Synthetic peptide를 이용한 mu-opioid receptor에 대한 항혈청의 생산과 검정 (Production and identification of antisera against mu-opioid receptor using synthetic peptide epitope)

  • In the present study we have analyzed the characteristics and distribution of the mu-opioid receptor(MOR) by raising anti-peptide antisera to the C-terminal peptide of MOR. The antisera against MOR was produced in New Zealand White rabbit against 15 residue corresponding to amino acids, 384-398 of the cloned rat MOR. The antigenic peptide was synthesized using an Applied Biosystems 432 solid-phase peptide synthesizer. The specificity and identification of the antisera were tested by analysis of transfected cells, epitope mapping and immunohistochemical method. COS-7 cells electroporated with MOR cDNA were used to evaluate the characteristics and subcellular distribution of MOR. MOR immunoreactivity was prodominent in the plasmalemma and subcellular compartments such as endoplasmic reticulum, Golgi apparatus and vesicle like structure. Furthermore, both tissue sections and transfected cell lines could be immunostained with these antisera and the immunoreactivity was abolished when anti-MOR sera were preincubated with the peptide against which they were raised. Based on epitope mapping analysis, all antisera appeared to have a similar epitope, which was determined to be within the last amino acid, 391-398. Moreover, immunohistochemistry showed that MOR immunoreactivity was observed in many brain areas including cerebral cortex, striatum, hippocampus, locus coeruleus and the superficial laminae of the dorsal horn. These stained spinal cord and brain areas showed the mirrored pattern observed in auto radiographic studies of mu-opioid binding as well as a pattern similar to that seen by is situ hybridization for MOR. Thus, several lines of evidence support the conclusion that the antisera produced in the present study most likely recognize mu-opioid receptor. These results suggest that MOR antisera may be utilized as useful tool to analyze the physiological and pharmacological studies for mu-opioid receptor in the future.

고분해능 X-선 분말 회절을 이용한 Cs-, Cd-, Pb-, Sr-으로 치환된 합성 모데나이트의 격자상수 비교 연구 (Comparative Crystal Chemistry of Exchanged by Cs-, Cd-, Pb-, and Sr-synthetic Mordenite Using High Resolution X-ray Powder Diffraction)

  • 본 연구는 합성 모데나이트(Na6.6Al6.6Si41.4O96·20.4H2O, Na-MOR)를 이용한 다양한 중금속의 포획 연구를 하기 위한 기초 단계로, 고분해능 X-선 분말 회절을 이용하여 치환체의 격자상수 및 부피 변화를 이해하기 위한 목적으로 실험을 진행하였다. 열중량 분석법(Thermogravimetric analysis, TGA)으로 측정한 결과, 1가 양이온 치환체(Cs-MOR, Na-MOR)는 단위포 당 19.4, 20.4개의 물분자가 존재하였으며, 2가 양이온 치환체(Pb-MOR, Sr-MOR, Cd-MOR)는 단위포 당 21, 23.1, 23.2개가 존재하는 것을 확인하였다. 측정한 모든 물질은 사방정계에 속하는 Cmcm의 공간군을 가지는 것으로 확인할 수 있었다. 치환 전 물질인 Na-MOR과 비교했을 때, 치환체의 (110)면과 (200)면의 회절강도가 명확하게 변화하였으나, 전체적인 피크의 위치는 거의 유사하게 나타나는 것을 확인할 수 있었다. 또한 Na-MOR에서는 확인이 되지 않았던 (220)면의 피크가 Pb-, Cd-, Sr-MOR에서 뚜렷하게 관찰되었다. 이를 통해서 양이온 치환에 따른 원자들의 분포 변화가 주로 ab-평면상에서 나타나지만, 격자상수의 변화는 미세할 것으로 짐작할 수 있었다. Whole profile fitting 방법을 사용하여 치환된 모데나이트의 미세한 격자상수의 변화를 관찰하였다. 치환체의 격자상수 및 격자부피의 변화는 치환된 양이온의 반경 및 전하수에 따라 서로 다른 경향성을 보였다. 1가 양이온의 경우, 이온반경이 증가할수록 a-축의 길이는 증가하지만, 반면에 b- 및 c-축의 길이는 감소하였다. 2가 양이온의 경우, 이온반경이 증가할수록 대체적으로 a-축의 길이가 감소하고, b- 및 c-축의 길이는 증가하였다. 격자부피는 1가 또는 2가 양이온 치환체들이 각각 독립된 경향성을 가지며, 이온반경에 따라 증가하는 것을 확인할 수 있었다.

체외성숙 과정 중 생쥐와 돼지 난자의 Malate Dehydrogenase(Mor2)의 기능에 대한 비교 분석 (Comparative Functional Analysis of the Malate Dehydrogenase(Mor2) during in vitro Maturation of the Mouse and Porcine Oocytes)

  • 생쥐의 난자는 특별한 첨가물이나 난구세포 없이 체외 배양해도 성숙율이 높은 반면 돼지 난자의 체외성숙율은 매우 낮다. 본 연구는 이러한 차이의 원인을 연구하기 위하여 생쥐와 돼지 난자의 에너지 생성에 관여하는 유전자인 malate dehydrogenase(Mor2)의 기능을 RNAi를 이용하여 비교 분석하였다. 생쥐와 돼지 각각의 Mor2 double-stranded RNA(dsRNA)를 제작하고, 생쥐는 난구세포를 제거한 미성숙(GV) 난자의 세포질에 mMor2 dsRNA를 미세 주입한 후 16시간 동안 배양하였고, 돼지는 난구세포를 제거하거나(DO) 혹은 그대로인 상태(COCs)로 pMor2 dsRNA를 미세 주입한 후 48시간 동안 배양하였다. 사용한 배양액은 M199에 10%의 porcine follicular fluid, pyruvate, p-FSH, EGF, cystein, estradiol-$17{\beta}$ 등이 첨가된 배양액을 사용하였고, 배양 후 난자의 형태학적 변화를 관찰하고, Mor2 mRNA양의 변화를 RT-PCR로 확인하여 RNAi 결과, 염기서열 특이적으로 Mor2 발현이 억제됨을 확인하였다. 돼지 난자의 pMor2 RNAi 결과, DO 난자에서는 약 58%의 난자가 MI에서 성숙이 억제되었으나, COCs에선 84.4%가 MII로 성숙하는 것을 볼 수 있었다. 이는 돼지의 난구세포가 난자 성숙에 매우 중요하며, 특히 malate 공급에 관여할 것임을 시사한다. 선행 연구 Mor2 RNAi 결과, 생쥐는 GV에서, 본 연구에서 돼지는 MI에서 성숙이 정지되었기 때문에 이러한 차이가 배양액의 차이인지 다시 mMor2 RNAi 후 생쥐 난자를 M199 배양액에 16시간 동안 배양한 후 성숙율을 관찰하였더니 MII로의 성숙율은 62%로 대조군과 비교하여 큰 차이가 없었다(non-injected: 76.8%, buffer-injected: 73.3%). 이는 mMor2 RNAi 결과가 배양액의 조성에 의해 극복될 수 있음을 보여준 결과다. 생쥐와 돼지에서 서로 다른 대사과정을 갖고 있음은 바로 이 두 종에서의 체외성숙율 차이의 원인이 될 수 있을 것이며, 앞으로 두 종에서의 난자와 난구세포간의 상호작용 및 대사 경로 등에 관한 심도 깊은 비교 분석 연구를 통하여 돼지의 체외성숙율을 증가시킬 수 있는 배양 시스템의 개발이 가능할 것으로 기대된다.

Nondestructive bending Strength Evaluation of Woodceramics Using Resonance Frequency Mode (I) - Carbonizing Temperature -

  • Nondestructive evaluation (NDE) technique method using a resonance frequency mode was carried out for woodceramics produced at different carbonizing temperatures (600℃, 800℃, 1000℃, 1200℃, 1500℃) at the phenol resin impregnation rate of 70%, for three kinds of species (Pinus densifora, Pinus koraiensis, Larix leptolepis), respectively. There was a poor relationship between density and static bending MOR. However, close correlations were found between dynamic MOEd and static bending MOR, and between static MOEs and MOR. Especially, the correlation coefficient was highest between MOEd and static bending MOR. Therefore, the MOEd using the resonance frequency mode is useful as a NDE method for predicting the MOR of woodceramics produced at different carbonizing temperatures.

Comparison of Somatostatin and Morphine Action on the Responses of Wide Dynamic Range Cells in the Dorsal Horn to Peripheral Noxious Mechanical and Heat Stimulation in Cats

  • The purpose of present study was to compare the effects of somatostatin (SOM) and morphine (Mor) on the responses of wide dynamic range (WDR) cells to peripheral noxious stimulation. Single neuronal activity was recorded with a carbon-filament electrode at the lumbosacral enlargement of cat spinal cord. After identifying WDR cells, their responses to peripheral noxious mechanical or thermal stimuli were characterized and the effects of SOM and Mor, applied either iontophoretically or intrathecally, were studied. In most cells SOM and Mor suppressed noxious stimulus-evoked WDR neuronal activity, though a few WDR neurons showed no change or were excited by SOM and Mor. Systemically applied naloxone, a non-specific opioid antagonist, always reversed the Mor induced suppression of neuronal activity evoked by noxious mechanical stimuli, but did not always reverse the suppression of neuronal activity elicited by SOM. The suppressive effect of Mor on thermal stimulus-evoked neuronal activity was partially reversed by naloxone, while that of SOM were not reversed at all. The above results suggest that both Mor and SOM exert an inhibitory effect on thermal and mechanical stimulus-evoked WDR neuronal activity in cat spinal dorsal horn, but the mechanisms are dependent upon the functional populations of dorsal horn nociceptive neurons.

Nondestructive Evaluation of Strength Performance for Heat-Treated Wood Using Impact Hammer & Transducer

  • Nondestructive evaluation (NDE) technique method using a resonance frequency mode was carried out for heat-treated wood under different conditions. The effect of heat treatment on the bending strength and NDE technique using the resonance frequency by impact hammer and force transducer mode for Korean paulownia, Pinus densiflora, Lidiodendron tulipifera and Betula costata were measured. The heat treatment temperature has been investigated at $175^{\circ}C$ and $200^{\circ}C$, respectively. There were a close relationship of dynamic modulus of elasticity and static bending modulus of elasticity to MOR. In all conditions, It was found that there were a high correlation at 1% level between dynamic modulus of elasticity and MOR, and static modulus of elasticity and MOR. However, the result indicated that correlation coefficient is higher in dynamic modulus of elasticity to MOR than that in static modulus of elasticity to MOR. Therefore, the dynamic modulus of elasticity using resonance frequency by impact hammer mode is more useful as a nondestructive evaluation method for predicting the MOR of heat-treated wood under different temperature and species conditions.

Relationship between Anatomical Properties and Modulus of Rupture (MOR) of Larix kaempferi Carr

  • Larix kaempferi is a tree with a major economic impact and is processed in large quantity every year in Korea. This study was carried out to collect basic data for the reasonable use of Larix kaempferi and to investigate the relation between anatomical properties and modulus of rupture (MOR) for heartwood and sapwood. As the length of earlywood tracheid and the radial wall thickness of earlywood tracheid and latewood tracheid increased, the modulus of rupture (MOR) increased, but decreased with increasing microfibril angle. Statistical analysis by the stepwise regression technique shows that the main factors affecting the modulus of rupture (MOR) of heartwood are the microfibril angle and the radial wall thickness of latewood tracheid, while those affecting MOR of sapwood are the length of earlywood tracheid and the microfibril angle.

The Effect of Exposure in Elevated Temperatures on Bending Properties of Wood

  • Temperature has important effect on mechanical properties of wood. These effect needs to be understood and taken into account in the structural use of wood. Furthermore, the effect of cooling after exposing to high temperature must be explained. In this study, the effect of temperature, exposure time, specific gravity, and cooling on bending properties were investigated. The boundary temperatures at which bending MOE and MOR reduced rapidly were approximately $200^{\circ}C$ and $150^{\circ}C$, respectively. This boundary temperature was nearly constant with independence of species(specific gravity), exposure time, and cooling. Above the boundary temperature, the effect of exposure time was increased with temperature and the reduction of bending MOE and MOR for Japanese Larch with relatively higher specific gravity was smaller than that of Hem-fir. The recovery of bending MOE and MOR after cooling was also more significant above the boundary temperature than below. The degree of cooling effect was larger for MOR than MOE. Consequently, bending properties of wood in elevated temperatures should be considered in terms of the boundary temperature, $200^{\circ}C$ for bending MOE, $150^{\circ}C$ for MOR, and these boundary temperatures must be considered an important factor. Furthermore, to evaluate the safety of structure, the recovery after cooling should be considered.

  • Intracerebroventricular (ICV) infusion of morphine (MOR) produces strong analgesia in man and animals. The analgesic effect is thought to be mediated by the centrifugal inhibitory control. But neural mechanisms of the analgesic effect of ICV morphine are not well understood. In the present study, we found that ICV MOR had dual actions on the activity of dorsal horn heurons: it produced both inhibition and excitation of dorsal horn neurons. Since MOR exerts its action via three different types of opioid receptors, we further sought to investigate if there are differential effects of opioid receptor agonists on dorsal horn neurons when administered intracerebroventricularly. Effects of ICV MOR were tested in 28 dorsal horn neurons of the spinal cord in the cat. ICV MOR inhibited, excited and did not affect the heat responses of dorsal horn neurons. ICV DAMGO and DADLE, $\mu$- and $\delta$-opioid agonist, respectively, exhibited the excitation of dorsal horn neurons. In contract, U-50488, a k-opioid agonist, exhibited both the inhibition and excitation of dorsal horn neurons. These results suggest that opioid receptors have different actions on activity of dorsal horn neuron and that the inhibitory action of k-opioid agonist may subserve the analgesia often produced by ICV MOR.

스트랜드/파티클 복합체의 기계적 성질에 관한 연구(I) - 단면구성이 기초물성에 미치는 영향 - (A Study on Mechanical Properties of Strand/Particle Composites(I) - Effect of Layer Constructions -)

  • 본 연구는 미이용 자원인 소경목(小經木) 조생수(早生樹) 등의 저질원료로부터 고성능을 지니는 구조용 보드를 제조하는 기술을 개발하는 것을 목적으로 하여 할렬 스트랜드 (S)/파티클(P) 복합체의 층구조와 S와 P의 혼합비율을 바꾼 단면구성에 따른 기초적 물성을 검토했다. 그 결과 스트랜드층을 포함하고 있는 SP 복합체의 경우, 전체적으로 휨성능(MOR, MOE)이 매우 높았다. 또 S단층 보드는 현저한 이방성을 나타내었지만 층구조가 PB에 가까워질수록 이방성은 감소하여, 7층 구조의 복합체는 이방성이 적었으며 특히 SP7은 직교방향도 휨강도가 높았다. 습윤시 휨강도 성능도 같은 경향을 나타내었다. 박리강도(IB)는 PB가 가장 높았고, 스트랜드층을 가진 복합체는 거의 같은 값을 나타내었다. 두께팽창율(TS) 은 PB가 가장 적은 값을 나타내었으며, 표층이 P층인 구조가 S표층구조보다 적은 경향을 나타내었다. 복합체의 표면특성은 표층 엘리먼트의 영향을 받아, 적고 미세한 엘리먼트인 P표층의 복합체가 크고 두꺼운 S표층 복합체보다 양호하였다. S와 P의 혼합비율의 영향은, SP비(比)를 증가시켜도 강도물성(MOR, MOE, IB)은 향상되지 않으며, 오히려 저하되는 것도 있었다. 두께팽창율(TS)은 SP 비(比) 1:1 이상의 복합체에서는 S만으로 제조한 보드와 같은 정도의 값을 나타내었다.
