Silage Yield of Korean Local Maize Lines(MET) with Many Tillers and Ears (다수다얼성 재래종 옥수수 계통의 청예수량)

  • From a series of studies conducted on the local maize lines at the Agr. College of the Chungnam National Univ., a few maize lines with many tillers and ears per plant were identified and tentatively named as MET. The purpose of the study was to evaluate the MET lines, which were selfed for five generations, for silage purpose under the different plant densities. A hybrid, Suwon #19 and a synthetic variety, Puyo #3${\times}$#2, were included for comparison. Plant height at harvesting times showed no significant varietal differences. However, the MET lines were very slow in early plant growth compared to the hybrid or synthetic variety, probably due to inbreeding depression of the MET lines. Total fresh weight at the harvesting times was highest in the MET 1 line. The MET 1 line was about 2,000 kgr. per 10a. higher than the hybrid at the harvesting time. The highest fresh weight was obtained when grown under the plant density of 60 ${\times}$ 20cm. Total dry weight per unit area showed the same tendency as the fresh weight. Total dry weight of MET 1 line was about 2.4 tons per 10a., which was about 10% higher than the hybrid, Suwon #19. As the fresh weight, the total dry weight was also highest in the plant density of 60 ${\times}$ 20cm. The grain yield per 10 are of MET 1 was comparable to the grain yield of the hybrid, especially in the low plant density, 60 ${\times}$ 40cm. The average number of effective tillers of MET lines were 4.5, while the mean tiller number of the hybrid or synthetic variety were none. However, the lodging was one of the problem for growing MET lines. The 100 kernel weight of MET lines was about 9 gr., while the 100 kernel weight of the hybrid or synthetic variety was about 30 grm.

Development of AFLP and STS Markers Related to Stay Green Trait in Multi-Tillered Maize

  • In order to develop molecular markers related to stay green phenotype, AFLP analysis was conducted using near-isogenic lines for either stay green or non stay green trait. Both lines have characteristics of multi-ear and tillers (MET). Two out of 64 primer combinations of selective amplification identified three reproducible polymorphic fragments in MET corn with stay green. Both of E+AGC/M+CAC and E+AAG/M+CAA primer combinations produced two and one specific polymorphic fragments linked to stay green trait, respectively. For the conversion of AFLPs to sequence tag sites (STSs), primers were designed form both end sequences of each two polymorphic fragments. One fragment, which was amplified with E+AAG/M+CAA primer combinations, possessed 298 bp long and showed a $91\%$ homology with maize retrotransposon Cinful-l. One out of two polymorphic fragments produced with E+AGC/M+CAC primer combination had 236 bp long and matched a $96\%$ homology with an intron region of 22kDa alpha zein gene cluster in Zea mays. One out of two PCR fragments amplified with MET2 primer set in the stay green MET was not produced in the non-stay green MET. The developed AFLP and STS marker could be used as an efficient tool for selection of the stay green trait in the MET inbred.

Investigation on Korean Local Maize Lines V. Variabilities of Plant Characters of Multi-eared and Tillered Lines(MET) (재래종 옥수수 수집종에 대한 특성조사 제5보 다수다벽 재래종 옥수수계통의 특성변이)

  • A maize line was selected in 1979 among 1000 Korean local maize lines collected in 1977. The selected maize line was characterized by having three to four tillers and eight to 10 ears on each individual plant. The line was assumed to have a great potential as a silage crop. The investigation was conducted as one of the serial studies on the Korean maize collected lines to provide basic information on the genetic variabilities of the multi-eared and tillered (MET) line and on other agronomic characters, prior to use the line as material for future breeding works for silage crop. The MET line and Suwon #19, single cross hybrid, as check variety were planted on May 1, 15 and 30, in three different levels of plant populations. The results obtained were summarized as follows: 1. The genetic variabilities of multi-ear and tillering habits were greater than environmental variabilities. 2. Total dry leaf weight of individual plant of MET line was also significantly higher than that of Suwon #19. 3. The mean number of tillers and ears bearing on the individual plant of MET line varied greatly with plant densities. The number of tillers and ears was on the average 2.9 and 7.0, respectively, when planted in 60cm. by 60cm. 4. The total dry matter and dried stem weight of the individual plant on MET line were comparable to those of Suwon #19. 5. The kernel weight from the individual plant of MET line was 5 to 40% less than that of Suwon #19, depending upon the plant densities. 6. The Kernel to stover ratio was higher for Suwon #19 than for the MET line. (41% to 35%). 7. The MET line had shown first tiller two weeks after planted on May 1. The second and third tillers appeared three to five days after the appearance of the first tiller. 8. The MET line was very specific in tillering habits. All the tillers were borne on the first few nodes of main stem below the soil surface. 9. The tillering habits of MET line were vigorous in the early part of the growing season, but less vigorous in the later part of the growing season. The number of efficient tillers bearing useable ears, was around two to three, when planted in 60cm. by 60cm. 10. The difference of plant height between main stem and first few tillers was around 10cm. 11. The ear size of MET line was around one-third of the major corn belt hybrids. The shape of ear of MET line was conical, with different diameter. 12. The kernel of the MET line was flinty with small soft starch patches on the endosperm part. 13. The 100 kernel weight was around 15gr., which is about one half of the major high yielding hybrids. 14. The ear height of MET line was comparatively higher than that of Suwon #19. 15. Significantly high and positive phenotypic correlation coefficients were obtained among major plant characters. 16. The growth rate of MET line was slower than that of Suwon #19. 17. MET line and Suwon #19 were both heavily infected with black streaked mosaic virus.

Cytoplasmic Inheritance of High Tillering and Earing Characters of a Korean Local Maize Line(MET) (다수.다얼성 옥수수(NET)의 세포질적 유전)

  • A Korean local maize line, MET, which has multi-ears and tillers has been proved as a potential source for silage production. However, no fundamental genetic nature for the line has been investigated. Therefore, this study was done to find genetic information on the multi-earing and -tillering habits of MET line. MET line and a hybrid. (Mo 17 ${\times}$ B68), with monoculm and single ear per plant were used for production of F$_1$(F$\_$1-12/ and F$\_$1-21/), F$\_$2-12/, F$\_$2-21/, BC$\_$1-12/ and BC$\_$1-21/ generations. From the comparison of reciprocal crosses, it was found that the tillering and earing habits of the MET line are controlled by cytoplasmic factors. The tiller and ear numbers, and barren ears were all characters associated with the MET cytoplasm. The cytoplasmic effect of MET on tiller and ear numb en was not evident in F$_1$ generation, probably because of suppressing effect of heterosis on appearance of tillers or ears. Genetic parameters for the gene action for both tiller and ear number also indicated a lack of mono- or digenic-chromosomal gene effects. The heritability (broad) was very low for both characters. Therefore, it is strongly concluded that the tillering and earing characters of MET line are due to cytoplasmic reasons.

Isozymic Characteristics of Multiple-Ear and Tiller Maize Lines (다수다얼성 옥수수의 동위효소 특성)

  • This experiment was conducted to determine the isozymic differences between normal maize and maize inbreds of multiple ears and tillers (MET). Two maize inbreds Euisung, Iri and their hybrid having tillers and multiple ears were compared with normal maize. With usual electrophoresis using 6% polyacrylamide gel, peroxidase and esterase enzymes were studied. Matured leaf, culm, leaf sheath, root and young ear tissues showed different isozymic patterns between METs and normal maize in peroxidase. The Euisung inbred grown for 7 days under dark condition showed typical peroxidase. bands compared with checks in the tissues of coleoptile and stele. Better observation of isozymic bands was made during early part of maize growth. Parental inbreds showed more active and apparent band differences than their hybrids in esterase. Bands for esterase were also apparently different in the stele, coleoptile and young ear tissues of the METs and the checks. The maize lines infected with black streaked dwarf virus showed obvious differences in peroxidase and esterase isozymes.

Plant Characters of Selfed MET Maize Line (자식(自殖)된 다수다얼성(多穗多蘖性) 옥수수(MET)의 특성(特性) 비교(比較) 시험(試驗))

  • The multiple ear and tiller line (MET), which was selected from a local maize collection and has been selected for earliness by selfing for five generations, was compared its plant characteristics with four U.S. leading hybrids. 1. The general plant growth was a little retarded by poor soil fertility and structure. 2. The fresh weight of the MET line was about 97.8% of the U.S. check varieties. 3. The dry weight of the MET line was also about 94.5% of the check U.S. hybrids (1,695 kg / 10a.). 4. The tillering and eating habits of the MET line had contributed toward increasing fresh and dry weight of the MET line. 5. The average plant height of the MET line was much less than those of the check varieties, due to the selfing depression manifested by the MET line, (234.7cm. of the MET vs 250.7cm. of the check) 6. Apparent differences of disease resistance was observed among varieties studied. The degree of plants infected with the black stripe mosaic virus was over 20% for the check U.S. hybrids, while less than 5% of the MET line was infected with the disease. 7. The grain yield per 10 a of the MET line was far below the check hybrids due to the inbreeding depression of the MET line. However, the kernel number per unit area of the MET line with small size kernel was much greater than that of the check hybrids. The 100 kernel weight of the MET line was around 10 grams. 8. The average fresh and dry matter weight of the three ($sorghum{\times}sudan$ grass) varieties was very much alike with those of the MET line. 9. In conclusion, the use of the MET line for silage production may have dual adventages because of its low seed price and of its small size kernels, which may reduce the amount of seeds required for planting in an unit area.

Investigation on Korean Local Maize Lines VI. Use of a Korean Local Maize Line for Silage Production (재래종 옥수수 수집종에 대한 특성조사 제6보 청예를 위한 재래종 옥수수의 이용에 관한 연구)

  • Silage yield and nutritional values of a locally collected Korean corn line with high tillering and earing characteristics were investigated at plant densities from 2778 to 8333 plants/ l0a. This line and a non-tillering and nonprolific corn as a check were grown at four plant densities and at three planting dates. The Korean local line abbreviated as MET had the highest dry matter per l0a. The highest dry matter of the MET line compared with the check hybrid was due to the highest dry leaf weight of the MET line. The highest dry leaf weight of the MET line was due to the increased number of tillers of the MET line. Other plant parts such as ear weight, kernel weight and cob weight of the MET line were lower than those of the check hybrid. The dry husk weight per l0a of the MET line was higher than that of the check hybrid, probably due to the increased number of ears in the MET line. The total embryo production per 10 a of the MET line was significantly higher than that of the check hybrid. The increased portion of embryo of the MET line is probably responsible for the higher TON values of the MET line. No interaction between variety x planting dates or planting density was found, indicating that the MET line and check hybrid were both the same in effects of planting dates and densities. Both line and hybrid showed the highest dry matter production when the planting density was high and planting dates was early. When silage was made from either MET line or check hybrid the nutritional values in terms of crude protein, crude fat, fiber, and ash contents of the MET line were similar to those of the check hybrid. But the TON of the MET line was higher than that of the check hybrid, while the OCP of the MET line lower than that of the check hybrid. Amino acid contents of the MET line were also comparable to those of the check hybrid, while lysine content of the MET line was 10% higher than that of the check hybrid.

Relative Performance and Immune Response in White Leghorn Layers Fed Liquid DL-methionine Hydroxy Analogue and DL-methionine

  • The relative performance and immune response was evaluated in White Leghorn layers fed liquid DL-methionine hydroxyl analogue-free acid (MHA-FA) relative to dry DL-methionine (DLM) in maize-soybean-sunflower based diets. Three graded levels of methionine (Met) from DLM or MHA-FA were added to the basal diet containing 0.27% Met on an equimolar basis to achieve 0.30, 0.36 and 0.42% Met in the diet. Each diet was fed ad libitum to 25 replicates of one bird (individual feeding) each, from 24 to 40 weeks of age. A regime of 16 h light was provided and all the layers were kept under uniform management throughout the experimental period. None of the parameters studied were influenced by the interaction between source and level of Met in diets. Similarly, the majority of parameters, except for daily feed consumption and immune response (influenced by level) and egg specific gravity and shell thickness (influenced by source), were not affected by either source or level of Met in the diets. Feed consumption was significantly lower in the birds fed a diet containing 0.42% Met compared to those fed lower levels of Met. The cutaneous basophilic hypersensitivity response to PHA-P and antibody titre (32 and 40 wk) to inoculation of sheep red blood cells increased significantly by increasing the concentration of Met in the diet from 0.30 to 0.36%. Thus, the Met requirement for immune competence was higher than for optimum production. The source of Met significantly influenced the egg specific gravity and shell thickness. The specific gravity and shell thickness of eggs increased significantly when MHA-FA was used as the source of Met in the diet compared to DLM. From the study it is concluded that Met requirement for immune competence (360 mg/b/d) is higher than for optimum production (300 mg/b/d). MHA-FA was comparable with DLM as a source of Met for production performance and immunity, when the bioavailability of MHA-FA was considered as 88% of DLM. Further, MHA-FA improved egg shell quality compared to DLM.

Effects of Intra-duodenal Infusion of Limiting Amino Acids on Plasma Insulin-like Growth Factor I, Growth Hormone, Insulin and Liver Insulin-like Growth Factor I mRNA in Growing Goat Fed a Maize Stover-based Diet

  • The effects of intra-duodenal infusion of methionine (Met), lysine (Lys) and leucine (Leu) on dry matter intake (DMI), the concentrations of insulin-like growth factor I (IGF-I), growth hormone (GH) and insulin in plasma, and liver IGF-I mRNA level were investigated in two experiments for Liuyang Black growing wether goats. In Experiment 1, three goats ($10.0{\pm}0.1$ kg) were fitted with ruminal, proximal duodenal and terminal ileal fistulaes to determine the infusion amounts of Met, Lys and Leu at the duodenum according to essential amino acid flows into the duodenum and their apparent digestibility. The infusion amounts were 0.77 g/d, 0.91 g/d and 0.58 g/d respectively. In Experiment 2, 4 groups of goats (($10.0{\pm}0.2$ kg) for each group, were cannulated at the duodenum, and were infused with a mixture of Met, Lys and Leu (Control), or mixtures with 21% Met, Lys or Leu replaced with glutamate respectively on a nitrogenous basis. The replacement of 21% Met, Lys or Leu with glutamate did not affect intakes of maize stover, concentrate or both (p>0.05) when compared with the control. The replacement of 21% Met or Lys significantly (p<0.05) reduced plasma GH, insulin and IGF-I concentrations and liver IGF-I mRNA level. The replacement of 21% Leu with glutamate reduced (p<0.05) plasma IGF-I concentration only, but not plasma insulin and GH, as well as liver IGF-I mRNA level (p>0.05). The close relationships between supplying Met and Lys in the lumen of the duodenum and plasma IGF-I, GH and insulin concentrations, as well as liver IGF-I mRNA level in this study indicate that the effects of the limiting amino acids on nutrition of animals are likely intermediated via their effects on these hormones, and these hormone profiles could be used as intermediate markers for the limiting order of amino acids.

Comparing Bio-efficacy of Liquid DL-methionine Hydroxy Analogue Free Acid with DL-methionine in Broiler Chickens

  • The present experiment was conducted to assess the efficacy of methionine hydroxy analogue free fatty acid (MHAFA) in comparison to DL-methionine (DL-Met) utilizing day-old commercial broiler chicks (n=300). The chicks were randomly distributed into 30 groups of 10 chicks each. Three dietary treatments, viz. D1-maize-soybean meal based basal diet (Control), D2- control diet supplemented with DL- methionine to meet its requirements and D3-control supplemented with MHA-FA @ 1.54 times of DL-methionine used in D2, were formulated. Each dietary treatment was offered to 10 replicated groups of chicks following completely random design (CRD). The chicks fed on supplemental DL-Met had significantly higher (p<0.01) gain in body weight, followed by MHA-FA group and control during 0-3 weeks of age. During overall growth period (0-6 weeks), chicks in DL-Met and MHA-FA groups grew better (p<0.01) than those in control. Feed conversion ratio (FCR) improved (p<0.01) on supplementation of either DL- Met or MHA-FA in the basal (Control) diet during 0-3 weeks of age. The FCR for overall period, however, did not differ statistically (p>0.05) amongst the treatments. The eviscerated yields emanated from diets with DL-Met or MHA-FA were higher (p<0.01) than that in Control. Abdominal fat pad was also more (p<0.01) in broilers fed control diet than in DL-Met or MHA-FA supplemented group. Breast yield was higher (p<0.05) in MHA-FA fed broilers than those fed DL-methionine supplemented or un-supplemented diets. The efficacy of MHA-FA in comparison to DL-Met for growth was 62.11, 64.82 and 63.88% and for feed efficiency was 62.98, 67.73 and 64.01% at 0-3, 3-6 and 0-6 weeks of age, respectively, while it was 65.85, 71.40 and 67.49% for eviscerated yield, abdominal fat pad reduction and breast yield at 6 weeks of age, respectively.
