8-Hydroxyquinoline is used as antibacterial agent and antioxidant based on its function inducing the chelation of ferrous ion present in host resulting in production of chelated complex. This complex being transported to cell membrane of bacteria and fungi exerts antibacterial and antifungal action. In this study, we have carried out in vitro genetic toxicity tests and microarray analysis to understand the underlying mechanisms and the mode of action of toxicity of 8-hydroxyquinoline. TA1535 and TA98 cells were treated with 8-hydroxyquinoline to test its toxicity by basic genetic toxicity test, Ames and two new in vitro micronucleus and COMET assays were applied using CHO cells and L5178Y cells, respectively. In addition, microarray analysis of differentially expressed genes in L5178Y cells in response to 8-hydroxyquinoline were analyzed using Affymatrix genechip. The result of Ames test was that 8-hydroxyquinoline treatment increased the mutations in base substitution strain TA1535 and likewise, 8-hydroxyquinoline also increased mutations in frame shift TA98. 8-Hydroxyquinoline increased micronuclei in CHO cells and DNA damage in L5178Y. 8-Hdroxyquinoline resulted in positive response in all three tests showing its ability to induce not only mutation but also DNA damage. 783 Genes were initially selected as differentially expressed genes in response to 8-hydroxyquinoline by microarray analysis and 34 genes among them were over 4 times of log fold changed. These 34 genes could be candidate biomarkers of genetic toxic action of 8-hydroxyquinoline related to induction of mutation and/or induction of micronuclei and DNA damage. Further confirmation of these candidate markers related to their biological function will be useful to understand the detailed mode of action of 8-hydroxyquinoline.

DIN CK35 (JIS S35CK) steels have been used as a material in fuel pump blocks for marine engines. Failures in the inner surface of a drilling hole, due to the initiation of fatigue cracks have been frequently reported. However, the mechanism initiating these cracks and growths has not been clearly diagnosed yet. This study was conducted using a scraped fuel pump block, containing an initiated fatigue crack in the inner surface of a drilling hole. Initially, the cracks and fractured surfaces inside the block were investigated using an optical microscope and a SEM (Scanning Electron Microscope). In addition, microstructure observation, fatigue life test and fatigue crack growth test were performed using a specimen, which was taken from the same block. Results from these tests are summarized as follows; (1) The early crack in the block was supposed to occur inside the inner surface of the drilling hole. (2) The fatigue endurance of this material was about 330 Mpa. (3) The early crack was generated in the cavitations created by the breakdown of a big inclusion, or separation between the big inclusion and the base metal, in which the fundamental ingredients of the inclusion were C, 5, and Mn. (4) In order to prevent these types of failures, the suppression of inclusions inflow by improving the casting process, formation of fine inclusions by applying a heat treatment process, and coating of the surface of the drilling hole were required.

This study was objected to evaluate the potential impact on the groundwater environment of the coal bottom ash used as fill materials on the land surface. From four coal-fired power plants, bottom-ashes were collected and analyzed through sequential extraction and column leaching tests following the meteoric water mobility procedure. The column tests shown leaching heavy metals including Pb, As, B, Cu, Zn, Mn, Ni, Ba, Sr, Sb, V, Cr, Mo, and Hg. The relatively high concentrations of B, Sr, Ba, and V in leachate were attributed to both the higher concentrations in the bottom ash and the relatively higher portion of leachable state, sorbed state, of metals. Bottom-ash samples from the D-plant only show high leaching potential of sulfate ($SO_4$), probably originated from the coal-combustion process, called the Fluidized Bed Combustion. Consequently, to manage recycling bottom ashes as fill materials, an evaluation system should be implemented to test the leaching potentials of metals from the ashes considering the absolute amount of metals and their state of existence in ashes, and the coal-combustion process.

The International Agency for Research on Cancer (IARC, 1989) has classified whole diesel exhaust as probably carcinogenic to humans. Diesel exhaust particulate matter (DPM) adsorbs different chemical substances including PAHs and nitroarenes. DPM is emphasized because it is a major component of diesel exhaust, it is suspected of contributing to a health hazard. Diesel exhaust is a complex mixture of carbon particles and associated organics and inorganics, and it is not known what fraction or combination of fractions cause the health effects [cancer effects, noncancer effects (respiratory tract irritation/inflammation and changes in lung function)] that have been observed with exposure to diesel exhaust. In order to identify which chemical classes are responsible for the majority of the observed biological activities, we performed a particular biological/chemical analysis. Respirable particulate matter (PM2.5: <2.5mm) was collected from diesel engine exhaust using a high-volume sampler equipped with a cascade impactor. Particulate oganic matter was extracted by the dichloromethane/sonication method and the crude extract was fractionated according to EPA recommended procedure into seven fractions by acid-base partitioning and silica gel column chromatography. We examined genotoxic potentials of diesel exhaust particulate matter using novel genotoxicity tests, which are rapid, simple and sensitive methods for assessing DNA-damage at the DNA and chromosomal level (comet assay, in vitro MN test and Ames test). Higher genotoxic potency was observed in non polar fractions and several PAHs were detected by GC-MS, such as 1,2,5,6 dibenzanthracene, chrysene, 1,2-benzanthracene, phenanthrene and fluoranthene.
A bioassay technique and organ bath study were performed to analyze the effects of extracellular $Ca^{2+}$ and $Ca^{2+}$-antagonists on endothelium-derived relaxing factor[s][EDRF] released from the endothelial cells of rabbit aorta. Transverse strips with intact endothelium or damaged endothelium were used for the mechanical contraction experiment using organ bath. Long segment including thoracic and abdominal aorta with endothelium [EDRF donor aorta] was perfused with Tyrode solution which was aerated with 95% $O_2-5%$$CO_2$ mixed gas and kept at 35oC. The perfusate was bioassayed with a transverse strip of thoracic aorta with damaged endothelium. The test strip was contracted with nor-epinephrine and acetylcholine was used to stimulate the release of EDRF from endothelial cells. The results obtained were as follows; 1] The endothelium-dependent relaxation[EDR] induced by acetylcholine was biphasic; an initial rapid relaxation followed by a slow relaxation. 2] EDR induced by acetylcholine was reduced gradually with the decrease in the concentration of extracellular $Ca^{2+}$. The effect of extracellular $Ca^{2+}$ on EDR was more prominent in the late slow relaxation phase. 3] EDR to acetylcholine was not altered by acute exposure to organic $Ca^{2+}$-antagonists. Pretreatment with verapamil to the EDRF donor aortic segment did not alter the magnitude of EDR. 4] Among the inorganic $Ca^{2+}$-antagonists $Mn^{2+}$ and $Cd^{2+}$ did not inhibit EDR, whereas $Co^{2+}$ and $La^{3+}$ inhibited EDR. 5] The inhibitory response of $Co^{2+}$ to EDR developed when infused directly on the test strip. That of $La^{3+}$, however, was evoked when added to solution perfusing the donor aortic segment. The above results suggest that $Ca^{2+}$-antagonists do not affect EDR and the inhibitory effect of $Ca^{2+}$ results from influencing the action of EDRF on vascular smooth muscle, whereas that of $La^{3+}$ results from its action on the release of EDRF from endothelial cells.

The purpose of this study was to evaluate a new sampling and analytical method for the determination of hexavalent chromium, Cr(VI) in mist from field plating operation. The Procedures of this new method (Shin & Paik's Method) are as the following: Airborne hexavalent chromium is collected on polyvinyl chloride (PVC) filter according to the National Institute for Occupational Safety and Health (NIOSH) Method 7600, and the filter sample is placed in a screw-capped vial and soaked with 2% NaOH/3% Na₂CO₃ solution immediately after sampling. The Cr(VI) sample is analyzed by ion chromatography/visible spectrophotometry (IC/VS) according to the U.S. Environmental Protection Agency (EPA) Method 218.6. The airborne Cr(VI) concentrations measured by this method were compared with those determined by three reference methods: One (NIOSH/EPA Method) consisted of sampling airborne Cr(VI) on PVC filters and storing the sample filters in screw-capped vials according to the NIOSH method, and analyzing Cr(VI) in samples using IC/VS according to the EPA method. The second method (Impinger Method/NaHCO₃) consisted of absorbing airborne Cr(VI) into 0.02 MN/sub a/Hco₃ solution in midget impinger, and analyzing the Cr(VI) in samples using IC/VS. The third method was the OSHA Method Id-215. Using these four different methods, four replicates of air samples were collected at an electroplating process and analyzed simultaneously. Two-way ANOVA and Paired t-test were used to test difference among values determined by the methods. There was no significant difference and a strong correlation (r/sup 2/=0.99) between Cr(VI) concentrations measured by the shin & Paik's Method and an impinger method (p>0.05). However, Cr(VI) concentrations determined by Shin & Paik's Method were Significantly different form those by the NIOSH/EPA Method (p<0.05) or the OSHA method (p<0.05). The Cr(VI) concentrations of Shin & Paik's Method were Significantly higher than those of the NIOSH/EPA Method or the OSHA method. We concluded that the Shin & Paik's Method could prevent Cr(VI) losses caused by reduction and give more reliable results of airborne Cr(VI) concentrations in work environments.

The process of cold rolling is becoming more severe with the increase in the production of high tensile steel strip as a result of increased demand. Consequently, there is a need to develop work roll materials with better resistance to wear and roll failure. DOOSAN has developed an improved in-house 5%Cr alloy, New HSR1, with properties superior to the existing in- house 5%Cr alloy, Old HSR1. Test alloys were designed with controlled amounts of Si and Mn based on Old HSR1 and an optimum alloy was chosen based on thermal shock tests. A prototype work roll was manufactured with New HSR1, and properties of test specimens were evaluated. The results indicated that New HSR1 has better properties than Old HSR1. After application of New HSR1 work rolls, productivity increased due to advanced resistance to wear and roll failure.
The object ofthis study was to test for STORMSYS process that composed Catch Basin and Stormsys(three units: vortex solids separator, filter media bed and vegetated filter strips). It could be applied to treat the first-flush non-point pollution materials on the road(especially, motorway). This study investigated that the runoff characteristics of non-point pollutions containing the heavy metal(Fe, Zn and Cu) by rainfall showed relatively high pollution concentration in the early-stage storm runoff on the road, which seems to be caused by the vehicular traffic, and showed the rapid reduction of pollution concentration on the basis of about 5mm rainfall volume. As the number of the non-rainy days were increased, the pollution concentration by storm runoff was increased, also. As a test result of this process, the average removal efficiency of BOD, $COD_{mn}$, SS, T-N and T-P over the testing period were 92.7%,88.6%,97.4%,93.0% and 93.3%, respectively. Also, the average removal efficiency of n-Hexane, Fe, Zn and Cu were 86.7%, 96.1%, 84.4% and 78.4%, respectively. As shown in the characteristics of storm runoff, the non-point pollution materials have high pollution concentration in the early-stage storm runoff on the road, the installation of STORMSYS process is expected to reduce considerable amount of non-point pollutions.
The aim of this study was to enhance of the Discfilter process (maximum treatment capacity - 500 ton/day) removal efficiency for small-scale wastewater reclamation under various influent wastewater conditions (with / without coagulation process, coagulant content and temperature). The result of chemical resistance test for fiber filter in the Discfilter that weight loss was obtained with 0.535% under pH 3, 0.092% under pH 9 and 0.028% under 10% NaClO. The removal efficiency test of Discfilter process on the with / without coagulation process showed that with coagulation process condition was occurred CODMn of 42.26 ± 0.61, BOD5 of 88.72 ± 0.44, T-P of 84.67 ± 0.72 and SS of 90.58 ± 0.61. The removal efficiency of Discfilter process on the coagulant content (4.5, 5.0 and 5.0 mL/min) and temperature (< 10℃, 10℃~ 20℃ and > 20℃) conditions were increased as coagulant content and temperature increased. This study demonstrated the potential application of Discfilter process for small-scale wastewater reclamation.

Rotary swaging is one of the incremental forming process which is a chipless process using the reduction of cross-sections of bars, tubes and wires. The TDS(Tube Drive Shaft) of monobloc used in automotive has been developed by the rotary swaging process. The mechanical characteristics of swaged parts such as the hardness, thickness and roughness are also estimated to conduct experimental analyses of rotary swaging process with the materials of 34Mn5 Furthermore the change in the vibration mode of TDS due to design parameters, which are the tube length, diameter and thickness, has been investigated and analysed. The weight of the TDS product is smaller by about $12.8\%$ than that of SDS with the same performance. It could be evidently found that the TDS is designed to be much lighter than SDS (Solid Drive Shaft). This advantage might give some possibility to improve the NVH (Noise-Vibration-Harshness) characteristics. A maximum torque and a total number of torsional repetitions for the TDS is checked and measured to know the torsional intensity and fatigue strength through the static torsion test and torsional durability test, respectively. A total number of the torsional repetitions up to the fracture for the TDS is greater than 250,000 times.
