목련 꽃 에센셜 오일의 항균 및 항염증 활성 (Antibacterial and Anti-inflammatory Effects of Essential Oil from the Magnolia kobus Flower)

  • 목련은 항산화효과와 진정작용과 같은 다양한 효능을 가진 것으로 알려져 있다. 본 연구에서는 목련 에센셜오일의 항균 활성 및 항염증 활성을 검증하고자 한다. 목련 에센셜 오일은 증류법으로 추출한 것을 사용하였으며, 항균 활성은 S. aureus와 E. coli, P. aeruginosa에 대하여 paper disk diffusion법과 최소저해농도(minimum inhibitory concentration, MIC) 값을 측정하여 확인하였다. 목련 에센셜 오일은 S. aureus 균주에 대하여 18 mm의 clear zone형성과 0.25 mg/ml의 MIC 값을 나타내는 우수한 항균효과를 보였다. P. aeruginosa 균주와 E. coli 균주는 clear zone 형성이 각각 14 mm와 17 mm로 나타났다. 또한, MIC값은 각각 1 mg/ml와 0.5 mg/ml로 측정되었다. 항염증 활성은 LPS 유도된 RAW264.7 세포에서 nitric oxide (NO)와 PGE2의 생성억제율을 조사하였다. 목련 에센셜 오일, 500 ㎍/ml의 농도에서 RAW264.7세포에 대한 세포 독성은 나타내지 않았지만, NO (37.7%)와 PGE2 (24.0%)억제를 보였다. GC-MS를 이용하여 목련 에센셜 오일을 분석하였다. 목련 에센셜 오일의 3 가지 주요 방향제 성분은 3-carene (77.07%), β-elemene (6.92%) 및 caryphyllene (2.86%)로 분석되었다. 이러한 결과는 목련 에센셜 오일이 항균 및 항염증 효과를 갖는 화장품 기능성 물질로서 잠재력을 가지고 있음을 시사한다.

충치균 (Streptococcus mutans)에 대한 다시마 추출물의 항균활성 (Antibacterial Activity of Sea-mustard, Laminaria japonica Extracts on the Cariogenic Bacteria, Streptococcus mutans)

  • 우리 나라 연안에서 채취한 해조류 27종의 $80\%$ ethanol 추출물이 Streptococcus mutans KCTC 3300의 증식에 미치는 영향을 검색하고, 다시마의 추출용매별 항균성과, $80\%$ ethanol 추출물을 용매분획하였을 때 획분별 MIC를 측정하였다. S. mutans KCTC 3300에 대한 해조류 추출물의 항균활성은 다시마가 가장 강하였고, 갈파래, 청각, 감태, 곰피 및 미역도 비교적 높은 활성을 나타내었으며 홍조류보다 갈조류와 녹조류에 속하는 종이 대체적으로 강하였다 다시마의 항균활성은 추출용매에 따라 차이가 있었으나 용매별 가용성 획분의 량과 항균성간에는 뚜렷한 관련이 나타나지 않았다. 5. mutans KCTC 3300과 S. sobrinus KCTC 3307에 대한 해조류의 $80\%$ ethanol 추출물의 MIC는 해조류의 종뿐만 아니라 동일한 해조류 추출물의 경우에도 균종에 따라 차이가 있었다. 다시마의 $80\%$ ethanol 추출물을 유기용매를 사용하여 순차 분획하였을 때 항균활성 물질의 대부분은 ether 획 분에 이행하여 이 획분의 MIC는 105$\mu$g/mL이었고, 수용성 획분은 약 5,500$\mu$g/mL의 농도에서도 균의 증식을 저지하지 못하였다.

베타-갈락토시데이즈를 이용하여 합성한 1, 2-Hexanediol Galactoside의 보습력과 항균력에 대한 연구 (Water-holding Capacity and Antimicrobial Activity and of 1, 2-Hexanediol Galactoside Synthesized by β-Galactosidase)

  • 재조합 대장균의 ${\beta}-galactosidase$ (${\beta}-gal$)을 이용하여 transgalactosylation 반응으로 1, 2-hexanediol galactoside (HD-gal) 합성을 수행하였다. 먼저, 합성 반응을 수행할 때의, ${\beta}-gal$의 양, 1, 2-hexanediol(HD)의 양, 반응 pH, 반응 온도 등 반응조건을 최적화하였다. ${\beta}-Gal$은 4.8 U/mL, HD는 75 mM, pH는 7.0, 온도는 $37^{\circ}C$의 조건이 최적 합성 조건이었으며, 이때, 약 96%의 HD가 HD-gal로 conversion 되었다. 그리고, 돼지 epidermis를 이용하여 HD와 HD-gal의 water holding capacity (WHC)를 비교 측정하여 보았는데, 이때, epidermis 표면에 HD와 HD-gal을 84.4, 126.6, 168.8, 211.0 mM을 처리하였으며, 84.4, 126.6, 168.8 mM의 HD와 HD-gal을 처리하였을 경우 HD-gal이 HD보다 더 큰 WHC 값을 보여 주었고, 168.8 mM의 경우에는 HD-gal이 HD와 비교하여 최대 약 20% 정도의 더 큰 WHC 값을 보여 주었다. 그러나, HD-gal의 MIC (minimal inhibitory concentration) 값을 측정해 본 결과, Enterococcus faecalis의 경우에는 HD의 MIC와 같았으나, Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus의 경우에는 약 3배에서 10배까지 HD-gal의 MIC 값이 HD에 비하여 증가되는 현상이 관찰되었다. 결론적으로, HD의 transgalactosylation 반응으로 인하여 합성된 HD-gal의 보습력은 HD에 비하여 증가되고, HD-gal의 항균력은 HD와 비교하여 감소하는 현상을 관찰하였다.

  • Pseudomonas aeruginosa와 Staphylococcus aureus는 만성 창상 감염의 원인이 되는 주요 병원체이며 상처부위에서 공존한다. 이러한 감염은 단일 감염에 비해 독성이 높아 환자에게 바람직하지 않은 결과를 초래한다. 복합미생물 감염내에서 미생물 간의 상호작용은 질병 진행을 악화시키는 것으로 알려져 있다. 호흡기, 상처, 당뇨병 발과 같은 질병 내 복합 미생물 감염은 다양한 미생물들을 포함하나, 녹농균과 황색포도상구균이 가장 일반적으로 확인된다. 본 연구는 그람음성균 P. aeruginosa와 그람양성균 S. aureus를 중심으로 독성화학물질의 농도구배에 따른 성장양상을 비교하고자 하였다. 세균 성장이 억제되는 농도를 의미하는 최소 억제 농도(MIC)는 특정화학물을 함유한 배지를 연속적으로 희석하여 성장 곡선을 평가하여 결정한다. 두 균주 모두 상기 방법을 통해 성장곡선을 확인하였고, 지수성장기를 적용하여 세균의 배가시간을 계산하였다. 각 독성 물질에 대한 MIC 결과로부터 그람양성균과 그람음성균 사이의 성장 속도 차이와 각 독성 물질에 대한 내성 차이를 식별하였다. 우리는 이 접근 방식이 세균 관련 감염의 혁신적인 치료법 개발에 대한 강력한 잠재력을 가지고 있다고 기대한다.

Antimicribial and Antioxidant Activities of Ethanol Extracts of Medicinal Plants

  • The objective of this study was to determine the radical scavenging activity, total phenolic content, antimicrobial activity, minimum inhibitory concentration (MIC) of ethanol extracts of 32 medical plant species that have been commonly used in medicinal plants. Total phenolic index of T. chebula exhibited the highest value (498.01㎎/g), followed by R. coreanus miquel (400.33㎎/g), Sanguisorba officinalis (368.25㎎/g), P. thumbergiana (259.74㎎/g) and Eugenia aromaticum (229.38㎎/g). Radical scavenging activity for the DPPH radical was highest in T. chebula (40.91%, p<0.01), followed by C. sappan (36.50%), S. officinalis (32.92%), R. coreanus miquel (26.54%) and P. thumbergiana (24.50%). The extracts from T. chebula, R. coreanus muquel, C. sappan, E. aromaticum, S. officinalis and C. japonica possessed outstanding antimicrobial activity against Escherichia coli, Salmonella Typhimurium, Pseudomonas aeruginosa, Staphylococcus aureus, Bacillus subtilis and Lactobacillus plantarum. MIC was determined on those extracts that showed high efficacy against the test organisms. The most potent MIC values were seen for T. chebula extract against P. aeruginosa, S. aurusa, E. coli, B. subtilis, L. plantarum and S. Typhimurium at 7.8, 7.8, 15.6, 7.8, 125 and 31.2㎍/mL, respectivley. Furthermore, the total phenolic content and radical scavenging activity were very closely correlated for all samples (r=0.78). The coefficient correlations between total phenolic index and antimicrobial activity were 0.91 (E. coli), 0.91 (B. subtillis), 0.79 (P. aeruginosa), 0.79 (S. Typhimurium) and 0.70 (L. plantarum).

황련과 Pelargonium sidoides 복합제제의 호흡기 감염 세균에 대한 항균 효과 (Anti-bacterial effect of fusion formulation of Coptis rhizoma and Pelargonium sidoides on the growth of bronchial diseases bacteria)

  • Objective : Coptis rhizoma is traditional herb in Korean medicine, and Pelargonium sidoides extract has been used for relief of acute bronchitis in Western medicine. The present study examined the antibacterial effect of fusion formulation of Coptis rhizoma extract and Pelargonium sidoides extract against bronchial diseases bacteria. Methods : Test sample, fusion formulation of Korean and Western medicine, was prepared by mixing Coptis rhizoma extract and Pelargonium sidoides extract at a ratio of 1:2 (w/w). Antimicrobial properties of test sample were determined by agar diffusion assay and minimum inhibitory concentration (MIC) against bronchus diseases bacteria. Results : In agar diffusion assay, the highest amount of test sample (4 mg/disk) exhibited antibacterial activity against all microorganisms tested. Test sample showed the high activity for S. aureus (19.5 mm), C. diptheriae (16.5 mm), A. fumigatus (19.3 mm), F. nucleatum (22.7 mm) and Mycobactrium sp. (17.3 mm), whereas it showed a low activity for K. pneumonia (9.7 mm). The MIC value was determined as $250{\mu}g/m{\ell}$ against C. diptheriae. Test sample showed better growth inhibitory effects against S. aureus and A. fumigatus with the MIC valus of $125{\mu}g/m{\ell}$. Conclusion : These results suggest the possibility of application to chronic respiratory diseases of fusion formulation of Korean and Western medicine, which was prepared with Coptis rhizoma extract and Pelargonium sidoides extract.

인천연안 Vibrio parahaemolyticus의 분포 및 유산과 과산화수소 처리에 의한 항균효과 (Antimicrobial Effect of Lactic acid and Hydrogen Peroxide and Distribution of Vibrio parahaemolyticus from the Incheon Adjacent Sea)

  • This study was carried out to investigate the distribution of Vibrio parahaemolyticus in the Incheon adjacent sea, and antimicrobial effect on growth of Vibrio parahaemolyticus in lactic acid and hydrogen peroxide and combination of lactic acid and hydrogen peroxide. The detected strains were compared geographical, months and sample types. The distribution of Vibrio parahaemolyticus was high at Ganghwa county with 66.1%(336 samples), on 7-9 months with 72.4%(386 samples) and from tireland with 75.0%(90 samples), respectively. The minimun inhibitory concentration (MIC) of lactic acid in Vibrio parahaemolyticus were 1250 ppm at pH 6.5 and 7.0, 625 ppm at pH 6.0. respectively. The minimun inhibitory concentration (MIC) of hydrogen peroxide in Vibrio parahaemolyticus were 25 ppm at pH 6.5 and 7.0, 12.5 ppm at pH 6.0, respectively. MICs of combined treatment of lactic acid and hydrogen peroxide in Vibrio parahaemolyticus were 625 ppm of lactic acid with 12.5 ppm of hydrogen peroxide. The correlations between MICs of lactic acid and hydrogen peroxide in Vibrio parahaemolyticus were obtained through the coefficient of determination($R^2$). $R^2$ value were 1.0000. The antimicrobial effect of lactic acid and hydrogen peroxide in Vibrio parahaemolyticus could be confirmed from the result of this experiment.

An Effective Antidandruff Agent-IPBC

  • The purpose of this study was to determine the effect of IPBC(3-lodo-2-propynylbutylcarbamate) on dandruff caused by the anthropophilic fungus Malassezia furfur. The effects of IPBC on dandruff were examined by evaluating (a) the MIC value of IPBC using broth dilution method; (b) the remnant antimicrobial activity of IPBC containing shampoo on skin disc; (c) the antidandruff efficacy of 1.0 % IPBC containing shampoo in double blind clinical trial. To investigate the remnant antimicrobial activity of IPBC against Malassezia furfur, guinea pig-skin disc was washed with antidandruff shampoo and then the diameter of inhibition zone per disc was measured. For clinical trial, thirty healthy volunteers, aged 25-35, participated in 4 week study. At 0, 2, 4 weeks, examinations of scaling, itching on scalp were carried out. The MIC(Minimun Inhibition Concentration) values of IPBC range from 0.10 to 1.00${\mu}g/ml$ and it seems that IPBC is more effective in the MIC values than zinc pyrithione, selenium disulphide, piroctone olamine and comparable to ketoconazole, climbazole. When the remnant antimicrobial activity of IPBC shampoo on skin disc was determined, 0.5% IPBC shampoo and 2.0% Ketoconazole shampoo resulted in similar antimicrobial effect. In addition, 1.0%, 2.0% IPBC shampoo was more effective than 2.0% ketoconazole shampoo. After two and four-weeks of 1.0% IPBC shampoo treatment, there was significant reduction of scaling, itching in test group compared to control group. On the basis of these results, it can be concluded that 1.0% IPBC is more effective than 2.0% Ketoconazole in reducing dandruff. It seems that strong capacity of drug binding to the stratum corneum plays a role in its antidandruff effect since adsorption of active ingredients on scalp is very important factor in reducing dandruff.

Antimicrobial Effects of Oleanolic Acid against Streptococcus mutans and Streptococcus sobrinus Isolated from a Korean Population

  • Oleanolic acid is a natural triterpenoid that exists widely in foods and some medicinal herbs. The purpose of this study was to determine the antimicrobial activity of oleanolic acid against Streptococcus mutans strains isolated from a Korean population. Antimicrobial activity against these bacteria was evaluated by minimal inhibitory concentration (MIC) and time kill curves. The tolerance of human gingival fibroblasts and human periodontal ligaments to oleanolic acid was tested using a methyl thiazolyl tetrazolium (MTT) assay. The $MIC_{90}$ value of oleanolic acid for both S. mutans and S. sobrinus isolated from Koreans was 8 ${\mu}g/ml$. Oleanolic acid showed bactericidal effects against S. mutans ATCC $25175^T$ and S. sobrinus ATCC $33478^T$ at $1\;{\times}\;MIC$ ($8{\mu}g/ml$) and had no cytotoxic effects against KB cells at this dose. The results suggest that oleanolic acid could be useful in the future development of oral hygiene products for the prevention of dental caries.

An Effective Antidandruff Agent - IPBC
