In the present study we have analyzed the characteristics and distribution of the mu-opioid receptor(MOR) by raising anti-peptide antisera to the C-terminal peptide of MOR. The antisera against MOR was produced in New Zealand White rabbit against 15 residue corresponding to amino acids, 384-398 of the cloned rat MOR. The antigenic peptide was synthesized using an Applied Biosystems 432 solid-phase peptide synthesizer. The specificity and identification of the antisera were tested by analysis of transfected cells, epitope mapping and immunohistochemical method. COS-7 cells electroporated with MOR cDNA were used to evaluate the characteristics and subcellular distribution of MOR. MOR immunoreactivity was prodominent in the plasmalemma and subcellular compartments such as endoplasmic reticulum, Golgi apparatus and vesicle like structure. Furthermore, both tissue sections and transfected cell lines could be immunostained with these antisera and the immunoreactivity was abolished when anti-MOR sera were preincubated with the peptide against which they were raised. Based on epitope mapping analysis, all antisera appeared to have a similar epitope, which was determined to be within the last amino acid, 391-398. Moreover, immunohistochemistry showed that MOR immunoreactivity was observed in many brain areas including cerebral cortex, striatum, hippocampus, locus coeruleus and the superficial laminae of the dorsal horn. These stained spinal cord and brain areas showed the mirrored pattern observed in auto radiographic studies of mu-opioid binding as well as a pattern similar to that seen by is situ hybridization for MOR. Thus, several lines of evidence support the conclusion that the antisera produced in the present study most likely recognize mu-opioid receptor. These results suggest that MOR antisera may be utilized as useful tool to analyze the physiological and pharmacological studies for mu-opioid receptor in the future.
This study aimed to fundamentally understand changes of cell parameters of cation-exchanged mordenites using high resolution X-ray powder diffraction for studies that immobilization of various heavy metal cation using synthesis mordenite (Na6.6Al6.6Si41.4O96·20.4H2O, Na-MOR). As a results of measurement by Thermogravimetric analysis (TGA), it was confirmed that 19.4, 20.4 water molecules per unit cell were present in monovalent-cation substituted MOR (Cs-MOR, Na-MOR), and 21, 23.1, 23.2 water molecules per unit cell were present in divalent-cation substituted MOR (Pb-MOR, Sr-MOR, Cd-MOR). The space group of all the samples were identified as Cmcm belonging to the orthorhombic crystal system. Compared to Na-MOR, starting material, relative peak intensity of (110) and (200) is significantly changed after cation substitution whereas peak position is almost similar. Also, (220) peak that was not found in Na-MOR was clearly observed in Pb-, Cd- and Sr-exchanged MOR. Thus, it was estimated that changes of atomic distribution usually occurred on ab-plane while changes of cell parameters were little. Detailed changes in the cell parameters of cation-exchanged mordenites were derived from whole profile fitting method using the GSAS suite program. Changes in the axial lengths and unit cell volume of cation substitution showed different relationship depending on ionic radius and charge number. In case of monovalent-cation substituted MOR, the length of a-axis increases whereas the length of b- and c-axis decrease by absorbed cation radius. In the case of divalent-cation exchanged MOR, the length of a-axis usually decreases while the length of b- and c-axis increases by cation radius. It was confirmed that unit cell volume of monovalent and divalent cation substituted MORs had an independent tendency by cation radius.

Contrast to mouse where its in vitro maturation rates are high without specific supplements or presence of the cumulus cells, there are some species, such as porcine, where its in vitro oocyte maturation rates are still very low. This comparative study was conducted to investigate the role of malate dehydrogenase(Mor2) during oocyte maturation by RNAi in the mouse and porcine. The Mor2 double-stranded RNA(dsRNA) was prepared speciesspecifically and microinjected into the cytoplasm of denuded germinal vesicle(GV) oocytes. Oocytes were cultured for 48 h(porcine) and 16 h(mouse) in M199 with 10% porcine follicular fluid, pyruvate, p-FSH, EGF, cystein, and estradiol-$17{\beta}$. We measured changes in oocyte morphology, maturation rates and mRNA levels after Mor2 RNAi. We confirmed gene sequence-specific knock down of Mor2 mRNA in both species after Mor2 RNAi. In contrast to our previous finding that mMor2 RNAi resulted in GV arrest in the mouse, we found that pMor2 RNAi resulted in MI arrest in denuded porcine oocytes(58%), but developed to MII(84.4%) in COCs. To determine whether this difference between mouse and porcine RNAi is due to differences in culture media, we cultured mouse oocytes in the M199 media for 16 h after mMor2 RNAi. Mouse oocytes were developed to MII stage(62%) and there was no statistical difference compared to that of non-injected(76.8%) and buffer-injected(73.3%) control groups. Therefore, we concluded that the mouse and porcine oocytes are having different metabolic systems in relation to malate dehydrogenase for oocyte maturation. This could be a basis for differences in maturation rates in vitro in two species. Further scrutinized studies on the metabolic pathways would led us in finding better culture system to improve oocyte maturation rates in vitro, especially in more challenging species like the porcine.
Nondestructive evaluation (NDE) technique method using a resonance frequency mode was carried out for woodceramics produced at different carbonizing temperatures (600℃, 800℃, 1000℃, 1200℃, 1500℃) at the phenol resin impregnation rate of 70%, for three kinds of species (Pinus densifora, Pinus koraiensis, Larix leptolepis), respectively. There was a poor relationship between density and static bending MOR. However, close correlations were found between dynamic MOEd and static bending MOR, and between static MOEs and MOR. Especially, the correlation coefficient was highest between MOEd and static bending MOR. Therefore, the MOEd using the resonance frequency mode is useful as a NDE method for predicting the MOR of woodceramics produced at different carbonizing temperatures.
The purpose of present study was to compare the effects of somatostatin (SOM) and morphine (Mor) on the responses of wide dynamic range (WDR) cells to peripheral noxious stimulation. Single neuronal activity was recorded with a carbon-filament electrode at the lumbosacral enlargement of cat spinal cord. After identifying WDR cells, their responses to peripheral noxious mechanical or thermal stimuli were characterized and the effects of SOM and Mor, applied either iontophoretically or intrathecally, were studied. In most cells SOM and Mor suppressed noxious stimulus-evoked WDR neuronal activity, though a few WDR neurons showed no change or were excited by SOM and Mor. Systemically applied naloxone, a non-specific opioid antagonist, always reversed the Mor induced suppression of neuronal activity evoked by noxious mechanical stimuli, but did not always reverse the suppression of neuronal activity elicited by SOM. The suppressive effect of Mor on thermal stimulus-evoked neuronal activity was partially reversed by naloxone, while that of SOM were not reversed at all. The above results suggest that both Mor and SOM exert an inhibitory effect on thermal and mechanical stimulus-evoked WDR neuronal activity in cat spinal dorsal horn, but the mechanisms are dependent upon the functional populations of dorsal horn nociceptive neurons.
Nondestructive evaluation (NDE) technique method using a resonance frequency mode was carried out for heat-treated wood under different conditions. The effect of heat treatment on the bending strength and NDE technique using the resonance frequency by impact hammer and force transducer mode for Korean paulownia, Pinus densiflora, Lidiodendron tulipifera and Betula costata were measured. The heat treatment temperature has been investigated at $175^{\circ}C$ and $200^{\circ}C$, respectively. There were a close relationship of dynamic modulus of elasticity and static bending modulus of elasticity to MOR. In all conditions, It was found that there were a high correlation at 1% level between dynamic modulus of elasticity and MOR, and static modulus of elasticity and MOR. However, the result indicated that correlation coefficient is higher in dynamic modulus of elasticity to MOR than that in static modulus of elasticity to MOR. Therefore, the dynamic modulus of elasticity using resonance frequency by impact hammer mode is more useful as a nondestructive evaluation method for predicting the MOR of heat-treated wood under different temperature and species conditions.
Larix kaempferi is a tree with a major economic impact and is processed in large quantity every year in Korea. This study was carried out to collect basic data for the reasonable use of Larix kaempferi and to investigate the relation between anatomical properties and modulus of rupture (MOR) for heartwood and sapwood. As the length of earlywood tracheid and the radial wall thickness of earlywood tracheid and latewood tracheid increased, the modulus of rupture (MOR) increased, but decreased with increasing microfibril angle. Statistical analysis by the stepwise regression technique shows that the main factors affecting the modulus of rupture (MOR) of heartwood are the microfibril angle and the radial wall thickness of latewood tracheid, while those affecting MOR of sapwood are the length of earlywood tracheid and the microfibril angle.
Temperature has important effect on mechanical properties of wood. These effect needs to be understood and taken into account in the structural use of wood. Furthermore, the effect of cooling after exposing to high temperature must be explained. In this study, the effect of temperature, exposure time, specific gravity, and cooling on bending properties were investigated. The boundary temperatures at which bending MOE and MOR reduced rapidly were approximately $200^{\circ}C$ and $150^{\circ}C$, respectively. This boundary temperature was nearly constant with independence of species(specific gravity), exposure time, and cooling. Above the boundary temperature, the effect of exposure time was increased with temperature and the reduction of bending MOE and MOR for Japanese Larch with relatively higher specific gravity was smaller than that of Hem-fir. The recovery of bending MOE and MOR after cooling was also more significant above the boundary temperature than below. The degree of cooling effect was larger for MOR than MOE. Consequently, bending properties of wood in elevated temperatures should be considered in terms of the boundary temperature, $200^{\circ}C$ for bending MOE, $150^{\circ}C$ for MOR, and these boundary temperatures must be considered an important factor. Furthermore, to evaluate the safety of structure, the recovery after cooling should be considered.
Intracerebroventricular (ICV) infusion of morphine (MOR) produces strong analgesia in man and animals. The analgesic effect is thought to be mediated by the centrifugal inhibitory control. But neural mechanisms of the analgesic effect of ICV morphine are not well understood. In the present study, we found that ICV MOR had dual actions on the activity of dorsal horn heurons: it produced both inhibition and excitation of dorsal horn neurons. Since MOR exerts its action via three different types of opioid receptors, we further sought to investigate if there are differential effects of opioid receptor agonists on dorsal horn neurons when administered intracerebroventricularly. Effects of ICV MOR were tested in 28 dorsal horn neurons of the spinal cord in the cat. ICV MOR inhibited, excited and did not affect the heat responses of dorsal horn neurons. ICV DAMGO and DADLE, $\mu$- and $\delta$-opioid agonist, respectively, exhibited the excitation of dorsal horn neurons. In contract, U-50488, a k-opioid agonist, exhibited both the inhibition and excitation of dorsal horn neurons. These results suggest that opioid receptors have different actions on activity of dorsal horn neuron and that the inhibitory action of k-opioid agonist may subserve the analgesia often produced by ICV MOR.
To develop the technology of producing structural board from low grade materials, an attempt was made to produce strand/particle composites from split wood strand(S) and particle(P) of (Cryptomeria japonica D. Don), which changed the layer construction and the ratio of S/P. The influence of layer construction on board properties was determined, focusing on the number and alignment of the S layers. The effect of weight ratio of S/P (3:7, 1:1, 7:3) on mechanical properties was also discussed on seven layered panel. Mechanical properties were determined from static bending tests to give parallel and perpendicular modulus of rupture (MOR) and modulus of elasticity (MOE), and the internal bond (IB) strength. In general, the surface strand layers contributed to the MOR and MOE. The parallel MOR and MOE values were the largest for the single layered S panel (only Slayers: S1), but the perpendicular MOR and MOE was the smallest. Perpendicular MOR and MOE were the largest for seven layered composite that had two cross oriented strand layers (SPSPSPS: SP7). Specimens retained more than half of their MOE and MOR after two hours in boiling water and one hour soaking. IB was the largest for the panel having only P layers, however, differences in IB strength were not identified among the other multi-layered composite panels thus the effect of layer construction on IB strength was small. Thickness swelling (TS) and surface roughness were smaller for the composite having P layers on the surface than for those having S layers. The addition of strands did not enhance the mechanical properties (MOR, MOE, IB). TS values for the panels, with which the S/P ratio was over than 1:1, was the similar to the value for the single layered S panels.
