역상 액체 크로마토그래피에 의한 Ni(II)-Isonitrosoethylacetoacetate Imine 유도체 킬레이트의 용리 메카니즘에 관한 연구 (A Study on the Elution Mechanism of Ni(II)-Isonitrosoethylacetoacetate Imine Chelates by Reversed Phase High Performance Liquid Chromatography)

  • 역상 액체 크로마토그래피에 의하여 Ni(IEAA-NH)(IEAA-NR)(R = H, $CH_3,\;C_2H_5,\;n-C_3H-7,\;n-C_4H9-,\;CH_5-CH_$) 킬레이트의 동시 분리 및 분석을 위한 최적조건을 Micropak MCH-5 분리관을 사용하여 조사하고, 이들의 용리거동에 대한 인자들과의 관계를 검토하였다. 용리액에서 methanol/water의 이성분 용매를 사용하였으며, 모든 금속 킬레이트의 용매 세기 인자는 $0{\le}logk'{\le}1$의 범위임을 확인하였고, 각 금속 킬레이트의 logk'값과 이성분용매계 중의 물의 부피분율을 도시한 결과 직선 관계가 성립하였다. 또한 뱃치법으로 측정한 분포비(Dc)값과 k'값을 도시하여 본 결과 직선 관계가 성립됨으로써 금속 킬레이트의 용리 메카니즘이 주로 소용매성 효과 즉 소수성효과에 기인함을 확인하였다. 한편 금속 킬레이트의 머무름에 미치는 용량 인자와 온도와의 관계를 조사한 결과 좋은 직선성을 나타내고 있으며, 뿐만 아니라 직선의 기울기로부터 엔탈피(${\delta}H$)와 엔탈피(${\delta}S$)를 구하고, 엔탈피와 용량 인자와의 값을 도시한 결과도 직선관계가 성립하였다. 직선의 기울기로부터 구한 보정 온도(${\beta}$)는 $773.47^{\circ}K$이었다. 이 결과 역상 HPLC에서 Ni(II)-Isonitrosoethylacetoacetate imine 킬레이트의 용리 메카니즘의 주로 소수성 효과에 기인함을 재확인 할 수 있었다.

랫드를 이용한 Acetanilide의 반복투여 및 생식/발생독성 병행시험 (Repeated Dose and Reproductive/Developmental Toxicities of Acetanilide in Rats)

  • The study was conducted to assess the repeated dose and reproduction and developmental toxicities of acetanilide, an intermediate for drug production, as a part of OECD Screening Information Data Set (SIDS) program. The test agent was administered by gavage at dose levels of 0, 22, 67, 200 and 600 mg/kg to Sprague-Dawley rats (12/group/sex) during pre-mating and mating period for males(up to 30 days) and females and pregnancy and early lactation period for females (up to 39-50 days). At 22 mg/kg, decreases in HGB, HCT (males) and MCHC (females), hyperplasia of spleen red pulp, hyperplasia of femur bone marrow (both sexes) were observed. At 67 mg/kg, salivation (males), reduced food consumption (both sexes), decreases in RBC, HGB, HCT and MCHC (males), increases in MCV (males) and spleen weight (males), hyperplasia of spleen red pulp and femur bone marrow (both sexes) were observed. At 200 mg/kg, decreases in locomotor activity and salivation (both sexes), reduced food consumption (both sexes), decreases in RBC, HGB, HCT and increases in MCV, MCH, BUN, T-BIL (males), enlargement of spleen (both sexes), increased weight of spleen (males), hyperplasia of spleen red pulp and femur bone marrow and extramedullary hematopoiesis of liver (both sexes), atrophy of thymus and corpus luteum hyperplasia of ovary (females) were observed. At 600 mg/kg, decreases in locomotor activity, cyanosis (both sexes), reddish tear, and salivation (males), mortality (4 out of 12 females), decreased body weight (females), reduced food consumption (both sexes), decreases in RBC, HGB, HCT and MCHC and increases in WBC, MCV, MCH, reticulocyte, neutrophil, lymphocyte, monocyte, AST, ALT, BUN, T-BIL, ALB, Ca and A/G ratio (males), enlargement of spleen, increased weights of spleen (both sexes), liver (males), kidney and ovary, decreased weights of thymus (females), hyperplasia of spleen red pulp, hyperplasia of femur bone marrow and extramedullary hematopoiesis of liver (both sexes), and atrophy of thymus and corpus luteum hyperplasia of ovary (females) were observed. Regarding the reproduction and development toxicities, there were no treatment-related changes in precoital time, mating index, fertility index and pregnancy index at all doses tested. At 22 and 67 mg/kg, there were no adverse effects on all the parameters observed. At 200 mg/ kg, decreased body weight of pups (day 4 p.p.) were observed. At 600 mg/kg, decreased body weight of pups (day 0 and 4 p.p.) and viability index (day 4 p.p.), increased incidence of newborns dead or with abnormal clinical signs were observed. The results suggest that the NOAELs for general toxicity are < 22 mg/kg, LOAELs are 22 mg/kg and the NOAELs for reproductive toxicity are 67 mg/kg.

시간활동 접근법에 의한 면 보건요원의 업무량 분석 (Measuring Myun Health Worker's Performance by Time-Activity Approach)

  • This study attempts to examine the performances of Myun Health Workers-the frontline workers in the Korean rural health care delivery system. The time-activity approach was mainly utilized as a measuring tool. This study was undertaken in September 1976 with 35 Myun Health Workers at the Kang Wha County. The pretested time-activity approach sheets were filled out daily for one month by those Myun Health Workers themselves. Statistical means and variances of analysis were utilized for statistical method in comparing some activities and functions converged into time distribution Findings: 1. The workers's average working hours derived in this study is 8 hours and 48 minutes per day, which takes half an hour longer than normal schedule. 2. They spend 56% working hour for direct services, in other words, the main function, 22% for supportive function, and 22% for other activities, the unrelated health services. 3. Considering the total working hours of main function, out-center activity is far more than in-center services with the ratio of 70% to 30% respectively, which proves, therefore, that the main activity of the workers is home visiting. 4. It takes 20 minutes purely for home visiting and takes 14 minutes for transportation. 5. This research also indicates that such factors as characteristics of the health workers and myun influence in shaping the structures of the worker's function and activity: a. The workers whose working site is located in myun office spend 15% among total working hours in carring out official myun activities, which is incidentally unrelated to health services, while the health subcenter have no rooms for administrative jobs for myun office. b. The workers whose office is in health subcenter contribute much time in doing main function and those working in special project distribute more time in performing supportive function. c. The types of workers are another dominant factor to influence the components of worker's functions and activities. MCH workers and MPW I spend much time for manipulating main function. d. MPW II, whose function is reorganized by special project in 2 myuns shows different pattern of time distribution compared to the TB worker orFP worker in the ordinary area. MPW II distributes their time evenly in performing MCH program, T.B. Program, F.P. program and education activity, while the unipurpose workers engage in carring out only their dominant role. e. Another variables which involve the variation of the worker's activity can be illustrated with the variables like target population, size of myun and convenience for transportation, among which the latter two are remarkable factors in determining the time for out-center service.

고지사육(高地飼育) 번식장해(繁殖障害) 유우(乳牛)의 혈액학치(血液學値)의 관(關)한 연구(硏究) (A study on the hematological values of the infertile dairy cattle raised in the high-land)

  • Observations were made on the blood picture of 50 multiparous and 98 infertile cows. The results obtained in this study were summarized as follows. 1. Mean values of RBC, Hb, PCV, MCV, MCH, MCHC and platelets for normal multiparous cows were $6.87{\pm}0.84{\times}10^6/mm^3$, $10.3{\pm}1.3g/100ml$, $31.4{\pm}3.7ml/100ml$, $46.1{\pm}6.9{\mu}^3$, $15.3{\pm}2.6pg$, $34{\pm}2.7%$, and $372.7{\pm}304.7{\times}10^3/mm^3$ respectively. 2. Mean values of RBC, Hb, PCV, MCV, MCH, MCHC and platelets for infertile cows were $7.00{\pm}0.98{\times}10^6/mm^3$, $10.6{\pm}1.2g/100ml$, $32.1{\pm}3.6ml/100ml$, $45.8{\pm}6.7{\mu}^3$, $15.3{\pm}1.8pg$, $33.9{\pm}3.3%$, and $382.7{\pm}157.5{\times}10^3/mm^3$, respectively. 3. Mean values of WBC count for normal multiparous and infertile cows were $7.92{\pm}1.72$ and $9.04{\pm}2.87{\times}10^3/mm^3$ respectively. 4. In the differential leukocytes count, mean values of neutrophils, lymphocytes, monocytes, eosinophils and basophils for normal multiparous cows were $29.7{\pm}7.9%$, $56.8{\pm}7.6%$, $2.9{\pm}1.9%$, $9.9{\pm}5.3%$ and $0.3{\pm}0.7%$, respectively, and mean values of neutrophils, lymphocytes, monocytes, eosinophils and basophils for infertile cows were $31.9{\pm}7.8%$, $57.5{\pm}7.9%$, $2.6{\pm}1.9%$, $7.5{\pm}4.8%$, and $0.4{\pm}0.2%$, respectively.

The Effects of Endurance Training on the Hemogram of the Horse

  • The purpose of this study was to evaluate the changes and readjustment capacity in the hematological characteristics of the horse during and after a prolonged training program. One pony and two hot-blooded horses were used in this study. Resting or basal blood parameters were assessed by collecting blood samples of the animals for 1 to 2 months prior to start of the training program. Each animal was subjected to arbitrary exercise for 30 min by an automatic hot trotter and was bled at 0, 15, 30, 45 (15 min of recovery), 60 (30 min of recovery), and 75 min (45 min of recovery) after onset of exercise. All animals were exercised 3 times a week over a fivemonth period. Hematological parameters including average white blood cell counts (WBC, ${\times}$$10^3$/$\mu$l), erythrocyte concentrations (RBC, ${\times}$$10^6$/$\mu$l), hematocrit (HCT, %), mean corpuscular volume (MCV, fl), number of platelets (PLT, ${\times}$$10^4$/$\mu$l), hemoglobin concentration (Hb, g/dl), mean corpuscular hemoglobin (MCH, pg), and mean corpuscular hemoglobin concentration (MCHC, g/dl) were analyzed using an automatic cell counter. All animals showed that RBC, WBC, and HCT were significantly (p<0.05) increasing from 7.09, 8.55, and 43.5 to 8.11, 9.67, and 49.5, respectively, during the 30 min of exercise and were back to or lower than the initial basis (resting and 0 min) 30 min after exercise. However, no significant differences were detected in MCV (50.3-51.3 fl), MCH (17.2-17.4 pg), and MCHC (33.7-34.4 g/dl) values (p>0.05) regardless of the training periods. Similar trends were observed after 1, 3, 4, and 5 months of training when compared to the resting state. When these parameters were analyzed by the effect of training periods (month), mean WBC concentrations significantly reduced in the fourth and fifth month after onset of training compared to that in resting condition or the first month of training program (p<0.05). The RBC values elevated at the second month (9.40) and reaching a significantly low level (p<0.001) at the fifth month (8.62) after training compared to the first month of training (7.89). In conclusion, a mild training program enhances blood parameters gradually in both the horse and the pony. Therefore, an optimized training program is beneficial in promoting the endurance performance of the horse.

배합비율(配合比率)에 따른 당귀보혈탕(當歸補血湯) 약침(藥鍼)이 빈혈백서(貧血白鼠)에 미치는 영향(影響) (Effects of Combination of Danggwibohyeol-tang Herbal-acupuncture on Anemic Rats)

  • Objective; This study was investigated to test the anti-anemic effects of Danggwibohyeol-tang for herbal acupuncture on rat with anemia induced by phenylhydrazine according to composition ratio. Methods; Danggwibohyeol-tang herbal-acupuncture was administered into Kyoksu(膈兪穴, BL17) and RBC, Hemoglobin, Hematocrit, MCV, MCH, MCHC, RDW, platelet, MPV, PDW, indirect bilirubin and lactate dehydrogenase were tested. Results; The obtained results are summarized as follows: (1) In control group RBC was significantly reduced in comparison with normal group. And in Danggwibohyeol-tang group A, B, C there was significant inhibitory effect on reduction in comparison with control group. The effect of Danggwibohyeol-tang group C was similiar to that of Danggwibohyeol-tang group B and was higher than that of Danggwibohyeol-tang group A. (2) In control group hemoglobin was significantly reduced in comparison with normal group. And in Danggwibohyeol-tang group A, B, C there was significant inhibitory effect on reduction in comparison with control group but there was no significant difference between Danggwibohyeol-tang group A, B, C. (3) In control group hematocrit was significantly reduced in comparison with normal group. And in Danggwibohyeol-tang group A, B, C there was significant inhibitory effect on reduction in comparison with control group. The effect of Danggwibohyeol-tang group C was higher than that of Danggwibohyeol-tang group A or B. (4) In case of MCV, MCH, MCHC and RDW there was no significant anti-anemic effect. (5) Danggwibohyeol-tang group significantly inhibited the growth of platelet. In case of Danggwibohyeol-tang A and B group there was more inhibitory effect than Danggwibohyeol-tang C group but there was no significant difference between Danggwibohyeol-tang A and B group. (6) In case of MPV, PDW and indirect bilirubin there was no significant difference between Danggwibohyeol-tang group and control group. (7) Danggwibohyeol-tang group significantly inhibited the growth of lactate dehydrogenase. In case of Danggwibohyeol-tang C group there was more inhibitory effect than Danggwibohyeol-tang B group and Danggwibohyeol-tang B group showed more inhibitory effect than Danggwibohyeol-tang A group. Conclusion; From above results Danggwibohyeol-tang for herbal acupuncture showed significant anti-anemic effect in some parameters. Further study is needed for the effect of Danggwibohyeol-tang according to different composition ratio.

다른 거리의 철인 3종 경기 시 체중, 나트륨 및 혈액 성분에 미치는 영향 (The Effect of Different Triathlon on Weight, Sodium and Hematological Changes)

  • 철인 3종 경기에 참가하는 선수들은 탈수, 저나트륨혈증, 용혈현상 및 연역반응과 같은 의학적인 문제에 직면할 수 있다. 본 연구는 철인 3종 경기 시 체중의 변화, 나트륨농도 및 혈액성분의 변화를 조사하기 위하여 서로 다른 Olympic Distance Triathlon (ODT)과 Ironman Distance Triathlon (IDT)에 참가한 선수들에게 체중과 나트륨 농도 및 혈액 성분이 어떠한 영향을 미치는가를 연구하였다. 체중은 ODT와 IDT 모두 유의하게 감소하였고, 나트륨 농도는 IDT에서만 증가하였다. RBC, Hb, Hct 및 MCH는 IDT에서 증가하였고, Hct는 ODT에서 감소하였다. IDT에서는 체중의 변화율과 나트륨 농도간의 유의한 상관관계가 있었으며, 체중 감소에 따른 나트륨농도의 증가현상이 나타났다. RBC, Hb 및 Hct의 연화는 거리에 따라 영향을 받는 것으로 조사되었다. 이러한 결과들을 종합해 볼 때 철인 3종 경기와 같은 장시간 지속되는 운동 시 체중 변화가 나트륨 농도에 영향을 미치는 것으로 파악되었으며, 경기 거리에 따른 혈액 성분의 변화에 차이는 있었지만 이러한 변화는 모두 정상범위인 것으로 나타났다.

잡종견(雜種犬)의 혈액학치(血液學値)에 관(關)한 연구(硏究) (Hematologic values of mixed breed dog in Korea)

  • The hematologic values were examined from 74 healthy mixed breed dogs in the area of Seoul. The results obtained are summarized as follows; 1. Mean${\pm}$SD values and ranges of red blood cell(RBC) count were $6.16{\pm}0.92{\times}10^6/{\mu}l$ and $4.40{\sim}8.62{\times}10^6/{\mu}l$, of hemoglobin(Hb) content $14.90{\pm}2.42g/100ml$ and 8.7~19.2g/100ml, of packed cell volume(PCV) $45.47{\pm}6.16ml/100ml$ and 30~57ml/100ml, of mean corpuscular volume (MCV) $74.80{\pm}6.83fl$ and 54.08~90.90fl, of mean corpuscular hemoglobin (MCH) $24.41{\pm}3.91pg$ and 14.19~32.97pg, of mean corpuscular hemoglobin concentration (MCHC) $32.07{\pm}3.24g/100ml$ and 22.23~39.76g/100ml, respectively. 2. The RBC count value in the age group of 3~4 years was higher (p<0.05) than the total RBC count value. The Hb content value in the age group of less than 6 months was lower (p<0.01) but in the age group of 1~2 years and the age group of 3~4 years were higher (p<0.05, p<0.01) than the total Hb content value. The PCV value in the age group of less than 6 months was lower (p<0.05) than the total PCV value. The MCHC values in the age group of 1~2 years and the age group of 3~4 years were higher (p<0.01, p<0.05) than the total MCHC value. 3. Mean${\pm}$SD values and ranges of white blood cell (WBC) count were $11.26{\pm}3.05{\times}10^3/{\mu}l$ and $6.30{\sim}18.4{\times}10^3/{\mu}l$, of band neutrophil $2.97{\pm}1.44%$ and 1~10%, of segmented neutrophil $62.81{\pm}4.92%$ and 42~70, of lymphocyte $30.55{\pm}5.69%$ and 17~50%, of monocyte $2.49{\pm}0.84%$ and 1~5%, of eosinophil $1.81{\pm}1.175$ and 1~8%, respectively. 4. The WBC count value in the age group of 7~12 months was lower (p<0.05) but in the age group of 3~4 years was higher (p<0.05) than the total WBC count value. The band neutrophil values in the age group of 1~2 years and the age group of 3~4 years were higher (p<0.05, p<0.01) than the total band neutrophil value.

역상 고성능 액체 크로마토 그래피에 의한 Pd(II) - isonitrosoethylacetoacetate imine 유도체 킬레이트의 용리거동에 관한 연구 (A Study on the Elution Behavior of Pd-Isonitroso ethylacetoacetate imine Chelates by Reversed-phase High Performance Liquid Chromatography)

  • 본 연구는 역상 HPLC를 이용하여 Pb(IEAA-NR)2 (R=H, CH3, C3H7, C6H5, C4H9) 킬레이트를 동시 분리하기 위한 최적 분리 조건을 조사하고 이들의 용리거동에 관련된 인자들과의 관계를 검토하였는데 그 결과는 다음과 같다. 1. 각 Pb(IEAA-NR)2 킬레이트의 분리에 대한 기기의 최적 조건을 실험적으로 측정한 결과는 다음과 같다. 분리관 : Micropak MCH-5 (4.0 mm I.D. $\times$ 15 cm L, Particle size 5 $\mu\textrm{m}$) 용리액 : MeOH / H20 (73/27) 흐름속도 : 0.7 ${\mu}\ell$/min. 시료주입량 : 4 ${\mu}\ell$ 검출기 : UV 검출기 (254 nm) 2. 용리액의 흐름속도는 0.7${\mu}\ell$/min. 일때 적당한 분리 시간과 좋은 분리도를 나타내었다. 3. 시료 용매가 킬레이트의 머무름 시간에는 큰 영향을 주지 않았으며 금속 킬레이트간의 겸침 현상이 줄어들고 봉우리 면적이 큰 CH3CN을 사용하였다. 4. 용리액의 조성은 MeOH/H2O 의 비가 73/27 으로써 대개의 경우에 lig k' 값이 0$\leq$log k' $\leq$1을 잘 만족하지 않았으며 전체적으로 적당한 분리시간과 좋은 분리도를 나타내었다. 5. Pb(IEAA-NR)2 킬레이트의 log k' 값을 이성분 용매계에서 물의 부피 분율에 대하여 plot한 결과 이들이 직선적인 관계를 가지는 것으로 보아 Pb(IEAA-NR)2 의 머무름은 소수성 효과(Hydrophobic effect)에 크게 기인 한다는 것을 알 수 있었다.

시안 노출에 의한 이스라엘잉어, Cyprinus carpio의 혈액화학성상의 변동 (Hemochemical Changes in Israel carp, Cyprinus carpio exposed to cyanide)

  • 유용 양식어류인 이스라엘잉어를 대상으로 유수식 방법에 의해 시안독성에 대하여 5주 동안 그들의 혈액성상 및 화학적 변동을 조사하였다. 시안노출에 의한 이스라엘잉어의 RBC수, Hb농도 및 Ht값은 시안농도 151$\mu$g/$\ell$ 이상에서 4주 후부터 유의한 감소를 나타내었다 MCH 및 MCV는 시안농도 289$\mu$g/$\ell$에서 5주 후부터 유의한 증가를 나타내었으나, MCHC의 변동은 관찰되지 않았다. 혈청 무기성분인 calcium농도는 5주 후에 시안농도 151 $\mu$g/$\ell$ 이상에서 유의하게 감소하였고, magnesium농도는 시안농도 289$\mu$g/$\ell$에서 4주 후부터 유의한 증가를 나타내었으며, inorganic phosphate농도는 모든 시안농도에서 유의한 변동이 관찰되지 않았다. 혈청 유기성분인 total protein, albumin 및 glucose농도는 시안농도 151 $\mu$g/$\ell$이상에서 4주 후부터 유의한 감소가 인정되었다. 혈청 GOT, GPT 및 LDH활성은 시안농도 151 $\mu$g/$\ell$이상에서 3주 후부터 유의한 증가를 나타내었고, 혈청 Ch-E 활성은 289$\mu$g/$\ell$에서는 4주 후부터 유의한 증가를 나타내었으나, Al-P 및 amylase활성의 특징적인 변동은 관찰되지 않았다. 결론적으로 151 $\mu$g/$\ell$이상의 시안농도는 이스라엘잉어의 혈액내 의 RBC수, Hb농도 및 Ht값, 혈청내의 calcium, magnesium, total protein, albumin 및 glucose농도, 혈청내의 GOT, GPT 및 LDH 활성변동에 영향을 미치고 있었다.
