FRC에서 Lymphotoxin β receptor의 자극은 MLCK와 ROCK의 이중 신호전달 경로를 통해 stress fiber 변화에 관여 (Signals of MLCK and ROCK Pathways Triggered via Lymphotoxin β Receptor are Involved in Stress Fiber Change of Fibroblastic Reticular Cells)

  • Lymphotoxin ${\beta}$ receptor ($LT{\beta}R$)는 TNF 계열로 림프조직의 미세구조와 기관형성에 중요한 역할을 한다. MLCK와 ROCK는 세포의 stress fiber 형성조절에 관여하는 주요 신호전달자이다. Fibroblastic reticular cell (FRC)에서 $LT{\beta}R$ 자극을 통한 이런 신호전달자들의 관련성을 알아보기 위해 ML-7 (MLCK 저해제)이 사용되었다. ML7 처리된 FRC에서 SF가 완전히 파괴되었고 anti-$LT{\beta}R$ antibody 처리 세포와 유사하게 ML7 처리 FRC에서 응축된 세포형태를 관찰 할 수 있었다. Y27632로 ROCK를 저해 했을 때 FRC의 액틴 세포골격과 세포형태 변화가 유도 되었다. FRC에서 p-MLC가 액틴과 함께 SF 구성성분을 이루었다. FRC세포 추출물로 Rho-guanosine diphosphate (GDP)/guanosine triphosphate (GTP) 교환활성을 확인했다. Agonistic anti-$LT{\beta}R$ antibody로 $LT{\beta}R$을 자극 했을 때 Rho-GDP/GTP 교환활성이 크게 감소했다. MLCK 저해처럼 $LT{\beta}R$ 자극은 MLC의 인산화를 감소시켰다. Agonistic anti-$LT{\beta}R$ antibody-treated FRC에서 세포골격 구성요소인 세포막과 세포골격 링커 역할을 하는 p-ezrin의 인산화는 감소 되었고 b- actin, 그리고 tubulin 발현도 줄었다. 이런 결과는 FRC의 $LT{\beta}R$ 신호전달을 통한 SF 조절에는 MLCK와 ROCK가 관여하고 있다는 것을 알 수 있었다.

  • 종양괴사인자 수용체 일종인 Lymphotoxin ${\beta}$ receptor ($LT{\beta}R$)은 림프 구조와 기관 형성에 중요한 역학을 한다. 우리는 fibroblastic reticular cell (FRC)에서 agonistic $anti-LT{\beta}R$ antibody로 $LT{\beta}R$을 자극하면 stress fiber (SF)에 변화가 생긴다는 것을 알았다. MLCK와 ROCK는 세포에서 SF 형성 기여에 중요한 역할을 한다. 본 연구는 MLCK 저해에 초점을 맞추어 $LT{\beta}R$ 신호 전달은 SF 조절로 항섬유화 효과에 대하여 조사하였다. SF 변화에 대한 $LT{\beta}R$의 기능 조사를 위해 agonistic $anti-LT{\beta}R$ antibody로 처리된 FRC와 세포 추출물을 이용하여 immunoblot, fluorescence assay와 Rho-guanosine diphosphate (GDP)/guanosine triphosphate (GTP) exchange 활성 분석법으로 분석하였다. 세포막과 세포골격 연결자 ezrin은 agonistic $anti-LT{\beta}R$ antibody 처리된 FRC에서 완전히 탈인사화가 유도되었다. Actomysoisn에 의한 SF를 확인하였고 인산화 myosin light chain (p-MLC)인 함께 co-localization 되는 것도 확인하였다. ML7 처리된 FRC에서 agonistic $anti-LT{\beta}R$ antibody 처리된 세포에서 관찰되는 유사한 현상인 SF분해, 세포막 응축과 쇠퇴 현상이 나타났다. ROCK 활성저해는 액틴 골격 변화는 유도하였으나 부분적으로 SF가 세포에 남아 있었다. 반면, ML7에 의한 MLCK저해는 SF를 완전히 분해하였다. 또한, $LT{\beta}R$ 자극은 MLC 인산화를 완전히 억제하였지만, Rho-GDP/GTP exchange 활성변화에서는 감소는 되었으나 활성이 완전히 없어지지는 않았다. 결론적으로 이런 결과들은 FRC에서 $LT{\beta}R$신호전달을 통해 유도되는 SF 조절에는 MLCK가 보다 더 강력한 역할을 한다는 것을 제시하고 있다.

Melatonin inhibits the Migration of Colon Cancer RKO cells by Down-regulating Myosin Light Chain Kinase Expression through Cross-talk with p38 MAPK

  • Background: Melatonin, which is mainly produced by the pineal gland, has a good inhibitory effect on cell growth of multiple cancer types. However, the underlying molecular mechanisms of anti-tumor activity for colon cancer have not been fully elucidated. In this study, we investigated the effects of melatonin on migration in human colon cancer RKO cells and the potential molecular mechanisms. Materials and Methods: The viability of RKO cells was investigated by MTT assay after treatment with melatonin, SB203580 (p38 inhibitor) and phorbol 12-myristate 13-acetate (PMA, MAPK activator) alone or in combination for 48h. The effects of melatonin, and ML-7, a selective inhibitor of myosin light chain kinase (MLCK), and SB203580, and PMA on the migration of RKO cells were analyzed by in vitro scratch-wound assay. The relative mRNA levels of MLCK was assessed by real-time quantitative RT-PCR. Western blotting analysis was performed to examine the expression of MLCK, phosphorylation of myosin light chain (pMLC) and p38 (pp38). Results: The proliferation and migration of human colon cancer RKO cells were inhibited significantly after treatment with melatonin. The expression levels of MLCK and phosphorylation of MLC of RKO cells were reduced, and real-time quantitative RT-PCR showed that melatonin had significant effects on suppressing the expression of MLCK. Furthermore, the phosphorylation level of p38, which showed the same trend, was also reduced when cells were treated by melatonin. In addition, ML-7 (25umol/l) could down-regulate the phosphorylation of p38. Conclusions: Melatonin could inhibit the proliferation and migration of RKO cells, and further experiments confirmed that p38 MAPK plays an important role in regulating melatonin-induced migration inhibition through down-regulating the expression and activity of MLCK.

A Novel All-trans Retinoid Acid Derivative N-(3-trifluoromethyl-phenyl)-Retinamide Inhibits Lung Adenocarcinoma A549 Cell Migration through Down-regulating Expression of Myosin Light Chain Kinase

  • Aim: To observe the effects of a novel all-trans retinoid acid (ATRA) derivative, N-(3-trifluoromethyl-phenyl)-retinamide (ATPR), on lung adenocarcinoma A549 cells and to explore the potential mechanism of ATPR inhibiting of A549 cell migration. Materials and Methods: The cytotoxicity of ATRA and ATPR on A549 cells was assessed using MTT assay. Wound healing assays were used to analyze the influences of ATRA, ATPR, ML-7 (a highly selective inhibitor of myosin light chain kinase (MLCK)), PMA (an activator of MAPKs) and PD98059 (a selective inhibitor of ERK1/2) on the migration of A549 cells. Expression of MLCK and phosphorylation of myosin light chain (MLC) were assessed by Western blotting. Results: ATRA and ATPR inhibited the proliferation of A549 cells in a dose- and time-dependent manner, and the effect of ATPR was much more remarkable compared with ATRA. Relative migration rate and migration distance of A549 cells both decreased significantly after treatment with ATPR or ML-7. The effect on cell migration of PD98059 combining ATPR treatment was more notable than that of ATPR alone. Moreover, compared with control groups, the expression levels of MLCK and phosphorylated MLC in A549 cells were both clearly reduced in ATRA and ATPR groups. Conclusions: ATPR could suppress the migration and invasion of A549 cells, and the mechanism might be concerned with down-regulating the expression of MLCK in the ERK-MAPK signaling pathway, pointing to therapeutic prospects in lung cancer.

Characteristics of Diprophylline-Induced Bidirectional Modulation on Rat Jejunal Contractility

  • In this study, we propose that diprophylline exerts bidirectional modulation (BM) on the isolated rat jejunal segment depending on its contractile state. The results supported the hypothesis. Diprophylline ($20{\mu}M$) exerted stimulatory effects on the contractility of jejunal segment in six low contractile states while inhibitory effects in six high contractile states, showing the characteristics of BM. Diprophylline-induced stimulatory effect was significantly blocked by atropine, indicating the correlation with cholinergic activation. Diprophylline-induced inhibitory effect was partially blocked by phentolamine, propranolol, and L-N-Nitro-Arginine respectively, indicating their correlation with sympathetic activation and nitric oxide-mediated relaxing mechanisms. Diprophylline-induced BM was abolished by tetrodotoxin or in a $Ca^{2+}$ free condition or pretreated with tyrosine kinase inhibitor imatinib, suggesting that diprophylline-induced BM is $Ca^{2+}$ dependent, and that it requires the presence of enteric nervous system as well as pacemaker activity of interstitial cells of Cajal. Diprophylline significantly increased the reduced MLCK expression and myosin extent in constipation-prominent rats and significantly decreased the increased MLCK expression and myosin extent in diarrhea-prominent rats, suggesting that the change of MLCK expression may also be involved in diprophylline-induced BM on rat jejunal contractility. In summary, diprophylline-exerted BM depends on the contractile states of the jejunal segments, requires the presence of $Ca^{2+}$, enteric nervous system, pacemaker activity of interstitial cells of Cajal, and MLCK-correlated myosin phosphorylation. The results suggest the potential implication of diprophylline in relieving alternative hypo/hyper intestinal motility.

  • Stress fiber (SF) 변화는 세포외부의 결합인자와 세포 수용체와 결합후 리모델링을 위해 액틴골격에 신호를 전달하며 일어난다. 이 연관은 결합장소에서 기계적 활동과 신호전달활동을 조절하는 다양한 스케폴드들과 신호 전달자에 의해 매게된다. Heterotrimeric transmembrane lymphotoxin α1β2 (LTα1β2)는 용해성 homotrimeric LT α를 포함하는 tumor necrosis factor (TNF) 계로 림프조직을 구성하는데 중요한 역할을 한다. LTα1β2와 LTβR의 결합은 fibroblastic reticular cell (FRC)에서 신호전달을 촉발한다. Agonistic anti-LTβR antibody 단독 혹은 LTα 그리고 TNFα의 조합으로 LTβR 자극은 세포의 액틴과 형태적 변화를 보았다. Agonistic anti-LTβR antibody의 FRC에서 작용을 통한 세포골격 재배열이 myosin과의 관련성을 확인하기위해 myosin light chain kinase (MLCK)의 저해제인 ML-7과 myosin light chains (MLC)와 myosin phosphatase target subunit 1 (MYPT1)의 인산화에 대한 효과를 확인하였다. MLCK 저해는 액틴 세포골격 재배열과 세포형태 변화를 유도하였다. 또한, MLC와 MYPT1인산화가 LTβR 자극에 의해 줄어드는 것을 확인하였다. DNA chip 분석은 myosin and actin 구성선분이 전사체 수준에서도 줄어드는 것을 보였다. 결론적으로 LTβR 자극은 FRC에서 SF변화는 myosin과 관련되어 있다는 것을 제시한다.

New Insights into 4-Amino-2-tri-fluoromethyl-phenyl Ester Inhibition of Cell Growth and Migration in the A549 Lung Adenocarcinoma Cell Line

  • Objective: The present study was designed to investigate the probable mechanisms of synthetic retinoid 4-amino-2-tri-fluoromethyl-phenyl ester (ATPR) inhibition of the proliferation and migration of A549 human lung carcinoma cells. Materials and Methods: After the A549 cells were treated with different concentrations of ATPR or all-trans retinoic acid (ATRA) for 72 h, scratch-wound assays were performed to assess migration. Immunofluorescence was used to determine the distribution of CAV1 and $RXR{\alpha}$, while expression of CAV1, MLCK, MLC, P38, and phosphorylation of MLC and P38 were detected by Western blotting. Results: ATPR could block the migration of A549 cells. The relative migration rate of ML-7 group had significantly decreased compared with control group. In addition, ATPR decreased the expression of a migration related proteins, MLCK, and phosphorylation of MLC and P38. ATPR could also influence the expression of RARs or RXRs. At the same time, CAV1 accumulated at cell membranes, and $RXR{\alpha}$ relocated to the nucleus after ATPR treatment. Conclusions: Caveolae may be implicate in the transport of ATPR to the nucleus. Change in the expression and distribution of $RXR{\alpha}$ may be implicated in ATPR inhibition of A549 cell proliferation. The mechanisms of ATPR reduction in A549 cell migration may be associated with expression of MLCK and phosphorylation of MLC and P38.

MLCK and PKC Involvements via Gi and Rho A Protein in Contraction by the Electrical Field Stimulation in Feline Esophageal Smooth Muscle

  • We have shown that myosin light chain kinase (MLCK) was required for the off-contraction in response to the electrical field stimulation (EFS) of feline esophageal smooth muscle. In this study, we investigated whether protein kinase C (PKC) may require the on-contraction in response to EFS using feline esophageal smooth muscle. The contractions were recorded using an isometric force transducer. On-contraction occurred in the presence of $N^G$-nitro-L-arginine methyl ester (L-NAME), suggesting that nitric oxide acts as an inhibitory mediator in smooth muscle. The excitatory composition of both contractions was cholinergic dependent which was blocked by tetrodotoxin or atropine. The on-contraction was abolished in $Ca^{2+}$-free buffer but reappeared in normal $Ca^{2+}$-containing buffer indicating that the contraction was $Ca^{2+}$ dependent. 4-aminopyridine (4-AP), voltage-dependent $K^+$ channel blocker, significantly enhanced on-contraction. Aluminum fluoride (a G-protein activator) increased on-contraction. Pertussis toxin (a $G_i$ inactivator) and C3 exoenzyme (a rhoA inactivator) significantly decreased on-contraction suggesting that Gi or rhoA protein may be related with $Ca^{2+}$ and $K^+$ channel. ML-9, a MLCK inhibitor, significantly inhibited on-contraction, and chelerythrine (PKC inhibitor) affected on the contraction. These results suggest that endogenous cholinergic contractions activated directly by low-frequency EFS may be mediated by $Ca^{2+}$, and G proteins, such as Gi and rhoA, which resulted in the activation of MLCK, and PKC to produce the contraction in feline distal esophageal smooth muscle.

High fat diet confers vascular hyper-contractility against angiotensin II through upregulation of MLCK and CPI-17

  • Obesity is a critical risk factor for the hypertension. Although angiotensin II (Ang II) in obese individuals is known to be upregulated in obesity-induced hypertension, direct evidence that explains the underlying mechanism for increased vascular tone and consequent increase in blood pressure (BP) is largely unknown. The purpose of this study is to investigate the novel mechanism underlying Ang II-induced hyper-contractility and hypertension in obese rats. Eight-week old male Sprague-Dawley rats were fed with 60% fat diet or normal diet for 4 months. Body weight, plasma lipid profile, plasma Ang II level, BP, Ang II-induced vascular contraction, and expression of regulatory proteins modulating vascular contraction with/without Ang II stimulation were measured. As a result, high fat diet (HFD) accelerated age-dependent body weight gaining along with increased plasma Ang II concentration. It also increased BP and Ang II-induced aortic contraction. Basal expression of p-CPI-17 and myosin light chain (MLC) kinase was increased by HFD along with increased phosphorylation of MLC. Ang II-induced phosphorylation of CPI-17 and MLC were also higher in HFD group than control group. In conclusion HFD-induced hypertension is through at least in part by increased vascular contractility via increased expression and activation of contractile proteins and subsequent MLC phosphorylation induced by increased Ang II.

EFFECTS OF CAFFEINE AND 2,5-DI-(tert-BUTYL)-1,4-BENZOHYDROQUINONE ON BLUE LIGHT-DEPENDENT $H^+$ PUMPING IN GUARD CELL PROTOPLASTS FROM Vicia faba L.

  • The sensory transduction processes of blue light in guard cells have been suggested the involvement of Ca$^{2+}$/calmodulin-dependent myosin light chain kinase (MLCK) or MLCK-like proteins. The source of Ca$^{2+}$ required for the signal transduction process was investigated in guard cell protoplasts (GCPs). The GCPs showed the typical H$^+$ pumping activity by blue light (200 $\mu$mol m$^{-2}$ s$^{-1}$) and fusicoccin (10 $\mu$M) under background red light (600 $\mu$mol m$^{-2}$ s$^{-1}$). The blue light-dependent H$^+$ pumping was not significantly affected by the externally changed Ca$^{2+}$ concentrations. The addition of 1 mM Ca$^{2+}$ in the bathing medium ratherly inhibited the H$^+$ pumping. In contrast, the blue light-dependent H$^+$ pumping was inhibited by caffeine and 2,5-di-(tert-butyl)-1,4-benzohydroquinone (BHQ), inhibitor of C$^{2+}$-ATPase in endoplasmic reticulum (ER) without inhibiting the H $^+$ pump. The inhibition by caffeine and BHQ was fully reversible. The extent of inhibition by caffeine and BHQ was larger when they were added together than when added separately. The results suggest that Ca$^{2+}$ required for the blue light-dependent H$^+$ pumping may be released from the intracellular Ca$^{2+}$ stores, probably ER in guard cells.
