX-5 protects mitochondrial oxidative phosphorylation capacity and preserves PGC1α expression levels during hypoxia/reoxygenation injury

  • Although the antioxidant and cardioprotective effects of NecroX-5 on various in vitro and in vivo models have been demonstrated, the action of this compound on the mitochondrial oxidative phosphorylation system remains unclear. Here we verify the role of NecroX-5 in protecting mitochondrial oxidative phosphorylation capacity during hypoxia-reoxygenation (HR). Necrox-5 treatment ($10{\mu}M$) and non-treatment were employed on isolated rat hearts during hypoxia/reoxygenation treatment using an ex vivo Langendorff system. Proteomic analysis was performed using liquid chromatography-mass spectrometry (LC-MS) and non-labeling peptide count protein quantification. Real-time PCR, western blot, citrate synthases and mitochondrial complex activity assays were then performed to assess heart function. Treatment with NecroX-5 during hypoxia significantly preserved electron transport chain proteins involved in oxidative phosphorylation and metabolic functions. NecroX-5 also improved mitochondrial complex I, II, and V function. Additionally, markedly higher peroxisome proliferator-activated receptor-gamma coactivator-$1{\alpha}$ ($PGC1{\alpha}$) expression levels were observed in NecroX-5-treated rat hearts. These novel results provide convincing evidence for the role of NecroX-5 in protecting mitochondrial oxidative phosphorylation capacity and in preserving $PGC1{\alpha}$ during cardiac HR injuries.

The Effect of Additional Elements on the Tailored Magnetic Properties of Electrochemically Prepared CoPtP-X Alloys (전기화학적으로 제조한 CoPtP-X합금의 첨가제 효과에 따른 맞춤형 자기적 성질)

  • Coptp films with the additive elements (X=Fe, Mn) of varying concentrations were prepared by in-situ electrodeposition, to tailor their magnetic properties. Alloys of CoPtP-X (X=Fe, Mn) were synthesized by changing the solution concentrations of Fe and Mn for electrodeposition. In the electrodeposited CoFePtP alloys, preferred orientation of the electrodeposited films changed from hexagonal (001) to (100) direction with increasing iron contents as revealed by X-ray diffraction, and these films exhibited various magnetic properties ranging from a typical hard magnetic to a soft magnetic property in accordance with microstructural variations. In the case of Mn addition, excellent hard magnetic property was observed at a specific Mn concentration of 0.0126 M in the electrolyte, with the coercivity of 4630 Oe and squareness of 0.856 and this was attributed to the fact that magnetization easy-axis (hexagonal c-axis) coincides with the preferred growth orientation of the film confirmed by transmission electron microscopy.

Missed Skeletal Trauma Detected by Whole Body Bone Scan in Patients with Traumatic Brain Injury

  • Objective : Unclear mental state is one of the major factors contributing to diagnostic failure of occult skeletal trauma in patients with traumatic brain injury (TBI). The aim of this study was to evaluate the overlooked co-occurring skeletal trauma through whole body bone scan (WBBS) in TBI. Methods : A retrospective study of 547 TBI patients admitted between 2015 and 2017 was performed to investigate their cooccurring skeletal injuries detected by WBBS. The patients were divided into three groups based on the timing of suspecting skeletal trauma confirmed : 1) before WBBS (pre-WBBS); 2) after the routine WBBS (post-WBBS) with good mental state and no initial musculoskeletal complaints; and 3) after the routine WBBS with poor mental state (poor MS). The skeletal trauma detected by WBBS was classified into six skeletal categories : spine, upper and lower extremities, pelvis, chest wall, and clavicles. The skeletal injuries identified by WBBS were confirmed to be simple contusion or fractures by other imaging modalities such as X-ray or computed tomography (CT) scans. Of the six categorizations of skeletal trauma detected as hot uptake lesions in WBBS, the lesions of spine, upper and lower extremities were further statistically analyzed to calculate the incidence rates of actual fractures (AF) and actual surgery (AS) cases over the total number of hot uptake lesions in WBBS. Results : Of 547 patients with TBI, 112 patients (20.4 %) were presented with TBI alone. Four hundred and thirty-five patients with TBI had co-occurring skeletal injuries confirmed by WBBS. The incidences were as follows : chest wall (27.4%), spine (22.9%), lower extremities (20.2%), upper extremities (13.5%), pelvis (9.4%), and clavicles (6.3%). It is notable that relatively larger number of positive hot uptakes were observed in the groups of post-WBBS and poor MS. The percentage of post-WBBS group over the total hot uptake lesions in upper and lower extremities, and spines were 51.0%, 43.8%, and 41.7%, respectively, while their percentages of AS were 2.73%, 1.1%, and 0%, respectively. The percentages of poor MS group in the upper and lower extremities, and spines were 10.4%, 17.4%, and 7.8%, respectively, while their percentages of AS were 26.7%, 14.2%, and 11.1%, respectively. There was a statistical difference in the percentage of AS between the groups of post-WBBS and poor MS (p=0.000). Conclusion : WBBS is a potential diagnostic tool in understanding the skeletal conditions of patients with head injuries which may be undetected during the initial assessment.

Reinterpretation of the protein identification process for proteomics data

  • Introduction: In the mass spectrometry-based proteomics, biological samples are analyzed to identify proteins by mass spectrometer and database search. Database search is the process to select the best matches to the experimental mass spectra among the amino acid sequence database and we identify the protein as the matched sequence. The match score is defined to find the matches from the database and declare the highest scored hit as the most probable protein. According to the score definition, search result varies. In this study, the difference among search results of different search engines or different databases was investigated, in order to suggest a better way to identify more proteins with higher reliability. Materials and Methods: The protein extract of human mesenchymal stem cell was separated by several bands by one-dimensional electrophorysis. One-dimensional gel was excised one by one, digested by trypsin and analyzed by a mass spectrometer, FT LTQ. The tandem mass (MS/MS) spectra of peptide ions were applied to the database search of X!Tandem, Mascot and Sequest search engines with IPI human database and SwissProt database. The search result was filtered by several threshold probability values of the Trans-Proteomic Pipeline (TPP) of the Institute for Systems Biology. The analysis of the output which was generated from TPP was performed. Results and Discussion: For each MS/MS spectrum, the peptide sequences which were identified from different conditions such as search engines, threshold probability, and sequence database were compared. The main difference of peptide identification at high threshold probability was caused by not the difference of sequence database but the difference of the score. As the threshold probability decreases, the missed peptides appeared. Conversely, in the extremely high threshold level, we missed many true assignments. Conclusion and Prospects: The different identification result of the search engines was mainly caused by the different scoring algorithms. Usually in proteomics high-scored peptides are selected and low-scored peptides are discarded. Many of them are true negatives. By integrating the search results from different parameter and different search engines, the protein identification process can be improved.
